Antioxidant activities and tyrosinase inhibitory effects of guava (Psidium guajava L.) leaf (구아바(Psidium guajava L.) 잎의 항산화 활성 및 tyrosinase 저해효과)

  • This research was designed to investigate the antioxidant activities and tyrosinase inhibitory effects of guava (Psidium guajava L.) leaf. Total phenol content was obtained from guava leaf extract of 19.0 (g/100g, D.W.). The crude extract exhibited significantly antioxidant activities (IC50value $102.5{\mu}g/ml$, free radical scavenging; $49.4{\mu}g/ml$, SOD like activity). The crude extract of guava leaf was fractionated into four partition layers; hexane (G-H), ethyl acetate (G-E), butanol (G-B) and water (G-W) layer. The extracts of G-E, G-B, G-W showed high radical scavenging activities of over 50% at $100{\mu}g/ml$. SOD like activities of G-E, G-B, G-W were revealed, as 81.8%, 84.7%, 65.3% at $100{\mu}g/ml$, while those of G-H did not showed the effectively. The crude extract of guava leaf showed high tyrosinase inhibitory effect as 60.8% at 1mg/ml, the measurement of G-E, G-B, G-W were 65.2%, 62.8%, 51.6% and that of G-H was not effective. These results indicate that useful bioactive substances exist in the guava leaf extracts, especially G-E, G-B. And the guava leaf has the potential of being developed into health related products.

The Relationship between the Progression of Chronic Kidney Disease and Beta Cell Function in Non-Diabetic Korean Adults (대한민국 비당뇨 성인에서 만성신장질환과 인슐린저항성 및 베타세포기능의 관련성)

  • This study examined the relationship between chronic kidney disease (CKD) and the homeostasis model assessment of insulin resistance (HOMA-IR) and beta-cell function (HOMA-B) in non-diabetic Korean adults. This study included 4,380 adults aged 20 or older (50.32±16.14) using the 2015 Korea National Health and Nutrition Examination Survey (KNHANES) data, which represents the national data in Korea. The present study had several key findings. First, in terms of HOMA-IR, after adjusting for the related variables (Model 4), the HOMA-IR (M±SE, 95% confidence interval [CI]) in group 1 (G1; estimated glomerular filtration rate [eGFR], ≥90 mL/min/1.73 ㎡), group 2 (G2; eGFR, 60~89 mL/min/1.73 ㎡), group 3a (G3a; eGFR, 30~59 mL/min/1.73 ㎡), and ≥group 3b (≥G3b; eGFR, <30 mL/min/1.73 ㎡) were 1.78±0.03 (1.73~1.83), 1.87±0.03 (1.81~1.93), 2.16±0.13 (1.91~2.42), and 2.59±0.24 (2.12~3.06), respectively. The HOMA-IR was positively associated with the progression of CKD (P<0.001). Second, in terms of the HOMA-B, after adjusting for the related variables (Model 4), the HOMA-B (M±SE, 95% CI) in G1, G2, G3a, and ≥G3b were 87.46±1.21 (85.08~89.84), 89.11±1.38 (86.40~91.81), 104.82±5.91 (93.23~116.42), and 123.97±10.87 (102.66~145.29), respectively. HOMA-B was positively associated with the progression of CKD (P<0.001). Both insulin resistance and the beta-cell function were positively associated with CKD in non-diabetic Korean adults.

  • Poncirus trifoliata (L.) Raf (Rutaceae) fruits (PTFE) has been used for the treatment of allergic disease. IgE is normally one of the least abundant immunoglobulin (Ig) isotypes in the serum of both humans and several species of experimental animals: however a number of different stimuli can result in profound increases in IgE levels relative to other isotypes. In rodents, infection with many parasitic helminths can cause approximately 100-fold elevation in IgE within 2 wks. Immunization of mice with small amounts of protein antigens on alum also results in 10-fold to fold increase in total serum IgE, much of it specific for the immunizing antigen. In this experiment, I investigated the effect of an aqueous extract of Poncirus trifoliata (L.) Raf (Rutaceae) fruits (PTFE) on a in vivo and in vitro IgE production. PTFE dose-dependently inhibited the serum levels of IgE induced by antigens. The regulation of IgE synthesis is influenced by T cells and T cell derived factors. IL -4, a T cell-derived cytokine, has been shown to stimulate murine IgE synthesis both in vitro and in vivo. Current evidence suggests that IL-4 induces IgE synthesis in the mouse by stimulating H chain isotype switch. Lipopolysaccharide (LPS) plus IL-4 cause about l00-fold increase in IgE secretion by murine B cells. The effects of PTFE on the IL-4-dependent IgE response by mouse whole spleen cells were studied. Whole spleen cells were cultured for 7 days in the presence of LPS plus IL-4 and PTFE and the supernatants were assayed for IgE. IL-4 dependent IgE production of LPS-stimulated whole spleen cells was inhibited by PTFE. Moreover, in the present study using U266Bl human IgE-bearing B cells, I found that PTFE inhibited the production of IgE activated by LPS plus IL-4. These results indicate that PTFE have antiallergic activity by inhibition of IgE production from B cells.

Prophylactic Uses of Probiotics as a Potential Alternative to Antimicrobials in Food Animals

  • The antagonistic activity of probiotic strains (Bifidobacterium animalis BB-12, Bifidobacterium bifidum A, Bifidobacterium longum B6, Lactobacillus acidophilus ADH, Lactobacillus paracasei ATCC 25598, and Lactobacillus rhamnosus GG) against nalidixic acid resistant ($NA^R$) Escherichia coli O157:H7 MF1847, E. coli O157:H7 H2439, E. coli O157:H7 ATCC 43894, and E. coli O157:H7 C7927 was investigated using the agar-overlay, well diffusion, and broth culture tests. L. paracasei ATCC 25598 was the most effective probiotic strain in terms of in vitro antagonistic activity against $NA^R$ E. coli O157:H7, followed by L. rhamnosus GG, B. longum B6, and L. acidophilus ADH. The use of selected probiotic strains could be an effective pre-harvest intervention strategy to reduce the risk of $NA^R$ E. coli O157:H7 by maintaining a balanced microflora in animals and might provide many potential benefits in lieu of using antimicrobials.

Effect of Methyl Jasmonate on the Root Growth and the Eleutheroside Accumulation in the Adventitious Root Culture of Eleutherococcus senticosus (가시오갈피나무 부정근(不定根) 배양(培養)에서 부정근(不定根)의 생장(生長)과 Eleutheroside류(類)의 생산(生産)에 미치는 Methyl jasmonate 처리(處理)의 영향)

  • This study was carried out to investigate the dose-dependent effect of methyl jasmonate on both the adventitious root growth and the accumulation of various eleutherosides in the bioreactor culture of Eleutherococcus senticosus adventitious roots. The highest biomass production (5.4 g DW/L) was observed in the absence of methyl jasmonate and the root growth was significantly decreased by increasing the methyl jasmonate concentration. However, methyl jasmonate stimulated the production of both eleutheroside B, E and $E_1$. The highest level of eleutheroside B (359.9 ${\mu}g$/g DW) was obtained at 40 ${\mu}M$ of methyl jasmonate, while eleutheroside E and $E_1$ was accumulated at the highest level by the addition of 10 ${\mu}M$ of methyl jasmonate. Total eleutheroside was increased up to 3818.1 ${\mu}g$ per liter when 10 ${\mu}M$ of methyl jasmonate was applied. In addition, when the adventitious roots were cultured with 20 ${\mu}M$ of methyl jasmonate, the highest levels of eleutheroside B, E and $E_1$ were observed at the 12th, 3th and 9th days of culture, respectively.

An Empirical Study on the Driving Force for Diffusion of Surrender B/L as an International Trade Payment Document (무역결제서류로서 Surrender B/L의 확산 동인에 관한 실증연구)

  • Electronic bill of lading, Letter of Guarantee, Surrender B/L, Seaway Bill, etc. have been proposed as alternative tools to overcome the crisis of bill of lading, but the most useful of these is by far the Surrender B/L. However, since the Surrender B/L has various problems due to the absence of a legal basis, studies so far have been conducted focusing on these legal limitations or problems in use of the Surrender B/L. This study tried to empirically identify the factors that affect the spread of Surrender B/L by expanding this point of research view. A hierarchical regression analysis was conducted by acquiring 190 valid samples centered on member companies of the Korea International Freight Forwarders Association. In addition, the R program was used to diagnose the research data and analyze the degree of spread by region. As a result of the empirical analysis, it was found that the utilization of Surrender B/L increased due to the convenience and cost reduction effect of Surrender B/L and the apprenticeship training of forwarding companies. In addition, as a result of regional analysis, the spread of Surrender B/L was notable not only in neighboring regions but also even in areas far from Korea.

Prevalence and Molecular Characterization of Quinolone Antibiotic Resistance in Escherichia coli Isolates from Raw Bulk Milk in Gyeonggi-do (원유시료에서 분리한 대장균의 퀴놀론 항생제 내성 기전)

  • The aim of this study was to investigate the prevalence of quinolone resistant E. coli from raw bulk milk and to characterize the resistance determinants. In this study, the gyrA, gyrB, parC, and parE quinolone resistance determining regions (QRDR) were sequenced from quinolone resistant E. coli isolates. Also, the presence of plasmid-mediated quinolone resistance (PMQR) and the expression of efflux pump genes based on quantitative real-time PCR (qRT-PCR) were investigated. Of the 487 coliform bacteria, 9 strains showed nalidixic acid resistance, and 6 of the 9 nalidixic acid resistant isolates were also ciprofloxacin resistant. These 9 strains had a single mutation at codon 83 (S83L) in gyrA, 2 of them had double mutations at codon 83 and 87 (S83L and D87N) in gyrA and 3 of the 9 isolates had single mutations at codon 80 (S80I) in parC. None of the 9 isolates harbored PMQR determinants. Compared with wild-type E. coli ATCC 25922, an over-expression of the acrB gene (2.15-5.74 fold), encoding the pump component of the AcrAB-TolC efflux pump was observed in 4 of 6 ciprofloxacin resistant isolates. This study identified the quinolone resistance mechanism of E. coli isolated from raw milk samples in Gyeonggi-do.

  • The addition reactions of cysteine without blocking amino and carboxyl groups to substituted and unsubstituted ${\beta}$-nitro-styrene derivatives were investigated. ${\beta}$-Nitrostyrene(1a), p-methyl-${\beta}$-nitrostyrene(1b), 3,4,5-trimethoxy-$[\beta}$ -nitrostyrene(1c), $[\varpi}$-3,4-methylenedioxy-${\beta}$ -nitrostyrene(1d), o-, m- and p-chloro-${\beta}$ -nitrostyrene (1e, 1f, 1g) and o-, m- and p-methoxy-${\beta}$-nitrostyrene (1h, 1i, 1j) easily undergo addition reactions with cysteine to form S-(2-nitro-1-phenylethyl)-L-cysteine(3a), S-[2-nitro-1-(p-methyl)phenyl-ethyl]-L-cysteine(3b), S-[2-nitro-1-(3',4',5'-trimethoxy) phenylethyl]-L-cysteine(3c), S-[2-nitro-1-($[\vatpi}$ -3',4'-methylenedioxy)phenylethyl]-L-cysteine(3d), S-[2-nitro-1-(o-chloro)phenylethyl]-L-cysteine(3e), S-[2-nitro-1-(m-chloro)-phenylethyl]-L-cysteine(3f), S-[2-nitro-1-(p-chloro)phenylethyl]-L-cysteine(3g), S-[2-nitro-1-(o-methoxy)phenylethyl]-L-cysteine(3h), S-[2-nitro-1-(m-methoxy)phenylethyl]-L-cysteine(3i) and S-[2-nitro-1-(p-methoxy)phenylethyl]-L-cysteine(3j), respectively. The structure of adducts were confirmed by means of UV-spectrum, IR-spectrum, molecular weight measurement and elemental analysis. The various factors effecting the yield of cysteine adducts to ${\beta}$-nitrostyrene derivatives were also studied.
