인삼(人蔘) Saponin이 혈장(血漿) Lipoproteins 및 Lecithin Dispersion과 Dextran Sulfate의 상호작용(相互作用)에 미치는 영향(影響) (Studies on the Effect of Panax ginseng on the Interactions of Human Plasma Lipoproteins and Lecithin Dispersions with Dextran Sulfate)

  • The effect of saponin fractions of Panax ginseng root on the interactions of human plasma lipoproteins and lecithin dispersions with dextran sulfate were studied in order to examine the effect of Panax ginseng on the lipid accumulation in the aorta. The total saponin fraction and protopanaxadiol glycosides of Panax ginseng root seemed to slightly enhance the interaction of low density lipoproteins with dextran sulfate in the absence of divalent metal ions. Protopanaxatriol glycosides remarkably inhibited the interaction of low density lipoproteins with dextran sulfate. However, all of these three saponin fractions of Panax ginseng root showed the tendency of inhibition to the interaction of high density lipoproteins with dextran sulfate in the presence of divalent metal ions by the order of protopanxatriol glycosides, protopanaxadiol glycosides and total saponin. Three saponin fractions of Panax ginseng exerted almost same tendency to the interaction of lecithin dispersions with dextran sulfate in the presence of divalent metal ions as the interaction of low density lipoproteins with dextran sulfate absence of divalent metal ions.

Fabrication and Evaluation of Polyelectrolyte Complexes of Dextran Derivatives for Drug Coating of Coronary Stents

  • The aim of this study was to fabricate a dextran polyelectrolyte multi-layer on a bare metal stent (BMS) and to evaluate bio-physical properties of the layer. Diethylaminoethyl-dextran (DEAE-D) as a polycation and dextran sulfate (DS) as a polyanion were successively coated on the bare metal stent by a well-known layer-by-layer procedure. The morphology of the stent surface and its cell adhesion were studied after each coating step by scanning electron microscopy. The stent showed more blotched and slightly rougher morphology after dextran-DS coating. The contact angle of the DEAE-DS group ($39.5{\pm}0.15^{\circ}$) was significantly higher than that of the BMS group ($45.16{\pm}0.08^{\circ}$), indicating the improvement of hydrophilic. The SMC proliferation inhibition in the DEAE-DS-coated stent group ($20.9{\pm}0.04%$) was stronger than that in the control group ($21.7{\pm}0.10%$ in DS-coated group only). The DEAE-DS coating is desired for stent coating materials with biocompatibility and anti-restenosis effect.

Dextran Sulfate Sodium으로 유도된 궤양성 대장염에 대한 작약의 개선 효과 (The Improving Effect of Paeoniae Radix on Dextran Sulfate Sodium-induced Colitis in Mice)

  • 염증성 대장질환에 대한 작약의 효능 연구가 미비하여 본 연구에서는 dextran sulfate sodium로 유도된 궤양성 대장염의 임상증상인 체중감소, 결장단축, 질병활성화 정도에 대한 작약의 효과를 관찰하였다. 또한 대장 조직 내 염증매개인자인 interleukin-6 및 cyclooxygenase-2의 생성에 대한 작약의 효과를 측정하여 다음과 같은 결과를 얻었다. 첫째 dextran sulfate sodium로 유도된 궤양성 대장염에서 체중 감소에 대한 작약의 효과를 관찰한 결과 유의적으로 억제함을 확인하였다. 둘째 DSS로 유도된 궤양성 대장염에서 결장 단축에 대하여 작약의 억제 효과가 유의성 있게 나타났다. 셋째 dextran sulfate sodium로 유도된 궤양성 대장염에서 임상증상인 설사, 잠혈 및 출혈 등의 질병활성화 정도에 대한 작약의 효과를 관찰한 결과 유의적으로 질병활성화 정도를 억제하였다. 넷째 대장 조직내 염증매개인자인 interleukin-6 및 cyclooxygenase-2의 생성증가에 대한 작약의 억제효과를 확인 하였다. 본 연구는 작약의 효능을 과학적으로 입증한 결과로, 작약의 궤양성 대장염의 치료제 개발의 소재로서의 가능성을 시사한다.

  • Dextran-5-(4-ethoxycarbonylphenylazo)salicylic acid ester(Dextran-5-ESA) was synthesized as a potential colon-specific prodrug of 5-aminosalicylic acid (5-ASA). No free 5-(4-eth oxycarbonylphenylazo) salicylic acid (5-ESA) was detected when the chemical stability of dextran-5-ESA was tested at pH 1.2, or pH 6.8 bath solution, Effects of the degree of substitution (DS) and molecular weight of dextran on the depolymerization by dextranase was investigated. Depolymerization(%) decreased with increasing DS, and was not affected by M.W. of dextran. The extent of prodrug conversion after incubation in the contents of various G.I. Tract segments of rats was evaluated. 5-ASA was released in the cecal contents, but not in the contents of proximal small intestine (PSI) or distal small intestine (DSI). No significant prodrug conversion was observed in the cecal contents of rats pretreated with kanamycin sulfate, which indicated that microbial enzymes were responsible for the cleavage of the prodrug.

상지추출물이 Dextran Sulfate Sodium으로 유도된 대장염 마우스 모델에 미치는 항염증 효능 (Anti-inflammatory effects of mulberry twig extracts on dextran sulfate sodium-induced colitis mouse model)

  • 본 연구는 DSS 유도 대장염 마우스 모델에서 MT열수 추출물의 항대장염 효능을 확인하고자 ICR 마우스에 5일 동안 3% DSS와 함께 MT추출물을 경구투여한 후 대장염의 임상적 증상 및 염증 지표들을 분석하였다. 그 결과 MT추출물의 투여는 DSS에 의해 유도된 마우스 대장염 증상의 지표들, 즉 체중 감소와 혈성 설사를 포함한 질병 활성도, 대장점막증식, 그리고 $TNF-{\alpha}$와 IL-6를 포함한 염증성 사이토카인량 증가를 개선시키는 것으로 나타났다. 또한 DSS에 의해 증가된 Cox-2, iNOS, p-Erk 등 염증 지표 단백질의 발현도 MT추출물 투여군에서 감소되었다. 이러한 결과들은 MT추출물이 항대장염 효능이 있음을 제시하며 향후 MT추출물에 포함되어 있는 생리활성물질의 효능과 관련기작이 규명된다면 상지를 물로 우려내어 섭취하는 상지차 형태의 대장염 예방 혹은 개선용 건강식품으로의 개발을 기대해 볼 수 있을 것으로 사료된다.

매실 혼합물이 DSS로 유도된 염증성 장질환 동물모델의 면역조절에 미치는 활성 (Anti-Inflammatory Effects of Prunus mume Mixture in Colitis Induced by Dextran Sodium Sulfate)

  • This study was conducted to investigate the anti-inflammatory effects of Pruns mume, Schisandra chinensis, Chaenomeles sinensis-- Prunus mume mixtrue (PM) treatment on colitis induced in mice by dextran sodium sulfate (DSS) treatment. A total of 25 male BALB/c mice (average weight $20.7\;{\pm}\; 1.6 \;g$) were divided into 5 treatment groups and fed a commercial diet (A), PM administration (B), commercial diet + induced colitis by DSS (C), PM administration + induced colitis by DSS (D) and sulfasalazine + induced colitis by DSS (E). We found that PM treatment (D) and sulfasalazine (E) decreased the expression of $TNF-{\alpha}$ and COX-2 compared to the DSS-induced colitis group (C). The expression of IL-4, STAT6, $IFN-{\gamma}$, STAT1 was decreased in group D and group E compared to the colitis group (C), COX-2 and STAT1 were more decreased in group D. The serum IgE levels decreased in the PM treatment groups (C and D) compared to the non-PM treatment groups (A and B) although there was no significant difference between the PM treatment groups. It is notable that a therapeutic application of the PM extracts ameliorated DSS-induced colitis in mice.

황련해독탕이 DSS로 유발된 흰쥐의 궤양성 대장염에 미치는 영향 (Effects of Hwangryunhaedoktang on DSS-induced Colitis)

  • Objectives : We examined the effect of Hwangryunhaedoktang(HH) on the experimental ulcerative colitis induced by dextran sulfate sodium (DSS). Methods : Experimental colitis was induced in rats by daily treatment with 5% DSS in the drinking water for 5 days. Afterward, the mts were divided into two groups: the control group was administered water and the sample group was administered HH for 7 days. Results : The sample group provided HH for 7 days demonstrated fast recovery of body weight compared with the control group. Histologic change showed fast regeneration of crypt and surface epithelial cells and decreased edema of the submucosa and decreased lymphatic follicle of mucosa compared with the control group. Immunohistochemical stain usingCOX-2 gene was decreased and there was localized Ki-67 positive reaction. Regeneration of surface epithelial cell and goblet cell in mucosa was observed by transmission electron microscope. These results indicate therapeutic effect of HH on DSS-induced colitis in rats.

DSS으로 유발된 생쥐의 대장점막손상에 대한 도체탕(導滯湯)의 효과 (Effects of Doche-tang on Colonic Mucosal Ulcer Induced by DSS in Mice)

  • Objectives : This study was carried out to investigate the effects of Doche-tang on colonic mucosal ulcer induced by dextran sulfate sodium(DSS). Method : The group was divided into three. The normal group consisted of mice that were not inflammation-induced. The control group was composed of untreated colitis elicited mice. The sample group was administered Doche-tang after colitis elicitation. The effects on colonic mucosal ulcers were evaluated by the morphological change of colonic mucosa, the anti-oxidant effect, HSP 70, $NF-{\kappa}B$, COX-1, COX-2 and iNOS. Results : In terms of immunohistochemical findings, the distribution of COX-1 in mice treated with Doche-tang noticeably increased more than that in the control group. The distribution of HSP70, $NF-{\kappa}B$, COX-2, iNOS in mioe treated with Doche-tang decreased more than that in the control group. Regeneration of surface epithelial cell and goblet cell in mucosa was observed by transmission electron microscope. Conclusion : According to the results, Doche-tang is practicable treatment for colonic mucosal ulcer.

  • ICR웅성마우스에 5% DSS로 궤양성 대장염을 유발시킨 후, 정상대조군, 궤양유발군, 양성대조군 및 식방풍 잎 추출물 투여군의 궤양성 대장염에 미치는 효과를 요약한 결과는 다음과 같다. 1. 식방풍 잎 추출물 투여군은 양성대조군에 비해 체중감소 완화 효과를 나타냈지만, 농도에 따른 유의적인 차이는 나타나지 않았다. 2. 식방풍 잎 추출물 투여군의 대장조직 및 비장변화는 양성대조군과 비슷한 개선효과를 나타냈다. 3. 식방풍 잎 추출물 투여군은 혈중내 염증성 사이토카인을 농도 의존적으로 감소시켰으며, 대장조직내 염증성 사이토카인 mRNA 발현을 억제하는 효과를 나타냈다.
