  • Pseudomonas sp. BK7, an alkalophile, displayed the highest growth and protease activity when grown in a fermenter which was controlled at a pH level of 9.0, and the enzyme production was significantly enhanced by the increase of agitation speed. Two forms of alkaline proteases (BK7-1 and BK7-2) were fractionated and purified to near homogeneity. Protease BK7-1 was purified through CM-Sepharose CL-6B and Sephadex G-75 column chromatographies, and Protease BK7-2 was purified through CM-Sepharose CL-6B, DEAE-Sepharose, and Sephadex G-75 column chromatographies. The molecular weights of proteases BK7-1 and BK7-2 determined by gel filtration chromatography were 20,700 and 40,800, respectively. The $K_m$ value, isoelectric point, and optimum pH of protease BK7-1 were 2.55 mg/ml, 11.0, and 11.0, respectively, whereas those of protease BK7-2 were 1.57 mg/ml, 7.2, and 10.0, respectively. Both proteases were practically stable in the pH range of 5-11. The optimum temperatures for the activities of both protease BK7-1 and BK7-2 were $50^{\circ}C$ and $45^{\circ}C$, respectively. About 56% of the original protease BK7-2 activity remained after being treated at $50^{\circ}C$ for 30 min but protease BK7-1 was rapidly inactivated at above $25^{\circ}C$. Both proteases were completely inhibited by phenylmethane sulfonyl fluoride, a serine protease inhibitor. Protease BK7-2 was stable against EDTA, EGTA, STP, and detergents such as SDS and LAS, whereas protease BK7-1 was found to be unstable.

  • The purpose of this study is to find out the better washing condition, especially on detergent contents to satisfy the consumer's concern on fabric hand using Drum Type Washer. The hand and preference of washed fabrics by various detergent contents were analyzed through subjective evaluation using questionnaire method in dry and wet state. Wine rank's semantic differential scale questions were developed with 27 kinds of adjective pairs and seven rank's scale questions were to evaluate preference of washed fabrics oil holistic touch, washing and rinsing effect and purchase intention of detergent. Group of trained panelists and untrained women panelists of $30{\sim}40$ years old were participated. The factors affecting consumer's taste for the washed fabrics were analyzed by SPSS 12.0. Smoothness showed relatively higher correlation with preference of washed fabrics on holistic touch, washing and rinsing effect and purchase intention of detergent. There were significant differences in preference of washed fabrics on holistic touch, washing and rinsing effect and purchase intention of detergent by detergent contents when tested in wet state. Fabrics washed with detergents of non-zeolite were appeared to be the preferred ones.

  • An alcohol oxidase from Hansenula polymorpha was strongly induced when cells were grown with 0.5% methanol supplementation as the carbon source. The induced Hansenula polymorpha alcohol oxidase was purified to electrophoretic homogeneity by using DEAE-Sephacel and Mono Q column chromatographys. The enzyme oxidized mainly primary aliphatic alcohols and exhibited high substrate specificity towards ethanol and methanol. The activity of the enzyme optimally proceeded at pH 8.5 and $50^{\circ}C$. The midpoint of the temperature-stability curve for the enzyme was approximately $52^{\circ}C$ and the enzyme was not completely inactivated even at $65^{\circ}C$ temperature. The enzyme showed resistance toward detergents and highly stable over 7 weeks of storage condition. This Hansenula polymorpha alcohol oxidase may be useful for the enzymatic determination of alcohol and for the industrial production of alcohols and aldehydes.

  • ATP synthase produces ATP, a major energy source for metabolic processes in organisms, from ADP and inorganic phosphate in cellular membranes. ATP synthase is known as a rotary motor, in which the c-subunit ring functions as a rotor. In this work, we have tried to develop a more general preparation procedure of thermophilic $F_oc$-ring ($TF_oc$-ring) for NMR measurements. The expression of $TF_oF_1$ is easily affected by various experimental conditions such as temperature, shape and size of a flask, a volume of medium, and shaking rate of an incubator. Accordingly, we have tried to optimize the expression conditions of $TF_oF_1$. $TF_oc$-rings were purified from $TF_oF_1$ according to a reported method. We modified purification procedures to improve purity and yield of $TF_oc$. On top of them, we found a new combination of detergents for the purification at anion-exchange column chromatography. To examine the effect of lipid environments on the structure, the $TF_oc$-rings were reconstituted into two kinds of lipid bilayers, namely, saturated and unsaturated lipid ones. Then, we have compared characteristics of the $TF_oc$-ring structures in these membranes with solid-state NMR.

  • Two putative genes, lip29 and est29, encoding lipolytic enzymes from the thermophilic bacterium Geobacillus thermocatenulatus KCTC 3921 were cloned and overexpressed in Escherichia coli. The recombinant Lip29 and Est29 were purified 67.3-fold to homogeneity with specific activity of 2.27 U/mg and recovery of 5.8% and 14.4-fold with specific activity of 0.92 U/mg and recovery of 1.3%, respectively. The molecular mass of each purified enzyme was estimated to be 29 kDa by SDS-PAGE. The alignment analysis of amino acid sequences revealed that both enzymes belonged to GDSL lipase/esterase family including conserved blocks with SGNH catalytic residues which was mainly identified in plants before. While Est29 showed high specificity toward short-chain fatty acids (C4-C8), Lip29 showed strong lipolytic activity to long-chain fatty acids (C12-C16). The optimal activity of Lip29 toward p-nitrophenyl palmitate as a substrate was observed at 50℃ and pH 9.5, respectively, and its activity was maintained more than 24 h at optimal temperatures, indicating that Lip29 was thermostable. Lip29 exhibited high tolerance against detergents and metal ions. The homology modeling and substrate docking revealed that the long-chain substrates showed the greatest binding affinity toward enzyme. Based on the biochemical and insilico analyses, we present for the first time a GDSL-type lipase in the thermophilic bacteria group.

  • Surface activities including CMC, surface tension, foaming power etc. of the rhamnolipid (RL), a biosurfactant fermented by microorganism were investigated. Both the RL and liquid detergent contained RL showed a good surface activity. For the detergency test, the liquid detergent with RL showed a slightly better performance than that of using an fatty acid. However for the foaming profile, the liquid detergent with RL exhibited a lot of foam volume and the suppression of the rinsing and foaming during rinsing cycles with the RL was not seen. Therefore it can be concluded that RL can be used as a main surfactant or co-surfactant in liquid detergents without functions of foaming suppression or rinse-aid.

  • Water pollution is drastically increasing day by day, because of the enhancement in use of livestock, urban sewage detergents and fertilizers. Moreover increase in concentration of phosphorous and nitrogen contained in sewage, leads to green tide problem in the rivers and causes marine pollution. For this problem to solve, several technologies are being researched and developed. Among them, this experiment is a study on Bench-scale STP based on internationally certified sewage treatment equipment by MEPC. 227(64) of IMO. The purpose of this study is to compare the biological treatment efficiency of phosphorus of Bench-scale STP. The ratio of C : N : P was set to 10 : 5 : 3 and 10 : 3 : 1 as the operating conditions. And the operation cycle was set to anoxic(mixed) 70 min - aeration 50 min (70-50), anoxic(mixed) 90 min - aeration 60 min (90-60). As a result, the phosphorous treatment efficiency was 88% at average, and the treatment efficiency was steady at 90-60 better than 70-50. The efficiency of the bench-scale STP has been verified through this experiment and additional experiments are required to derive the optimal operating conditions.

  • Blood-sensitive reagents may exhibit false positives or negatives under the influence of substances other than blood. Since these reactions lead to the misinterpretation of blood evidence, it is essential to investigate the possibility of false-positive and -negative reactions of blood-sensitive reagents. Acidic hydrogen peroxide (AHP) is a recently discovered blood-sensitive reagent, and it is not yet known whether it causes false-positive or -negative reactions. To confirm this, 20 µL of blood was placed on metal surfaces, plastic surfaces, paper surfaces, paint surfaces, foods, vegetable oils, detergents, and petroleum hydrocarbons, and then AHP was applied. The blood was observed through an orange filter under a 505-nm light source, and no false-positive or false-negative reactions were observed with any of the substances/materials. However, it was confirmed that polyethylene terephthalate surfaces, polyvinylchloride surfaces, some paint surfaces, and foods exhibit their own photoluminescence under the conditions of blood observation, which interferes with blood observation.

  • The purpose of this research is to improve sustainable clothes care by comparing household washer's standard course and quick course. Detergency at each course was classified by laundry weight, detergent concentration, and soils. Also, fabric damage from each course was compared. Washing experiments were carried out using two types of washing machines and three types of detergents. Using the standard soiled fabrics of EMPA 108 set, detergency was compared by laundry weight, soil, and detergent concentration. Additionally, fabric damage was evaluated using the mechanical action of MA-40. The results of the research were as follows. First, a standard course, having more working time exhibited better detergency than a quick course. However, the detergency deviation under 6kg laundry weight was as low as 9.0%. Second, detergency by the type of soil was more effective in standard course than in a quick course, but hydrophilic protein soils had a small detergency deviation at 7.6%. Moreover, hydrophobic oil, complex, and particulate soils had a higher deviation at 19.7% Third, fabric damage was in proportion to operating time. Fourth, a quick course showed approximately 80% detergency regardless of the type of detergent. in the case of using 50% of the recommended allowance by the detergent manufacturer. In conclusion, reducing the operating washing time and detergent concentration is in accordance with increasing sustainability, in the case of washing with lightly soiled fabrics under 6kg of laundry weight.

  • In this study, the efficacy of ultrasonic washing in cotton and wool fabrics was compared and evaluated against conventional washing in terms of cleaning properties and fabric deformation. Factors such as washing temperature, time, liquid ratio, and detergent concentration were kept varied, and the cleaning properties of sebum-soiled fabrics were assessed using different detergents such as alcohol ethoxylate, linear alkylbenzenesulfonate, and IEC 60456 Reference Detergent A*. In addition, the effects and emulsification power of enzymes and oxygen bleach were examined. To compare the cleaning properties with general washing, a launder-O-meter was used. To investigate fabric deformation during the washing process, the loosening test cloth, shrinkage test cloth, and mechanical strength test cloth were compared between ultrasonic washing machines and household drum washing machines. The results indicate that ultrasonic washing exhibits superior cleaning properties than launder-O-meter when the temperature is low and the washing time is short. Furthermore, there is less deformation and damage during the washing process. It was also observed that the activity of the detergent increases when ultrasonic waves are applied to the washing process. Considering the increasing tendency to pursue convenience and simplicity in clothing management as well as the anticipated commercialization of smart clothing with built-in electric circuits, ultrasonic laundry could serve as a new alternative to existing laundry methods.
