MICROTENSILE BOND STRENGTH OF SELF-ETCHING AND SELF-ADHESIVE RESIN CEMENTS TO DENTIN AND INDIRECT COMPOSITE RESIN (간접 복합레진 합착 시 자가부식형과 자가접착형 레진시멘트의 상아질에 대한 미세인장 결합강도)

  • The purpose of this study was to evaluate the microtensile bond strength (${\mu}TBS$), failure modes and bonding interfaces of self-etching and three self-adhesive resin cements to dentin and indirect composite resin. Cylindrical composite blocks (Tescera, Bisco Inc.) were luted with resin cements (PA: Panavia F 2.0, Kuraray Medical Inc., RE: RelyX Unicem Clicker, 3M ESPE., MA: Maxem, Kerr Co., BI: BisCem, Bisco Inc.) on the prepared occlusal dentin surfaces of 20 extracted molars. After storage in distilled water for 24 h, $1.0\;mm\;{\times}\;1.0\;mm$ composite-dentin beams were prepared. ${\mu}TBS$ was tested at a cross-head speed of 0.5 mm/min. Data were analyzed with one-way ANOVA and Tukey's HSD test. Dentin sides of all fractured specimens and interfaces of resin cements-dentin or resin cements-composite were examined at FESEM (Field Emission-Scanning Electron Microscope). In conclusion, PA and RE showed higher bond strength and closer adaptation than MA and BI when indirect composite blocks were luted to dentin using a self-etching and three self-adhesive resin cements.

EFFECT OF AN INTERMEDIATE BONDING RESIN AND FLOWABLE RESIN ON THE COMPATIBILITY OF TWO-STEP TOTAL ETCHING ADHESIVES WITH A SELF-CURING COMPOSITE RESIN (자가 중합 복합 레진과 두 단계 산 부식 접착제의 친화성에 대한 중간 접착제와 흐름성 레진의 효과)

  • This study compared the effect of an activator, intermediate bonding resin and low-viscosity flowable resin on the microtensile bond strength of a self-curing composite resin used with two-step total etching adhesives. Twenty extracted permanent molars were used. The teeth were assigned randomly to nine groups (n=10) according to the adhesive system and application of additional methods (activator, intermediate adhesive, flowable resin). The bonding agents and additional applications of each group were applied to the dentin surfaces. Self-curing composite resin buildups were made for each tooth to form a core, 5mm in height. The restored teeth were then stored in distilled water at room temperature for 24h before sectioning. The microtensile bond strength of all specimens was examined. The data was analyzed statistically by one-way ANOVA and a Scheffe's test. The application of an intermediate bonding resin (Optibond FL adhesive) and low-viscosity flowable resin (Tetric N-flow) produced higher bond strength than that with the activator in all groups. Regardless of the method selected, Optibond solo plus produced the lowest ${\mu}TBS$ to dentin. The failure modes of the tested dentin bonding agents were mostly adhesive failure but there were some cases showed cohesive failure in the resin.

EFFECT OF A DESENSITIZER ON DENTINAL BOND STRENGTH IN CEMENTATION OF COMPOSITE RESIN INLAY (레진 인레이 합착시 지각과민처리제의 사용이 상아질 결합강도에 미치는 영향)

  • The purpose of this study was to evaluate the effect of a desensitizer on dentinal bond strength in cementation of composite resin inlay. Fifty four molar teeth were exposed the occlusal dentin. Class I inlay cavities were prepared and randomly divided into six groups. Control group: no agent, Group 1 : Isodan, Group 2 : One-step, Group 3 : All-Bond SE, Group 4 : Isodan + One-step, Group 5 : Isodan + All-Bond SE. Desensitizing agent and dentin bonding agents were applied immediately after the completion of the preparations. Impressions were then made. The composite resin inlays (Tescera, Bisco) were fabricated according to the manufacturers' guidelines. Cementation procedures followed a standard protocol by using resin cement (Bis-Cem, Bisco). Specimens were stored in distilled water at $37^{\circ}C$ for 24 hours. All specimens were sectioned to obtained sticks with $1.0{\times}1.0\;mm^2$ cross sectional area. The microtensile bond strength (${\mu}TBS$) was tested at crosshead speed of 1 mm/min. The data was analyzed using one way ANOVA and Tukey's test. Scanning electron microscopy analysis was made to examine the details of the bonding interface, 1. Group 1 showed significantly lower ${\mu}TBS$ than other groups (p<0.05). 2. There was no significant difference between the ${\mu}TBS$ of Group 3 and Group 5. 3. The ${\mu}TBS$ of Group 4 showed significantly lower than that of Group 2 (p<0.05). In conclusion, a desensitizer (Isodan) might have an adverse effect on the bond strength of composite resin inlay to dentin.

Studies on the Screening and Development of Antibiotics in the Mushroom -The Screening of Antifungal Components in Basidiomycetes (I)- (버섯중 항균물질의 검색 및 개발에 관한 연구 -버섯중 항진균활성 물질의 검색 (I)-)

  • For the searching of antibiotics in the mushroom, we studied on the screening of antiyeast and antifungal components. The powder of fruiting body of each mushroom was extracted with petroleum ether, 80% ethanol, and distilled water in that order, respectively. The antifungal activity of each extract from 32 different mushrooms were tested. The petroleum ether or ethanol extracts from fruiting bodies of Boletus auripes, Leccinum extremiorientale, Gomphus floccosus, Phaeolus schweinitzii, Pycnoporus cinnabarinus and Marasmius maximus showed antiyeast activities. The ethanol extract of Gyrophora esculenta showed an antiyeast activity against Cryptococcus neoformans, and its minimal inhibitory concentration (MIC) was $600\;{\mu}g/ml$. The petroleum ether or ethanol extracts from fruiting bodies of Amanita citrina, Leccinum extremiorientale, Gomphus floccosus, Phaeolus schweinitzii, Pycnoporus cinnabarinus, Marasmius maximus and Gyrophora esculenta showed antifungal activities. The ethanol extract of Gomphus floccoscus showed an antifungal activity against Microsporum gypseum, and its MIC was $1,000\;{\mu}g/ml$. The ethanol extract of Gyrophora esculenta showed antifungal activities against Trichophyton mentagrophytes, Microsporum gypseum and Pyricularia oryzae, and theirs MIC were $300\;{\mu}g/ml,\;600\;{\mu}g/ml\;and\;600\;{\mu}g/ml$, respectively. The petroleum ether extract of Gyrophrora esculenta also showed an antifungal activity against Microsporum gypseum, and its MIC was $300\;{\mu}g/ml$.

Effect of Deep Seawater on Expression of μ-Opioid Receptor in Cultured Rat Hippocampal Neurons (배양된 쥐 해마신경세포에서 μ-아편양 수용체의 발현에 대한 해양심층수의 영향)

  • Deep seawater (DSW) generally refers to seawater at depths equal to or greater than 200 meters. DSW is rich in inorganic materials which have attracted attention for its various applications. In this study we investigated the effects of the DSW upwelled from the East Sea, offshore Yang Yang (KangWon-do, Korea), on the expression of ${\mu}$-opioid receptor (MOR) of cultured rat hippocampal neurons. Neurons were grown in a minimal essential medium containing 10% (v/v) fetal bovine serum and either 25% (v/v) distilled water, or hardness (H) 800, or H 1000 DSW. Cultures grown in the presence of DSW with H 800 and H 1000 exhibited robust MOR immunoreactive signals in both neurons and astrocytes. Interestingly, the increase in MOR immunoreactive signals was more dramatic in astrocytes than in neurons. Statistical analysis revealed that the relative intensities for MOR clusters increased approximately 4-fold in astrocytes cultured in H 800 and H 1000 media. These increases were statistically very significant (p<0.001). In contrast, the increase in intensities for MOR immunoreactive signals was relatively less dramatic in neurons, where only the increase in the H 1000 culture was statistically very significant (p<0.001). These results indicated that DSW promotes expression of MOR in both neurons and astrocytes, and more significantly in the latter.

Effects of Mole Crickets (Gryllotalpa orientalis) Extracts on Anti-oxidant and Anti-inflammatory Activities. (땅강아지(Gryllotalpa orientalis) 추출물의 항산화 및 항염증 활성)

  • Tremendous natural product extracts were used as a herb medicine remedy or therapy for centuries. Because these extracts have various biological activities, we examined the effects of Gryllotalpa orientalis extract for anti-oxidant and anti-inflammation activity by 1,1-diphenyl-2-picrylhydrazyl (DPPH), ferric reducing ability of plasma (FRAP), hydroxy radical scarvenging and cyclooxygenase-2 promoter assays. Gryllotalpa orientalis extracts were prepared from solvents such as distilled water (DW), dimethyl sulfoxide (DMSO), ethanol and methanol. The results showed that Gryllotalpa orientalis extracts have potent DPPH (methanol extract), FRAP (DW extract) and hydroxy radical scavenging (DW and methanol extracts) activity than any other extracts used. A significant inhibition of cyclooxygenase-2 (Cox-2) promoter activity was detected in the presence of DMSO extract or ethanol extract. Collectively, the present results suggested that Gryllotalpa orientalis extract could be used for anti-oxidant and/or anti-inflammation agent for human or agricultural purposes.

Esterification of Lactic Acid with Alcohols (젓산과 알코올간의 에스테르화 반응)

  • Esterification of lactic acid with alcohols catalyzed by Amberlyst-type ion exchange resins and sulfuric acid was carried out in a batch reactor with total /or partial recycle of distilled condensates, respectively. The esterification of lactic acid in the total-recycling reactor (n-butanol/lactic acid = 4, $100^{\circ}C$) was promoted by decreasing the residual water and increasing the mole ratio of n-butanol/lactic acid. Also, it was confirmed that methanol with simple structure and tert-butanol with superior substitution reactivity were more effective in increasing the conversion of esterification reaction, compared to ethanol, n-butanol, and iso-butanol. In a partial-recycling reactor (n-butanol/ammonium lactate = 4, $115^{\circ}C$), the conversion of ammonium lactate into butyl lactate with 1.0 wt% Amberyst-type resins was higher in comparison to that with 0.2 mol $H_2SO_4$ (per 1.0 mol ammonium lactate). The esterification was gradually occurred during the initial stage of reaction in the presence of solid catalyst, whereas the initial addition of $H_2SO_4$ did not affect the initial rate of esterification reaction because of ammonium sulfate formation by the neutralizing reaction of ammonium lactate with sulfuric acid.

  • Over 90 of lysine producing auxotrophs were obtained from Corynebacterium sp. S-27-12, Brevibacterium flavum ATCC 15168 and Micrococcus glutamicus ATCC 13032 by UV light, $Co^{60}$ irradiation and N-methyl-N'-nitro-N-nitrosoguanidine treatment. One of the mutant, Brev. flavum U46-N59, was identified as a leucine auxotroph and accumulated lysine during flask (500 ml) cultivation (180 strokes/min.) up to 21.6 mg per ml of broth at pH 7.5 and $28^{\circ}C$ after 4 days. The medium consisted of glucose, 100; urea, 10; corn steep liquor, 40; $KH_2PO_4,\;2;\;K_2HPO_4,\;0.5;\; MgSO_4.\;7H_2O,\;0.4;\;antifoam\;S-57,\;1g;\;Fe_2(SO_4)_3.XH-2O,\;10;\; MnCl_2,\;4H_2O,\;10mg;\;biotin,\;30;\;thiamine-HCl,\;100{\mu}g$in 1l of distilled water, and 40 U/ml of penicillin was added after 36 hrs fermentation.

Effect of varying ethanol concentrations on the extraction properties and physiological activity of Artemisia annua L. (에탄올 농도 조건이 개똥쑥 추출 및 생리활성에 미치는 영향)

  • The present study was undertaken to compare the effects exerted by different extraction solvents on the extraction of active components, such as polyphenols and flavonoids, from the dried leaves of Artemisia annua L. Different extracts were prepared using a heating mantle. The extraction solvents used were distilled water, and 20, 40, 60, 80, and 99.5% ethanol solution. It was observed that the 40% ethanol solution yielded the most significant results in the extraction of various phytochemicals with phenol concentration of 154.8±0.28 mg of gallic acid equivalent/g and flavonoid content of 25.28±0.01 mg quercetin equivalent/g. However, based on the extraction solvent used, varying trends were observed in the antioxidant, enzyme inhibition, and bacterial inhibition analyses. It was concluded that the extraction solvent should be selected based on the purpose of use of the dried leaves of A. annua L.

Effect of Cultivation Method, Harvest Season and Preservative Solution on the Quality and Vase Life of Cut Rose 'Rote Rose' (롯데로제 장미의 재배방법, 수확시기 및 보존제 종류가 절화 품질과 수명에 미치는 영향)

  • Experiments were conducted to evaluate quality and vase life of cut rose 'Rote Rose' cultivated in soil or hydroponically in rockwool. Rose flower stems harvested in commercial greenhouses in Kimhae on May 27 and June 14, 1998 were transported for about two hours to a laboratory and recut in water to an uniform stem length of 45cm. The rose flowers harvested on the same day were displayed at a density of $10cm{\times}10cm$ and were subjected to the same environmental conditions in a growth chamber. The stems were put in four different preservative solutions, 0.5% Chrysal RVB, BS (2% sucrose+200ppm 8HQS+0.3% Chrysal RVB), Sonk1 (BS+0.1mM ethionine), and double distilled $H_2O$. Flower stems harvested on May 27 were displayed at $18{\pm}1^{\circ}C$, RH 60-70%, and light intensity of 220lux provided by fluorescent lamps for $16h{\cdot}d^{-1}$. Flower stems harvested on June 14 were displayed at $25{\pm}1^{\circ}C$, RH 70-80%, and light intensity of 220lux provided by fluorescent lamps for $16h{\cdot}d^{-1}$. Fresh weight and flower diameter were affected by cultivation method, and were greater in hydroponically-grown roses than in soil-grown roses. Among the preservative solutions, BS and Sonk1 were superior to Chrysal RVB in terms of prolonging vase life. Vase life extension in Chrysal RVB, BS and Sonk1 over the control was about one day in both display temperatures. At $18^{\circ}C$ vase life was maintained for three to four additional days as compared to that at $25^{\circ}C$.
