To investigate the applicability of hot water extract (PLW) and ethanol extract (PLE) from Phellinus linteus as functional food and cosmeceutical materials, its total flavonoids content, total phenolics content, electron donating ability (EDA), nitrite-scavenging ability (NSA), SOD-like activity, inhibitory effect of tyrosinase and elastase were examined. Total flavonoids contents of PLW and PLE were 17.31 mg/g and 42.61 mg/g, respectively, and total phenolics contents were estimated as 149.92 mg/g for PLW and 432.42 mg/g for PLE. The EDA of PLW and PLE were $6.49{\sim}92.98%$ and $22.61{\sim}94.28%$. The EDA and total phenolics contents had a high correlation of 0.83. The NSA was pH dependent, and was highest at pH 1.2 and lowest at pH 6.0. The NSA of PLE was higher than that of PLW. The SOD-like activities of PLW and PLE were $14.36{\sim}35.21%$ and $17.27{\sim}81.84%$, respectively, and the activity was dependent on the sample concentration. The tyrosinase inhibitory activity was the highest in PLE ($10.51{\sim}80.93%$) while that of PLW was $4.77{\sim}43.69%$. Finally, the elastase inhibitory activity was $10.01{\sim}76.02%$ at PLE. Based on the above results, we deemed that the ethanol extract of Phellinus linteus was the most pertinent for use as functional food and cosmeceutical materials.

The purpose of this study was to investigate the role of the Moringa oleifera (M. oleifera) extract as a cosmetic additive. The tyrosinase and elastase inhibitory effects showed 47% and 39% at $1,000{\mu}g/mL$ concentration, respectively. Also, the collagenase inhibition effect was 31% at $500{\mu}g/mL$ concentration. A cell viability test, measured on macrophage cell (RAW 264.7) and melanoma cell (B16F10) by ethanol extract of M. oleifera, showed 94.2% and 94.8% at $100{\mu}g/mL$ concentration, respectively. In order to confirm anti-inflammatory activity, we examined the inhibitory effects on the production of lipopolysaccharides (LPS)-induced NO in RAW 264.7 cells by Griess assay. As a result, the M. oleifera extract showed a concentration-dependent inhibition of NO production. The protein expression inhibitory effects of M. oleifera extract were measured by western blot at 25, 50, $100{\mu}g/mL$ concentration and the ${\beta}-actin$. Results showed that the expression inhibition rates of the iNOS, COX-2, MITF, TRP-1, TRP-2, tyrosinase protein were decreased by 85.8%, 57.5%, 80.7%, 30%, 29.9%, 23.6% at $100{\mu}g/mL$ concentration, respectively. It was concluded that M. oleifera extracts had the anti-inflammatory and whitening effects and thus could be applied for cosmetics as a natural ingredient.
In this study, graviola leaf extracts (GLE) was investigated for the effect of antioxidant, antibacterial, whitening, anti-wrinkle. The antioxidant effect of GLE was measured by an electron donating ability assay. As a result, GLE increased the electron donating ability in a concentration-dependent manner. The antibacterial effect of GLE was found to show the higher antibacterial effect in Methicillin-resistant Staphylococcus aureus CCARM3115 compared with that of ampicillin by a paper disc method. The whitening effect of GLE was also measured by tyrosinase inhibition assay, and it was found that the tyrosinase activity of GLE decreased as the concentration increased. The inhibition activity of tyrosinase involved in hydroxylation reaction which is related to converting L-tyrosine to (DOPA) was higher than that of arbutin's at the concentration ranging from 125 to $250{\mu}g/mL$. In addition, GLE reduced melanin contents of B16-F10 melanoma cells in a dose-dependant manner and decreased to about 76.7% at a concentration of $100{\mu}g/mL$ Regarding wrinkling formation of GLE, an elastase inhibition assay was performed. As a result, GLE and ursolic acid were 10.5% and 56.5%, respectively under the identical concentration. These results suggest that GLE has significant antioxidant and whitening activities, and also may be potentially used as a therapeutic agent for hyperpigmentation treatment as an ingredient of whitening cosmetics.
The purpose of this study was to research about the ingredients for anti-oxidation and anti-wrinkle effects of the solvent fractions from Agrimonia pilosa L. hot water (AW) and 70% ethanol (AE) extracts. The electron donating ability of the solvent fractions from AW and AE extracts showed 84.9, 92.5% in ethyl acetate layer of AW and AE at 1,000 ppm. The superoxide dismutase like activity of solvent fractions from AW and AE extracts showed 61.8% in ethyl acetate layer of AW extracts and 58.0% in buthanol layer of AE extracts at 1,000 ppm. For anti-wrinkle effect, elastase inhibition effect of the solvent fractions from AW and AE extracts showed 55.2, 70.1% in ethyl acetate layer of AW and AE extracts at 1,000 ppm. And collagenase inhibition effect of the solvent fractions from AW and AE extracts showed highest inhibition effect as 90.6% in ethyl acetate layer of AW extract and 88.6% in $n$-butyl alcohol layer of AE extract at 1,000 ppm. All these findings suggested that solvent fractions from AW and AE extracts has an anti-oxidation and anti-wrinkle effects.
Perilla leaves cultivated in greenhouses (Jan., May) and in the fields (Aug.) of Geumsan province were investigated for their extract yields and physiological functionalities. The yield was highest in 30% ethanol extracts of the August perilla leaves. The highest fibrinolytic activity (8.2 U) was observed in 30% ethanol extracts of the May perilla leaves, while the HMG-CoA reductase inhibition level, which is related to the inhibition of cholesterol biosynthesis, was 83% in water extracts of the August perilla leaves. Anti-hypertensive ACE inhibitory activity was 64.5% in the water extracts of the January perilla leaves, and antioxidative electron donating ability was the highest (69%) in 30% ethanol extracts of the August perilla leaves. Elastase inhibitory activity, which is related to the inhibition of skin aging, was highest (47.5%) in 30% ethanol extracts of the May perilla leaves. However, SOD-like activity, nitrite scavenging activity, and tyrosinase inhibitory activity were not detected were very weak in all samples.
The purpose of this study was to research the biological activity of ethanol extract from Smilax china L. which is a vine shrub belonging to the lily family. For antiwrinkle effects, elastase inhibition effect of ethanol and water extracts from S. china L. showed 41.1% and 16.3% at $1,000{\mu}g/ml$ concentration. The collagenase inhibition effect of ethanol and water extracts from S. china L. showed more than 96.6% and 60.0% at $1,000{\mu}g/ml$ concentration. As a result of having fibroblast measured cell viability on fibroblast cell of ethanol extract from S. china L., it showed 71.7% with cell viability at $100{\mu}g/ml$ concentration. At $50{\mu}g/ml$ concentration, the procollagen biosynthesis effect of ethanol extract from S. china L. was 139.86%. At the same concentration, the matrix metalloprotease (MMP)-1 inhibition effect of the ethanol extract was 74.9%. According to the results of Western blot of ethanol extract from S. china L., the expression of the MMP-1 protein was decreased by 35% at $50{\mu}g/ml$ concentration. Reverse transcription-polymerase chain reaction (PCR) of ethanol extract from S. china L. showed that the expression of MMP-1 mRNA was decreased by 45% at $50{\mu}g/ml$ concentration. The findings suggest that 70% ethanol extract from S. china L. (SC) has great potential as a cosmeceutical ingredient with antiwrinkle effects.

In this research, water and ethanol extract of Momordica charantia shells, fruits and seeds were tested to see possibility as natural functional cosmetic agent. Water and ethanol extract showed 69.45 mg/g and 70.87 mg/g polyphenol concentration, respectively. Momordica charantia water and ethanol extracts did not indicate cell toxicity up to $1,000{\mu}g/ml$ concentration in MTT assay. Tyrosinase inhibition effects of water and ethanol extract were lower than arbutin, however, ethanol extract showed better DOPA oxidation inhibition effect than arbutin. Elastase inhibition effects of ethanol extract displayed similar efficacy with adenosine at higher concentrations. Solution formulations (5% extract) were stable for 28 days in both extracts, however, lotion formulation (1% extract) showed considerable variation in viscosity whereas ethanol extraction indicated relative stability. In conclusion, water and ethanol extract of Momordica charantia shells, fruits and seeds indicated strong possibility for whitening and antiwrinkle functional cosmetic ingredient.
In order to develop a potent antidementia $\beta$-secretase inhibitor from phytochemicals, $\beta$-secretase inhibitory activities of extracts from many medicinal plants and herbs were determined. Water extracts from Rubus coreanus showed the highest $\beta$-secretase inhibitory activity of 84.5%. After purification of the $\beta$-secretase inhibitor from R. coreanus using systematic solvent extraction, ultrafiltration, Sephadex G-10 column chromatography, and reverse-phase high performance liquid chromatography (HPLC), a purified $\beta$-secretase inhibitor with $IC_{50}$ inhibitory activity of $6.3{\times}10^3\;ng/mL$ ($1.56{\times}10^{-6}\;M)$ was obtained with a 0.08% solid yield. The molecular mass of the purified $\beta$-secretase inhibitor was estimated to be 576 Da by liquid chromatography-mass spectrometry (LC-MS) and $\beta$-secretase inhibitor also is a new tetrapeptide with the sequence Gly-Trp-Trp-Glu. The purified $\beta$-secretase inhibitory peptide inhibited $\beta$-secretase non-competitively and also show less inhibition on trypsin, however no inhibition on other proteases such as $\alpha$-secretase, chymotrypsin, and elastase.
Antielastic fragment in GBⅠ-Ⅱ differ on $P_1$ site with that of antitryptic fragment in LBI. To obtain further understanding of the role of amino acid residue near the reactive site, specificity of $P_1$ site and loop size, Tyr substituted cyclic nonapeptide and cyclic pentapeptide were synthesized and the inhibition constants for some proteinases were calculated by Dixon method. Cyclic nonapeptide showed no inhibition for chymotrypsin but appeared low inhibitory activity for trypsin and elastase and that of cyclic pentapeptide possessed inhibitory activity for chymotrypsin.
We observed that treatment with dimethyl α-ketoglutarate (DMK) increased the amount of intracellular α-ketoglutarate significantly more than that of α-ketoglutarate in HaCaT cells. DMK also increased the level of intracellular 4-hydroxyproline and promoted the production of collagen in HaCaT cells. In addition, DMK decreased the production of collagenase and elastase and down-regulated the expression of selected matrix metalloproteinases (MMPs), such as MMP-1, MMP-9, MMP-10, and MMP-12, via transcriptional inhibition. The inhibition of MMPs by DMK was mediated by the suppression of the IL-1 signaling cascade, leading to the attenuation of ERK1/2 phosphorylation and AP-1 transactivation. Our study results illustrate that DMK, an alkylated derivative of α-ketoglutarate, increased the level of 4-hydroxyproline, promoted the production of collagen, and inhibited the expression of selected MMPs by affecting the IL-1 cascade and AP-1 transactivation in HaCaT cells. The results suggest that DMK might be useful as an anti-wrinkle ingredient.
