Sequence analysis of segment A gene of a very virulent infectious bursal disease virus recently isolated in Korea (최근 국내 분리 고병원성 infectious bursal disease virus의 segment A 유전자 특성)

  • Infectious bursal disease virus (IBDV) is a member of the Avibirnavirus genus of the Birnaviridae family which genome consists of two segments (A and B) of double stranded RNA. Segment A gene of KNU08010 isolate, which was isolated from a 15-day-old chicken flock in 2008, was sequenced and compared with other IBDV isolates including SH/92 strain, the first Korean very virulent (vv) IBDV isolate. The amino acid sequences of segment A gene showed that KNU08010 had 99.2% homology with SH92 strain. KNU08010 isolate had specific amino acids A222, I242, I256, I294 and S299 which are highly conserved among vvIBDV strains. Phylogenetic analysis based on the nucleotide sequences of variable region of the VP2 gene of 18 IBDV strains revealed that KNU08010 was grouped with vvIBDVs and was closely related to Korean vvIBDVs isolated from wild birds.

Postnatal Expression of Growth/Differentiation Factor-8 (GDF-8) Gene in European and Asian Pigs

  • Myostatin (growth differentiation factor (GDF)-8), is one member of the transforming growth factor $\beta$ superfamily. Investigations of GDF-8 null mice and double-muscled cattle revealed that GDF-8 has a profound influence upon skeletal muscle growth. Therefore, the GDF-8 effect upon the productive performance of pigs is worth exploring. In the present study, the nucleotide sequences and expression levels of GDF-8 genes in European pigs (Landrace and Duroc) and Asian pigs (Taoyuan and Small-ear) were evaluated. Based upon their genetic background these breeds possess significantly distinct growth rate and muscle productionphenotypes. Our sequence data showed that the nucleotide sequences of European and Asian pigs were 100% similar. Postnatal expression of GDF-8 gene in skeletal muscles, from birth to 12 mo of age, among different breeds was measured. GDF-8 expression levels in the longissimus muscle of neonatal European breed littermates were the highest, however it declined significantly (p<0.05) at 1 and 3 mo, and then increased gradually at 6 to 12 mo. The Asian breeds, however, GDF-8 expression level increased markedly at 3 mo and maintained a constant level thereafter. The results indicate that rather than polymorphism within the GDF-8 functional sequence between European and Asia breeds, it was relative to the gene regulation in postnatal muscle growth.

Genetic Diversity in the Coat Protein Genes of Prune dwarf virus Isolates from Sweet Cherry Growing in Turkey

  • Sweet cherry is an important fruit crop with increasing economical value in Turkey and the world. A number of viruses cause diseases and economical losses in sweet cherry. Prune dwarf virus (PDV), is one of the most common viruses of stone fruits including sweet cherry in the world. In this study, PDV was detected from 316 of 521 sweet cherry samples collected from 142 orchards in 10 districts of Isparta province of Turkey by double antibody sandwich-enzyme linked immunosorbent assay (DAS-ELISA). The presence of PDV in ELISA positive samples was confirmed in 37 isolates by reverse transcription- polymerase chain reaction (RT-PCR) method. A genomic region of 862 bp containing the coat protein (CP) gene of PDV was re-amplified from 21 selected isolates by RT-PCR. Amplified DNA fragments of these isolates were purified and sequenced for molecular characterization and determining genetic diversity of PDV. Sequence comparisons showed 84-99% to 81-100% sequence identity at nucleotide and amino acid level, respectively, of the CP genes of PDV isolates from Isparta and other parts of the world. Phylogenetic analyses of the CP genes of PDV isolates from different geographical origins and diverse hosts revealed that PDV isolates formed different phylogenetic groups. While isolates were not grouped solely based on their geographical origins or hosts, some association between phylogenetic groups and geographical origins or hosts were observed.

  • The formation process of a current sheet is important for solar flare from a viewpoint of a space weather prediction. We therefore derive the temporal development of the spatial and statistical distribution of electric current density distributed in a flare-producing active region to describe the formation of a current sheet. We derive time sequence distribution of electric current density by applying a nonlinear force-free approximation reconstruction to Active Region 12158 that produces an X1.6-class flare. The time sequence maps of photospheric vector magnetic field used for reconstruction are captured by a Helioseismic and Magnetic Imager (HMI) onboard Solar Dynamic Observatory (SDO) on 10th September, 2014. The spatial distribution of electric current density in NLFFF model well reproduce observed sigmoidal structure at the preflare phase, although a layer of high current density shrinks at the postflare phase. A double power-law profile of electric current density is found in statistical analysis. This may be expected to use an indicator of the occurrence of a solar flare.

Species and Hybrid Identification of Genus Coreoleuciscus Species in Hwnag-ji Stream, Nakdong River Basin in Korea (낙동강 상류 황지천에 서식하는 쉬리속(genus Coreoleuciscus) 어류 집단의 종 동정 및 잡종 판별)

  • A natural hybrid of interspecific between the Coreoleuciscus splendidus and C. aeruginos (Cypriniformes: Cyprinidae) was captured in the Hwang-ji Stream, a tributary of the Nakdong River basin in Korea. An interspecific hybrid between C. splendidus and C. aeruginos was genetically identified based on morphological characteristics and the sequence analysis of nuclear recombination activating gene 1 (RAG1) gene (1,334 bp) and mitochondrial cytochrome c oxidase subunit 1 (CO1) gene (1,551 bp). As a result of morphological variations, the natural hybrid appeared to have an intermediate character between two parental species (C. splendidus and C. aeruginos) in three variations of black array (s) on dorsal, caudal and anal fin rays. Phylogenetic analysis inferred from RAG1 and CO1 sequence data revealed that Coreoleuciscus populations from Hwang-ji stream consist of two pure Coreoleuciscus species and a hybrid individual group. The individuals were clearly identified the cross and reciprocal hybrid by CO1 gene analysis. In RAG1 gene, 13 nucleotide variation loci were detected and the hybrid individuals displayed the double peaks of sequence chromatograms at the 9 diagnostic positions. In this study, molecular data and morphological variations were clearly demonstrated that hybridization did occur between C. splendidus and C. aeruginos. However, F2 hybrid generation and reproductive capacity of F1 hybrid individuals were not demonstrated.

A GNSS Code Tracking Scheme Based in Slope Difference of Correlation Outputs (상관 함수의 기울기 차에 기반한 GNSS의 부호 추적 기법)

  • The global navigation satellite system (GNSS) is using a direct sequence/spread spectrum (DS/SS) modulation. In order to recover the information data, the DS/SS system first performs a two-step synchronization process: acquisition and tracking. The acquisition process adjusts the phase difference between the received and locally generated acquisition sequences within ${\pm}T_c/2$ or less, where $T_c$ is the chip period. The tracking process performs fine synchronization. In this paper, we focus on the tracking issue. The single delta delay locked loop($\Delta$-DLL) is the optimal tracking scheme for a GNSS in the absence of multipath signals, where $\Delta$ means the spacing between the early and late correlation time offset. In the multipath environments, however, the $\Delta$-DLL suffers from huge estimation bias(denoted by $\beta$) caused by distorted correlation values. Although some modified schemes such as a $\Delta$-DLL with a narrow $\Delta$ and a double delta DLL (${\Delta}^{(2)}$-DLL) were proposed to reduce the estimation bias, they cannot remove the estimation bias completely and need more accurate acquisition process. This paper proposes a novel tracking scheme that can dramatically reduce the estimation bias, using the maximum slope change among the correlation outputs.

  • DNA double-strand breaks (DSBs) occur commonly in the all living and in cycling cells. They constitute one of the most severe form of DNA damage, because they affect both strand of DNA. DSBs result in cell death or a genetic alterations including deletion, loss of heterozygosity, translocation, and chromosome loss. DSBs arise from endogenous sources like metabolic products and reactive oxygen, and also exogenous factors like ionizing radiation. Defective DNA DSBs can lead to toxicity and large scale sequence rearrangement that can cause cancer and promote premature aging. There are two major pathways for their repair: homologous recombination(HR) and non-homologous end-joining(NHEJ). The HR pathway is a known "error-free" repair mechanism, in which a homologous sister chromatid serves as a template. NHEJ, on the other hand, is a "error-prone" pathway, in which the two termini of the broken DNA molecule are used to form compatible ends that are directly ligated. This review aims to provide a fundamental understanding of how HR and NHEJ pathways operate, cause genome instability, and what kind of genes during the pathways are associated with head and neck cancer.

Simultaneous and Systemic Knock-down of Big Defensin 1 and 2 gene Expression in the Pacific Oyster Crassostrea gigas using Long Double-stranded RNA-mediated RNA Interference

  • RNA interference (RNAi)-mediated transcriptional knock-down of Crassostrea gigas big defensin 1 and 2 genes (Cg-BigDef1 and Cg-BigDef2) was investigated. The cDNA sequences of Cg-BigDef1 and Cg-BigDef2 were identical, excluding an additional fragment of 20 nucleotides in Cg-BigDef1; thus, a long double-stranded RNA (dsRNA) targeting the mRNA of Cg-BigDef2 effectively downregulated both Cg-BigDef2 and Cg-BigDef1. In addition, long dsRNA targeting green fluorescent protein (GFP) did not affect transcription of the two big defensin genes. These results suggest that the transcriptional downregulation of Cg-BigDef1 and Cg-BigDef2 was mediated by sequence-specific RNA interference (RNAi). Despite injection of long dsRNA targeting Cg-BigDef2 into only the adductor muscle, knock-down of Cg-BigDef1 and Cg-BigDef2 was observed in the adductor muscle, hemocytes, mantle, and gills, suggestive of systemic spread of RNAi in C. gigas. Furthermore, the inhibitory effect of dsRNA persisted until 72 h post-injection, indicative of a long-lasting RNAi-mediated knock-down of target genes.

Correlation Analysis of Flow Characteristics Downstream of a Double Bent Pipe and Mounting Positions of Ultrasonic Flowmeter (곡관하류의 유동특성과 초음파유량계 설치위치의 상관관계 분석)

  • In this study, the establishment of the criteria for accurate measurement is investigated via a statistical analysis of experimental results. The magnitude of influence on measurement errors is severely affected by the number of paths, mounting angle of sensor, straight pipe length in sequence, and Reynolds number. Three-dimensional numerical analysis has been conducted to understand the flow patterns downstream of a double bent pipe. Numerical analysis shows that the results well agreed with the experimental ones in case of a sensor mounting angle of $0^{\circ}$ and L/D = 3, 5 of $45^{\circ}$, $135^{\circ}$ in a single path. As a result, when the Reynolds number is 700,000-1,400,000, the sensor error of a single-path ultrasonic flowmeter is reduced with the mounting condition of L/D = 3, $45^{\circ}$.

In the film, the dress style is of extreme importance for the personality creation of the character as well as the development of the plot. With this background, I have looked over the Femme Fatale dress style that appears in the movie . First, it expresses Femme Fatale which stresses the feminine trait by a dress style with enlarged chest and hip parts and long, slim silhouette with tight waist and knee line. Second, the Femme Fatale with pretended purity presents narcissistic satisfaction like a saint virgin by wearing a neck-high one-piece dress embellished with frills, but the use of mirror represents self-alienation and the vanity of purity. Third, the Femme Fatale with its military dress style presents authoritative sternness which leads seduced male into ruin with its destructive power. Fourth, wearing jersey type or knitted wear that sticks to the body presents sensual Femme Fatale. Fifth, the tulle, chiffon, and white handkerchief used for the creation of grotesque Femme Fatale represents harmlessness and prohibition of immoral behavior, and the black gloves, pillbox, and the number on the tulle present her uneasy psychological state from the fear of being discovered after murdering her husband. On the other hand, the see-through tulle presents alluring sensual beauty. Sixth, the combination of the dialogue in the final sequence and drapery type as in Greek goddess represents the saint virgin of salvation who acknowledges her own fault and returns to her purity again. In addition, accessories and others made from shining material represent hidden conspiracy through simple dress and contradictory dynamics, and shoes embellished with bonbon, etc represent sexual implications in connection with male.
