$CO_2$ 분리를 위한 PEGDA/PETEDA dendrimer 막의 제조 (Preparation of PEGDA/PETEDA Dendrimer Membranes for $CO_2$ Separation)

  • PEGDA/PETEDA dendrimer 막은 PEGDA에 5~15 wt%의 PETEDA dendrimer를 첨가하고 UV를 조사하여 제조하였다. 제조된 막의 특성은 FT-IR, $^1H$-NMR 그리고 DSC를 통해 분석하였다. PEGDA/PETEDA dendrimer 막의 유리전이온도($T_g$)는 PETEDA dendrimer의 함량이 증가할수록 감소하였다. $CH_4$에 대한 $CO_2$ 투과 특성은 PETEDA dendrimer의 함량과 압력을 변화시키며 조사하였다. 10 wt%의 PETEDA dendrimer를 포함하는 막의 투과도는 162.2 barrer였으나 $CO_2/CH_4$ 이상분리 인자는 31.8로 가장 높게 나타났다.

Streptomycin-anionic linear globular dendrimer G2: Novel antibacterial and anticancer agent

  • Recent researches demonstrated well promising anticancer activities for antibiotics. Such effects would be significantly increased while nanoparticle based delivery systems were applied. In this study, the goal was aim to improve anticancer and antitoxic effects of Streptomycin by loading on special kind of dendrimer (anionic-linear-globular second generation). In the current study, Size and zeta potential as well as AFM techniques have been used to prove the fact that the loading was performed correctly. The Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) of the drug loaded on dendrimer nanoparticle were determined and compared with both of dendrimer alone and free drug with respect to staphylococcus aureus as the test microorganism. The anticancer activity among three groups including Streptomycin, Streptomycin -G2 dendrimer, and control was measured in vitro. In vitro studies showed that G2 anionic linear-globular polyethylene-glycol-based dendrimer, which loaded on Streptomycin was able to significantly improve the treatment efficacy over clinical Streptomycin alone with respect to proliferation assay. Maximal inhibitory concentration (IC50) was calculated to be $257{\mu}g/mL$ for streptomycin alone and $55{\mu}g/mL$ for Streptomycin -G2 dendrimer. In addition, Streptomycin -G2 dendrimer conjugate prevented the growth of MCF-7 cancerous cells in addition to enhance the number of apoptotic and necrotic cells as demonstrated by an annexin V-fluorescein isothiocyanate assay. Streptomycin -G2 dendrimer conjugate was able to increase Bcl-2/Bax ratio in a large scale compared with the control group and Streptomycin alone. Based on results a new drug formulation based nano-particulate was improved against S. aureus with sustained release and enhanced antibacterial activity as well as anticancer activity shown for functional cancer treatment with low side effects.

Modulation of Electroosmotic Flow through Skin: Effect of Poly(Amidoamine) Dendrimers

  • The objective of this work is to evaluate the effect of polyamidoamine (PAMAM) dendrimers on electroosmotic flow (EOF) through skin. The effect of size and concentration of dendrimer was studied, using generation 1, 4 and 7 dendrimer (G1, G4 and G7, respectively). As a marker molecule for the direction and magnitude of EOF, a neutral molecule, acetoaminophen (AAP) was used. The visualization of dendrimer permeation into the current conducting pore (CCP) of skin was made using G4-fluorescein isothiocyanate (FITC) conjugate and confocal microscopy. Without dendrimer, anodal flux of AAP was much higher than cathodal or passive flux. When G1 dendrimer was added, anodal flux decreased, presumably due to the decrease in EOF by the association of G1 dendrimer with net negative charge in CCP. As the generation increased, larger decrease in anodal flux was observed, and the direction of EOF was reversed. Small amount of methanol used for the preparation of dendrimer solution also contributed to the decrease in anodal flux of AAP. Cross-sectional view perpendicular to the skin surface by confocal laser scanning microscope (CLSM) study showed that G4 dendrimer-FITC conjugate (G4-FITC) can penetrate into the viable epidermis and dermis under anodal current. The permeation route seemed to be localized on hair follicle region. These results suggest that PAMAM dendrimers can permeate into CCP and change the magnitude and direction of EOF. Overall, we obtained a better understanding on the mechanistic insights into the electroosmosis phenomena and its role on flux during iontophoresis.

Synthesis and Characterization of A Carbosilane Dendrimer Containing Allylic End Groups

  • A silane dendrimer with 48 allylic end groups has been synthesized in excellent yield, via repetitive alkenylation-hydrosilation cycles. Starting with hexaallylethylenedisilane ((CH2=CHCH2)3SiCH2)2 as the core molecule (G0), a succession of alternate platinum catalyzed hydrosilation (Pt/C, 10% platinum content on active carbon) of all allyl groups with methyldichlorosilane and the allylation of all SiCl2 groups with allylmagnesium bromide in THF provided 3rd generation (G3) as carbosilane dendrimer. All the generations of dendrimer have been characterized by 1H and 13C NMR spectroscopy as well as elemental analysis.

PAMAM Dendrimers Conjugated with L-Arginine and γ-Aminobutyric Acid as Novel Polymeric Gene Delivery Carriers

  • In this study, we synthesized functional dendrimer derivatives as nonviral gene delivery vectors. Poly(amidoamine) dendrimer (PAMAM, generation 4) was modified to possess functional amino acids to enhance gene transfection efficiency. PAMAM G4 derivatives conjugated with L-arginine (Arg) and ${\gamma}$-aminobutyric acid (GABA) showed higher transfection efficiency and lower cytotoxicity compared to the native PAMAM G4 dendrimer. The polyplex of the PAMAM G4 derivative/pDNA was evaluated using an agarose gel retardation assay and Picogreen reagent assay. Additionally, the MTT assay was performed to examine the cytotoxicity of synthesized polymers. All PAMAM G4 derivatives showed lower cytotoxicity than PEI25kD. Particularly, PAMAM G4-GABA-Arg displayed enhanced transfection efficiency compared to the native PAMAM G4 dendrimer.

  • Infection is one of the major mortality causes throughout the globe. Nuclear medicine plays an important role in diagnosis of deep infections such as osteomyelitis, arthritis infection, heart valve and heart prosthesis infections. Techniques such as labeled leukocytes are sensitive and selective for tracking the inflammations but they are not suitable for differentiating infection from inflammation. Anionic linear-globular dendrimer-G2 was synthesized then conjugation to gemifloxacin antibiotic. The structures were identified by FT-IR, 1H-NMR, C-NMR, LC-MS and DLS. The toxicity of gemifloxacin and dendrimer-gemifloxacin complex was compared by MTT test. Dendrimer-G2-gemifloxacin was labeled by Technetium-99m and its in-vitro stability and radiochemical purity were investigated. In-vivo biodistribution and SPECT imaging were studied in a rabbit model. Identify and verify the structure of the each object was confirmed by FT-IR, 1H-NMR, C-NMR and LC-MS, also, the size and charge of this compound were 128 nm and -3/68 mv respectively. MTT test showed less toxicity of the dendrimer-G2-gemifloxacin than free gemifluxacin (P < 0.001). Radiochemical yield was > %98. Human serum stability was 84% up to 24 h. Biodistribution study at 50 min, 24 and 48 h showed that the complex is significantly absorbed by the intestine and accumulation in the lungs and affects them, finally excreted through the kidneys, biodistribution results are consistent with results from full image means of SPECT/CT technique.

Sequential Conjugation of 6-Aminohexanoic Acids and L-Arginines to Poly(amidoamine) Dendrimer to Modify Hydrophobicity and Flexibility of the Polymeric Gene Carrier

  • We synthesized a novel cationic dendrimer consisting of a poly(amidoamine) dendrimer (PAMAM, generation 4) backbone with both L-arginine (Arg) at the termini and 6-aminohexanoic acid (Ahx) between the original core polymer and the peripheral Arg units. The sequential chemical modification of PAMAM G4 with Ahx and Arg resulted in higher transfection efficiency with much less cytotoxicity. PAMAM G4-Ahx-Arg formed stable polyplexes at weight ratios of 8:1 or higher (polymer: plasmid DNA), and the mean polyplex diameter was $180{\pm}20nm$. PAMAM G4-Ahx-Arg showed much higher transfection ability than PAMAM G4 or PAMAM G4-Ahx. Furthermore, PAMAM G4-Ahx-Arg was much less cytotoxic than PEI25KD and PAMAM G4-Arg. In addition to Arg grafting of the PAMAM dendrimer, which endows a higher transfection capability, the addition of Ahx spacer increased dendrimer hydrophobicity, introduced flexibility into the conjugated amino acids, and reduced cytotoxicity. Overall, it appears that the concomitant modification of PAMAM with Ahx and Arg could lead to new PAMAM conjugates with better performances.

  • Dendrimers are well-defined macromolecules exhibiting a tree-like structure, first derived by the cascade molecule approach. Peculiar features of the dendritic geometry are the large number of end groups as well as the shape persistence in higher generations, approaching spherical geometry. And one of the most peculiar characteristics of dendritic macromolecules is their controlled molecular structure and orientation, which means that they have a practical application in achieving a highly organized molecular arrangement. We attempted to fabricate a dendrimer LB films containing 48 pyridinepropanol functional end group. As the pyridinepropanol functional group could form a complex structure with metal ions. We investigated the surface activity of dendrimer films at air-water interface compared with pure dendrimer and its complex with $Fe^{2+}$ ions into subphase. We though that metal ions are contributed to networking or branching reaction between dendrimers. And we expected that it can result in the differences on the electrical properties. We have studied the electrical properties of the ultra thin dendrimer LB films investigated by the current-voltage characteristics of metal dendrimer LB films/metal (MIM) structure.

Balb/c 마우스에서 Keyhole limpet hemocyanine (KLH)의 항원성에 대한 PAMAM dendrimer 의 면역증강 효과 (Adjuvant Effect of PAMAM Dendrimer on the Antigenicity of Keyhole Limpet Hemocyanin in Balb/c Mice)

  • 본 연구에서는 Keyhole limpet hemocyanin (KLH)에 대한 체액성 및 세포성 면역반응 유도에 대한 PAMAM dendrimer G4 (PAMAM)의 증강 효과를 조사하였다. PAMAM을 KLH와 2주 간격으로 2회 피하주사로 면역한 후, KLH에 대한 특이항체를 측정한 결과, KLH+PAMAM 면역 그룹은 KLH만을 단독으로 면역한 그룹에 약 30배이상 높은 유의한 항체가(IgG+IgA+IgM) 상승을 나타냈다. ELISA 분석에 의해 KLH 특이적인 면역글로부린의 isotype을 측정한 결과, PAMAM를 혼합하여 면역함으로서 IgG1, IgG2a, IgG2b, IgG3 및 IgM 항체의 역가가 유의하게 증가하는 것으로 확인되었다. 또한 면역 개시 7주째에 면역동물에 KLH 항원을 피하주사하고 swelling reaction을 통해 세포성 면역반응인 지연형 과민반응(DTH)을 측정한 결과, KLH+PAMAM으로 면역한 그룹에서 KLH만을 단독으로 면역한 그룹에 비해 높은 DTH 유도활성이 관찰되었다. 한편 면역동물의 비장세포를 취하여 in vitro에서 KLH로 재자극한 후 림프구 증식반응과 사이토카인 유도활성을 측정한 결과, PAMAM을 혼합하여 면역한 그룹에서 KLH 단독 면역 그룹에 비해 림프구의 증식반응에 유의하게 증가하였으며, Th1 type (IFN-γ)과 Th2 type (IL-4) 사이토카인의 생성도 모두 상승하는 것으로 확인되었다. 이상의 결과로부터 PAMAM dendrimer는 함께 투여된 항원물질에 의해 유도되는 세포성 면역과 체액성 면역을 상승시키는 활성이 있는 것으로 확인되었으며, 이는 PAMAM dendrimer가 면역 adjuvant로서 응용 가능한 소재임을 입증하는 것이다.

G4-48PyP 덴드리틱 거대분자 박막의 금속이온 착체에 의한 전기적 특성 (Electrical Properties by Effect of Metal Complex of G4-48PyP Dendritic Macromolcules Thin Films)

  • We attempted to fabricate a dendrimer Langmuir-Blodgett(LB) films containing 48 pyridinepropanol functional end group. As the pyridinepropanol functional group could form a complex structure with metal ions. In this study the samples for electrical measurement were fabricated to two types metal complexes with $Pt^{4+}$ and $Fe^{2+}$ ions by LB method. And we have investigated the surface activity at the air-water interface as well as the electrical properties for the monolayers of pure G4-48PyP dendrimer and its complex with metal ions($Pt^{4+}$ and $Fe^{2+}$ ions). In the surface pressure-area($\pi-A$) isotherms of the dendrimers, the stable condensed films formed at the air-water interface and the metal ions effect showed the difference on molecular behavior. We have studied the electrical properties of the ultra thin dendrimer LB films investigated by the current-voltage(I-V) characteristics of metal/dendrimer LB films/metal(MIM) structure. In conclusion, it is demonstrated that the metal ion around G4-48PyP dendrimer can contribute to make formation of network structure among dendrimers and it result from the change of electrical properties.
