Effect of seaweed addition on enhanced anaerobic digestion of food waste and sewage sludge

  • To investigate the effect of seaweed (SW) addition on anaerobic co-digestion of food waste (FW) and sewage sludge (SS), batch experiments were conducted at various substrate concentrations (2.5, 5.0, 7.5, and 10.0 g volatile solids (VS)/L) and mixing ratios ((FW or SS):SW = 100:0, 75:25, 50:50, 25:75, and 0:100 on a VS basis). The effect of SW addition on FW digestion was negligible at low substrate concentration, while it was substantial at high substrate concentrations by balancing the rate of acidogenesis and methanogenesis. At 10 g VS/L, $CH_4$ production yield was increased from 103 to $350mL\;CH_4/g$ VS by SW addition (FW:SW = 75:25). On the other hand, SW addition to SS enhanced the digestion performance at all substrate concentrations, by providing easily biodegradable organics, which promoted the hydrolysis of SS. $k_{hyd}$ (hydrolysis constant) value was increased from 0.19 to $0.28d^{-1}$ by SW addition. The calculation showed that the synergistic $CH_4$ production increment by co-digesting with SW accounted for up to 24% and 20% of total amount of $CH_4$ production in digesting FW and SS, respectively.

Effect of Inoculum and Carbon Sources Difference on Characteristics of Anaerobic Digestion (접종원 및 탄소원의 차이가 혐기소화 특성에 미치는 영향)

  • This study was conducted to investigate the effects of inoculum and carbon sources on anaerobic digestion characteristics. The treatments were combinations of inoculum (digestate of cattle manure and rumen fluid) with carbon sources (starch, cellulose, and xylan). Anaerobic digestion was performed in triplicate at $37^{\circ}C$ for 18 days at 100 rpm. Sampling was performed at 0, 1, 2, 3, 4, 5, 7, 9, 12, 15, and 18 days to measure pH, ammonia-N, volatile solids reduction, the cumulative methane content, and the cumulative methane production. There was a significant difference in methane content depending on the carbon source and there was a significant difference in pH, ammonia-N, methane production, and methane content depending on the inoculum (P < 0.05). The results of methane production were higher in the digestate of cattle manure treatment than in the rumen fluid treatment (P < 0.05). In this study, different digestive patterns depending on the type of carbon source could be used as basic research data to set the hydraulic residence time of anaerobic digestion facilities. In addition, the use of ruminal fluid as an inoculum may help accelerate the hydrolysis and acid production steps.

Effect of Ammonia Load on Microbial Communities in Mesophilic Anaerobic Digestion of Propionic Acid (암모니아 부하에 따른 프로피온산 중온 혐기성 소화 미생물 군집 변동 조사)

  • The present study investigated the effect of ammonia load on microbial communities in mesophilic anaerobic digestion of propionic acid. A laboratory-scale continuous anaerobic digester treating propionic acid as a sole organic substrate was operated under non-inhibitory condition and inhibitory conditions with ammonia (1.5 g and 3.5 g ammonia-N/L, respectively), and bacterial and archaeal communities in the steady states of each ammonia condition were analyzed using high-throughput sequencing. Thirteen bacterial families were detected as abundant bacterial groups in mesophilic anaerobic digestion of propionic acid. Increase in ammonia concentration resulted in significant shifts in microbial community structures. Syntorophobacter, Pelotomaculum, and Thermovigra were determined as the dominant groups of (potential) propionate oxidizing bacteria in the non-inhibitory condition, whereas Cryptanaerobacter and Aminobacterium were the dominant groups of (potential) propionate oxidizing bacteria in the ammonia-inhibitory condition. Methanoculleus and Methanosaeta were the dominant methanogens. Acetate-oxidation coupled with hydrogenotrophic methanogenesis might be enhanced with increases in the relative abundances of Methanoculleus and Tepidanaerobacter acetatoxydans under the ammonia-inhibitory condition. The results of the present study could be a valuable reference for microbial management of anaerobic digestion systems that are exposed to ammonia inhibition and propionic acid accumulation.

Effects of hydrothermal pretreatment on methane potential of anaerobic digestion sludge cake of cattle manure containing sawdust as bedding materials

  • Objective: The purpose of this study was to analyze the effect of the hydrothermal pretreatment of anaerobic digestion sludge cake (ADSC) of cattle manure on the solubilization of organic matter and the methane yield to improve the anaerobic digestion efficiency of cattle manure collected from the sawdust pens of cattle. Methods: Anaerobic digestion sludge cake of cattle manure was thermally pretreated at 160℃, 180℃, 200℃, and 220℃ by a hydrothermal pressure reactor, and the biochemical methane potential of ADSC hydrolysate was analyzed. Methane yield recovered by the hydrothermal pretreatment of ADCS was estimated based on mass balance. Results: The chemical oxygen demand solubilization degree (CODs) of the hydrothermal hydrolysate increased to 63.56%, 67.13%, 70.07%, and 66.14% at the hydrothermal reaction temperatures of 160℃, 180℃, 200℃, and 220℃, respectively. Considering the volatile solids content obtained after the hydrothermal pretreatment, the methane of 10.2 Nm3/ton-ADSC was recovered from ADSC of 1.0 ton, and methane yields of ADSC hydrolysate increased to 15.6, 18.0, 17.4, and 17.2 Nm3/ton-ADSC. Conclusion: Therefore, the optimal hydrothermal reaction temperature that yielded the maximum methane yield was 180℃ based on mass balance, and the methane yield from cattle manure containing sawdust was improved by the hydrothermal pretreatment of ADSC.

Thermophilic Anaerobic Digestion of Polyhydroxybutyrate with and without Thermo-alkaline Pretreatment (열적-알칼리성 전처리 유무에 따른 폴리하이드록시부티레이트의 고온 혐기성 소화 영향 연구)

  • The study investigated the effect of thermo-alkaline pretreatment on the solubilization of polyhydroxybutyrate (PHB) and its potential to enhance of thermophilic anaerobic digestion, focusing on biochemical methane potential (BMP) and methane production rate, using two different particle sizes of PHB (1500 ㎛ and 400 ㎛). Thermo-alkaline pretreatment tests were conducted at 90 ℃ for 24 hours with varying NaOH dosages from 0-80% (w/w). BMP tests with untreated PHB exhibited methane production ranging from 150.4~225.4 mL CH4/g COD and 21.5~24.2 mL CH4/g VSS/d, indicating higher methane production for smaller particle sizes of PHB, 400 ㎛. Thermo-alkaline pretreatment tests achieved a 95.3% PHB solubilization efficiency when 400 ㎛ PHB particles were treated with 80% NaOH dosage at 90 ℃ for 24 hours. BMP tests with pretreated PHB showed substantial improvement in thermophilic anaerobic digestion, with an increase of up to 112% in BMP and up to 168% in methane production rate. The results suggest that a combined pretreatment process, including physical (400 ㎛ PHB particles) and thermo-alkaline (90 ℃, 40-80% NaOH dosage, and 24 hours reaction time), is required for high-rate thermophilic anaerobic digestion of PHB with enhanced methane production.

  • For the laboratory diagnosis of Sarcocystis infections especially in domesticated food animals, several antificial digestion techniques were applied for the musculature specimens and several staining techniques was applied for the bradyzoites of Sarcocystis species isolated. The digestion technique using trypsin(0.5%) and sodium chloride(0.85%) mixed solution was regarded as the most valuable for the detection of asexual stages of Sarcocystis in bovine musculature specimens. Optimal time for digestion was approximately one to four hours. The trypsion digestion technique with Giemsa's stain could be helpful for the detection of Sarcocystis prolferative forms and for the observation of the nucleus of the parasite. A systematic detection was also performed in an autopsy for a bovine carcass naturally infected with Sarcocystis species, and the asexual stages such as metrocytes and bradyzoites were observed in the specific organs, respectively.

In Situ Ruminal Digestion Kinetics of Forages and Feed Byproducts in Cattle and Buffalo

  • The relative disappearance and rate of degradation of dry matter (DM) and neutral detergent fiber (NDF) of nine different feedstuffs were determined by simultaneously suspending groups of substrates, using the nylon bags, in the rumen of males of Sahiwal cattle and Nili-Ravi buffalo. The digestion kinetics of leguminous forages (Lucerne, berseem and cowpeas) and feed byproducts (cotton seed cake, wheat bran and wheat straw) did not differ between the two species. However, the DM and NDF digestibilities and rates of digestion of grasses and wheat straw were greater in buffalo than in cow bulls, indicating that buffaloes are better converters of poor quality roughages than are Sahiwal. The lag time for DM of grasses did not differ between these two species but the NDF lag time was lower in buffalo than in cows, indicating that both the rate and lag time of digestion may be reliable indicators for assessing the NDF quality.

  • Since the anaerobic rumen fungi were discovered in the rumen of a sheep over two decades ago, they have been reported in a wide range of herbivores fud on high fibre diets. The extensive colonisation and degradation of fibrous plant tissues by the fungi suggest that they have a role in fibre digestion. All rumen fungi studied so far are fibrolytic. They produce a range of hydrolytic enzymes, which include the cellulases, hemicellulases, pectinases and phenolic acid esterases, to enable them to invade and degrade the lignocellulosic plant tissues. Although rumen fungi may not seem to be essential to general rumen function since they may be absent in animals fed on low fibre diets, they, nevertheless, could contribute to the digestion of high-fibre poor-quality forages.

  • 2004년말 현재 국내에서 발생되는 음식물쓰레기의 재활용량은 전체 발생량의 $87.7\%$인 10,015톤/일이며, 전체 재활용량의 $6.47\%$인 640톤/일 정도가 혐기성소화 방법에 의해 처리 및 자원화 되고 있다. 국내에서 적용되고 있는 음식물쓰레기 혐기성소화 기술의 대부분은 습식소화(Wet digestion) 공법이며, 처리 방법별로 2상 혐기성소화(Two-phase anaerobic digestion)와 하수슬러지 흑은 축산분뇨와 함께 혼합처리 하는 통합소화(Co-digestion) 공정으로 구분되고 있다. 음식물쓰레기의 자원화 방법에 있어 혐기성소화는 사료화, 퇴비화에 비하여 폐기물의 효과적인 감량화와 자원화 효과뿐만 아니라 유용 에너지원인 메탄가스의 회수가 가능하기에 최근에 주목을 받는 biotechnology중의 하나로 자리매김 하고 있으며, 또한 유기성폐기물의 자원순환형 관리 시스템 구현에 있어 적절한 대안으로 고려되고 있다.

Effects of Solubilization Pretreatment of Wastewater Sludge on Anaerobic Digestion (하수슬러지의 혐기성 소화에 미치는 가용화 전처리의 효과)

  • Solubilization pretreatments were conducted to enhance the anaerobic digestion of the waste activated sludge. Four pretreatment techniques including heating, sonication freezing and thawing, and enzyme addition were employed to solubilize the waste activated sludge under various conditions. Thermal pretreatment by heating showed the highest efficiency compared with other methods, and freezing and thawing was confirmed as a feasible alternative of solubilization as well as the pretreatment of dewatering. There is a clear correlation between the solubilization efficiency of the waste activated sludge and the gas production. Batch digestion results showed the cumulative gas production as much as four times after thermal pretreatment as compared with that by the control sludge without pretreatment. As a result, hydrolysis or solubilization pretreatment might play a significant role in the high rate digestion of the waste activated sludge.
