This experiment was conducted to investigate the mass propagation system of Aster glehni FR. and to obtain the basic data for improvement of germination and seedling. The following was the results of experimentation about temperature and light conditions, and priming treatment as kinds of chemicals, their concentration and treated periods affect germination of Aster glehni FR. The germination percentage of Aster glehni FR. seed was higher in 20 and 25$^{\circ}C$ than the others, but it dropped rapidly at 30$^{\circ}C$ and didn't germinate at 35$^{\circ}C$. The first day to germination was the slowest at 15$^{\circ}C$. The germination rate of Aster glehni FR seed increased with increasing with temperatures from 15 to 25$^{\circ}C$. But the seed was rotten easily in high temperature. The germination rate was shown highest in 25$^{\circ}C$, and next was 15, 20, 30$^{\circ}C$ in order. Light treatment enhanced germination percentage, the first day to germination, germination rate and T50, but there was no significant difference. The 3 hours priming treatments had more effect on germination percentage than 30 minutes treatments as comparing averages. Aster glehni FR seeds primed in $KNO_3+K_3PO_4$ for 3 hours had most eHective on germination percentage (83.3%) and also showed shortest $T_{50{\cdot}}T_{50}$ and day of first germination was better in 30 minutes than 3 hours treatments, and most of priming treatments were better than control(non-priming seeds). While priming seeds showed shorter day of first gemination than control, there was no significant difference between 30 minutes and 3 hours treatments.
The purpose of this study is to investigate changes of isoflavone and nutrients during germination of soybean varieties and to determine the optimal germination condition for soybean processing. After 5 days' germination, water content of the soybeans increased in the order of Poongsannamoolkong> Sinpaldal#2> Black#1> Jinpumkong. Ash content was higher in the order of Poongsannamoolkong> Sinpaldal#2> Jinpumkong> black#1 soybean. All varieties of the soybean samples increased in ash content during germination. Crude fat content was higher in the order of Poongsannamoolkong> Black#1> Jinpumkong> Shinpaldalkong. Crude protein content was higher in the order of Shinpaldalkong> Black#1> Jinpumkong> Poongsannamoolkong. Polypeptide subunits separated from the SDS-PAGE of the entire proteins were showing comparatively high level in the range of 2-16KD in Jinpumkong and 2-21KD in Poongsannamoolkong till the first day of germination, 2-25KD in Sinpaldal#2 till the second day, and 6-27KD in Black#l till the third day. Changes of total isoflavone contents during germination showed different patterns in soybean varieties: Jinpumkong and Black#1 had the highest total isoflavone content on the fourth day of germination, Poongsannamoolkong and Sinpaldal#2 on the first and the second day of germination, respectively.
Diospyros mespiliformis is a highly valued and threatened tree species within the Sahelo-Sudanian zone of Africa, but its seed germination requirements under cultivation are not well researched. In a first experiment which aimed at determining germination response of seeds to dehydration, fresh seeds were dried at room temperature for 26 days during which their moisture content, their germinability, and their viability were monitored at two-day intervals. In the second experiment, 14 pre-germination treatments were tested for their effect on the germination of dried seeds. Results showed that fresh seeds had 52.7% moisture and achieved 97.7% germination. As seeds were dried, percentage germination gradually decreased with decreasing moisture content and reached 0% when moisture content had dropped to 18%. Meanwhile, seed viability remained at 100% over drying duration. Seeds that were not germinated after air dry also recorded 100% viability. The most effective treatment for inducing germination of dried seeds was scarification using 98% sulfuric acid for 30 min which resulted in 96.6% germination. This study reports for the first time in D. mespiliformis seeds a desiccation-induced dormancy which can be efficiently alleviated by acid scarification. This study provides useful information that will contribute to efficient management of D. mespiliformis seed resources for propagation.
This study was conducted to use fundamental data about functional foods based on pigmented rice varieties-2 types of normal rice, 6 types of black rice-to analyze changes in chemical component activity during germination. The results are summarized as follows: Germination rates of the pigmented rice varieties were the highest at $30^{\circ}C$. There was a tendency for crude protein contents to increase under the influence of germination periods, and the crude protein contents were the highest on the $6^{th}$ day. Crude fat contents were the highest on days 6-8. Nevertheless, the crude fat contents in pigmented rice decreased during the first 2 days, and subsequently, the contents increased steadily. Crude fiber contents were the highest on the $8^{th}$ day. However, there was no meaningful tendency. Sucrose, glucose, and fructose contents in the pigmented rice increased during germination. In normal rice, glucose content increased up to the $8^{th}$ day. It was higher than that of brown rice. However, sucrose and fructose contents showed no meaningful tendency. There was a general tendency for GABA, alanine, glutamic acid, asparagine, aspartic acid, and lysine contents in the pigmented rice to increase under the influence of germination periods. Aspartic acid and asparagine contents were shown to decrease, compared with that in pre-germinated normal rice. In all varieties, lysine was detected from the second germination day onwards, and on the $6^{th}$ day after germination, its contents were 6.24 mg/100 g and 15.58 mg/100 g in normal rice and pigmented rice, respectively.
The fate of Telfairia occidentalis seed Agglutinin, TOA, has been monitored during germination. While the level of the Agglutinin in the cotyledons decreased sharply in the first three days to about half of the initial level, it stabilises at this level for the following twelve days. In this interval, Agglutinin activity becomes manifest in the radicle on the fourth day, peaking on the fifth and decreasing rapidly thereafter. In the plumule, the lectin activity becomes manifest on the sixth day, peaks on the seventh and decreases rapidly thereafter. No lectin activity is detectable in any plant tissue including the rump of the cotyledons twenty-seven days after germination. The implications of these observations on the possible role of lectins in plants are discussed.
Ten strains of the entomopathogenic fungi Metarhizium anisopliae and Beauveria bassiana were evaluated to find the most effective strain for optimization studies. The first criterion tested for strain selection was the mortality (> 50%) of Spodoptera litura larvae after inoculation of the fungus for 4 days. Results on several bioassays revealed that B. bassiana BNBCRC showed the most virulence on mortality S. litura larvae (80% mortality). B. bassiana BNBCRC also showed the highest germination rate (72.22%). However, its conidia yield ($7.2{\times}10^8$ conidia/mL) was lower than those of B. bassiana B 14841 ($8.3{\times}10^8$ conidia/mL) and M. anisopliae M6 ($8.2{\times}10^8$ conidia/mL). The highest accumulative radial growth was obtained from the strain B14841 (37.10 mm/day) while the strain BNBCRC showed moderate radial growth (24.40 mm/day). M. anisopliae M6 possessed the highest protease activity (145.00 mU/mL) while M. anisopliae M8 possessed the highest chitinase activity (20.00 mU/mL) during 96~144 hr cultivation. Amongst these criteria, selection based on virulence and germination rate lead to the selection of B. bassiana BNBCRC. B. bassiana B14841 would be selected if based on growth rate while M. anisopliae M6 and M8 possessed the highest enzyme activities.

This study was conducted to investigate the germination characteristics through the temperature change treatment compared to the general room temperature immersion treatment of the native species, the Larix kaempferi, and the Berula platyphyllavar. japonica seeds, and to obtain basic data for the production of healthy seedlings with high productivity and efficiency. As a result, the germination rate of the larch seeds showed a significant difference according to room temperature and temperature immersion treatment, and the average germination days of the birch seeds showed a significant difference according to room temperature immersion and temperature immersion treatment. On the other hand, there was no significant difference in germinal uniformity. The results of the study are as follows. First, the results of the analysis of the fallen leaves were the highest germination rate in the treatment of 1.5 days of hot water and 1.5 days of cold water, the fastest germination rate, the lowest average germination day, and the highest germination uniformity. This is the best result in most items, and the change temperature immersion treatment of hot and cold baths was superior to the conventional room temperature immersion treatment. Second, the results of birch tree showed that the germination rate was the highest in 1.5 days of hot water and 1.5 days of cold water. The average germination day is 1.5 days of cold water 1.5 days of hot water 1.5 days, but the difference between 1.5 days of hot water and 1.5 days of cold water is 0.01 days. The highest germinated uniformity was found in 1.5 days of cold water and 1.5 days of hot water. As a result, in the case of larch and birch, the seed temperature immersion (1.5 days of hot water, 1.5 days of cold water) treatment is superior to the existing three-day immersion treatment in various analysis methods, and it is expected that productivity and efficiency can be improved at a low cost in a short period of time at the seedling production site through seeds.
Mobilization of storage lipids is critical for the germination of oil seeds, as they supply carbon and energy until photosynthesis commences in cotyledons. In this study, we determined the levels of plant carnitine and associated changes in these levels from seed germination to cotyledon senescence. We also examined changes in the content of unsaturated fatty acids throughout seedling development. Carnitine levels peaked on day 3 at 14.5 nM in cotyledons and decreased sharply to 7.2 nM on day 4. On development day 3 carnitine levels were maintained at around 3 nM until day 7. The unsaturated fatty acid content dropped by half at the same time as carnitine peaked (day-3), and storage lipids were almost depleted by day 5. Thereafter, carnitine was hardly detected until the second stage of cotyledon senescence, at which stage the carnitine content was 6.8 nM, similar to that on day 4 at the time of fatty acid depletion in the cotyledons. Unsaturated fatty acids levels remained constant in green cotyledons but slightly increased in the senescing cotyledons. The latter can be explained by intracellular breakdown of membrane lipids. This is the first such discovery in developing cotyledons and may offer clues regarding other roles of the acetyl unit transport system in plants. The expression of BOU was closely associated with carnitine metabolism during seed germination and cotyledon development. The results provide support for the possibility of carbon re-routing during the glyoxylate cycle in the supply of energy for early germination and development.
An electron beam or gamma-rays(0-5 kGy) were applied to two popular commercial sprout seeds, those of radish and red cabbage, and both physiological and microbiological qualities during germination were examined. Total bacterial counts of radish and red cabbage seeds were 5.02 and 2.86 log CFU/g, respectively, and were reduced by 2-3 logs after irradiation, but nonirradiated and irradiated samples showed similar microbial populations on day 5 after germination, although day 3 levels of irradiated samples were $23{\pm}2%$ lower. Coliforms were absent in both seed types. However, they were detected from the first day of germination, and rose to 4-5 log CFU/g on the fifth day. Coliforms were reduced by about 1-2 log CFU/g in 1 kGy-irradiated samples and were absent in the samples irradiated at 3 kGy. Irradiation at 1 and 3 kGy decreased germination by about 7% and 18%, respectively. Sprout yield and length were also significantly affected by irradiation, with no apparent difference between samples treated with the electron beam and gamma-rays.
Research was initiated to investigate early establishment characteristics and germination pattern of fine-textured fescues (FF). Six varieties from Chewings fescue ($Festuca$$rubra$ L. ssp. $commutata$ Gaud., CF), creeping red fescue ($F.$$rubra$ L. ssp. $rubra$ Gaud., CRF), hard fescue ($F.$$ovina$ ssp. $longifolia$ Thuill., HF) and sheep fescue ($F.$$ovina$ L., SF) were evaluated in the study. An alternative environmental condition requiring a FF germination test by International Seed Testing Association (ISTA) was applied in the experiment, consisting of 8-hr light at $25^{\circ}C$ and 16-hr dark at $15^{\circ}C$ (ISTA conditions). Daily and cumulative germination patterns were measured and analyzed on a daily basis. Significant differences were observed in germination pattern, days to the first germination, days to 50% germination, days to 60% germination, and germination rate. The final germination percentage was variable with species and varieties, being 40.25 to 82.00% at the end of study. There were considerable variations in early germination characteristics and germination pattern among FF species. The first germination in all entries except HF was initiated between 5 and 6 DAS (days after seeding) under ISTA conditions, while HF between 6 and 7 DAS, being 1 day later. It was 8 to 10 DAS in days to the 50% germination, which was 2 to 4 days after the first germination date. Days to the 60% germination were 9.10 to 14.80 DAS under ISTA conditions, being 5.70 days in differences among the entries. CF 'Jamestown II' and 'Shadow II' and HF 'Aurora Gold' were the fast varieties. The slowest one was HF 'Rescue 911'. Among FF species, turf establishment speed was becoming faster in CRF, SF, HF and CF in this order. Information on differences in germination characteristics and pattern from this study would be usefully applied for golf course design and construction, when established with FF.
