Preliminary Study on a Microsporidian Isolate Occurring in the Lamerin Breed of the Silkworm Bombyx mori L. in India

  • The silkworm, Bombyx mori L. is prone to infection of various pathogenic organisms. Pebrine, one of the deadliest disease of silkworm caused by highly virulent parasitic microsporidian, Nosema bombycis has been understood since long. Infections of the disease range from chronic to highly virulent and can result in complete lose to the sericulture industry. Several strains and species of microsporidians have since been isolated from the infected silkworms. A new microsporidian spore was isolated from Lamerin breed of the silkworm B. mori have been studied under scanning electron microscope, found to be different in spore size (length 4.36$\pm$0.06 ${\mu}{\textrm}{m}$, width 2.14$\pm$0.01${\mu}{\textrm}{m}$) and shape (ova cylindrical with slight depression) from standard strain N. bombycis (length 3.08$\pm$0.21 ${\mu}{\textrm}{m}$, width 2.01$\pm$0.05 ${\mu}{\textrm}{m}$ and ovidal respectively). In immunological test, the silkworm breed Lamerin isolated micrisporidian spore does not react to different antibody (N. bombycis, M$_{11}$ and M$_{12}$) sensitized latex particle and thus appeared to be a different strain of microsporidian parasitic to the Lamerin breed of the silkworm B. mori.i.i.

Evaluation of Reciprocal Cross Design on Detection and Characterization of Mendelian QTL in $F_2$ Outbred Populations

  • A simulation study was conducted to evaluate the effect of reciprocal cross on the detection and characterization of Mendelian QTL in $F_2$ QTL swine populations. Data were simulated under two different mating designs. In the one-way cross design, six $F_0$ grand sires of one breed and 30 $F_0$ grand dams of another breed generated 10 $F_1$ offspring per dam. Sixteen $F_1$ sires and 64 $F_1$ dams were randomly chosen to produce a total of 640 $F_2$ offspring. In the reciprocal design, three $F_0$ grand sires of A breed and 15 $F_0$ grand dams of B breed were mated to generate 10 $F_1$ offspring per dam. Eight $F_1$ sires and 32 $F_1$ dams were randomly chosen to produce 10 $F_2$ offspring per $F_1$ dam, for a total of 320 $F_2$ offspring. Another mating set comprised three $F_0$ grand sires of B breed and 15 $F_0$ grand dams of A breed to produce the same number of $F_1$ and $F_2$ offspring. A chromosome of 100 cM was simulated with large, medium or small QTL with fixed, similar, or different allele frequencies in parental breeds. Tests between Mendelian models allowed QTL to be characterized as fixed (LC QTL), or segregating at similar (HS QTL) or different (CB QTL) frequencies in parental breeds. When alternate breed alleles segregated in parental breeds, a greater proportion of QTL were classified as CB QTL and estimates of QTL effects for the CB QTL were more unbiased and precise in the reciprocal cross than in the one-way cross. This result suggests that reciprocal cross design allows better characterization of Mendelian QTL in terms of allele frequencies in parental breeds.

mtDNA Diversity and Origin of Chinese Mongolian Horses

  • In order to learn the origin of the Chinese Mongolian horse, we analyzed polymorphisms within the mtDNA D-loop variable region in 305 horses of 6 types of 3 different breeds, including one imported breed, one cultivated breed and 4 types of one local breed. We detected 13 different haplotypes, and subsequent sequence analysis showed that all 6 horse types were genetically diverse. By constructing a cladogram of mtDNA D-loop sequences from the 6 horse types along with homologous sequences from several other horse types obtained from GenBank, we showed that Chinese Mongolian horses have a close genetic relationship with other horse types from Mongolia. We also speculate that several Chinese Mongolian horses descended from Przewalskii horse. Additionally, the 13 haplotypes were dispersed throughout the cladogram, suggesting that Chinese Mongolian horses likely originated from multiple female ancestors. A phylogenetic map of the 6 horse types showed that the genetic relationship between the local Wuzhumuqin and Wushen types were the closest. The Xinihe and Baerhu were also closely related to each other, and slightly more distantly related to the cultivated Sanhe breed. All five of the local Chinese horse types had a much more distant relationship with the imported Thoroughbred breed.

Growth- and Breed-related Changes of Fetal Development in Cattle

  • Breed differences in adult animals are determined during fetal development. If interventions are to be developed that influence growth of muscle and fat, it is important to know at which time during gestation breed differences appear and are fixed. The objective of this study was to characterize fetal development in cattle of different breeds. Pregnant cows of 4 cattle breeds with different growth impetus and muscularity were slaughtered under normal processing conditions and the fetuses were removed. German Angus, a typical beef cattle; Galloway, a smaller, environmentally resistant beef type; Holstein Friesian, a dairy type; and Belgian Blue, an extreme type for muscle growth were used. Fetuses of each breed were investigated at 3, 6, and 9 mo of gestation. Fetuses were weighed and dissected into carcass, organs, and muscles. Body fat weight was obtained using the Soxhlet extraction method. Fetal weight increased most rapidly in the third trimester of gestation mainly due to the accelerated muscle and fat deposition. The organ weight to body weight (BW) ratios decreased and the muscle and fat weight to BW ratios increased. At 3 mo of gestation, Galloway fetuses had the significantly smallest BW, half-carcass weight, leg weight, organ weight, muscle weight and shortest leg length. In contrast, Holstein fetuses had the significantly greatest BW, liver, kidney, and lung weights and significantly longest leg length among the 4 breeds, but no differences between Holstein Friesian and Belgian Blue were detected in half-carcass and leg weight. Indeed, Belgian Blue fetuses had the significantly greatest half-carcass weight, leg weight, and muscle weight at 9 mo of gestation, and Galloway had a significantly greater body fat to BW ratio than Holstein Friesian and Belgian Blue. These differences were not evident at 3 and 6 mo of gestation. These data show that the profound increase of tissue and organ weights occurred in later gestation in cattle fetuses even though breed differences were evident as early as 3 mo of gestation. Depending on the tissue of interest, impacting fetal growth likely needs to occur early in gestation before the appearance of breed-specific differences.

Comparison of Faecal Microbial Community of Lantang, Bama, Erhualian, Meishan, Xiaomeishan, Duroc, Landrace, and Yorkshire Sows

  • The objective of this study was to investigate differences in the faecal microbial composition among Lantang, Bama, Erhualian, Meishan, Xiaomeishan, Duroc, Landrace, and Yorkshire sows and to explore the possible link of the pig breed with the gut microbial community. Among the sows, the Meishan, Landrace, Duroc, and Yorkshire sows were from the same breeding farm with the same feed. Fresh faeces were collected from three sows of each purebred breed for microbiota analysis and volatile fatty acid (VFA) determination. Denaturing gradient gel electrophoresis (DGGE) analysis revealed that samples from Bama, Erhualian, and Xiaomeishan sows, which from different farms, were generally grouped in one cluster, with similarity higher than 67.2%, and those from Duroc, Landrace, and Yorkshire sows were grouped in another cluster. Principal component analysis of the DGGE profile showed that samples from the foreign breeds and the samples from the Chinese indigenous breeds were scattered in two different groups, irrespective of the farm origin. Faecal VFA concentrations were significantly affected by the pig breed. The proportion of acetate was higher in the Bama sows than in the other breeds. The real-time PCR analysis showed that 16S rRNA gene copies of total bacteria, Firmicutes and Bacteroidetes were significantly higher in the Bama sows compared to Xiaomeishan and Duroc sows. Both Meishan and Erhualian sows had higher numbers of total bacteria, Firmicutes, Bacteroidetes and sulphate-reducing bacteria as compared to Duroc sows. The results suggest that the pig breed affects the composition of gut microbiota. The microbial composition is different with different breeds, especially between overseas breeds (lean type) and Chinese breeds (relatively obese type).

ADDITIVE AND HETEROSIS EFFECTS ON MILK YIELD AND BIRTH WEIGHT FROM CROSSBREEDING EXPERIMENTS BETWEEN HOLSTEIN AND THE LOCAL BREED IN BANGLADESH

  • Data from purebred and crossbred cattle involving Holstein and the Local breed in Bangladesh were used to estimate the genetic effects on average daily milk yield and birth weight A total of 877 records on average daily milk yield for 4 types of breed groups and a total of 418 records on birth weight for 5 breed groups were analyzed. Two different methods were applied in this study; the least squares analysis of variance approach and the linear regression approach. Breed group effects were highly significant for both average daily milk yield and birth weight. The result showed that straightbred Holstein produced the highest milk yield and the 7/8 crosses ranked highest in birth weight For the two traits, the additive breed effect was highly significant, whereas the individual heterosis effect was not significant. Furthermore, this study showed a negative maternal heterosis for average daily milk yields and a positive maternal heterosis for birth weight Comparing the breed least squares means obtained from the linear regression approach revealed that straightbred Holstein produced the highest average milk yield and the 3/4 crosses were predicted to have the largest birth weight. It is indicated that the linear regression approach can adequately separate the genetic component of performance, estimate unknown crossbreeding parameters and predict unknown performance of crosses which are not include in the original data.

  • Value of excellent breeding animals is important in livestock industry, but their economic life time is limited. And, many countries have been trying procuration of genetic resource in good animals. Therefore, this study was conducted to determine embryo production and to test efficiency of embryo transfer via non-surgical artificial insemination (AI) in different breed of superior sows. A total of 17 sows were used in this experiment (Duroc, n=10; Landrace, n=4; Yorkshire, n=3). The sows were artificially inseminated by semen of same breed boars. After 4 or 5 days following the AI, the embryos were obtained from the sows and then transferred to Landrace and Yorkshire recipients (n=3, respectively) by non-surgical method. The corpora lutea tended to be increased in Yorkshire and Landrace than Duroc(28 and 26 vs. 17, respectively). The recovery of embryo was 78.8% in Landrace, 65.4% in Duroc and 51.4% in Yorkshire. Duroc showed lower morulaes and early blastocyst embryos than 2, 4, 8 and 16 cell. The morula in Yorkshire was higher (P<0.05) than that of Duroc (4.7 vs. 3.4). Similarly, the morulaes and early blastocyst embryos presented greater (P<0.05) in Landrace compared with other breed sows. The recipient sows were pregnant in a Landrace only. This reason may be due to little embryos inserted in the recipients. In addition, pregnancy results were limited because of the little sows. In conclusion, ovulated ovum in sows can be affected by different breed. Also, further study needed pregnant test by using the many embryo in each breed.

Evaluation of Reciprocal Cross Design on Detection and Characterization of Non-Mendelian QTL in $F_2$ Outbred Populations: I. Parent-of-origin Effect

Eight $F_1$ sires and 32 $F_1$ dams were randomly chosen to produce 10 $F_2$ offspring per $F_1$ dam, totaling 320 $F_2$ offspring. Another mating set comprised three $F_0$ grand sires of B breed and 15 $F_0$ grand dams of A breed to produce the same number of $F_1$ and $F_2$ offspring. A chromosome of 100 cM was simulated with large, medium or small QTL with fixed or different allele frequencies in parental breeds. A series of tests between Mendelian and POE models were applied to characterize QTL as Mendelian, paternal, maternal or partial expression QTL. The overall detection powers were similar between the two mating designs. However, the proportions of paternally expressed QTL that were declared as paternal QTL type were greater in the reciprocal cross design than in the one-way cross, and vice versa for Mendelian QTL. When QTL alleles were segregating in parental breeds, a significant proportion of Mendelian QTL were spuriously declared POE QTL, suggesting that care must be taken to characterize imprinting QTL in a QTL mapping population with a small number of $F_1$ parents.

Comparative Meat Qualities of Boston Butt Muscles (M. subscapularis) from Different Pig Breeds Available in Korean Market

  • This study aimed to determine the effects of breed on meat quality characteristics of porcine Boston butt muscles (M. subscapularis) from three different pig breeds: Landrace×Yorkshire×Duroc (LYD), Berkshire, and Ibérico available in Korean market. Ibérico showed significantly higher fat content, yellowness (CIE b⁎), cooking loss, and lower shear force values than LYD and Berkshire. Moreover, the contents of oleic acid (18:1) and palmitic acid (16:0) were significantly higher in Ibérico breed, but stearic acid (18:0) was higher in LYD. As linoleic acid (18:2) and arachidonic acid (20:4) were higher in Berkshire sows as compared to the other breeds, atherogenicity and thrombogenicity indexes were significantly lower in Berkshire sow. Ibérico had lower the ω-6/ω-3 fatty acids ratio, and higher taurine and free amino acids compared with the others. Ibérico also showed significantly greater lipid oxidation, lower antioxidant capacity, and higher hypoxanthine contents, whereas the Berkshire had higher inosine-5'-monophosphate and lower K-index value as compared to the Ibérico. The breed did not impart any significant effect on the size and density of muscle fibers. Thus, quality characteristics of Boston butt varied from breed to breed, and certain consumer preferences for Ibérico can be explained, in part, by the unique quality characteristics imparted by higher contents of intramuscular fat, oleic acid, and free amino acids.

CT Evaluation of Age-Related Changes in Epaxial Muscle Attenuation and Cross-Sectional Area for Sarcopenia and Myosteatosis in Small Breed Dogs

  • Sarcopenia and myosteatosis can increase the risk of adverse effects in dogs and humans. However, such imaging study results for evaluating sarcopenia and myosteatosis in small dogs have not yet been available. The objective of this study was to assess age-related changes according to the breed in epaxial muscle cross-sectional area (CSA) and fat infiltration using CT to evaluate sarcopenia and age-related myosteatosis in small breed dogs. In 144 dogs (92 Maltese dogs, 27 Poodle dogs, and 25 Shih Tzu dogs), Hounsfield Unit (HU) values and CSA of left epaxial muscle were measured at the thirteenth thoracic vertebral level on non-contrast transverse CT images. Differences in HU values and CSA according to age and breed were analyzed. The geriatric group (≥12 years) had significantly lower HU values of epaxial muscle than mature adult group (2 to 6 years) of all breeds. The geriatric group had significantly lower CSA of epaxial muscle than mature adult and senior groups (7 to 11 years) of Maltese dogs. HU values of epaxial muscle were not significantly different among all age groups of all breeds. Maltese dogs had significantly lower CSA of epaxial muscle than Poodle and Shih Tzu dogs in all age groups. Results of this study showed that as age increased in small breed dogs, muscle mass and density decreased.
