Enterotoxin Productivity and Antimicrobial Susceptibility of Bacillus cereus BY06 Isolated from Pigs with Diarrheal Disease (자돈 설사 분변에서 분리한 Bacillus cereus BY06의 장 독소 생성 및 항균제 감수성)

  • The enterotoxin production and antimicrobial susceptibility on hemolytic strains from stools of diarrheal pigs was investigated in this study. Through morphological observation, gyrB nucleotide sequence, and API kit analysis, the selected potential pathogenic strain BY06 was identified as Bacillus cereus. Because the characteristic of enterotoxin symptoms were widely caused by Bacillus cereus strains, a PCR test was carried out in order to check the enterotoxin genes (hblA) in this strain. According to the results, this strain was an enterotoxin positive strain containing the hblA gene. The minimum inhibitory concentrations of 10 different antimicrobial agents were screened by the agar dilution test, indicating that this strain was resistant to penicillin G and intermediate to erythromycin; however, it susceptible to cephalothin, vancomycin, clindamycin, fusidic acid, gentamicin, ciprofloxacin, tetracycline and rifampin. These results suggest that the B. cereus BY06 isolated from pig feces has a potential risk of producing enterotoxin and is resistant to penicillin G, but susceptible to various antimicrobial agents.

The Effect of Rotating Magnetic Field on Enterotoxin Genes Expression in Staphylococcus Aureus Strains

  • Staphylococcus aureus cultures exposed to rotating magnetic field (RMF) were studied in order to analyse the possible induced changes in staphylococcal enterotoxin genes (se) expression. Liquid cultures of S. aureus strains carrying different se were exposed to the RMF of magnetic frequency 50 Hz and magnetic induction 34 mT for 10 h at $37^{\circ}C$. Three time points of bacterial growth cycle were considered for RNA extractions. Gene expression analyses were evaluated using real-time quantitative PCR method. The present study confirmed, that the RMF can stimulate the growth rate of S. aureus cultures in comparison to the unexposed controls, while the stimulation is not strain dependent. The studies have also shown, that the RMF, depending on the exposure time but regardless the bacterial strain, can influence on the expression of various se. In general, except for sea, as a result of bacterial exposure to the RMF through subsequent growth phases, the expression of se decreased, reaching the values below results recorded for unexposed controls. In the case of sea expression remained at a lower level as compared to the control, regardless the time of exposition.

Prevalence and Toxin Characteristics of Bacillus thuringiensis Isolated from Organic Vegetables

  • The prevalence and toxin characteristics of Bacillus thuringiensis isolated from 39 organic vegetables were investigated. B. thuringiensis was detected in 30 out of the 39 organic vegetables (76.9%) with a mean value of 2.60 log CFU/g. Twenty-five out of the 30 B. thuringiensis isolates (83.3%) showed insecticidal toxicity against Spodoptera exigua. The hblCDA, nheABC, and entFM genes were found to be the major toxin genes, but the ces gene was not detected in any of the tested B. thuringiensis isolates. The hemolysin BL enterotoxin was detected in all 30 B. thuringiensis isolates (100%). The non-hemolytic enterotoxin complex was found in 27 out of 30 B. thuringiensis isolates (90.0%). The B. thuringiensis tested in this study had similar toxin gene characteristics to B. cereus, which possessed more than one toxin gene. B. thuringiensis could have the potential risk of foodborne illness based on the toxin genes and toxin-producing ability.

  • The purpose of this study was to provide basic data necessary for the prevention of food poisoning and safe food management. We examined 872 food samples for B. cereus in accordance with the MFDS Food Code and investigated characteristics of their harboring toxin genes. We detected and isolated 113 strains of B. cereus from 78 food samples (8.9%), and the average detection level was 48 CFU/g. B. cereus isolates carried at least 1 toxin gene among the emetic toxins and 5 enterotoxin genes. The toxin gene profiles of B. cereus were classified into 18 different types of isolates showing genetic diversity. Among the strains, 34 (30.1%) had all 5 enterotoxin genes (Cytk-nheA-entFM-bceT-hblC), accounting for the highest percentage. The entFM and nheA genes were major enterotoxin genes, while the emetic toxin gene, CER, was the least detected in B. cereus isolated from food samples.

  • Staphylococcus aureus is spread worldwide and can result in food poisoning outbreaks. Among samples collected from soil, water, protected houses, packing houses, employees, strawberries, and leaves, and analyzed for S. aureus contamination, 16% samples 'showed S. aureus contamination, particularly on employees' hands, scissors, and strawberries. Examination of enterotoxins A, B, and C genes of S. aureus by PCR revealed sea and seb in 92 and 38% of total strains, respectively, whereas sec was not detected. In conclusion, implementation of Good Agricultural Practice is necessary for preventing food-borne diseases of staphylococcal origin, thereby ensuring the safety of farm-to-table products.

  • Objectives: This study was undertaken to analyze Staphylococcus aureus from cultivation environments for agricultural products and to confirm antibiotic resistance and enterotoxin genes for the isolated S. aureus. Methods: A total of 648 samples were collected from apple, peach, ginseng and balloon flower farms. S. aureus was isolated from soil, agricultural water, personal hygiene elements (hands, gloves and clothes) and work utensils (boxes). Results: S. aureus was detected in a total of 25 samples and 72 strains were isolated. The resistance rate of the isolated S. aureus strains was confirmed at 33.3%, with 24 resistant strains among the total of 72. Fourteen different patterns types were found, and three pattern types (NV, OX, VA) were confirmed most frequently. As result of the detection of enterotoxin gene type, four gene types (sea: 1, sed: 4, seg: all isolated S. aureus, sei: all isolated S. aureus) were analyzed among a total of nine types. Conclusions: This study demonstrates that personal hygiene techniques should be properly managed, such as washing and sterilization before or after work, because agricultural contamination by S. aureus frequently developed through improper management.

Biological Characteristics of Staphylococcus aureus Isolated from Food-Borne Patients in Seoul (식중독 환자에서 분리한 황색포도상구균의 생물학적 특성)

  • Staphylocorccus aureus is gram positive, facultatively anaerobic, non-sporulative coccus, and positive for coagulase and DNase. The food-poisoning outbreak of Staphylococcus aureus increases in the world, and third occurrence happened in our country. Of 105 isolates (25.4%) obtained 413 focal samples of food-poisoning suspicious patients. In those cases, the enterotoxins were detected from a total of 45 isolates (42.9%), 9 isolates(20.0%) were A type, 33 isolates (73.3%) were H types, 2 isolates (4.4%) were G type and 1 isolate was a I type enterotoxin. Among the isolates possessing staphylococcal enterotoxins, 29 isolates had H type only(64.4%), 5 isolates had A type only and 4 isolates had both A and H type. Two isolates had G type only and 1 isolate had I type only. In the antibiotic susceptibility, 48 isolates (46%) had at least one antibiotic resistance among 105 isolates, 34 isolates (70.8%) were resistant to penicillin. 1 isolate (2.1%) to ampicillin, 3 isolates (6.3%) to erythromycin and kanamycin. Seven were resistant to more than two antibiotics and especially 1 isolate was resistant to penicillin-ampicillin-nitrofurantoin.

Screening of MRSA (Methicilline Resistant Staphylococcus Aureus) and seb Gene in Producing Strains Isolated from Food Service Environment of Elementary Schools (초등학교 급식 환경에서의 메치실린 내성 황색포도상구균(MRSA)과 seb gene의 검색)

  • Most of food poisoning is frequently raised from mass catering. Especially, staphylococci takes the large part of pathogenic agents which are related to the hygienic condition. Among total 98 samples, four staphylococci were isolated from food service environment such as drinking water (A), hands (D), refrigerator and apron (E) of 5 elementary school (A, B, C, D, E) in Gyeongnam Province. These isolated strains are characterized as 1 MRCNS (Methicilline Resistant Coagulase Negative Staphylococcus aureus) and 3 MSCPS (Methicilline Sensitive Coagulase positive Staphylococcus aureus). Also, production of enterotoxin B (sob gene) were examined by PCR which has known as a big problem because of their temperature resistance. Hence, PCR was performed on isolated 4 staphylococci. The all 4 isolated Staphylococcus aureus have 477 bp of seb gene. Antibiotics susceptibility test was completed on PCR detected strains. All strains were fully resistance to ampicillin and penicillin. The drinking water of A place has resistance to oxacilline, therefore this strain turned out to be MRSA (Methicilline Resistant Staphylococcus Aureus).

  • Salmonella encodes an enterotoxin (Stn) which possesses biological activity similar to the cholera toxin. Stn contributes significantly to the overall virulence of S. typhimurium in a murine model. The production of Stn is enhanced in a high-osmolarity medium and by contact with epithelial cells. In the present study, the in vitro and in vivo transcriptional regulations of the sin promoter revealed two promoters, P1 and P2. The P1 promoter identified by a primer extension analysis of stn mRNA exhibited a switching mechanism in vivo. Depending on the growth stage, transcription was initiated from different start sites termed $P1_S\;and\;P1_E$. $P1_S$, recognized by RNA polymerase containing ${\sigma}^S(E{\sigma}^S),\;and\;P1_E$, recognized by $E{\sigma}^70$, were activated during the stationary and exponential phases, respectively, while $P1_S\;and\;P1_E$ were both negatively regulated by CRPㆍcAMP and H-NS. Results revealed that $P1_S$ was the responsible promoter activated under a high osmolarity and low pH. The P2 promoter was identified 45 nucleotides downstream from $P1_E$ and negatively controlled by CRPㆍcAMP in vitro. No P2 activity was detected in vivo. The regulation of stn expression monitored using a Pstn::egfp fusion indicated that $E{\sigma}^S$ was required for the induction of stn and various factors were involved in stn regulation inside animal cells.

  • The FimH subunit of type 1-fimbriated Escherichia coli has been determined as a major cause of urinary tract infection. To produce a possible vaccine antigen against urinary tract infection, the fimH gene was genetically linked to the Itxa2b gene, which was then cloned into the pMAL -p2E expression vector. The chimaeric construction of pMALfimH/Itxa2b was transformed into Escherichia coli TB1 and its N-terminal amino acid sequence was analyzed. (omitted)
