Effects of free alkali and moisture on sucrose polyesters (SPE)-possible non calorie fat substitute-synthesis were investigated using a model system composed of sodium oleate, sucrose, potassium carbonate and methyl oleate. Trace amounts of free alkali in sodium oleate were found to interefere with SPE synthesis. When free alkali content in sodium oleate was varied gradually from 0% to 5%(w/w), the yield of SPE production was reduced from 92% to 45.5%. The moisture absorbed in sodium oleate, sucrose and potassium carbonate during storage also interefered with SPE synthesis. The yield (92%) of SPE production with dried ($105^{\circ}C$.6 hrs) reactants and catalysts was higher than that (89%) of SPE production with non-dried. Soybean oil fatty acid sodium soaps (FASS) not containing free alkali could be manufactured with slightly less than molar ratio of sodium hydroxide to soybean oil fatty acid methyl esters (FAME). Practically, 91.7% yield of soybean oil SPE production was outcomed by minimizing free alkali and moisture which were remaining in sucrose, potassium carbonate, soybean oil FASS and soybean oil FAME.

The function of genes related to spectinomycin biosynthesis (spcD, speA, speB, spcS2) from Streptomyces spectabilis ATCC 27741, a spectinomycin producer, was analyzed. Each gene was subcloned from a spectinomycin biosynthetic gene cluster and overexpressed in E. coli BL21 (DE3) using pET vector. After incubating each purified protein with its possible substrates, the final products were analyzed using high-performance liquid chromatography (HPLC). From these results, spcD, speA, and speB have been identified to be dTDP-glucose synthase, myo-inositol monophosphatase, and myo-inositol dehydrogenase, respectively. In addition, the results suggest that the spcS2 gene product functions downstream of the speB gene product in the biosynthetic pathway of spectinomycin. Taken together, the present study elucidates the early steps of the biosynthetic pathway for 6-deoxyhexose (6-DOH) part (actinospectose) and aminocyclitol part (actinamine) of spectinomycin.

We have performed Liquid Chromatography-Solid Phase Extraction-NMR (LC-SPE-NMR) analysis for liquid crystalline mixture and elucidated the structures of selected components by NMR spectra. Combining the results of one-dimensional 1H experiments as well as homonuclear and heteronuclear two-dimensional experiments, we could analyze the molecular structure of the liquid crystal singles whose structure had not been interpretable by mass spectrometry alone.

SpeROD, the optical design program. is developed far specular reflectors of fluorescent luminaries. SpeROD can save much tirre when to design specular reflectors. SpeROD calculates conditions 2-dimensionally. The conditions for designing reflectors considered in this program are the intensity distribution, reflector type, and Tangent Spiral Design method. Using SpeROD, uniform intensity distribution type and bat-wing type reflectors are simulated in this paper, and alrrnn(T8) diarreter fluorescent lamp and lmnm width reflector are used for the setting. The sarre setting was simulated in FIElD, the intensity distribution prediction program. and the' result represented the intensity distribution and the efficacy simulated in SpeROD were satisfactory.actory.

The aim of this study was to optimize a simple, fast, and economic analytical method for the simultaneous determination of various pesticides (aldrin, p,p'-DDT, o,p'-DDT, p,p'-DDE, ${\alpha}$-endosulfan, ${\beta}$-endosulfan, dieldrin, heptachlor, permethrin, chlordane, deltamethrin, diazinon, bifenthrin, methoprene, propargite, fenpropathrin, cypermethrin, fenvalerate, and fenpropathrin) in milk by using dispersive solid phase extraction (SPE). In this study, two different extraction methods (low temperature cleanup and liquid-liquid partitioning), which were followed by a cleanup process based on dispersive-SPE, were evaluated and compared for the 19 pesticides. The results for all the pesticides were determined by gas chromatography mass spectrometry (GC/MS) with selected-ion monitoring mode, and the matrix effect of the method was evaluated. Comparison of these approaches yielded higher and more consistent recoveries of most pesticides at fortification levels of $1{\mu}g/mL$ using low-temperature fat precipitation, followed by cleanup process based on dispersive-SPE with PSA and C18 as sorbents, than other preparation process. The relative standard deviation was <20 % and the combination of this method were very effective for the cleanup.
Here we report on the first results of sensitivity evaluation of the gadolinium-aluminum-gallium- garnet (GAGG) scintillation detector with SiPM readout to fast and slow neutrons and, to the natural background and Co-60 γ-radiation as well. Data on sensitivity were obtained using certified dosimetry benches, so it can be utilized in the calculation of detection limits of neutron flux with such type of detectors. It was concluded that use of GAGG scintillator has a good prospect for neutron monitoring in different parts of nuclear research reactors and power plants.

Xanthium sibiricum is used as a traditional folk medicine for the treatment of cancer, fever, headache, nasal sinusitis, and skin pruritus. This study aimed to identify components from Xanthium sibiricum extracts using an SPE-800MHz NMR-MS hyphenated system. The simultaneous acquisition of MS and NMR spectra from the same chromatographic peaks significantly increases the depth of information acquired for the compound and allows the elucidation of structures that would not be possible using MS or NMR data alone. LC -NMR analysis was conducted using a HPLC separation system coupled to 800 MHz spectrometer equipped with a cryoprobe, and a SPE unit was used to automatically trap chromatographic peaks using a HPLC pump. LC-MS analysis was conducted with a Q-TOF MS instrument using ESI ionization in the negative ion mode. Using the hyphenated analysis, several secondary metabolites were identified, such as 3',5'-O-dicaffeoylquinic acid, 1',5'-O-dicaffeoyl- quinic acid, and ethyl caffeate. These results demonstrate that the SPE-800MHz NMR-MS hyphenated system can be used to identify metabolites within natural products that have complex mixtures.

The chitinase producing Lysobacter enzymogenes C-3 has previously been shown to suppress plant pathogens in vitro and in the field, but little is known of the regulation of chitinase production, or its role in antimicrobial activity and biocontrol. In this study, we isolated and characterized chitinase-defective mutants by screening the transposon mutants of L. enzymogenes C-3. These mutations disrupted genes involved in diverse functions: glucose-galactose transpoter (gluP), disulfide bond formation protein B (dsbB), Clp protease (clp), and polyamine synthase (speD). The chitinase production of the SpeD mutant was restored by the addition of exogenous spermidine or spermine to the bacterial cultures. The speD and clp mutants lost in vitro antifungal activities against plant fungal pathogens. However, the gluP and dsbB mutants showed similar antifungal activities to that of the wild-type. The growth of the mutants in nutrient rich conditions containing chitin was similar with that of the wild-type. However, growth of the speD and gluP mutants was defective in chitin minimal medium, but was observed no growth retardation in the clp and dsbB mutant on chitin minimal medium. In this study, we identified the four genes might be involved and play different role in the production of extracellular chitinase and antifungal activity in L. enzymogenes C-3.
In this paper, durability evaluation and surface damage mechanism were investigated through solid particle erosion (SPE) test after applying hot-dip aluminizing (HDA) technology for the purpose of maintenance of marine economizer tube. Damaged surface shape was analyzed using SEM and 3D microscope. Compositional changes and microstructure of the HDA layer were analyzed through EDS and XRD. Durability was evaluated by analyzing weight loss and surface damage depth after SPE. HDA was confirmed to have a two-layer structure of Al and Al5Fe2. HDA+HT was made into a single alloy layer of Al5Fe2 by diffusion treatment. In the microstructure of HDA+HT, void and crack defect were induced during the crystal phase transformation process. The SPE damage mechanism depends on material properties. Plastic deformation occurred in the substrate and HDA due to ductility, whereas weight loss due to brittleness occurred significantly in HDA+HT. As a result, the substrate and HDA showed better SPE resistance than HDA+HT.
Composite scintillation screens on a base of Gd1.2Y1.8Ga2.5Al2.5O12:Ce (GYAGG) scintillator have been evaluated for neutron detection. Besides the powdered scintillator, the composite includes 6LiF particles; both are merged with a binder and deposited onto the light-reflecting aluminum substrate. Results obtained demonstrates that screens are suitable for use with a silicon photomultiplier readout to create a prospective solution for a compact and low-cost thermal neutron sensor. Composite GYAGG/6LiF scintillation screen shows a pretty matched sensitivity and γ-background rejection with a widely used ZnS/6LiF screens however, possesses forty times faster response.
