To study the effect of dietary docosahexaenoic acid (DHA) enrichment on the expression of hepatic genes in pigs, weaned, crossbred pigs (30 d old) were fed diets supplemented with either 2% tallow or DHA oil for 18 d. Hepatic mRNA was extracted. Suppression subtractive hybridization was used to explore the hepatic genes that were specifically regulated by dietary DHA enrichment. After subtraction, we observed 288 cDNA fragments differentially expressed in livers from pigs fed either 2% DHA oil or 2% tallow for 18 d. After differential screening, 7 genes were found to be differentially expressed. Serum amyloid A protein 2 (SAA2) was further investigated because of its role in lipid metabolism. Northern analysis indicated that hepatic SAA2 was upregulated by dietary DHA enrichment (p<0.05). In a second experiment, feeding 10% DHA oil for 2d significantly increased the expression of SAA2 (compared to the 10% tallow group; p<0.05). The porcine SAA2 full length cDNA sequence was cloned and the sequence was compared to the human and mouse sequences. The homology of the SAA2 amino acid sequence between pig and human was 73% and between pig and mouse was 62%. There was a considerable difference in SAA2 sequences among these species. Of particular note was a deletion of 8 amino acids, in the pig compared to the human. This fragment is a specific characteristic for the SAA subtype that involved in acute inflammation reaction. Similar to human and mouse, porcine SAA2 was highly expressed in the liver of pigs. It was not detectable in the skeletal muscle, heart muscle, spleen, kidney, lung, and adipose tissue. These data suggest that SAA2 may be involved in mediation of the function of dietary DHA in the liver of the pig, however, the mechanism is not yet clear.
The Bacterial community structure and its complexity of the enrichment culture during the isolation and screening of imidacloprid-degrading strain were studied using denaturating gradient gel electrophoresis analysis. The dominant bacteria in the original tea rhizosphere soil were uncultured bacteria, Rhizobium sp., Sinorhizobium, Ochrobactrum sp., Alcaligenes, Bacillus sp., Bacterium, Klebsiella sp., and Ensifer adhaerens. The bacterial community structure was altered extensively and its complexity reduced during the enrichment process, and four culturable bacteria, Ochrobactrum sp., Rhizobium sp., Geobacillus stearothermophilus, and Alcaligenes faecalis, remained in the final enrichment. Only one indigenous strain, BCL-1, with imidacloprid-degrading potential, was isolated from the sixth enrichment culture. This isolate was a gram-negative rod-shaped bacterium and identified as the genus Ochrobactrum based on its morphological, physiological, and biochemical properties and its 16S rRNA gene sequence. The degradation test showed that approximately 67.67% of the imidacloprid (50 mg/l) was degraded within 48 h by strain BCL-1. The optimum conditions for degradation were a pH of 8 and $30^{\circ}C$. The simulation of imidacloprid bioremediation by strain BCL-1 in soil demonstrated that the best performance in situ (tea soil) resulted in the degradation of 92.44% of the imidacloprid (100 mg/g) within 20 days, which was better than those observed in the ex situ simulations that were 64.66% (cabbage soil), 41.15% (potato soil), and 54.15% (tomato soil).
현재 우리나라 정보과학영재의 교육과정은 비실용적이고 체계적이지 못하여 정보과학영재 교육 운영에 매우 어려운 실정이며 그 문제점은 다음과 같이 요약된다. 첫째, 중등 영재교육과정에 대한 기존 연구가 많이 부족한 상태이다. 둘째, 운영 개발되어 있는 영재교육과정은 창의성, 문제해결력, 논리적 사고력과 통찰력 등을 신장시키기 힘들다. 셋째, 영재의 능력, 적성 소질에서의 개인차를 고려한 교육과정이 없는 실정이다. 따라서 본 연구에서는 중등 정보과학영재의 창의적 문제해결능력과 계산적 사고를 향상시킬 수 있는 단계별 심화 교육과정을 설계하여 제안하고자 한다. 이를 위해 먼저 정보과학영재 교육에 대한 기존 연구와 교육기관의 실태를 조사하여 문제점을 분석하였다. 그리고 이를 근거로 실용적인 중등정보과학영재 교육목표에 부합하는 세부적인 단계별 심화 교육과정을 설계하였다.
The purpose of this research is to investigate the enrichment and distribution patterns of naturally occurring potentially toxic elements in soils derived from black shales and slates. Soil samples were collected from the Chubu area covered with uranium-enriched black shales and slates of the Changri Formation of the Okchon Super Group, and analysed for multiple-elements using INAA, ICP-AES and AAS. Soil pH and loss-an-ignition were also measured. Trace element contents in black shale and slate of the Chubu area are relatively lower than those in black shales, and higher than those in black slates reported elsewhere (Chon, Jung, 1991; Chon et al., 1996). Soil pH values range from 3.5 to 6.5, and loss-an-ignition values are in the range from 3 to 10%. Potentially toxic elements including As, Ba, Cr, Cu, Mo and U are highly enriched in residual soils with maximum content of $540{\mu}g/g$, $35,000{\mu}g/g$, $280{\mu}g/g$, $300{\mu}g/g$, $240{\mu}g/g$ and $860{\mu}g/g$, respectively. Significant concentrations of those elements were found in soils taken from the Tojangkol, the Chubu tunnel and the Meokti areas. Soils derived from black shales and slates, calcareous and phyllitic rocks, and intrusions were discriminated in terms of As, Ba, Co, Cr, Cu, Mo, Sc and U contents. Enrichment index was calculated using the concentrations of As, Ba, Cr and Mo, and enrichment index map shows very similar trend with U distribution in soils.
Reducing carbon dioxide ($CO_2$) exhaust has become a major issue for society in the last few years, especially since the initial release of the Kyoto Protocol in 1997 that strictly limited the emissions of greenhouse gas for each country. One of the primary sectors affecting the levels of atmospheric greenhouse gases is agriculture where $CO_2$ is not only consumed by plants but also produced from various types of soil and agricultural ecosystems including greenhouses. In greenhouse cultivation, $CO_2$ concentration plays an essential role in the photosynthesis process of crops. Optimum control of greenhouse $CO_2$ enrichment based on accurate monitoring of the added $CO_2$ can improve profitability through efficient crop production and reduce environmental impact, compared to traditional management practices. In this study, a sensor-based control system that could estimate the required $CO_2$ concentration considering emission from soil for cucumber greenhouses was developed and evaluated. The relative profitability index (RPI) was defined by the ratio of growth rate to supplied $CO_2$. RPI for a greenhouse controlled at lower set point of $CO_2$ concentration (500 ${\mu}mol{\cdot}mol^{-1}$) was greater than that of greenhouse at higher set point (800 ${\mu}mol{\cdot}mol^{-1}$). Evaluation tests to optimize $CO_2$ enrichment concluded that the developed control system would be applicable not only to minimize over-exhaust of $CO_2$ but also to maintain the crop profitability.
The real-time enrichment and detection of pathogens are serious issues and rapidly evolving field of research because of the ability of these pathogens to cause infectious diseases. In general, bacterial detection is accomplished by conventional colony counting or by polymerase chain reaction (PCR) after DNA extraction. As colony counting requires considerable time to cultivate, PCR is an attractive method for rapid detection. A small number of pathogens can cause diseases. Hence, a pretreatment process, such as enrichment is essential for detecting bacteria in an actual environment. Thus, in this study, we developed a microfluidic chip capable of performing rapid enrichment of bacteria and the extraction of their genes. A lectin, i.e., Concanavalin A (ConA), which shows binding affinity to the surface of most bacteria, was coated on the surface of magnetic particles to nonspecifically capture bacteria. It was subsequently concentrated through magnetic forces in a microfluidic channel. To lyse the captured bacteria, magnetic particles were irradiated by a wavelength of 532nm. The photo-thermal effect on the particles was sufficient for extracting DNA, which was consequently utilized for the identification of bacteria. Our device will help monitor the existence of bacteria in various environmental situations such as water, air, and soil.
This study compared the survival rates, growth, and fatty acid content of larval Pacific cod Gadus macrocephalus fed with rotifers raised on different enrichment diets. We used four commercial rotifer enrichment products (one domestic, ER1, and three imported, ER2, ER3 and ER4). Twelve 200-L tanks were used, with three replicates per treatment. Larvae were fed rotifers 3 times daily at a rate of 5 rotifers/ml/feeding from 7 to 21 days post-hatch. At the end of the feeding trial, the survival rate and total length of larvae fed ER3 were greater than those of larvae fed ER1, ER2, or ER4. ER3 had the highest proportions of eicosapentaenoic acid (EPA), docosahexaenoic acid (DHA), and polyunsaturated acid (PUFA). Partially reflecting this composition, larvae fed ER3 had the highest proportions of DHA and PUFA. These results show a positive effect of rotifer DHA and PUFA proportions on the survival and growth rates of Pacific cod larvae.

The effective and accurate assessment of the total microbial community diversity is one of the primary challenges in modem microbial ecology, especially for the detection and characterization of unculturable populations and populations with a low abundance. Accordingly, this study was undertaken to investigate the diversity of the microbial community during the biodegradation of cis- and trans-dichloroethenes in soil and wastewater enrichment cultures. Community profiling using PCR targeting the l6S rRNA gene and denaturing gradient gel electrophoresis (PCR-DGGE) revealed an alteration in the bacterial community profiles with time. Exposure to cis- and trans-dichloroethenes led to the disappearance of certain genospecies that were initially observed in the untreated samples. A cluster analysis of the bacterial DGGE community profiles at various sampling times during the degradation process indicated that the community profile became stable after day 10 of the enrichment. DNA sequencing and phylogenetic analysis of selected DGGE bands revealed that the genera Acinetobacter, Pseudomonas, Bacillus, Comamonas, and Arthrobacter, plus several other important uncultured bacterial phylotypes, dominated the enrichment cultures. Thus, the identified dominant phylotypes may play an important role in the degradation of cis- and trans-dichloroethenes.
본 연구에서는 단일반응조에서 포기/비포기 시간에 따른 하수 내 유기물질 및 질소화합물을 변화양상을 살펴보고자 하였다. 실험 결과 C/N비 3 : 1, 포기/비포기 20/40 min 구간에서부터 90% 이상의 안정적인 유기물 및 질소 제거가 이루어짐을 알 수 있었다. 포기/비포기 구간의 비율을 길게 유지하는 것이 탈질에 더욱 효과적이었으며 이는 비포기 구간을 유지하는 동안 반응조 내 미생물의 군집변화에 기인하는 것으로 판단하였다. PCR-DGGE를 한 결과, 유기물 및 질소화합물의 산화에 관여하는 미생물로 Dysgonomonas mossii strain Melo40, Eubacterium sp. oral clone JN088, Uncultured bacterium clone SPESB2_718과 Bacterium enrichment culture clone LE이 관찰되었고 탈질에 관여하는 미생물은 Uncultured Acidobacteria bacterium clone AKYG487, Lactobacillus harbinensis strain FQ003, Erythrobacter litoralis strain Gi-3, Phytobacter diazotrophicus strain Ls8, Mycobacterium sp. enrichment culture clone GE10037biofNNA로 나타났다.
Purpose: To investigate and analyze effects of environmental enrichment(EE) and nerve stimulation that follows in application times with change of functional recovery(1, 3, 7 and 14 days). Methods: Focal ischemic brain injury was produced in 100 Sprauge-Dawley rats through middle cerebral artery occlusion(MCAO). Neurobehavioral assessment were selected, such as tilting plane testing, horizontal wire testing, vestibulomotor function testing and complex neuromotor function test, then they were randomly divided into five groups; Group I : Sham group, Group II: MCAO group, Group III: MCAO and ES group, Group IV: MCAO and EE group, Group V: MCAO and EE and ES group. Results: In neurobehavioral assessment, group V were significantly difference from other groups on between-subject effects. Conclusion: Our findings suggest that in focal ishemic brain injury, combined environmental enrichment and peripheral nerve electrical stimulation is more improved that the improvement of exercise function recovery than non treatment group.
