Variation of Growth and Allylisothiocyanate contents of Wasabia japonica Matsum. Cultivar (고추냉이 품종별 생육 및 Allylisothiocyanate 함량 변이)

  • This study was conducted to select the optimum cultivar of wasabi in water condition. In three cultivar, Daruma, Simanesarai and Daihoichigo, the Daruma was superior to the others. Though total rhizome weight was similar to Daruma and Daihoichigo, rhizome of main stem of Daruma was bigger than that of Daihoichigo, specially distribute in $80{\sim}159g$ size. The allylisothiocyanate content was higher in rhizome than that of leaf, lateral rhizome and petiole. In three cultivar, the allylisothiocyanate content of rhizome, Daihoichigo, Daruma and Simanesairai was 0.3389, 0.6332, 0.3956(mg/g), respectively.

Study on Reduction of Microbial Contamination on Daruma by Combination Treatment of Strong Acidic Hypochlorous Water and Ultrasonic Waves (강산성차아염소산수와 초음파를 병용처리한 조미오징어 반가공품의 미생물 오염도 저감화에 관한 연구)

  • This study was performed to develop treatment method for reducing microbial contamination on Daruma (a semi-processed product of seasoned and dried squid) by combination of strong acidic hypochlorous water (SAHW) and ultrasonic waves (UW). The available chlorine concentration, oxidation reduction potential (ORP) and pH of SAHW were $69.67{\pm}0.58ppm$, $1071.33{\pm}4.16mV$ and 2.79, respectively. The 1.49 log CFU/g of viable cell count and 1.32 log CFU/g of Staphylococcus aureus was reduced, and Escherichia coli was reduced below detection limit when the Daruma was treated with 20 times (w/v) of sodium hypochlorite solution (SHS) for 120 min. The 3.62 log CFU/g of viable cell count and 3.22 log CFU/g of Staphylococcus aureus was reduced, and Escherichia coli was reduced below detection limit when the Daruma was treated with 20 times (w/v) of SAHW for 120 min. The antibacterial effects of SAHW were stronger than those of SHS at same available chroline concentration. SAHW treatment after washing strongly alkalic electrolyzed water (SAEW) showed better bactericidal effects than SAHW treatment only. The 4.0 log CFU/g of viable cell count was reduced, S. aureus was reduced below regulation limit (Log 2.0 CFU/g), and E. coli was reduced below detection limit when the Daruma was treated with 20 times (w/v) of SAHW for 90 min after washing with 20 times (w/v) of SAEW for 60 min. The viable cell number was reduced below detection limit and S. aureus was reduced below regulation limit when the Daruma was treated with 20 times (w/v) of SAHW for 60 min in ultrasonic washer. E. coli was reduced below detection limit when the Daruma was treated with 20 times (w/v) of SAHW for 10 min in ultrasonic washer. These results suggest that combination of SAHW and UW may be a good technique to reduce the microbial contamination in daruma.

  • Field experiment was conducted to find out the chemical characteristics of 12 Korean local varities, Local varities were compared with oriental, flue-cured and burley tobacco. Among tested varities, Hyangcho and Matskawa were desirable in respect of flavor, leaf color and texture. Hyangcho and Matskawa were higher in the content of nicotine and petroleum ether extract. Gajacho, Byeolcho and Kukjuo were higher in the content of reducing sugar, but lower nicotine, total nitrogen and petroleum ether extract. The nicotine and total nitrogen content of Kwangcho, Daruma and Catterton were lower than that of Br. 21. In the content of volatile organic acid which were major characteristic component of oriental tobacco, the Korean local varities were lower in the content of 3-methyl butanoic and 3-methyl pentanoic acid, but hirities, Hyangcho, Matskawa and Daruma were higher in the content of volatile organic acid. Korean local varities, especially small leaf group, were higher in the content of benzyl alcohol and furfural than any other type tobacco.

Microbial Contamination of Seasoned and Dried Squid Dosidicus gigas during Processing (조미오징어(Dosidicus gigas)의 가공 공정 중 미생물 오염도 및 오염원에 관한 연구)

  • This study examined microbial contamination during seasoned and dried squid Dosidicus gigas processing, including the apparatus, machines, and employee's gloves at each step in processing at two companies. The numbers of bacteria floating in air in each processing area were also examined. The numbers of Staphylococcus aureus (3.6-6.0 log CFU/g) and Escherichia coli (1.3-1.4 log MPN/100 g) in domestic and imported daruma (a semi-processed product of seasoned and dried squid) at companies A and B exceeded the regulatory limits of the Food Sanitary Law of Korea (S. aureus, ${\leq}2.0$ log CFU/g; E. coli, negative). S. aureus in both daruma was reduced to below the detection limit or 3.6 log CFU/g after the roasting step, but increased again to 3.3 and 5.5 log CFU/g after the mechanical tearing step at companies A and B, respectively. E. coli showed similar tendencies at both companies. The surfaces of the apparatus, machines, and employee's gloves that contacted daruma were also contaminated with S. aureus (1.0-5.5 log CFU/$m^2$) and E. coli (negative-to 3.5 log MPN/$m^2$). The numbers of bacteria floating in air were high (1.7-5.1 log CFU/$m^3$) at both companies. These results suggest that sanitation standard operating procedures (SSOP) must be developed to control of microbial cintamination in seasoned and dried squid.

Growth and Yield of Wasabi Cultivated in Cold Water Drained from Trout Nursery (송어양식장 배출냉수 이용 재배시 고추냉이의 생육 및 수량)

  • We investigated rhizome yield and growth characteristics of wasabi variety, Daioichigo and Daruma cultured by flowing water from Trout nersery in Pyong Chang. Average temp. of air and water during a year was 14.3, 12.3$^{\circ}C$, respectively in PE film house. Wasabi growth was good and healthy in May and October. Rhizome weight of main stem in Daioichigo was 78.78g per plant and 608kg per l0a, while those of the Daruma was 37.1g per plant and 287kg per l0a. Rate of marketable rhizome was 90% in Daioichigo and 37.5% in Darurna. Rhizome weight of main stem in Daioichigo was increased, up to 21% of the control by the remove of floral axis with two times a year.

Intraspecific Genetic Relation of Wasabia japonica Matsum. Based on RAPD Analysis (RAPD를 이용한 고추냉이의 유연관계 분석)

  • The genetic variation and intraspecific relationships between 10 individuals of seven cultivars and one Ulleungdo native of Wasabia japonica were investigated using RAPD (Randomly Amplified Polymorphic DNA) analysis. The 21 primers out of 50 random primers were amplified for all tested plants. The 68 (47.2%) among 144 bands derived from 21 primers showed polymorphism, and 3.2 bands per primer were observed. Number of bands per primer was ranged from 2 to 13, and average numbers were 6.8. The phenograms for 11 analyzed individuals by RAPD markers were not matched well with those of the result by morphological characters since they were clustered monophyletic at the similarity coefficient value ranged from 0.81 to 0.96. The Ulleungdo native individual was clustered sister to Daruma, Simanesairal, Sawa, and Hujidaruma cultivars. The RAPD markers were not useful to evaluate the intraspecific variations in Wasabia japonica cultivars, therefore need to more specific molecular phylogenetic characters such as AFLP technology and gene sequence of nuclear and chloroplast DNA.

Flowering Characteristics and Optimal Harvest Time in Wasabia japonica Mastum (고추냉이의 개화(開花) 특성(特性) 및 채종적기(採種適期))

  • Flowering characteristics and optimal harvest time for seed of wasabi variety. Daruma, were investigated from Dec. 1995 to June 1996 in film house of Suwon. The number of peduncle was $10.2{\pm}2.29$ and most of the peduncle emerged between Jan. 20 and Feb. 10. Average peduncle length was $124{\pm}29.60cm$ and the peduncle grew most rapidly in the middle of march. Flowering started on Jan. 27 and lasted until June 4 with a peak at march 21 and flowering period was $99{\pm}21.32$ days. Flowers which bloomed later than the middle of March set seed but those bloomed earlier became sterile because days with minimum temperature below freezing occurred until the middle of March. Maturing period was $37.5{\pm}2.60$ days and total seed set was $43.2{\pm}8.77%$ in the natural culture condition. Optimal harvest time for seed was the last ten days of May and seed yield per $m^2$ was $79.0{\pm}19.94g%$(11,177 seeds).
