The Single-Cell Revelation of Thermogenic Adipose Tissue

  • The past two decades have witnessed an upsurge in the appreciation of adipose tissue (AT) as an immunometabolic hub harbouring heterogeneous cell populations that collectively fine-tune systemic metabolic homeostasis. Technological advancements, especially single-cell transcriptomics, have offered an unprecedented opportunity for dissecting the sophisticated cellular networks and compositional dynamics underpinning AT remodelling. The "re-discovery" of functional brown adipose tissue dissipating heat energy in human adults has aroused tremendous interest in exploiting the mechanisms underpinning the engagement of AT thermogenesis for combating human obesity. In this review, we aim to summarise and evaluate the use of single-cell transcriptomics that contribute to a better appreciation of the cellular plasticity and intercellular crosstalk in thermogenic AT.

Nature Activities in Urban Parks to Encourage Curiosity and Scientific Problem-Solving Ability in Kindergarteners

  • This study was conducted based on the fact that children in institutions for early childhood education located in cities lack the opportunity to experience nature. Therefore, urban parks are where it is possible to observe nature and natural environment, through which we examined the effects of nature activities on kindergarten children's curiosity and scientific problem-solving ability. The subjects of this study were 5-years old kindergarten children in attending public kindergartens in Cheonan and Asan and 42 children were randomly selected. The pretest and posttest were conducted on curiosity and scientific problem-solving ability before and after nature activities. The results showed that nature activities in urban parks had significant effects on improvement of kindergarten children's curiosity and scientific problem-solving ability (p <.05). Therefore, nature activities in urban parks had positive effects on preference for unknown and exploratory behavior, which are sub-factors of kindergarten children's curiosity. Nature activities also had positive effects on discovery and statement of the problem, creation and application of ideas, and conclusion to problem solving.

미적분 문제해결 과정에서 수학적 사고력 향상을 위한 몰입적 사고의 적용 (Application of Long-term Slow Thinking(Flow) to Improve Mathematical Thinking Ability in the Process of Solving a Basic Calculus Problem)

  • 수학적 사고력이 학생 스스로 문제를 해결하는 과정에서 가장 발달한다는 주장과 함께 이를 구현하는 방법론에 대한 연구도 꾸준히 이루어져왔다. 최근에 그 방법론으로 몰입적인 사고를 통한 학생의 학습 방법이 제안되었다. 이에 본 연구에서는 몰입적인 사고를 적용하여 학생들이 스스로 수학문제를 해결해 나갈 수 있는지를 알아보았다. 연구는 고등학교 교과과정인 미적분에 대한 선행학습을 하지 않은 중학교 3학년 학생들 10명을 대상으로 몰입적 사고를 통해서 학생 스스로 미적분 문제를 해결할 수 있는지와 그 과정에서 학생이 경험하는 수학학습 성취에 대한 탐구로 진행되었다. 학생들은 주어진 미적분 문제를 3일 동안 몰입적 사고를 적용하여 풀었다. 그 결과 2명이 스스로 해결하였고 7명이 힌트를 받고 해결하였다. 연구 결과 상당수의 학생이 장시간의 몰입적인 사고를 통하여 배우지 않은 문제들을 스스로의 능력으로 해결할 수 있음을 알게 되었다. 이 과정에서 학생들의 수학적 사고력이 발달되었고 학생들은 수학하는 즐거움과 성취감을 경험했을 것으로 기대되었다. 본 연구 결과는 몰입적 사고를 도입함으로써 교실에서 학생들 스스로 문제를 푸는 교수법의 개발에 하나의 가능성을 제시하였다고 볼 수 있으며 몰입을 통한 훈련으로 수학적 사고력 발달을 통한 실제 문제해결력에도 기여할 수 있음을 시사하고 있다.

Caenorhabditis elegans: A Model System for Anti-Cancer Drug Discovery and Therapeutic Target Identification

  • The nematode Caenorhabditis elegans (C. elegans) offers a unique opportunity for biological and basic medical researches due to its genetic tractability and well-defined developmental lineage. It also provides an exceptional model for genetic, molecular, and cellular analysis of human disease-related genes. Recently, C. elegans has been used as an ideal model for the identification and functional analysis of drugs (or small-molecules) in vivo. In this review, we describe conserved oncogenic signaling pathways (Wnt, Notch, and Ras) and their potential roles in the development of cancer stem cells. During C. elegans germline development, these signaling pathways regulate multiple cellular processes such as germline stem cell niche specification, germline stem cell maintenance, and germ cell fate specification. Therefore, the aberrant regulations of these signaling pathways can cause either loss of germline stem cells or overproliferation of a specific cell type, resulting in sterility. This sterility phenotype allows us to identify drugs that can modulate the oncogenic signaling pathways directly or indirectly through a high-throughput screening. Current in vivo or in vitro screening methods are largely focused on the specific core signaling components. However, this phenotype-based screening will identify drugs that possibly target upstream or downstream of core signaling pathways as well as exclude toxic effects. Although phenotype-based drug screening is ideal, the identification of drug targets is a major challenge. We here introduce a new technique, called Drug Affinity Responsive Target Stability (DARTS). This innovative method is able to identify the target of the identified drug. Importantly, signaling pathways and their regulators in C. elegans are highly conserved in most vertebrates, including humans. Therefore, C. elegans will provide a great opportunity to identify therapeutic drugs and their targets, as well as to understand mechanisms underlying the formation of cancer.

청소년기업가정신교육 효과성 검증에 관한 탐색적 연구: 디자인씽킹(Design Thinking)을 활용한 청소년기업가정신교육을 중심으로 (A study on the Effectiveness of Youth Entrepreneurship Education Program: Focusing on the Youth Entrepreneurs Education Program based on Design Thinking)

  • 본 연구는 청소년 대상 기업가정신 교육의 효과성을 구체적으로 살펴보고 이의 수준을 제고 하기 위한 교육 방법론을 탐색하는데 주된 목적이 있다. 이를 위해 '식품(푸드)'을 주제로 '디자인 씽킹' 기반의 기업가정신 교육을 경기도 소재 중학교 및 고등학교 각 1개교에서 재학생들을 대상으로 실시하였다. 선행연구를 통해 기업가정신교육 효과성 측정에서 강조되는 9가지 영역(기회발견, 기회 활용, 창의성 역량, 진로준비 행동, 기업가정신, 창업 의지, 셀프리더십, 사회적 문제해결력, 교육프로그램 목표)을 기초로 '청소년기업가정신교육' 효과성 측정을 위한 탐색적 연구결과는 다음과 같다. 첫째, 디자인씽킹을 활용한 청소년기업가정신교육 후 9개 차원의 모든 측정값이 통계적으로 유의미하게 높아졌는데, 특히 '기회 활용' 차원과 '창업 의지' 차원이 가장 높게 상승했다. '기회 활용' 차원이 유의미하게 두드러진 배경에는, 금번 교육에 참여한 학생들이 '디자인 씽킹' 방법론을 '식품(푸드)' 이라는 구체적인 비즈니스 주제와 결합하여 적용한 실제적 경험을 처음 해보았기 때문에 보인 반응으로 추론된다. '창업 의지'의 경우는 선행연구에서와 같이 교육 참여 여부 또는 경험에 따라 변화의 정도가 가장 큰 만큼, 향후 '청소년기업가정신교육'의 효과성에서 중요한 변수로 다루어져야 함을 확인하였다. 둘째, 9개 차원에 대한 교육 효과분석을 위해 성별, 수강 동기별, 창업동아리 경험 여부 등 응답자특성별로 분석했다, 그 결과 종합지수의 경우 성별, 학교, 기업가정신교육참여 경험 여부에 상관없이 사전-사후 측정값이 통계적으로 유의미하게 상승했다. 특히 수강동기별로 살펴보면 본인 관심과 주위 추천이 통계적으로 유의미하게 높게 나타났다. 이 결과에 대한 해석을 위해 교육이 수행되었던 학교의 교사를 대상으로 심층 인터뷰를 실시한 결과 학생들이 자율적으로 참여하는 동아리 활동에 교사의 영향력(추천)이 크기 때문으로 판단했다. 향후 연구에서는 기업가정신교육이 9개 차원에 어떠한 경로로 영향을 미쳤는지 경로 메커니즘에 관한 연구가 수행되어야 할 것이다.

임상간호사의 임파워먼트와 직무만족에 대한 체계적 문헌고찰 (Empowerment and Job Satisfaction among Clinical Nurses in South Korea: Systematic Review)

  • Purpose: To synthesize and identify the studies that delineated the relationship between empowerment and job satisfaction in clinical nurses in Korea. Methods: Electronic databases were searched, including KmBase, KoreaMed. riss4u, National Assembly Digital Library and National Discovery for Science Leaders (NDSL) The search terms included nurse, satisfaction and empowerment. Only papers published in Korean were included. Results: Twelve studies, from 156 references screened, were included the final analysis. All studies were non-experimental and used correlational analysis. A significant positive relationship between structural or psychological empowerment and job satisfaction was found. Older nurses with longer clinical experience and higher education background were more likely to have high level of structural and psychological empowerment and job satisfaction. Conclusion: The study results indicate that to produce an empowering work environment, nurse managers have to create a supportive organizational culture, and provide access to resources and opportunity as well as to strengthen emotional support.

Study on the global distribution of far-ultraviolet emission in our Galaxy

  • FIMS/SPEAR is a dual-channel far-ultraviolet imaging spectrograph on board the Korean microsatellite STSAT-1, which was launched on 2003 September 27. The primary mission goal of FIMS was to conduct a survey of diffuse far UV emissions in our Galaxy. For this purpose, FIMS completed a survey of about 84% of the sky during its operation of a year and a half. The present study aims to analyze this survey data made in the far UV wavelengths to understand the global evolution of our Galaxy. The far UV wavelength band is known to contain important cooling lines of hot gas: hence, the study will show how the hot gas in our Galaxy, produced by stellar winds and supernova explosion, evolves globally to cool down and become mixed with ambient cooler medium. One of the main findings from previous analyses of the FIMS data is that molecular hydrogen exists ubiquitously in our Galaxy. This discovery leads to another important scientific question: how is molecular hydrogen distributed in our Galaxy and how does it affect globally the evolution of our Galaxy as a cold component? Hence, the present study will cover both the hot and cold components of the ISM, which will also provide the opportunity to investigate the interactions between the two.

  • The purpose of this study is to development of venture business curriculum by FGI interview to venture CEO and related field employer and suggest to venture related structure of liberal art, major part of undergraduate and graduate school. Research methods are 2 step interview with 325 venture business CEO and related employer. They selected practical course to motivate venture entrepreneurship among 3 subsidiary (entrepreneurial condition, psychology of entrepreneur, competency of entrepreneurial technology, competency of entrepreneurial knowledge, competency of managing growing business). Major findings are as follows; the selected course category of entrepreneurial condition of liberal art are venture mind and entrepreneurship, venture biz and spearhed industry. The selected course category of psychology of entrepreneur are leadership, personal relationship, and creativity, negotiation skill, etc. The selected course category of competency of entrepreneurial technology are venture technology, introduction to technology trends. The selected course category of competency of entrepreneurial knowledge are biz validity, entrepreneurship, the discovery of business opportunity, utilization of monetary resource etc. The selected course category of competency of managing growing business are operating strategy of venture business, service marketing and technical innovation and technology management.

  • 유비쿼터스 비즈니스 환경은 기존의 e-비즈니스, m-비즈니스와는 다른 특성을 가지고 있다. 이는 u-비즈니스 모델을 개발할 때 유비쿼터스 컴퓨팅 기술의 특성과 유비쿼터스 비즈니스 환경을 고려해야 한다는 것을 의미한다. 본 논문에서는 유비쿼터스 비즈니스 패러다임의 핵심인 가치의 공동 창출 경험에 초점을 맞춘 u-비즈니스 모델 개발 프레임워크를 제시했다. 본 논문의 u-비즈니스 모델 개발 프레임워크는 8단계로 구성되어 있고 각 단계별 세부 절차를 구체적으로 제시했다. 새로운 비즈니스 패러다임 하에서 성공적인 비즈니스 모델을 발굴하고 수행하고자 하는 기업들에게 본 논문에서 제안한 u-비즈니스 모델 개발 프레임워크는 유용한 가이드라인을 제공할 수 있을 것으로 기대된다.

Genetic Risk Prediction for Normal-Karyotype Acute Myeloid Leukemia Using Whole-Exome Sequencing

  • Normal-karyotype acute myeloid leukemia (NK-AML) is a highly malignant and cytogenetically heterogeneous hematologic cancer. We searched for somatic mutations from 10 pairs of tumor and normal cells by using a highly efficient and reliable analysis workflow for whole-exome sequencing data and performed association tests between the NK-AML and somatic mutations. We identified 21 nonsynonymous single nucleotide variants (SNVs) located in a coding region of 18 genes. Among them, the SNVs of three leukemia-related genes (MUC4, CNTNAP2, and GNAS) reported in previous studies were replicated in this study. We conducted stepwise genetic risk score (GRS) models composed of the NK-AML susceptible variants and evaluated the prediction accuracy of each GRS model by computing the area under the receiver operating characteristic curve (AUC). The GRS model that was composed of five SNVs (rs75156964, rs56213454, rs6604516, rs10888338, and rs2443878) showed 100% prediction accuracy, and the combined effect of the three reported genes was validated in the current study (AUC, 0.98; 95% confidence interval, 0.92 to 1.00). Further study with large sample sizes is warranted to validate the combined effect of these somatic point mutations, and the discovery of novel markers may provide an opportunity to develop novel diagnostic and therapeutic targets for NK-AML.
