Changes in physicochemical components of Astragalus membranaceus fermented with Phellinus linteus (상황버섯균 접종 황기의 배양 중 이화학적 성분변화)

  • This study analyzed the changes in physicochemical components of Astragalus membranaceus (AM) fermented with Phellinus linteus. Moisture content, pH, total acidity, total reducing sugar content, extraction yield, free sugar content, free amino acid and isoflavonoid (calycosin, formononetin) were investigated. The moisture content was increased during fermentation with Phellinus linteus. The pH level increased while the total acidity significantly decreased during fermentation. The reducing sugar content were in the range of 0.32~0.61%. The extraction yield using water was higher than that using 80% ethanol. The major free sugars were identified as glucose, fructose, sucrose and the content of free sugars decreased through fermentation. However, the glucose and sucrose contents of the water extracts were increased. In addition, the free amino acid increased significantly during fermentation. Finally, calycosin and formononetins contents in water extracts of after 30 days of AM fermentaion with Phellinus linteus were (3.91 mg/100 g) and (1.38 mg/100 g), respectively. These results suggest that fermentation with Phellinus linteus could be used to increase the bioactivity of AM. The mycelium-fermented AM could be a valuable source of functional material and edible resource for industry.

Comparison of chewing ability and quality of life before and after the dental implantation (임플란트 시술환자의 시술 전.후의 저작능력과 삶의 질 비교)

  • Statement of problem: Recently the populations of patients receiving implant surgery are greatly increase for maintaining oral health. Purpose: This study was conducted for implanted patients to assess the chewing ability patient satisfaction level and changes in quality of life before and after the implant surgery. Material and methods: The current study subjected 109 adult patients, older than 20 years of age, who received implant surgery from December, 2006 to October, 2007 at the 6 dental clinics located at Daegu and Ulsan metropolitan cities. Twice of surveys were conducted for the patients before and after receiving the dental implant surgery. Results: As the motivation of receiving implant surgery, 45.9% of the patients selected the surgery for the chance o "f chewing function recovery", and " failure of treatment and complications" was found to be the most worrisome at the time of surgery by recording 38.5%.The satisfaction level before the implant surgery scored 30.37, while the score was increased to 45.01 after the surgery by showing a significant difference before and after receiving the surgery(P<.001). Regard on the surgery, 91.8% of the patients responded as "Satisfy", and 89% of the study subjects responded that they have willingness to recommend the surgery to their families and friends. The chewing ability score measured by using the surveys on edible foods, the score before the surgery was 15.24, while the score was increased to 19.11 after the surgery by showing a significant difference before and after receiving the surgery(P<.001). The quality of life score was also found to be increased to 11.17 after the surgery from 9.99 before the surgery by showing a significant difference(P<.001). Conclusion: In a future, the studies on the numbers of implanted loss teeth and the location of tooth loss are necessary, more long-term follow study are needed, and it is thought to be necessary to enlarge the sample size of subjects in conducting the studies.

Changes in quality of fried rice with crab meat depending on the storage period and temperature (저장기간 및 저장온도에 따른 꽃게풍미 볶음밥의 품질변화)

  • The edible crab is among the rich resources that can be used as materials for food products, but living crabs and crabs marinated in soy sauce are the only widely utilized materials for food products. For seafood development, research is needed to promote the use of crabs in various products. In this study, the quality changes of fried rice prepared with crab meat, crab emulsion sauce and crab cooker effluent were investigated. The pH and acidity values did not show any significant difference at $-20^{\circ}C$. The acidity of the fried rice continuously increased during storage at 4 and $25^{\circ}C$. The VBN and TBA values of the fried rice stored at 4 and $25^{\circ}C$ were significantly higher than those of the fried rice stored at $-20^{\circ}C$ during the storage period. The viable cell count of the fried rice stored at $-20^{\circ}C$ changed little during the storage period while that of the fried rice stored at 4 and $25^{\circ}C$ rapidly increased over 2 weeks. For the fried rice stored at $-20^{\circ}C$, the overall quality did not show significant differences during the storage period. In conclusion, the best storage temperature for both quality and safety was found to be $-20^{\circ}C$. At such storage temperature, the quality of the stored instant rice using crab meat did not change, and through research the expanding possibilities of using crab materials were confirmed.

Flora and Vegetation of the Southern Slope Area at Mt. Chongok and Mt. Tuta(Kangwon-do) (청옥산 - 두타산 남사면 일대의 식물상과 식생)

  • Floristic composition and phytosociological studies of Mt. Chongok and Mt. Tuta were investigated, and that was compared with the previously published report in 1993. Vascular plants were composed of 100 families, 358 genera, 573 species, 95 varieties, and 18 formae, totaling 686 taxa. The vegetation was relatively well conserved based on Pteridophyta calculation (Pte-Q), 1.13. Compared with the vascular plants of the southern and northern slope area, the vascular plants of the southern slope area were composed of 87 families, 287 genera, 419 species, 73 varieties, and 11 formae, totaling 503 taxa, and those of the northern slope area consisted of 94 families, 293 genera, 427 species, 73 varieties, and 12 formae, totaling 512 taxa, respectively. Also, compared with the taxa in each side, both sides were composed of 332 species in common, southern sides, 172 species and northern sides, 182 species, respectively. The number of species of 11 families belonged to the higher level among total families taxa was composed of 328 species(47.8%). Among them, Compositae and Rosaceae were included much more species than remnant families. Korean endemic species were composed 16 families, 24 genera, 20 species, 8 varieties and 2 formae, totaling 30 species(4.4%). Compared with the Korean endemic taxa in each side, both sides were composed of 14 species in common, southern sides, 11 species and northern sides, 5 species, respectively. A naturalized plants were 20 species, correspond to 9.2% of totaling 218 species appeared in South Korea. Among them,12 species were appeared commonly in both sides, southern sides, 16 species and northern sides, 16 species, respectively. Life form spectra was H-D1-R5-e type and, useful resources plants are as follows; edible source(42.4%), medicinal source(31.5%), ornamental source(15.6%) and pasture source (13.3%) in the total region. The forest vegetation of the southern slope was classified into 1 order, 1 alliances and 5 communities; Rhododendro-Quercetalia mongolicae, Lindero-Quercion mongolicae, Quercus mongolica Typical community, Populus davidiana-Quercus mongolica community, Pinus koraiensis-Taxus cuspidata community, Pinus densiflora-Carex humilis var. nana community, Betula costata-Betula ermanii community. It is considered that the slight difference of the flora and vegetation in the northern and southern slope is mostly due to the topographical and climatic difference. Even closer investigation is required for the more accurate comparison in this area.

Distribution of Vascular Plants at the Ecological Landscape Conservation Area Heoninlleung in Seoul (서울시 생태.경관보전지역 헌인릉의 관속식물 분포)

  • To clarify the distribution of vascular plants and their usefulness in Heoninlleung, Ecological Landscape Conservation Areas of Seoul, we investigated it from April, 2006 to June, 2009. Total 313 taxa; 68 families, 191 genera, 264 species, 41 varieties and 8 forma were distributed in Heoninlleung. Among them, 37 taxa were highly abundant everywhere (3A), 16 taxa were highly abundant locally (3B), 70 taxa were moderately abundant everywhere (2A), 96 taxa were common in certain regions locally (2B), 9 taxa were rare but observed everywhere with low frequency (2A) and 85 taxa were rare and observed locally (1B). The economic plants were 293 taxa. There were 156 taxa of edible source, 223 taxa of medicinal source, 141 taxa of ornamental source, 69 taxa of pastoral source, 12 taxa of industrial, and 8 taxa of timber source. Twelve Korean endemic plants were collected. Based on the list of rare plants by the Korea National Arboretum and Ministry of Environment, 2 rare species were found. The specific species of I~V grades by phytogeography were 19 taxa. And twentyfour taxa of naturalized plant species were distributed. Naturalization Index was 7.7% and Urbanization Index was 8.4% in the investigated area.

Total Polyphenol Contents and Antioxidant Activities of Methanol Extracts from Vegetables produced in Ullung Island (울릉도산 산채류 추출물의 총 폴리페놀 함량 및 항산화 활성)

  • To discover new functional materials using edible plants, antioxidant activities of methanol extracts from various parts of seven wild vegetables were investigated in vitro. Total polyphenol contents, determined by Folin-Denis method, varied from 16.74 to $130.22{\mu}g/mg$. Radical-scavenging activities of methanol extracts were examined using ${\alpha},\;{\alpha}-diphenyl-{\beta}-pirrylhydrazyl$ (DPPH) radicals and 2,2'-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) (ABTS) assay. Inhibition effects on peroxidation of linoleic acid determined by ferric thiocyanate (FTC) method and on oxidative degradation of 2-deoxy-D-ribose in Fenton-type reaction system were dose-dependent. Athyrium acutipinulum Kodama (leaf and rood), Achyranthes japonica (Miq.) Nakai (seed), and Solidago virga-aurea var. gigantea Nakai (root) showed relatively high antioxidant activities in various systems.

  • In order to evaluate the utility of the anthocyanins of Amaranthys tricolor L. as an edible pigment, the present study was undertaken to investigate the effects of pH. temperature, ascorbic acid, sugars and their degradation products, quercetin, thiourea, sodium pyrophosphate and metal ions on the stability of the anthocyanins in the model systems. The results obtained from this study were as follows. 1. The degradation of total anthocyanins was retarded as the pH levels decreased from 8.0 to 1.0. At pH 1.0, however. the initial degradation reaction proceeded faster than at pH 2.0 to 3.0 2. On heating in buffered aqueous solution at $80^{\circ}C$, the total anthocyanin content was higher at pH 2.0 than other pH levels. Increasing the storage temperature accelerated greatly the pigment degradation. In darkness at $40^{\circ}C$, after 10 days, only 19% of the original amount was left, while at $2^{\circ}C$, under the same conditions of storage, approximately 90% of the pigment was retained. The half-life of the pigment, 63.0 days at $2^{\circ}C$, shortened to 1. 7 days at $40^{\circ}C$. 3. An increase in ascorbic arid concentration from 0. 15 to 0.50 mg/ml lowered the anthocyanin retention. 4. There was no significant difference between glucose and fructose in anthocyanin degradation effect. Furfural was more effective than other sugar degradation products, formic acid or levulinic acid in accelerating anthocyanin breakdown. 5. Neither quercetin nor sodium pyrophosphate had a protective effect on the anthocyanins in the presence of ascorbic acid, while, in the systems 0.5 or 1 mg/ml of thiourea with $150{\;}{\mu}g/ml$ of ascorbic acid, the loss of anthocyanins was significantly reduced. 6. Both mercuric and cupric ions in 30 ppm greatly accelerated anthocyanin degradation.

Quality Characteristics of Black Soybean Paste (Daemaekjang) Prepared with Bacillus subtilis HJ18-4 (Bacillus subtilis HJ18-4를 이용하여 제조한 대맥장의 품질 특성)

  • Black soybean paste (BSP) is a bealmijang manufactured with barley flour and black soybeans. This study investigated the quality characteristics of BSP prepared with Bacillus subtilis HJ18-4 (HJ18-4). ${\alpha}$-Amylase and protease activities of BSP rapidly increased on day 6 and day 9, respectively, ($41.45{\pm}0.3-42.72{\pm}0.16$, $21.84{\pm}0.24-23.11{\pm}0.35unit/g$) after which the activities were constant or decreased slightly. Amino type nitrogen contents increased until day 23 ($173.78{\pm}3.51-195.63{\pm}1.51mg\;%$), whereas ammonia type nitrogen sharply decreased on day 9. On day 30, lactic acid bacteria counts were higher compared with the initial count ($7.91{\pm}0.05-8.17{\pm}0.02log\;CFU/g$). Meanwhile, B. cereus and total aerobic counts in HJ18-4 added BSP were lower than that in the control. These results implied that BSP could be edible after 30 days of fermentation, and the addition of HJ18-4 in meju could contribute to the safety of the black soybean paste by the inhibition of B. cereus growth.

Activation of Pro-Apoptotic Multidomain Bcl-2 Family Member Bak and Mitochondria-Dependent Caspase Cascade are Involved in p-Coumaric Acid-Induced Apoptosis in Human Jurkat T Cells (p-Coumaric acid에 의해 유도되는 인체 Jurkat T 세포의 에폽토시스 기전)

  • The apoptogenic effect of p-coumaric acid, a phenolic acid found in various edible plants, on human acute leukemia Jurkat T cells was investigated. Exposure of Jurkat T cells to p-coumaric acid (50-$150{\mu}M$) caused cytotoxicity and TdT-mediated dUTP nick-end labeling (TUNEL)-positive apoptotic DNA fragmentation along with Bak activation, ${\Delta}{\psi}m$ loss, activation of caspase-9, -3, -7, and -8, and PARP degradation in a dose-dependent manner. However,these apoptotic events were completely abrogated in Jurkat T cells overexpressing Bcl-2.Under these conditions, necrosis was not accompanied. Pretreatment of the cells with the pan-caspase inhibitor (z-VAD-fmk) could prevent p-coumaric acid-induced sub-$G_1$ peak representing apoptotic cells, whereas it failed to block ${\Delta}{\psi}m$ loss, indicating that the activation of caspase cascade was prerequisite for p-coumaric acid-induced apoptosis as a downstream event of ${\Delta}{\psi}m$ loss. FADD- and caspase-8-positive wild-type Jurkat T cell clone A3, FADD-deficient Jurkat T cell clone I2.1, and caspase-8-deficient Jurkat T cell clone I9.2 exhibited similar susceptibilities to the cytotoxicity of p-coumaric acid, excluding an involvement of Fas/FasL system in triggering the apoptosis. The apoptogenic activity of p-coumaric acid is more potent in malignant Jurkat T cells than in normal human peripheral T cells. Together, these results demonstrated that p-coumaric acid-induced apoptogenic activity in Jurkat T cellswas mediated by Bak activation, ${\Delta}{\psi}m$ loss, and subsequent activation of multiple caspases such as caspase-9, -3, -7, and-8, and PARP degradation, which could be regulated by anti-apoptotic protein Bcl-2.

Environmental Condition for the Butt-Rot of Conifers by Cauliflower Mushroom (Sparassis crispa) and Wood Quality of Larix kaempferi Damaged by the Fungus (꽃송이버섯에 의한 침엽수 심재부후 발생환경 및 낙엽송 피해목의 재질 특성)

  • Cauliflower mushroom (Sparassis crispa) is recently recognized as a new edible and/or medicinal mushroom cultivated with conifers. By the way, the mushroom is notorious as a brown-rot fungus that causes a buttrot of larch. So, there should be a careful consideration to apply the mushroom cultivation in coniferous stand. This study was conducted to clarify the seriousness of heartwood decay on conifers such as larch by cauliflower mushroom with surveying the mushroom producing environment and to examine whether the cultivation of cauliflower mushroom produce any problem in conifer stands or not. The mushroom occurred in various coniferous stands such as Larix kaempferi, Pinus koraiensis, P. densiflora and Abies holophylla on fertile soils with adequate moisture. Soil texture of the mushroom producing site was comparatively fine compared to general forest soils; sandy loam, loam and silty loam. Soil pH ranged from 4.6 to 5.2, and organic matter contents were 4~11%, which showed relatively wide range. We could find S. crispa by a DNA technique from the wood that seemed to have no heartwood decay by naked eyes. The damaged wood showed 30% higher moisture contents than that of sound wood, while the compressive strength was 30% lowered down compared to that of sound wood. The fungus may invade conifers through the scars occurred on roots or stems, in this case spore dispersion of the mushroom takes a great role. Thus, we concluded that forest tending activities need to be applied with considering the invasion of S. crispa, and cultivation of cauliflower mushroom in forest should be attempted very carefully. By the way, we also infer that conifer stands can be nurtured without heartwood decay by S. crispa if the stand be managed in good aeration conditions by proper silvicultural practices such as sanitary thinning.
