Control of Seed-borne Infection of Ustilago nuda and Pyrenophora graminea on Barley (맥류(麥類)에 발생(發生)하는 Ustilago nuda 및 Pyrenophora graminea에 대한 종자(種子) 소독(消毒)에 관한 연구(硏究))

  • Effects of seed disinfectants on loose smut (Ustilago nuda) and leaf stripe (Pyrenophora graminea) of barley were investigated in field tests. For these experiments, seed samples carrying natural infection of Ustilago nuda and Pyrenophora graminea were used and the following fungicides were used: Baytan, Baytan U, Benlate T, Busan 30, KAC-7703, P 242, Panoctine, Sisthane Ec, Sisthane Wp, Sisthane Ds, Sodium Omadine, Terracoat Zn, Vitathiram and Zinc Omadine, respectively. Results have shown that Sisthane and Benlate T have equal effect to Vitathiram against Ustil­ago nuda and Pyrenophora graminea. Baytan U was effective against loose smut but inferior to leaf stripe of barley. P 242 was effective against leaf stripe but inferior to loose smut of barley. Busan 30 have shown moderately inferior effect to Vitathiram against loose smut and leaf stripe of barley. A mild inhibition of seed germination and seedling growth of barley, naked barley and wheat seeds was observed when high concentration of Sisthane and Baytan U were treated in seedling box placed in green house conditions, but no symptom observed in field conditions.

Application of Multiparametric Flow Cytometry (FCM) to Enumerate the Diagnosis of Pseudomonas aeruginosa and Escherichia coli

  • In this study, multiparametric flow cytometry (FCM) was installed to enumerate the diagnosis of Pseudomonas aeruginosa ATCC 10145 and Escherichia coli K12 (IFO 3301). The nucleic acids (DNA/RNA) were double stained by a LIVE/DEAD bacLight viability kit, involving green SYTO 9 and red propidium iodide (PI), based on the permeability of two chemicals according to the integrity of plasma membrane. As the results showed, the gate for dead bacteria was defined as the range of $0.2{\times}10^0$ to $6.0{\times}10^1$ photo multiplier tube (PMT) 2 fluorescence (X-axis) and $2.0{\times}10^0$ to $2.0{\times}10^2$ PMT 4 fluorescence (Y-axis), and the gate for live bacteria was defined as the range of $6.0{\times}10^0$ to $6.0{\times}10^2$ PMT 2 fluorescence (X-axis) and $2.0{\times}10^0$ to $4.0{\times}10^2$ PMT 4 fluorescence (Y-axis). In the comparison of the number of the tested bacteria detected by FCM (viability assessment) and plate culture (cultivability assessment), the number of bacteria detected by FCM well represented the number of bacteria that was detected by the colony forming unit (CFU) counting method when bacteria were exposed to isopropyl alcohol and silver/copper cations. Consequently, it is concluded that the application of FCM to monitor the functional effect of disinfectants on the physiological status of target bacteria can offer more rapid and reliable data than the plate culture colony counting method.

Rice Seed Infection of Fusarium spp. at Paddy Field and Selection of Seed Disinfectants (포장(圃場)에서 Fusarium속균(屬菌)의 종자(種子) 감염(感染)과 소독제(消毒劑)의 선발(選拔))

  • F. roseum "Graminearum" and F. moniliforme were isolated from hulls of seeds collected from fields. F. roseum "Graminearum" was isolated from brown rice of all the varieties tested. F. moniliforme was isolated from Nagdongbyeo, Seomjinbyeo, Sinsunchalbyeo, Milyang 23, Baegyangbyeo and Samnambyeo. F. moniliforme was isolated from the most of the seeds damaged by sparrow and it formed sporodochia of this fungus. Five species including F. moniliforme were isolated from plant hopper collected from paddy fields in September. To select seed a disinfectant, twelve fungicides were tested and P242 was one of the best effective fungicidetested. After seed treatment with the fungicide, F. moniliforme was not isolated from hulls but was frequently isolated from brown rices.

Activation of Multiple Transcriptional Regulators by Growth Restriction in Pseudomonas aeruginosa

  • Growth restriction by antibiotics is a common feature that pathogenic bacteria must overcome for survival. The struggle of bacteria to escape from growth restriction eventually results in development of antibiotic-resistance through the expression of a set of genes. Here we found that some physiologically important transcriptional regulators of Pseudomonas aeruginosa including QscR, a quorum sensing (QS) receptor, SoxR, a superoxide sensor-regulator, and AntR, a regulator of anthranilate-related secondary metabolism, are activated by various growth-restricted conditions. We generated the growth-restricted conditions by various methods, such as overexpression of PA2537 and treatment with antibiotics or disinfectants. The overexpression of PA2537, encoding an acyltransferase homologue, tightly restricted the growth of P. aeruginosa and significantly activated QscR during the growth restriction. Similarly, treatments with gentamycin, tetracycline, and ethanol also activated QscR near their minimal inhibitory concentrations (MICs). Some non-QS regulators, such as AntR and SoxR, were also activated near the MICs in the same conditions. However, LasR and PqsR, other QS receptors of P. aeruginosa, were not activated, suggesting that only a specific set of transcriptional regulators is activated by growth restriction. Since paraquat, a superoxide generator, significantly activated QscR and AntR, we suggest that the oxidative stress generated by growth restriction may be partly involved in this phenomenon.

Evaluation of Efficiency of Livestock Vehicle Disinfection Systems Using Water-Sensitive Paper (감수 시험지를 활용한 축산시설 차량소독시스템의 소독액 분사 효율성 평가)

  • The livestock infections had been happened seasonally, but they have gradually changed to be irrelevant to seasons and have an aspect to rapidly spread after outbreak. Especially in Korea, proactive disinfection measures are very important because the livestock farms are located densely so high as to accelerate the spread of disease between farms. livestock disease outbreaks like HPAI and FMD occurred with high probability due to vehicles visiting the farms, this study is to evaluate the efficiency of livestock vehicle disinfection systems by investigating the disinfectant coverage according to the type of vehicle disinfection system and the type of vehicle quantitatively. In field experiments, water-sensitive papers (WSPs) were attached to 21 locations on the surface of four vehicles (sedan, SUV, truck, and feed transport), respectively, and exposed to disinfectants while the vehicle was sprayed in two vehicle disinfection systems (tunnel type and simplified type). The WSPs were scanned and image-processed to calculate the disinfectant coverage. The results showed that the tunnel-type vehicle disinfection system had a better disinfection performance with an average coverage of 90.27% for all vehicles compared to 32.62% of the simplified type system. The problem of the simplified system was a wide coefficient of variation (1.05-1.31) of the disinfectant coverage between 21 locations indicating a need for further improvement of nozzle location and arrangement.

Administrators' Experience of Using Service Provided by Center for Children's Foodservice Management among Home-based Child Care Centers in Seoul (서울지역 가정어린이집 원장의 어린이급식관리지원센터 이용 경험 분석)

  • This study examined the experience of administrators of home-based child care centers about CCFSM (Center for Children's Foodservice Management) service using a qualitative approach. Eight administrators of home-based child care centers in Seoul with more than one year's experience of CCFSM were interviewed regarding their opinions on support activities, standards, and foodservice management. The interviews were semi-structured with open-ended questions and they were transcribed and classified according to the subjects. There were negative opinions regarding the indications after a hygiene inspection leaving photographs or records behind, but most positive opinions were about round visits for foodservice sanitation and nutrition management. The participants thought that it is not enough that the number of nutrition education sessions for children be twice a year, and there was a suggestion to increase the rentals of nutrition educational materials. There was a complaint about the menus in that were difficult to prepare for lunch time or to obtain food ingredients for. The administrators poorly recognized how to use the standardized recipes and chlorine-based disinfectants, so they could not be applied properly. They also pointed out the problems of joint purchasing as low quality and high prices in the food service operation. They felt discriminated against compared to with larger scale child care centers because of their size and expressed concerns regarding the fact that many home-based child care centers were not included due to the lack of publicity and budget. Through the results, the CCFSM should provide a differentiated service and management by creating a dedicated team or staff for home-based child care centers so they will not feel left out. In addition, It is also important to constantly gather opinions to improve the menus and to use standardized recipes practically. In addition, it will be necessary to develop nutrition educational materials corresponding to the infants' age for home-based child care centers and increase the rent to expand nutrition education.

Chemical Control of Rice White-tip Nematode (Aphelenchoides besseyi C.) by Seed-disinfectant and in the Paddy Field (벼잎선충(線蟲)에 대(對)한 종자소독(種子消毒) 및 본답(本畓) 엽제방제(葉劑防除)에 관(關)한 연구(硏究))

  • Experiments were caried out to invesstigate the effects of seed disinfectant and the chemical control method in the paddy field for the rice white-tip nematode, Aphelenchoides besseyi. All three seed disinfectants tested, MEP 50% EC, Fenthion 50% EC, Dasuzin 34% EC showed effective control without any symptom of phytotoxicity. Mixed treatments with fungicides, Benoram 40% Wp, proraz-25% EC, TCM 30% EC also showed the effective control and no phytotoxicity. Effective chemical control methods for the rice white-tip nematode in paddy fields was seed disinfectant before seeding and Carbofuran 3% G. on the day before the transplanting, or seed disinfectatnt + Carbofuran 3% G. water surface treatment on the early stage of injury.

  • Conformity to legal permission standard of $5\;log_{10}$ CFU/mL reduction of foodborne pathogens or spoilage bacteria such as Escherichia coli, Staphylococcus aureus, Salmonella typhimurium, Pseudomonas aruginosa, and Enterococcus hirae was examined in 11 domestic commercial sanitizers and disinfectant. One chlorine compound, two iodophor compounds, two peroxide compounds, and three quaternary ammonium compounds (QACs) met advised standard concentration (100%), showing $7\;log_{10}$ CFU/mL reduction, and met legal standard by $5-6\;log_{10}$ CFU/mL reduction at 75% of advised standard concentration. At 10% dilution, one chlorine compound, one iodophor compound, two peroxide compounds, and two QACs satisfied legal standard.

Reflection on Kinetic Models to the Chlorine Disinfection for Drinking Water Production

  • Experiments for the characterization of inactivation were performed in a series of batch processes with the total coliform used as a general indicator organism based on the chlorine residuals as a disinfectant. The water samples were taken from the outlet of a settling basin in a conventional surface water treat- ment system that is provided with the raw water drawn from the mid-stream of the Han River, The inactivation of total coliform was experimentally analysed for the dose of disinfectants contact time, filtration and mixing intensity. The curves obtained from a series of batch processes were shaped with a general tailing-off and biphasic mode of inactivation, i.e. a sharp loss of bacterial viability within 15 min followed by an extended phase. In order to observe the effect of carry-over suspended solids on chlorine consumption and disinfection efficiency, the water samples were filtered, prior to inoculation with coliforms, with membranes of both 2.5$\mu$m and 11.0 $\mu$m pore size, and with a sand tilter of 1.0 mm in effective size and of 1.4 in uniformity coefficient. As far as the disinfection efficiency is concerned, there were no significant differences. The parameters estimated by the models of Chick-Wat-son, Hom and Selleck from our experimental data obtained within 120 min are: log(N/N$\_$0/)=-0.16CT with n=1, leg(N/N$\_$0/)=-0.71C$\^$0.87/ with n 1 for the Chick-Watson model, log (N/N$\_$0/)=-1.87C$\^$0.47/ T$\^$0.36/ for the Hom model, log (MHo)=-2.13log (1+CT/0.11) for the Selleck model. It is notable that among the models reviewed with regard to the experimental data obtained, the Selleck model appeared to most closely resemble the total coliform survival curve.

Reduction of Bacillus cereus in Cooked Rice Treated with Sanitizers and Disinfectants

  • This study aimed to identify effective washing and sanitation programs to minimize the contamination of cooked rice by B. cereus. As such, the effectiveness of five sanitizers, including QAC, alcohol, chlorine, CaO, and $H_2O_2$, was evaluated in relation to the survivability of B. cereus spores in cooked rice and resulting sensory properties of the rice. The water-treated cooked rice showed remaining B. cereus spores at 1.09 $log_{10}CFU/g$. In contrast, treatment with the minimum inhibitory concentrations of the sanitizers, such as 200 ppm of QAC, 50% of alcohol, 100 ppm of chlorine, 650 ppm of CaO, and 500 ppm of $H_2O_2$, destroyed all the spores in the cooked rice below a non-detection limit (ND< 0.15 CFU/g). The sensory properties of the sanitizer-treated (1,000 ppm of $H_2O_2$, 100 ppm of chlorine, and 800 ppm of CaO) cooked rice did not differ significantly from those of the water-treated cooked rice. As a result, 500 ppm of $H_2O_2$, 650 ppm of CaO, and 100 ppm of chlorine were found to effectively eliminate B. cereus spores in rice while cooking.
