This study includes the transformation of genes such as ORE7, the increase of gene expression, and the use of the bar gene as a selectable marker that shows herbicide resistance with Agrobacterium tumefaciens using hypocotyls from the oilseed rape "Youngsan" cultivar. To establish an Agrobacterium transformation system for the production of oilseed rape with a high-yield trait, infection time and co-cultivation period with Agrobacterium were tested. Therefore, when hypocotyls from the oilseed rape "Youngsan" cultivar were infected with Agrobacterium for 20 min and co-cultivated for 3 days, approximately 32-36 putatively transformed hypocotyls with shoots including roots survived from 100 inoculated hypocotyls after 4 weeks of transformation on a selection medium containing 20 mg/L of phosphinothricin (PPT) as a selectable agent. Additionally, a PCR assay was performed to confirm the insertion of target genes and showed the presence of the ORE7 gene as a high-yielding trait and the bar gene as a selectable marker. Treatment with 0.5% (v/v) Basta solution as a selectable agent for 6 days with leaves from transformed oilseed rape expressed the bar gene. Therefore, this study can contribute to the development of special oilseed rapes containing agriculturally useful traits such as herbicide resistance, drought tolerance, high yielding traits, and high oleic acid content.
Korea has plentiful precipitation but rainfall events concentrate on several months of rainy season in her weather condition. Korea, therefore, experiences drought for a given period every year. Moreover the soil has usually low water holding capacity, as it is composed coarse particles originated from the granite. Response of several oaks and the Korean red pine (Pinus densiflora) on water stress showed that water budget was significant factor determining vegetation distribution. In addition, dehydration level due to cold resistance mechanism of several evergreen plants during the winter season was closely related to their distribution in natural condition. Experimental result under water stress showed that the Korean red pine was very tolerant to desiccation but the seedlings showed high mortality during the dry season. The mortality tended to proportionate to soil moisture content of each site. A comparison between soil moisture content during June when it is severe dry season and moisture content of the culture soil when the pine seedlings reached the permanent wilting point due to water withheld proved that high mortality during the dry season was due to water deficit. Water potential of sample plants measured during the exposure experiment to the air pollutant showed a probability that water related factors would dominate the occurrence of visible damage and the tolerance level of sample plants. In both field survey and laboratory experiment, plants exposed to air pollution showed more rapid transpiration than those grown in the unpolluted condition. The result would due to injury of leaf surface by air pollutants. Aluminum (Al/sup 3+/) increased in the acid soil not only inhibits root growth but also leads to abnormal distribution of root system and thereby caused water stress. The water stresses due to air pollution and soil acidification showed a possibility that they play dominating roles in inducing forest decline additionally to the existing water deficit due to weather and soil conditions in Korea. Sludge, which can contribute to improve field capacity, as it is almost composed of organic matter, showed an effect ameliorating the retarded growth of plant in the acidified soil. The effect was not less than that of dolomite known in widely as such a soil ameliorator. Litter extract contributed also to mitigate the water stress due to toxic Al/sup 3+/. We prepared a model showing the potential interaction of multiple stresses, which can cause forest decline in Korea by synthesizing those results. Furthermore, we suggested restoration plans, which can mitigate such forest decline in terms of soil amelioration and vegetation restoration.
Although the result of the tree height growth after water treatment, Fraxinus rhynchophylla has little affection by the artificial water treatment, the growth showed decreased tendency as the soil moisture decreased, and F. mandshurica showed high growth in relatively high soil moist $78{\sim}90%$ treated area. The growth of root collar diameter of F. rhynchophylla and F. mandshurica also showed decreased tendency as soil moisture decreased. The changes of biomass according to dry weight of root, stem, leaves of F. rhynchophylla demonstrating statistical significance as moisture contents of soil is lower showing decreased biomass tendency and in the treatment of $78{\sim}90%$(A) moisture content showed more than double the higher biomass compare to the treated area of $18{\sim}30%$(D) moisture contents. Also F. mandshurica showed statistical significance in A and D treatment demonstrating differences among each treatment. This can be purported to have physiological effects like weakening of seedling and softening of tissues including leaves as soil moisture decreased. Ultimately it is regarded to the main reason of unsatisfactory growth for F. rhynchophylla and F. mandshurica that are weak to drought resistance. SLA, which is one of the special traits of leaf area of F. rhynchophylla, didn't show statistical significance between moisture process, it demonstrated decreased tendency as the moisture content interval is minimal. LAR and LWR showed increased tendency while moisture content didn't show statistical significance between treatments as they are minimal.
In order to develop the multi-functional rhizobacteria that can exert positive effect on the growth of plants growing in the coastal sand dune located along East Coast of Korea, rhizospheral bacteria of 11 different plants from this area were isolated 1,330 rhizobacteria. Among these, 23 strains were able to produce auxin and had spectrum of antagonism toward various phytopathogenic microbes. To know the mechanism of this antifungal activity, these 23 strains were subjected to further analyses; 19 strains of these produced siderophore as determined by color reaction on CAS-blue plate, 4 strains produced antifungal cellulase as judged by color change on CMC-Congo red plate, 17 strains were able to utilized insoluble phosphate salts, also determined by clear zone formation on PVK medium. Identification of the strain was assigned to all 23 strains by l6s rDNA sequence analysed, and all were identified to be in the genus of Bacillus and Pseudomonas. One strain of these, denoted Pseudomonas fluorescens IB4-14, showed ACC deaminase activity which is known to be involved in the resistance of environmental stress such as salt and drought. Also, P. fluorescens IB4-l4 showed the germination stimulation and roots growth promoting activity on the in vivo assay of Lysimachia mauritiana Lam. (spoonleaf yellow loosestrife).
This study was carried out to suggest an experimental base to maintain the sustainability of extensive green roof as considering moisture demand by the type of the green roof plant before construction. The changes of soil moisture content, chlorophyll content, coverage of Sedum kamtschaticum and Dianthus chinensis var. senperflorens in two type of perlite soil and mix soil, were investigated under the condition of no rainfall and no irrigation during one month. The result shows that with the increase of stress time, soil moisture content and chlorophyll content on leaf were in a downward trend; After 30 days, Soil moisture content were lowest in 10 cm depth perlite soil system planted Dianthus chinensis. and 20 cm depth mix soil system planted Sedum kamtschaticum had the highest soil moisture. Generally soil moisture contents of soil system planted Dianthus chinensis var. senperflorens were lower than that of soil system planted Sedum kamtschaticum. In 10 cm depth soil system, state of plant growth was better than 20 cm depth soil system both perlite soil and mix soil type. It can be confirmed.

The objectives of this study were to understand the tolerance mechanism of woody plants to water stress and tolerance changes in relation to mycorrhizal formation. Lespedeza cyrtobotrya Miq. commonly used for erosion control in slopes were raised from seeds and transplanted to 120 plastic pots. Sixty pots received the top soil of a Fraxinus americana forest, while remaining 60 pots received the autoclaved top soil. The forest soil contained 1,200 spores per 100g of arbuscular endomycorrhizal fungus, mostly Glomus sp. The plants were raised outside with regular supply of water and mineral nutrients. Two kinds of water deficit treatment and a control were started at the middle of July : cyclic water deficit treatment with 3 cycles of sequential water stress at the point of xylem water potential of about -0.6, -0.6, and -1.7 MPa and recovery, and non-cyclic water deficit treatment with single water stress at about -1.5 MPa. The non-stressed plants received plenty of water throughout the period. In late August the plants were harvested for measurements of dry weight, N, P, carbohydrate contents, net photosynthesis and superoxide dismutase(SOD) activities. Both cyclic and non-cyclic water deficit treatments reduced dry weight by 60% and 40%, respectively, and reduced nitrogen absorption, while increased SOD activities. Water-stressed plants also showed increased carbohydrate contents in the leaves and lowered stomatal conductance. Mycorrhizal inoculation resulted in an average of 40% infection of roots and 2-3 times increase in P absorption in water-stressed as well as non-stressed plants. Mycorrhizal formation also increased shoot-root ratio. The results that SOD activities of water-stressed plants with mycorrhizal infection were significantly lower than those of non-mycorrhizal plants suggest the possibility of improvement of water-stressed condition by mycorrhizal formation. It was concluded that endomycorrhizal formation increased tolerance of Lespedeza cyrtobotrya seedlings to water stress.

Background: Cowpea (Vigna unguiculata [L.]) is a legume cultivated throughout most tropical countries and is valued as food and feed for human and livestock respectively. The search for an improved cowpea variety has been on-going with the aim of improving traits such as grain yield, drought and pest resistance. But no information exist on the feeding value of these improved varieties. Phosphate (P) fertilizer application is recommended to augment grain yield in grain legumes but data on the effect of P fertilizer on haulm quality is limited. Results: Two separate experiments were conducted to determine the effect of P fertilizer dose on the nutritive value of haulms from different cowpea varieties (V). In experiment 1, effect of three P doses (30, 60 and 90 kg $P_2O_5/ha$) on in vitro gas production (IVGP) characteristics, concentrations of digestible organic matter (DOM), crude protein (CP), acid detergent fiber (ADF) and neutral detergent fiber (NDF) of haulms from five cowpea varieties (Zaayura-SARC 4-75, Songotra-IT97K-499-35, Hewale-IT93K-192-4, IT99K 573-1-1 and Asomdwe-IT94K-410-2) were investigated using the $3(P){\times}5(V)$ factorial treatment arrangements in a completely randomized design. In experiment 2, the effects of two P doses (30 and 90 kg $P_2O_5/ha$) and two varieties (Zaayura-SARC 4-75 and Hewale-IT93K-192-4) on the voluntary feed intake, live weight, haematology and carcass characteristics of Djallonke lambs were also assessed using a $2(P){\times}2(V)$ factorial treatment arrangement. The $V{\times}P$ interaction significantly affected CP, NDF and ADF with CP concentration increasing with increase in P doses in Zaayura-SARC 4-75 and Asomdwe-IT94K-410-2. Whilst an increase (P < 0.05) in NDF was observed in Songotra-IT97K-499-35and Asomdwe-IT94K-410-2 as P doses increased, the other V only increased from P dose 30 to 60 kg/ha and declined at P dose 90 kg/ha. The ADF decreased (P < 0.05) with increase in P dose for all V with the exception of Songotra-IT97K-499-35. There was a significant V effect on DOM with the highest reported in Zaayura-SARC 4-75 (43.44 %). Daily DM intake, carcass length and blood urea nitrogen of the lambs were significantly affected by the V x P interaction. There was a significant V effect on globulin and P effect on live weight at slaughter, dressed weight, chuck, leg, loin, rib and flank and liver and lungs. Conclusion: It can be concluded that nutrient concentrations of cowpea haulms were positively influenced by different P dose and varieties with favorable effects on growth, haematology and carcass composition of lambs. Varieties Zaayura-SARC 4-75 and Hewale-IT93K-192-4 at P dose at 90 kg/ha are recommended to enhance growth performance and carcass yield of Djallonke lambs.

Determining plant moisture characteristics is an essential study not only for cultivation, but also for ex-situ conservation. In this study, employing pressure-volume curve we examined moisture characteristics of Rhododendron micranthum, known as rare plant, with the aim of its ex-situ conservation. Several individuals growing in Mt. Worak, Youngwol-gun Yeonha-ri and Bongwa-gun Seokpo-ri were selected for this study, from which we collected leaves. The original bulk osmotic pressure at maximum turgor(${{\Psi}_o}^{sat}$)was -1.5 MPa in those of Mt. Worak and Seokpo-ri, which is somewhat lower than that of Yeonha-ri(-1.2 MPa). It appeared that the osmotic pressure at incipient plasmolysis(${{\Psi}_o}^{tlp}$) of leaves collected in both Mt. Worak and Seokpo-ri were -1.29 MPa, and -1.26 MPa, respectively, which are lower than that of Yeonha-ri(-1.02MPa). Maximum bulk modulus of elasticity($E_{max}$) was 14.0 MPa, 8.67 MPa in leaves collected from both Seokpo-ri and Mt. Worak, respectively, those value of which were approximately 3 times higher than that of Yeonha-ri(4.00 MPa). The values of $RWC_{tlp}$(Relative water content at incipient plasmolysis) of leaves collected in three areas, were roughly 83%, suggesting that Rhododendron micranthum has relatively high capability of containing water. Our finding on moisture characteristics of Rhododendron micranthum is similar to those of other Rhododendron spp. We suggest that individuals growing in both Worak and Seokpo-ri, are preferable to those in Yeonha-ri for ex-situ transplantation since those individuals are found to have better drought resistance.
To exploit plant growth promoting bacteria in the roots of Miscanthus sacchariflorus, a biomass energy crop, total 64 bacteria were isolated. For the investigation of plant growth promoting effects from the isolated bacteria, production of indole acetic acid (IAA) and 1-aminocyclopropane-1-carboxylic acid (ACC) deaminase activities were tested and other cultural conditions were examined. As results, 8 isolates showed plant growth promoting effects on the M. sacchariflorus and an isolate designated Agrobacterium sp. BE516 has the highest activity by enhancing the shoot elongation over 2-fold than the control. Agrobacterium sp. BE516 produced 64 ${\mu}g$ IAA per mL and showed ACC deaminase activity which is involved in the resistance to environmental stress such as high salt and drought. It could grow at low temperature in the range from 4 to $15^{\circ}C$, at pH 4.0 and at 4% NaCl. These results indicate that the Agrobacterium sp. BE516 can be useful as a bio-fertilizer for M. sacchariflorus under the stressed conditions.
Superintendents have used a silicate fertilizer to improve a resistance of turfgrass against several diseases, drought damage and wear stress. This study was conducted to evaluate the effect of liquid fertilizer containing silicate (LFSi) on changes of turfgrass quality and growth by investigating visual quality, chlorophyll content-chlorophyll a, chlorophyll b, and total chlorophyll, root length, shoot length, dry weight of clipping, and nutrient content in leaves tissue. Treatments were designed as follows; control fertilizer (CF), SiF-1 (CF + $1ml\;m^{-2}$ LFSi), SiF-2 (CF + $2ml\;m^{-2}$ LFSi), and SiF-3 (CF + $4ml\;m^{-2}$ LFSi). As compared with CF, soil chemical properties, visual turfgrass quality, chlorophyll content, and dry weight of clipping of LFSi treatments were not significantly. Contrastingly, shoot density, root length, and the content of nitrogen or potassium were increased by application of LFSi. The content of Si in the tissue was positively correlated with potassium content or shoot length, and similarly shoot density positively with chlorophyll content or visual quality, respectively. These results suggested that the application of LFSi improved the turfgrass quality by increasing shoot density or K content in leaf tissue of creeping bentgrass.
