Effect of Different Spray Dried Plasmas on Growth, Ileal Digestibility, Nutrient Deposition, Immunity and Health of Early-Weaned Pigs Challenged with E. coli K88

  • A total of 96 piglets were weaned at 19 and 13 days in Exp. 1 and 2, respectively, and allotted to one of four diets: three with different spray dried plasmas (SPs) and one with hydrolysed casein (HC). SPs were from pigs (SPP), mixed origin (SMP), and mixed origin with standardized level of immunoglobulins (SMPIG). All the diets contained 1.7% total lysine, 25% of the test protein source, 45% corn starch, 15% lactose, 2% sucrose, 7% soybean oil. At d 4 and d 2 in Exp. 1 and 2, respectively, piglets were perorally challenged with $10^{10}$ CFU E. coli K88. Growth performance, immunity, and health condition were measured for 15 days and 14 days in Exp. 1 and 2, respectively. To investigate apparent ileal digestibility and nutrient deposition, all piglets were sacrificed at d 14 in Exp. 2. In 1. 3 piglets died in HC diet and 1 in SPP diet. HC diet showed higher mortality (p<0.01) than other diets. In Exp. 2, no clinical sign of infection was detected, no difference for the content of E. coli K88 was found in feces at 4 and 6 days after the infection, and no E. coli K88 was found in the jejunum at the end of experiment. In both experiments, feed intake was lower for HC diet and ADG was 96, 106, 122 and 155 for HC, SPP, SMP and SMPIG diet, respectively (HC vs others, p<0.05; SMPIG vs other SP, p<0.01). Heal apparent digestibility of nitrogen in sacrificed piglets was higher for HC diet (p<0.05). After the challenge, K88-specific titers in saliva (Exp. 1) and in plasma (Exp. 2) were reduced in SMP and SMPIG. The piglets positive to the adhesion of the used E. coli strain to the intestinal brush borders had a significantly reduced growth (p<0.01) and a higher K88-specific IgA titer in plasma, in comparison with negative ones. This effect was independent of the diet. The data show the relevance of spray dried plasma sources and particularly of SP with standardized level of immunoglobulins for the feeding of early-weaned at the risk of infection by enterotoxigenic bacteria.

  • This research evaluated how the composition of Yak-sun(oriental diet therapy), taken as a nutritional supplement with daily meals, can effect the medical condition of those suffering from modern diseases like obesity and hyperlipideia. We produced Yak-sun tea, determined its nutrient composition and physical characteristics and evaluated how this tea effects affects the active oxygen concentration inside the body by clinical practices. The study results indicated that this tea significantly decrease active oxygen concentration. We think that scientific and objective evaluation was done on the components of Yak-sun tea prescription. We concluded that we could apply the components not only in the form of tea, but also in other food forms. The information we received from this conclusion will provide basic information for the application of oriental medicinal resources into other foods and will also be a steppingstone in the field of functional food research for the development of medicinal herbs which is already an important research field world-wide.

Effect of High Dietary Carbohydrate on the Growth Performance, Blood Chemistry, Hepatic Enzyme Activities and Growth Hormone Gene Expression of Wuchang Bream (Megalobrama amblycephala) at Two Temperatures

  • The effects of high carbohydrate diet on growth, serum physiological response, and hepatic heat shock protein 70 expression in Wuchang bream were determined at $25^{\circ}C$ and $30^{\circ}C$. At each temperature, the fish fed the control diet (31% CHO) had significantly higher weight gain, specific growth rate, protein efficiency ratio and hepatic glucose-6-phosphatase activities, lower feed conversion ratio and hepatosomatic index (HSI), whole crude lipid, serum glucose, hepatic glucokinase (GK) activity than those fed the high-carbohydrate diet (47% CHO) (p<0.05). The fish reared at $25^{\circ}C$ had significantly higher whole body crude protein and ash, serum cholesterol and triglyceride, hepatic G-6-Pase activity, lower glycogen content and relative levels of hepatic growth hormone (GH) gene expression than those reared at $30^{\circ}C$ (p<0.05). Significant interaction between temperature and diet was found for HSI, condition factor, hepatic GK activity and the relative levels of hepatic GH gene expression (p<0.05).

Health and Nutritional Status of Industrial Workers (근로자의 근무유형별 건강상태와 영양섭취상태 비교 연구)

  • The study was curried out to collect information to establish a framework for nutrition education for the prevention of chronic degenerative disease. We analyzed differences in diet quality, food habits and health status of workers by work condition. Anthrometric parameters of height, weight and body fat were measured and biochemical parameters including glucose, total cholesterol, GOT, GPT and hemoglobin were determinded for 194 subjects. To assess the nutrient intake and diet quality of workers, dietary intake was measured by the day 24-hour recall method, Average daily nutrient intake, except for phos-phorous and vitamin C was lower than Korean RDA. The obesity related behavior score was significantly better in laborers than in office workers, while chronic degenerative diseases related to food habit score was significantly better in laborers than in office workers, while chronic degenerative diseases related to the food habit score was beet in offices workers than in laborers. Blood pressure, blood glucose levels were significantly higher in laborer than in office workers. Dietary variety score (DVS) food composition group score(FCGS), mean adequacy ratio(MAR) of office worker were better than those of labor workers. When diet quality was evaluated by FCGS(food composition group score) 16.0% of the subjects acquired 5 points and 14.4% of the subjects acquired 2 points. MAR and INQ showed a significantly positive correlation with DVS and FCGS . This results indicated that the onset possibility of hypertension and diabetes mellitus among chronic degenerative disease was higher in laborers than in office workers, while the onset possibility of obesity was higher in office workers than in laborers. In conclusion the overall diet quality of office workers is betters than that of laborers, therefore, nutrition education for prevention of chronic degenerative disease of industrial workers needs to be more focused on the improvement of the health status of laborers.

  • In the previous studies, one of our author had observed the obligatory nitrogen losses through urine and feces in young Korean male adults, and utilization of whole egg protein in Korean young adults with ordinary intake of energy as a basic study for estimating protein requirements of Korean. This study has tried to determine the protein requirement and utilization when subjects ate the Korean mixed diet at the energy level of 45 kcal per Kg body weight per day. Six healthy Korean young female adults, college students, aged $20{\sim}25$ years old, and $45{\sim}61Kg$ of body weight participated as the study subjects. They were given isocaloric diets with four different protein levels for five days each successively. These diets contained protein levels of 0.45, 0.60, 0.75 and 0.90g per Kg of body weight per day, respectively. It was attempted to observe energy and protein intake, urinary and fecal nitrogen losses, true digestibility of protein, net protein utilization of protein in the body over last two days of period of eating each diet and nitrogen balances were calculated. The body weight change and hematological observation were also performed. The results obtained were summarized as follows ; 1) True digestibility of protein of Korean mixed diets ranged from 83.1% to 86.5% and the average of which was $84.7{\pm}1.7%$. 2) Net protein utilization rate of Korean mixed diets was range of 49% to 55% and the average of which was $52{\pm}3%.$ 3) The body weight of subjects were slightly increased and the values of hemoglobin and hematocrit of the blood were also little increased during the experimental periods of twenty days. 4) Protein requirements of Korean mixed diet with the energy level of 45 kcal/Kg body weight per day of Korean young female adults were estimated as 1.00g/Kg/day on this experimental condition.

Effects of Dietary Inclusion of Various Additives on Growth Performance and Immune Responses in Juvenile Olive Flounder(Paralichthys olivaceus) (사료내 다양한 첨가제가 넙치 (Paralichthys olivaceus) 치어의 성장 및 비특이적 면역반응에 미치는 영향)

  • This study was conducted to investigate the effects of dietary inclusion of various additives Solid sulfur, fucoidan, and glucan on growth performance and immune responses in Juvenile olive flounder paralichthys olivaceus. Six experimental diets supplemented with : no additives (CON); 0.5 and 1.0% solid sulfur (S); 0.2 and 0.4% fucoidan (F); 0.1% glucan (G) of these additives diet on a dry-matter basis. Fish averaging $8.2{\pm}0.17g$ were fed one of six experimental diets in triplicate groups for 6 weeks. By the end of the feeding trial, weight gain, feed efficiency, specific growth rate, protein efficiency ratio, hepatosomatic index and condition factor of fish fed diet CON were significantly lower then those of fish fed the all additives diets (P < 0.05). In challenge test, fish were infected by intraperitonel injection of 0.1 ml bacterial suspension with Edwardsiella tarda per fish after the feeding trial. As a result fish fed All additive diet showed a lower cumulative mortality then did fish fed CON diet throughout the challenge test. In conclusion, these results indicated that solid sulfur, fucoidan and glucan enhanced the growth, feed efficiency and non-specific immune activity of juvenile flounder and protect the fish against microbial infections.

  • This treatise was sat up as a basic study of the artificial diets of silk worm, both nursed and wild, by storing the natural leaves in defarmentizing solution and the results obtained were as followings. 1. Defarmentizer C, a kind of defarmentizer, was developed by the author in order to store any natural diet by soaking them in the solution. 2. The defarmentizer was workable for aerophobic bacteriums, but was not effectable for aerophilic bacteriums or fungus. 3. The defarmentized diet could feed silk worm during winter season. 4. The storing method has found the importance of growthness of the diet leaves and the perfect riped leaves were found as the best condition for the purpose. 5. The perfect riped leaves were softened properly during the soaking to be fed for silk worm. 6. The defarmentizing solution has held almost the same pH value of protein isoelectric point so that the protein extraction be ceased during the soaking process. 7. The utilization of defarmentizer has found an economical value. 5. The analytical results of various wild plant leaves were found to be usable as the artificial diet mixture for general silkworm.

Impact of 25-hydroxyvitamin D3 on productive performance of gestating sows

  • The primary goals of this research were to evaluate the impact of diet supplemented with 25-hydroxyvitamin $D_3$ ($Hy{\cdot}D(R)$) on sow's body condition and reproduction performance. A total of sixteen multiparous sows [(Landrace ${\times}$ Yorkshire), average parity = $3.79{\pm}0.32$] and their litters were randomly allotted to 2 treatments to give 8 replicates per treatment. Diet treatments were randomized to receive a non-active (ND) or active 25-hydroxyvitamin $D_3$ (AD) diet (0.36 mg cholecalciferol/g) during pregnancy. The results of this experiment were observed at the gestation of d 58 - 75, d 76 - 95, d 96 - 110, and d 111 - 115. A corn-soybean meal-based diet was formulated to meet or exceed the nutrient requirements recommended by NRC (2012). Results indicated that the sows' farrowing duration was shortened (4.71, 5.38 h), and the average number of mummified fetuses decreased significantly (p < 0.05) in AD treatment compared with ND treatment (0.1, 0.5) while birth weight was significantly (p < 0.05) improved (1.44, 1.18 kg). There were no significant effects on body weight, backfat thickness, and fecal score during the gestation of sows in different phases (p > 0.05). And the total birth, stillbirth, live birth, and survival rates of the litter did not change (p > 0.05). The results of this study suggest that the farrowing duration of sow pigs will be shortened and the number of mummies will be decreased while their litters' body weight may be improved, if fed active 25-hydroxyvitamin $D_3$ (0.36 mg/g) during pregnancy phase.

Development of Anti-Obesity Agent from Resource Plants

  • Obesity is a physical condition that results from excessive storage of fat in the body. The present study examined the anti-obesity effects of the selected natural medicine, Galla rhois extract (GRE) and solvent fractions on 3T3-L1 preadipocytes and in vivo studies. Here, we show that EtOAc fraction of Galla rhois inhibits the differentiation of the 3T3-L1 preadipocytes induced by differentiated medium in a dose-dependent manner. To investigate the effect of the GRE-EtOAc fraction on obesity in high fat diet-fed C57BL/6 mice, which included a normal diet (ND), high-fat diet (HFD) and HFD+GRE concentration-dependent, were fed to the mice for 6 weeks. The GRE-EtOAc fraction was inhibited the highest adipocyte differentiation in vitro, the GRE supplement significantly decreased body weight and visceral fat mass compared to the HFD group. The total cholesterol and triglyceride levels in the plasma were significantly decreased by GRE supplementation compared with those of the HFD group. Also, we aimed to determine the differentiation inhibition and the modulation of differentiation genes brought about by the Galla rhois in adipocyte. A cDNA microarray-based method was introduced for the high contents screening (HCS) of gene expressions. This technology has revolutionized gene expression studies by providing the means to measure mRNA levels in thousands of genes simultaneously in simple and complex biological samples. 13 genes were founded to be affected in their expression levels by more than 5-fold up-regulation after 4 days treatment with the EtOAc fraction from Galla rhois. Otherwise, 21 genes were founded to be affected in their expression levels by more than 5-fold down-regulation treated with the EtOAc fraction. Therefore, Galla rhois extract may be considered for use in a therapeutic agent to control obesity.

Spent Mushroom Substrate Influences Elk (Cervus Elaphus Canadensis) Hematological and Serum Biochemical Parameters

  • The objective of this study was to evaluate the effect of spent mushroom substrate (SMS) derived from Pleurotus eryngii on the hematological and biochemical blood properties of elk. A total of 18, two and three-year-old elk were fed three different levels of SMS (0, 15 and 20%) in a corn-wheat bran diet for 80 days. The results indicated significantly high levels of blood monocytes, hemoglobin (Hb), and hematocrit (HCT) in elk fed 15% or 20% SMS (p<0.05) compared to control animals. Serum blood urea nitrogen (BUN) and glucose concentrations were also significantly elevated in elk fed both 15% and 20% SMS. The inclusion of SMS in the elk diet did not affect serum total cholesterol, triglyceride, or low density lipoprotein (LDL)-cholesterol concentrations; however, high density lipoprotein (HDL)-cholesterol concentration was significantly increased in SMS-fed groups. In addition, 20% SMS in the diet increased serum iron and testosterone concentrations in elk. These results indicate that adding SMS to the diet of elk can increase their Hgb, serum BUN, glucose, and HDL-cholesterol concentration; therefore, diets containing SMS may enhance the physiologic condition of elk during growth.
