Effects of Alternate Treatment of Estrogen Receptor Antagonist and Agonist on Morphology of Male Reproductive Organs of Adult Mice

  • ICI 182,780 (ICI) is known as an estrogen receptor antagonist, whereas propyl pyrazole triol (PPT) is an estrogen receptor agonist. In this study, ICI or ICI added with PPT was injected into adult male mice. Body and reproductive organ weights were reduced in the ICI added with PPT group compared to the control group. Further, the ICI and ICI added with PPT groups both showed increases in luminal areas of the seminiferous tubules of the testis, whereas cell heights of efferent ductules and the initial segment of the epididymis were reduced. Sperm count in the caudal epididymis was reduced in the ICI and ICI added with PPT groups. These results show that reproductive tissues were more deeply affected in the ICI added with PPT group. We also demonstrated that treatment with ICI resulted in histological changes in the testis, efferent ductule, and epididymis. Further, alternate treatment with ICI and PPT induced abnormalities in reproductive organs. These results indicate that a high concentration of PPT together with ICI may cause histological abnormalities instead of histological restoration in reproductive organs.

Localization of Methyl Mercuric Chloride in the Reproductive Tract of Male Mice

  • Localization of mercury compounds was investigated in selective regions of the male reproductive tract using autometallography. The results demonstrated that mercury was observed in Sertoli and Leydig cells in testis, but not in the epithelial cells of rete testis and germ cells. In the efferent ductule, mercury compounds were observed in the cytoplasmic compartments of epithelial cells in the proximal and common regions, while they were observed in the supranuclear cytoplasmic compartments in the conus region. In the epididymis, the compounds were observed in the cytoplasmic compariments of narrow and basal cells, but not in the principal cells of the initial segment. In contrast, the compounds were evenly detected in the cytoplasmic compartments of principal cells in the caput. In the corpus and caudal epididymis, the compounds were observed in the basal region of principal cells. The data shows that mercury is differentially accumulated in the male reproductive tract in a region-specific manner.

수컷 생쥐 생식기관의 형태에 미치는 에스트로겐 수용체 알파의 선택적 촉진제의 농도별 영향 (Concentration Effect of Estrogen Receptor-${\alpha}$ Selective Agonist on the Morphology of Reproductive Organs of the Male Mice)

  • 암컷호르몬으로 알려져 있는 에스트로겐이 실제로는 수컷 생식기관에도 영향을 미친다. 에스트로겐은 수용체와 결합하여 그 작용을 나타낸다. Propyl pyrazole triol (PPT)은 에스트로겐 수용체 베타에 비해 상대적으로 알파에 410배 혹은 1,000배 더 높은 선택적 친화성으로 결합하는 에스트로겐 수용체 알파 촉진제이다. 본 연구에서는 성체 수컷 생쥐를 대상으로 castor oil에 각각 0.01 mg, 0.1 mg, 1 mg, 4mg의 농도로 희석한 PPT를 1주에 1회씩 8주 동안 피하주사 하였다. 대조군은 castor oil을 주 1회씩 동일한 기간 동안 생쥐에 피하주사 하였다. 정소, 수출소관, 부정소의 조직학적 변화를 현미경으로 관찰하였다. PPT 4mg의 고농도 투여군에서는 체중, 정소, 부정소의 무게가 뚜렷하게 감소되었다. 실험기간 동안 PPT에 의해 세정관의 직경, 부정소의 상피세포 높이가 감소되었다. 부정소에 붙어 있던 지방세포의 크기는 PPT에 의해 줄어들었다. PPT 4 mg 투여군의 경우, 부정소의 꼬리 부위에 저장되는 정자가 관찰되지 않았다. 결론적으로 고농도의 PPT 투여로 인해 성체 수컷 생쥐의 정자형성 억제와 같은 생리학적 변화가 유발되었고 동시에 생식기관의 조직학적 변화도 유발되었다.

  • 본 연구에서는 남성 생식 기관인 efferent ductules(ED)에서 monocarboxylate transporter (MCT) isoform과 Basigin(Bsg)의 mRNA 발현을 탐구하고, MCT1의 발현이 에스트로젠 수용체 α에 의해 조절되는지를 연구하였다. 생후 여러 연령대의 백서 ED에서 MCT isoform과 Bsg의 mRNA 발현 여부는 real-time PCR에 의해 조사 되었고, 정상 생쥐와 에스트로젠 수용체 α knockout 생쥐를 사용하여 에스트로젠 수용체 α에 의한 MCT1의 발현 조절 여부는 immunohistochemistry 방법에 의해 간접적으로 알아 보았다. 본 연구 결과는 백서의 ED에서 MCT1, 2, 3, 4와 8 그리고 Bsg 유전자의 발현은 연령에 따라 다르게 나타나며, MCT1의 발현이 ED의 ciliated 세포의 basolateral 지역에서 발현되고 어떤 측면에서 ERα에 의해 조절되어 질 수 있음을 보여 주었다. 따라서 본 연구 결과는 MCT가 monocarboxylate의 세포 내, 외부로 운반을 조절함으로써 남성 생식기의 일부인 ED의 역할을 조절하는데 관여함을 시사한다.

생후 발달과정동안 숫 백서의 Efferent Ductules에서 Connexins 발현 양상 연구 (Expression Profiling of Connexins in the Efferent Ductules of Male Rats During Postnatal Development)

  • 본 연구의 목적은 생후 발달과정에서 숫 백서 생식기의 일부인 efferent ductules (ED)에서 다양한 connexin 이형질체의 발현과 양상을 알아보았다. 1주, 2주, 1개월, 3개월 그리고 6개월령의 백서의 ED로부터 total RNA을 분리하였으며, semi-quantitative RT-PCR 방법을 사용하여 총 14개 connexin 이형질체의 mRNA 발현 양상을 알아보았다. 백서의 ED에서 연구된 14개 중 6개 connexin 이형질체의 발현이 보여졌으며, connexin 26과 30의 mRNA 발현은 연령에 따라 심한 변화를 보여주었다. Connexin 31.1의 발현은 1달령의 ED에서 가장 낮게 나타났으며, connexin 37과 45는 주로 초기 발달 과정에서 높은 수준으로 발현됨을 보여주었다. 또한, connexin 43의 발현은 1주령에서 가장 낮았으며 2주령 이후에는 큰 변화없이 일정한 수준을 유지하였다. 본 연구는 백서의 ED에서 다양한 종류의 connexin 이형질체가 발현되며, 이러한 발현은 생후 연령에 따라 서로 다르게 조절됨을 보여준다.

  • 본 연구는 생후 발달과정에서 숫 흰쥐의 생식기인 Efferent ductules(EDs)과 부정소에서 cytochrome P450 aromatase(Cyp19) 유전자의 발현과 연령별 발현 양상을 알아보는데 목적이 있다. 조직으로부터 추출된 total RNA는 역 전사 반응을 통해 cDNA로 바뀌어진 후 real-time PCR 기법을 사용하여 부위별 그리고 연령별로 Cyp19 유전자 발현을 알아보았다. Cyp19 유전자의 발현은 EDs에서 90일령을 제외한 7일, 14일, 그리고 30일령 등 성숙기 이전의 모든 연령에서 나타났으며, 부정소에서는 7일령을 제외한 모든 연령에서 보여졌다. 특히 부정소에서 Cyp19의 발현은 부정소의 각 부위별로 특이한 양상을 보였다. 본 연구 결과를 통해 남성 생식기의 EDs와 부정소의 각 부분에서 Cyp19 유전자 발현은 연령과 부위에 따라 차별적으로 조절되는 것을 알 수 있었다.

오리 부고환(副睾丸) 및 정관(精管)의 주령별(週齡別) 조직학적(組織學的) 및 조직화학적(組織化學的) 연구(硏究) (Histological and Histochemical Studies on the Epididymal Region and Deferent Ducts of the Drakes by the Age in Weeks)

  • This study was made for the better information of the male reproductive system on the meat-type drake, Cherry Belly X White Golden. The epithelium of ductules of epididymal region and deferent duct were observed histologically and histochemically with the progress of their development. India-ink absorbability on the luminal epithelium was also investigated after the administration of India-ink. The results are as follows; 1. Rete testis and various round ductules in immature form appeared in epididymis within 6 weeks after hatching, and simple cuboidal and simple columnar epithelium were found in the epithelia of the ductules within 8 weeks after hatching. Larger ductules were found on epididymal surface which was in the developing stage near to the immature efferent ductule. From 10th to 20th week, various ductules appeared in epididymis, and developing form of efferent ductules were much more increased on epididymal surface. The luminal epithelium of the ductules were composed of ciliated simple columnar and pseudostratified ciliated columnar cells. At the same time, deferent duct appeared. From the 21th week, various ductules in epididymis became abruptly matured. Lumen of rete testis was lined by simple squamous or simple cuboidal epithelium, and that of efferent ductules, having many folds and being larger than any others were lined by pseudostratified ciliated columnar epithelium in which ciliated columnar cells, non-ciliated cells(clear cells) and basal cells were noted. Connecting tubules of star shaped lumen were composed of pseudostratified ciliated columnar epithelium in which ciliated columnar cells, nonciliated cells, and basal cells were observed. The luminal surface of epididymal ducts was smooth and has thick pseudostratified columnar epithelium which was composed of high columnar cells and basal cells. From 26th week after hatching, sperm pooling was started in various ductules. 2. From 4th to 10th week, simple cuboidal epithelium of deferent duct transformed to simple columnar epithelium with the progress of aging. At the basement of epithelium, clear round cells were noted. From 12th to 20th week, high columnar cells with enlongated nucleus were noted on the luminal border of deferent ducts, forming folds of pseuclostratified columnar epithelium. From 20th week, the deferent duct started to have septa in it's lumen and composed mainly of pseudostratified columnar epithelium, and round cells disappeared. From 20th week, the lumen diameter of deferent duct became wider with the progress of aging, but there was no difference among the values of lumen diameter in upper, middle, and lower part of deferent ducts. At 26th week, the pooling period of sperms in deferent ducts, the lumen diameter became rapidly widen, especially in the lower part of deferent ducts. Thickness of muscular layer of ductus deferens showed gradual growth within 24 weeks but did abrupt thickening from 26th week. 3. Saliva resistant PAS granules were dotted on the top of nucleus in efferent ductules epithelium but the amount of the granules were little in the connecting ductules's epithelium. The granules reactive to acid phosphatase were abundant in the some epithelial cells of efferent ductules and connecting ductules, especially above the nucleus of cells. The granules reactive to alkaline phosphatase were noted on the luminal border of efferent ductules. Parts of free border of efferent ductules and middle portion of deferent ducts were stained slightly by alcian blue technique. India ink granules were found mainly in the epithelium of efferent ductules but were few in that of connecting ductules.
