In order to product the furofuranoid lignans, (+)-eudesmin which is one of the secondary products from Magnolia sieboldii. through cell suspension cultures; various culture media, initial sucrose concentration, elicitations, shaking speeds, and inoculum sizes. Among the culture media tested, MS medium had a pronounced effect on suspension cell growth and (+)-eudesmin contents. The maximum dry cell weight (DCW) of 3.71 g per flask was obtained at inoculum size of 0.5 g and in MS medium supplemented with $3\%$ sucrose plus 0.5 mg/L 2,4-D after 8 weeks. (+)-Eudesmin biosynthesis was stimulated with high initial sucrose concentration ,and the maximum (+)-eudesmin production of $3.2{\mu}g/g$ DCW was achieved at 200mg/L chitosan and $5\%$ initial medium sucrose. The optimal shaking speeds for dry biomass accumulation and (+)-eudesmin contents was 130 rpm. This work is considered to be helpful for large-scale bioprocessing of Magnolia sieboldii suspension cell cultures in bioreactor.
This study was performed to investigate the optimum microalgal culture conditions using flask culture and to find the feasibility of using the flue gas of the rice husk incinerator for cultivating the microalgae. The optimum initial pH of media was 4.5 for the microalgae culture, and the intermittently illuminated culture was more effective than the continuous illuminated culture. Thus, the balance between photosynthesis and formative metabolism must be considered thoroughly to cultivate microalgal cells. The optimum CO2 concentrations were in the range of 7 to 10%, and the optimum temperature was about 35$^{\circ}C$ in both the daytime and the nighttime for the culture. When flue gas of the rice husk incinerator was applied to the microalgae culture using stirred photobioreactor, the dry cell weight was 0.026 g dry biomass/hr$.$l. The results obtained in experiments indicated that the flue gas was effective for microalgae culture without any limitations.
$H_2$ production by Chromatium sp., a large purple sulfur bacterium blooming in lake Kaiike, under various environmental conditions was examined. Chromatium sp. produced $H_2$ only in the presence of light and $H_2$. Maximum $H_2$ production ($0.01\;{\mu}mol/hr/(mg\;dry\;cell\;weight)$) was obtained in the solution of 20 mg $H_2S-S/l$ under low light intensity (1000 lux) at $30^{\circ}C$. $H_2$ production was severely inhibited by the presence of $N_2\;or\;NH_4^+$. The rate observed for Chromatium sp. was relatively low compared to that of other phototrophic bacteria. Chromatium sp. is probably a most potent Ha producing species in lake Kaiike, since the bacterium readily produced $H_2$ photoautotrophically even at low light intensities by the application of suboptimal $H_2$ concentrations. Based on the photoautotrophic characteristics of bacterial $H_2$ production, it is suggested that Chromatium sp. can be an economic and practical species for biological $H_2$ production system, particularly in temperate region.

The algal bloom, resulting from eutrophication, has caused serious water quality problems in river and lake. Therefore, it has to be removed by any means including physicochemical or biological treatment for preserving water quality. This study was conducted to investigate the microalgae removal and energy production using combined electro-flotation and anaerobic hydrogen fermentation processes. The result showed that algae removal efficiency based on chlorophyll a removal increased with the current. At a current of 0.6A, the maximum microalgae removal efficiency of 95.9% was achieved. The treatability of anaerobic hydrogen fermentation was investigated to recover energy from microalgae removed by electro-flotation. The ultimate hydrogen yields of algae before and after ultrasonic pretreatment were 17.3 and 61.1 ml $H_2/g$ dcw(dry cell weight), respectively. The ultrasonic pretreatment of algae led to 3.4-fold higher $H_2$ production due to the increase of hydrolysis rate.
A gene(GeneBank AF 135796) coding for a trehalose synthase from Thermus caldophilus GK24 was cloned into Escherichia coli K12 using five vector systems. The constitutive expression system(pHCETS) which shows the highest trehalose synthase activity from flask culture of recombinant E. coli was selected for the production of trehalose from maltose. For the shake flask culture, the final dry cell weight was 0.9 g/L and the trehalose synthase activity was 25 U/mL. Fed-batch culture of recombinant E. coli harboring plasmid pHCETS which uses the glycerolas a carbon source was performed in jar fermentor: the dry cell weight of 20 g/L and the trehalose synthase activity of 13.7 U/mL were attained in 48 h.
An alternative method to remove and recover heavy metals is biosorption based on metal-sequestering properties of natural or biological origin. In this study, the effects of factors such as temperature, pH, initial concentration of lead, and initial amount of biomass on biosorption of lead using Aureobasdium pullulans were investigated. A. pullulans has an excellent selectivity to remove lead than other heavy metals such as cadmium, chromium, nickel in pure and mixed solution. The optimum temperature of biosorption with A. pullulans was $40^{\circ}C$ and the amount of removal increased at high pH. The higher initial lead concentration or the lower cell dry weight, the higher amount of lead was adsorbed. The adsorption isotherm of lead was accorded with Freundlich model. The adsorption capacity and initial adsorption rate of living A. pullulans were about twice higher than that of dead one.
In this work, mass production of Bacillus licheniformis SCD121067 through medium optimization by statistical experimental method was studied. First, galactose, yeast extract and potassium phosphate dibasic were selected as carbon, nitrogen and phosphate sources for mass production of B. licheniformis SCD121067 by using one factor at a time method. Second, according to the result of Plackett-Burman experimental design, key factors was yeast extract and $K_2HPO$. Finally, the response surface methodology was performed to obtain the optimum concentrations of two selected variables. The optimized medium composition consisted of 20 g/L galactose, 36 g/L yeast extract, 0.41 g/L $K_2HPO4$, 0.25 g/L $Na_2CO_3$, 0.4g/L $MgSO_4$ and 0.01g/L $CaCl_2$. Dry cell weight (15.4 g/L) by optimum production medium were increased 10 times, as compared to that determined with basic production medium (1.5 g/L). Fermentation conditions were examined for the mass production of B. licheniformis. The effect of temperature, agitation speed, pH and aeration rate on the mass production of B. licheniformis were also studied in a batch fermenter which was carried out in a 2.5 L bioreactor with a working volume of 1.5 L containing optimized production medium. As a result, dry cell weight of batch culture was 30.7 g/L at $42^{\circ}C$, 300 rpm, pH 8.0 and 2 vvm.
Experiment has been performed to investigate the thermal and magnetic properties of Mn-ferrite by electrolysis. Using the 0.2%C mild steel as soluble anode and SUS 304 stainless steel as cathode, Mn-ferrite could be made from the sulfuric acid leaching of the wasted manganese dry cell and $MnSO_4$reagent by electrolysis. As the result of X-ray diffraction, thermal analysis and magnetic measurement, Mn-ferrite was the spinel type in $Mn_{x}Fe_{3-x}O_4$ (X=1), the weight loss rate of $Mn_{x}Fe_{3-x}O_4$ were linearly increased up to the $200^{\circ}C$. Ms, Mr and Hc values were decreased with increasing Mn content and heating temperature. When Mn-ferrite was formed by $MnCl_2$reagent electrolysis, Ms values were higher than those formed from the sulfuric acid leaching of the wasted manganese dry cell and $MnSO_4$reagent by electrolysis. In Mn-ferrite, which was formed from the sulfuric acid leaching of the wasted manganese dry cell by electrolysis, Ms and Mr values were higher, Hc values were lower than which was formed by $MnSO_4$ reagent electrolysis at $200^{\circ}C\;and\;300^{\circ}C, while the same values at $100^{\circ}C$. The shape of particles was spherical type, the sizes of them were about $0.1{\mu}m$ sub-micron in $MnSO_4$reagent electrolysis, $0.5{\mu}m$ in the sulfuric acid leaching of the wasted manganese dry cell by electrolysis.

A bacterial strain C-02 using methanol as a carbon source was isolated from Gumi Industrial Estate and selected based on its rapid growth and capability of poly-$\beta$-hydroxybutyrate accumulation. Characteristics of strain C-02 showed that it belongs to the Methylococcaceae family, Type II subgroup. Strain C-02 could incorporate valerate into the PHB chain to form 3-hydroxybutyrate and 3-hydroxyvalerate (P(3HB-co-3HV)). Among various nutrient limitation tests, the nitrogen limitation test resulted in the highest content of P(3HB-co-3HV) per dry cell weight, 50$%$. Under the nitrogen limited condition, the average molecular weight of P(3HB-co-3HV) obtained was determined to be approximately $2.8\times 10^5$ daltons.

The formation of managanese ferrite has been performed to investigate some properties according to the variation of compositions, pH, current density by electrolysis. It has been found that the amount of oxidized weight of anode were increased with increasing current density. The amount of oxidized weight of anode were most in pH10. As the result of X-ray diffraction Mn Fe2O4 crystal composition in pH13. When the particles of Mnx Fe3-x O4 were heated at 30$0^{\circ}C$, it has been shown typical MnFe2O4(JCPDS Card No. 10-319) in X-1 composition. As the result of SEM observation, the size of MnFe2O4 particles were about 0.1$\mu\textrm{m}$, the shape of particles were spherical type. According to the above mentioned experimental condition, 0.1-0.5$\mu\textrm{m}$ sub-micron particles of manganese ferrite were formed from the wasted manganese dry cell, through washing longrightarrow reduction longrightarrow electrloysis.
