Quantitative determination of 3-chloro-4-(dichloromethyl)-5-hydroxy-2(5H)-furanone (MX) in chlorinated drinking water using sample enrichment followed by liquid-liquid extraction and GC-MS (시료 농축 후 액-액-추출과 GC-MS를 이용한 염소 소독 음용수중 3-chloro-4-(dichloromethyl)-5-hydroxy-2(5H)-furanone(MX)의정량 분석)

  • This study explores the means by which MX can be effectively extracted from chlorinated water 3-Chloro-4-(dichloromethyl)-5-hydroxy-2(5H)-furanone (MX), a potent mutagen commonly found in chlorinated drinking water at concentrations of up to a few hundred ng/L, was quantitatively determined using sample enrichment followed by liquid-liquid extraction (LLE), derivatization to methylated form, and analysis with GC-MS. A 4-L water sample was enriched to a concentration of 0.4 L using a vacuum rotary evaporator at 30 ℃. MX in the water was extracted using ethyl acetate (100 mL × 2) as a solvent and MX in the extract was methylated with 10 % H2SO4 in methanol. MX was recovered at a rate of 73.8 %, which was higher than that (38.1 %) for the resin adsorption method. The limit of quantification and repeatability (as relative standard deviation) were estimated to be 10 ng/L and 2.2 %, respectively. This result suggested that LLE can be used for the determination of MX in chlorinated water as an alternative to more time-consuming resin adsorption method.

Implementation of Active Sound Enrichment Control for Improving Engine Sound Quality Inside the Cabin of a Passenger Car (차량 실내공간의 가속 시 엔진음 음질 향상을 위한 실시간 능동음향증강 제어 구현)

  • In this study, a concept of active sound enrichment (ASE) control system was implemented and demonstrated for improving engine sound quality inside the cabin of a passenger car during acceleration. Unlike the active noise control cancels the noise for disturbance rejection, the ASE adds additional sound to the noise for tracking control. This approach requires a new algorithm to provide additional artificial sound to the original engine sound using active control strategy to achieve a target sound profile, which is predefined to satisfy required interior sound quality. The ASE algorithm was implemented in a digital controller dSPACE DS1401 and real-time control experiment was accomplished in an actual car. The ASE control results show that the actively enriched sound of each engine order against RPM tracks the target profiles precisely and quickly and improves the discontinuity, the level ratios and the sound pressure level of each engine order. Thus it is anticipated the ASE system can be applied for the improvement of the engine sound quality inside the cabin during acceleration.

Enrichment of Coffee Flavors with Supercritical Carbon Dioxide (초임계 이산화탄소를 이용한 커피의 향기 성분 증진)

  • In this study, the supercritical $CO_2$ extraction was applied for the enrichment of coffee flavors. The extraction efficiency of coffee flavors was dependent on the pressure and temperature and optimized as 350 bar and 80$^\circ$C. Five flavors of high aroma values were analyzed. The flavors extracted by the supercritical $CO_2$ extraction were 76.6 times higher than those by the traditional method, espresso extraction. The modified headspace system proved the enhanced efficiency of supercritical $CO_2$ extraction. The coffee beverage containing the extract of supercritical $CO_2$ resulted 2.3 times of flavor enrichment when it was compared to that without the supercritical extract.

The Importance of Intertidal Benthic Autotrophs to the Kwangyang Bay (Korea) Food Webs: ${\delta}^{13}$C analysis

  • The importance of phytoplankton, benthic vegetation, vascular marsh plants (primarly Phragmites communis and Salix gracilstyla) and riverine particulates inputs to the coastal bay food web was studied in Kwangyang Bay, Korea using stable carbon isotope ratios. Vascular marsh plants (${\delta}^{13}$C=-27.4${\pm}$0.8%o) and riverine particulates (-26.0${\pm}$0.8%o) were isotopically distinct from phytoplankton (-20.7${\pm}$0.8%o), microphytobenthos (-14.2${\pm}$0.6%o) and seagrass (8.8%o). The ${\delta}^{13}$C values of consumers in the study site ranged from -20.2 to -11.3olo suggesting the assimilation of carbon derived from both phytoplankton and benthic vegetation (including algae and seagrass), The relative importance of both pelagic and benthic origins of food sources was likely to vary depending on feeding habit of the consumers. The isotopic difference between pelagic and benthic consumers indicated that plankton-derived carbon was used mostly by pelagic consumers, but the carbon derived from intertidal benthic vegetation was incorporated into food webs through benthic consumers. The ${\delta}^{13}$C values of consumers in the present study differed noticeably from published values of the phytoplankton-based ecosystem, particularly in the $^{13}$C enrichment of benthic grazers, deposit-feeders and demersal feeders of fishes. This tendency of the $^{13}$C enrichment was also found in suspension-feeding bivalves. Taking the biomasses of benthic vegetation into consideration, benthic microalgae was likely to account for the consumer $^{13}$C enrichment. Role of terrestrially derived riverine carbon was limited to the riverine system and was not evident within the bay systems. Phragmites, despite their important biomass, appeared to be of little importance as consumer diet.

  • Carboxymethylated polyamine-polyurea resin loaded with rubeanic acid (RCCPPI resin) was obtained by 1 step chemical reaction between chlorocarboxymethylated polyamine-polyurea(CCPPI) resin as matrix polymer and rebeanic acid. This resin was confirmed with infrared spectrometry, elemental analysis, and thermal analysis(DSC). The adsorption characteristics of the heavy metal's on the resin were studied by measuring distribution coefficient($K_d$) with changing pH of the solutions and frontal chromatography. The enrichment, recovery, and analysis of trace heavy metals, such as cadmium, cerium, copper, nikel, lead, and zinc, in the presence of high concentrations of sodium, calcium, and acetate salts was possible quantitatively by a column packed with the resin at each optimum pH. Preconcentration factors were more than 25. To elute the adsorbed heavy metals on the resin, 0.025M EDTA solution(pH 9.0) was used.

Development of Bioreactors for Enrichment of Chemolithotrophic Methanogen and Methane Production (독립영양형 메탄생산세균의 농화 및 메탄생산 반응기의 개발)

  • A gas-circulating bioreactor was used for enrichment of autotrophic methanogens. Mixture of hydrogen and carbon dioxide (5:1) was used as a sole energy and carbon source. Anaerobic digestive sludge isolated from wastewater treatment system was inoculated into the gas-circulating bioreactor. The enrichment of two chemolithotrophic methanogens, Methanobacterium curvum and Methanobacterium oryzae was accomplished in the gas-circulating bioreactor. The enriched bacteria were cultivated in a bioreactor equipped with hollow-fiber hydrogen-supplying system (hollow-fiber bioreactor), and a hybrid-type bioreactor equipped with hollow-fiber hydrogen-supplying system and electrochemical redox control system. The methane productivity was maximally 30% (V/V) in the hollow-fiber bioreactors and 50% (V/V) in the hybrid-type bioreactor.

Analysis of Key Genes and Pathways Associated with Colorectal Cancer with Microarray Technology

  • Objective: Microarray data were analyzed to explore key genes and their functions in progression of colorectal cancer (CRC). Methods: Two microarray data sets were downloaded from Gene Expression Omnibus (GEO) database and differentially expressed genes (DEGs) were identified using corresponding packages of R. Functional enrichment analysis was performed with DAVID tools to uncover their biological functions. Results: 631 and 590 DEGs were obtained from the two data sets, respectively. A total of 32 common DEGs were then screened out with the rank product method. The significantly enriched GO terms included inflammatory response, response to wounding and response to drugs. Two interleukin-related domains were revealed in the domain analysis. KEGG pathway enrichment analysis showed that the PPAR signaling pathway and the renin-angiotensin system were enriched in the DEGs. Conclusions: Our study to systemically characterize gene expression changes in CRC with microarray technology revealed changes in a range of key genes, pathways and function modules. Their utility in diagnosis and treatment now require exploration.

  • Currently, the robot is widely utilized in various educational settings such as after-school classes, and special classes for gifted students. The robot is widely recognized as a useful tool for helping students solve problems. The core activities of programming learning with robot need to provide various problem contexts to the students and guide students' problem solving process. Students gain cognitive and affective benefits when they solve problems with robots. This paper describes the impact of programming learning using robot based on schoolwide enrichment model on elementary school students' creative potential. As a result, the students of experimental group than the students of the control group improved creative personality and ideational behavior, and the gifted students of experimental group than the gifted students of control group improved ideational behavior.

N-Region Addition in Immunoglobulin Kappa Light Chains in B Cell Subsets in Rheumatoid Arthritis: Evidence for Over-expression of TDT in B Lineage

  • Background: Unusually high amounts of N region addition and CDR3 length diversity were found in immunoglobulin (Ig) light chain Vk and Jk joins in patients with rheumatoid arthritis (RA). We sought to determine whether this finding is due to excessive activity of the enzyme responsible for N region addition (terminal deoxynucleotidyl transferase [TdT]) in B lineage cells in bone marrow or from positive antigenic selection of B cells with long CDR3 lengths. Methods: We used FACS to isolate $IgM^+/IgD^+$ B cells (predominantly naive) and $IgM^-/IgD^-$ B cells (predominantly class-switched) B cells from peripheral blood of a patient with RA known to have enrichment for long Vk CDR3s and from that of two normal controls. RT-PCR of VkIII transcripts was performed, followed by sequencing of individual cDNA clones. We analyzed the CDR3 lengths and N region additions in 97 clones. Results: There was enrichment for long CDR3 lengths (11 or 12 amino acids) in both $IgM^+/IgD^+$ and $IgM^-/IgD^-$ B cells in RA compared to B cell subsets in the normal controls. The $IgM^+/IgD^+$ B cell subset in RA was markedly enriched for N region addition and was similar to that seen in the $IgM^-/IgD^-$ subset. Conclusion: These data suggest that enrichment for N region addition and long CDR3 lengths in RA may result from unusually high or prolonged activity of TdT in bone marrow.

  • Recent research has focused on alternative fuel to improve engine performance and to comply with emission regulation. Finding an alternative fuel and reducing environment pollution are the main goals for future internal combustion engines. The purpose of this study is to obtain low-emission and high-efficiency by hydrogen enriched CNG fuel in SI engine and is to clarify the effects of hydrogen enrichment in CNG fuelled engine on exhaust emission and performance. An experimental study was carried out to obtain fundamental data for performance and emission characteristics of hydrogen enrichment in SI engine. The experiment was conducted at 2500 rpm, bmep 2 bar, 4 bar conditions while CNG fuel was mixed with 10, 20 and 30% hydrogen blends. From the experimental results, combustion duration was shortened due to rapid flame propagation velocity of hydrogen and these were attributed to the burning velocity increasing exponentially with increasing hydrogen blending ratio. Hydrogen has much wider flammable limit than methane, gasoline and the minimum ignition energy is about an order of magnitude lower than for other combustion. By adding hydrogen, $CO_2$ and HC were reduced. However, $NO_X$ was increased dut to high rate of heat release for hydrogen substitutions.
