본 연구는 고종시 감나무 부위별 수용성 추출물의 항산화 및 항염증에 대한 효과를 조사하였다. 감꼭지와 감잎은 DPPH, ABTS 및 NO 등 라디칼을 효과적으로 제거하는 활성뿐만 아니라 환원력도 우수한 항산화 활성을 보였다. 또한 LPS에 의해 활성화된 설치류 유래 대식세포주인 RAW 264.7 세포에서 NO 생성억제는 감꼭지 추출물이 가장 우수하였지만, $PGE_2$ 생성의 경우 모든 부위의 추출물에서 탁월한 억제 효과가 있었다. 더욱이 활성화된 RAW 264.7 세포에서 감꼭지, 감잎 및 감껍질 추출물은 농도에 의존적으로 TNF-${\alpha}$와 IL-$1{\beta}$를 효과적으로 억제하였다. 이러한 결과는 고종시 감나무 부위별 추출물 중 감꼭지와 감잎은 항산화제뿐만 아니라 항염증에 효과적인 물질이라는 것을 제시해주었고, 감껍질의 경우 비록 항산화 효과는 낮았지만, 우수한 항염효과를 나타내는 물질임을 제시해주었다. 그러므로 고종시 감나무 수용성 추출물은 항산화와 항염 기능을 발휘할 수 있는 식품으로 활용될 수 있을 것이라 사료된다.
Viola tianshanica Maxim, belonging to the Violaceae plant family, is traditionally used in Uighur medicine for treating pneumonia, headache, and fever. There is, however, a lack of basic understanding of its pharmacological activities. This study was designed to observe the effects of the ethanol extract (TSM) from Viola tianshanica Maxim on the inflammation response in acute lung injury (ALI) induced by LPS and the possible underlying mechanisms. We found that TSM (200 and 500 mg/kg) significantly decreased inflammatory cytokine production and the number of inflammatory cells, including macrophages and neutrophils, in bronchoalveolar lavage fluid. TSM also markedly inhibited the lung wet-to-dry ratio and alleviated pathological changes in lung tissues. In vitro, after TSM ($12.5-100{\mu}g/ml$) treatment to RAW 264.7 cells for 1 h, LPS ($1{\mu}g/ml$) was added and the cells were further incubated for 24 h. TSM dose-dependently inhibited the levels of proinflammatory cytokines, such as NO, $PGE_2$, $TNF-{\alpha}$, IL-6, and $IL-1{\beta}$, and remarkably decreased the protein and mRNA expression of $TNF-{\alpha}$ and IL-6 in LPS-stimulated RAW 264.7 cells. TSM also suppressed protein expression of $p-I{\kappa}Ba$ and p-ERK1/2 and blocked nuclear translocation of $NF-{\kappa}B$ p65. The results indicate that TSM exerts anti-inflammatory effects related with inhibition on $NF-{\kappa}B$ and MAPK (p-ERK1/2) signaling pathways. In conclusion, our data demonstrate that TSM might be a potential agent for the treatment of ALI.
Objectives Hataedock method is a Korean medical therapy which removes fetal toxin by orally administering herbal decoction to neonates. This study was to observe skin damage and anti-inflammatory effect via regulating IL-4 activity in NC/Nga mice which were induced atopic dermatitis (AD)-like skin lesion by Dermatophagoides (D.) farinae after applying Douchi Hataedock method. Methods NC/Nga mice with 3 weeks of gestational age were used. Each 10 mice were allocated to the control group (Ctrl), the AD-induced group (AE), and the group which induced AD after administering Douchi extract (GT). After 4 weeks from administering Douchi extract to the mice, the primary AD was induced by applying D. farinae extract 6 times per week for 3 weeks and then the secondary AD was induced by the same method after 1 week from the primary AD induction. To identify the skin damage and anti-inflammatory effect, we observed LxR, IL-4, Fc ${\varepsilon}$ receptor, substance P, and $NF-{\kappa}B$. Results The GT group showed alleviation of skin injury and decrease in capillary angiogenesis. Stratum corneum damage, epithelial cell hyperplasia, lymphocyte infiltration, and capillary distribution relatively decreased in the GT group. LxR-positive reaction in the GT group were increased by 53% than that of the AE group. IL-4 production, $Fc{\varepsilon}$ receptor activity, and substance P-positive reaction in the GT group were decreased by 82%, 42%, and 82% respectively compare to those of the AE group. $NF-{\kappa}B$-positive reaction in the GT group were decreased by 15% compare to that of the AE group. Conclusions Hataedock method with Douchi extract alleviated AD via reducing inflammatory cytokines secreted at the early stage of AD. Thus, Douchi Hataedock method has a beneficial effect for the prevention and treatment of AD.
본 동물실험은 STZ로 유도한 C57BL/6에게 5% sodium butyrate를 급여했을 때 항당뇨 및 항염증 효과를 연구하고자 하였다. 본 연구에서 STZ로 당뇨를 유발한 마우스에게 5% sodium butyrate를 급여했을 때 체중과 식이섭취량에서는 크게 유의적 차이가 없음을 확인하였다(p<0.05). STZ에 의한 당뇨 쥐는 인슐린의 분비가 감소되면서 당대사의 불균형을 초래하며 간 등이 비대해진다고 알려져 있으나, 본 연구에서는 간의 장기 무게에서는 크게 실험군 간에 유의적인 차이가 없었다(p<0.05). 또한 비장과 흉선의 무게는 0.5% sodium butyrate 첨가 식이군에서 유의적으로 낮아짐을 알 수 있었다(p<0.05). 당뇨병은 염증 상태로서 고혈당으로 인하여 monocyte에서는 여러 염증성 사이토카인이 분비가 활성화된다. TNF-${\alpha}$, IL-6 등은 염증성 사이토카인으로서 혈관염증의 중요한 마커로 인식되고 있고, 당뇨병 환자들은 이러한 염증성 사이토카인이 높은 수준으로 활성화 된다. STZ 처리 시 마우스 혈청에서의 염증성 사이토카인의 분비 및 발현이 증가되었으나, 5% sodium butyrate를 급여했을 때 염증성 사이토카인의 분비 및 발현이 저해됨을 확인할 수 있었다. 본 연구는 sodium butyrate 보충은 당뇨병이 유발된 동물모델에서 혈청지질 농도 및 혈당 조절, 염증 상태를 개선에 다소간의 효과가 있는 것으로 나타났다. 이에 따라 당뇨병과 같은 만성적인 대사질환 개선에 sodium butyrate가 효과적인 식이인자가 될 것으로 생각된다. 그러나 앞으로 더 명확한 효능을 탐색하기 위해서 시료 첨가수준의 다각화 및 여러 가지 보완연구가 필요할 것으로 생각 된다.
본 연구에서는 아로니아 열매 추출물(AF-H)의 항염증 활성을 조사하기 위하여 LPS 자극에 의해 유도된 RAW 264.7 macrophage의 염증반응에서 AF-H의 염증매개인자 및 염증성 사이토카인 분비 억제활성과 이에 관련된 세포 내 작용기전 해석을 수행하였다. LPS($1{\mu}g/mL$)로 RAW 264.7 세포를 24시간 자극하는 염증모델에서 세포독성을 나타내지 않는 안전한 농도의 AF-H($0{\sim}500{\mu}g/mL$)를 LPS 처리 12시간 전에 처리하여 NO 및 PGE2의 분비 억제활성을 측정하였다. 그 결과 AF-H 처리에 의해 NO와 PGE2의 생성이 처리 농도에 의존하여 유의하게 억제되었으며, 이들 염증매개인자의 생합성 효소인 iNOS 및 COX-2의 세포 내 발현도 현저하게 억제되는 것으로 관찰되었다. 또한 AF-H의 처리에 의해 염증성 사이토카인인 $TNF-{\alpha}$와 IL-6의 분비도 유의하게 억제되는 것으로 확인하였다. 이러한 AF-H에 의한 항염증 활성의 세포 내 기전을 해석하기 위하여 LPS 자극에 의해 유도되는 MAPK와 $NF-{\kappa}B$ 전사인자의 활성화에 대한 억제 효과를 조사하였다. 그 결과 AF-H는 MAPK의 인산화에는 별다른 영향을 미치지 않고 $NF-{\kappa}B$의 활성화($I{\kappa}B$ 인산화)를 효과적으로 억제하는 것으로 확인되었다. 한편 LPS에 의한 in vivo 패혈증 모델에서 AF-H에 의한 패혈증 억제활성을 측정한 결과 비록 통계학적으로 유의하지는 않으나 AF-H 투여에 의해 생존율과 50% 사망률의 연장 효과가 관찰되었다. 이들 결과를 종합해 보면 아로니아 열매 열수추출물은 $NF-{\kappa}B$의 활성화 억제를 통해 NO, PGE2, $TNF-{\alpha}$ 및 IL-6 등의 염증매개인자와 사이토카인의 생성을 억제하는 항염증 활성을 지니는 것으로 확인되었다.
Objectives : Gamisoyosan (GMS) is a useful prescription for treating insomnia, dysmenorrhea and infertility induced by a stress. Also, GMS has been used traditionally to improve systemic circulation and biological energy production. The purpose of this study was to assess the anti-oxidant activity and anti-inflammatory effects of Gamisoyosan Formulation (Soft extract, GMS-SE). Methods : The biological activities such as anti-oxidant and anti-inflammatory effects were measured through cell line-based in vitro assay. We investigated the anti-oxidant properties of GMS-SE on the 1,1-diphenyl-2-picryhydrazyl (DPPH) radical, contents of total flavonoid and polyphenol. GMS-SE compared to butyl hydroxy anizole (BHA). Furthermore, based on this result the anti-inflammatory effects of GMS-SE have verified by mechanism from LPS- treated Raw264.7 macrophages. Results : The anti-oxidant activities of GMS-SE increased markedly, in a dose-dependent manner. The GMS-SE showed significant scavenging activity (GMS-SE $500{\mu}g/m{\ell}$ : $32.77{\pm}1.65%$, GMS-SE $1000{\mu}g/m{\ell}$ : $45.06{\pm}1.04%$ and BHA $100{\mu}g/m{\ell}$ : $39.25{\pm}2.41%$ for DPPH assay). and, The total phenolic compound and flavonoids contents of GMS-SE were $73.93{\pm}6.87{\mu}g/mg$ and $698.75{\pm}6.78{\mu}g/mg$. GMS-SE which is LPS has diminished in the LPS-induced release of inflammatory mediators (NO, iNOS, COX2 and PGE2) and pro-inflammatory cytokines (TNF-${\alpha}$, IL-6 and IL-$1{\beta}$) from the RAW264.7 macrophages. Moreover, GMS-SE inhibited the activation of phosphorylation of p38 and ERK MAPKs by induced LPS. Conclusion : The present results indicate that GMS-SE has an anti-oxidant and anti-inflammatory properties, therefore may be beneficial in diseases which related to oxidative stress-mediated inflammatory disorders.
This study was performed to compare the effects of immuno-modulating activities of Rhodiola sachalinensis A. Bor. fractionized by consecutive solvent separation. The Cytotoxicity of all fractionized extracts on human kidney cell (HEK293) was lower than crude extracts. Generally, the butanol and chloroform extracts showed less cytotoxicity on about 10.07% and 9.67% than the crude extracts. For human immune B and T cell growth, chloroform fraction showed the highest cell growth compared to the control. The secretion of cytokines (IL-6, $TNF-\alpha$) on human B and T cells were increased by adding chloroform extracts. Also, NK cell growth was significantly improved up to nearly 30% by adding the supernatant of B cell medium grown with the chloroform fraction. It was also found that chloroform fraction could yield higher nitric oxide production from macrophage than untreated control cells. Differentiation of HL-60 cells was increased up to 131.9% after treatment with chloroform fraction extracts, compared to the control. These results indicate that the chloroform fraction of R. sachalinensis have high immune activation activity than others fractions and the crude extracts, implying that this chloroform fractions could be used a new functional material.
Background: Vitamin D is considered to exert a protective effect on various renal diseases but its underlying molecular mechanism remains poorly understood. This study aimed to determine whether paricalcitol attenuates inflammation and apoptosis during lipopolysaccharide (LPS)-induced renal proximal tubular cell injury through the prostaglandin $E_2$ ($PGE_2$) receptor EP4. Methods: Human renal tubular epithelial (HK-2) cells were pretreated with paricalcitol (2 ng/mL) for 1 hour and exposed to LPS ($1{\mu}g/mL$). The effects of paricalcitol pretreatment in relation to an EP4 blockade using AH-23848 or EP4 small interfering RNA (siRNA) were investigated. Results: The expression of cyclooxygenase-2, $PGE_2$, and EP4 were significantly increased in LPS-exposed HK-2 cells treated with paricalcitol compared with cells exposed to LPS only. Paricalcitol prevented cell death induced by LPS exposure, and the cotreatment of AH-23848 or EP4 siRNA offset these cell-protective effects. The phosphorylation and nuclear translocation of p65 nuclear factor-kappaB ($NF-{\kappa}B$) were decreased and the phosphorylation of Akt was increased in LPS-exposed cells with paricalcitol treatment. AH-23848 or EP4 siRNA inhibited the suppressive effects of paricalcitol on p65 $NF-{\kappa}B$ nuclear translocation and the activation of Akt. The production of proinflammatory cytokines and the number of terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling-positive cells were attenuated by paricalcitol in LPS exposed HK-2 cells. The cotreatment with an EP4 antagonist abolished these anti-inflammatory and antiapoptotic effects. Conclusion: EP4 plays a pivotal role in anti-inflammatory and antiapoptotic effects through Akt and $NF-{\kappa}B$ signaling after paricalcitol pretreatment in LPS-induced renal proximal tubule cell injury.
본 연구는 제주도에서 재배된 레드 비트(B. vulagaris) 뿌리의 항산화 및 항염 효과를 알아보기 위하여 70% EtOH 추출물과 순차적 용매 분획물들을 확보하여 총폴리페놀 함량 및 ABTS 라디칼 소거 활성 측정을 통한 항산화 효능평가와 대식세포인 RAW264.7 세포에 LPS를 자극한 후 iNOS/NO, $COX-2/PGE_2$ 및 전염증성 cytokine을 유도하여 염증 억제 효과를 알아보았다. 그 결과, 총 폴리페놀이 37.02 mg GAE/g로 가장 높게 나온 EtOAc 분획물이 라디칼 소거활성도 가장 우수하게 나타났으며($IC_{50}$$42.9{\mu}g/mL$), 대조군으로 사용한 BHT($IC_{50}$$57.4{\mu}g/mL$)보다 좋은 활성을 보였다. 항염 활성 분석을 위하여 NO/iNOS, $PGE_2/COX-2$, 및 염증성 cytokine($TNF-{\alpha}$, IL-6, $IL-1{\beta}$)의 생성 억제 효능을 분석한 결과, 총 폴리페놀 함량 결과와는 유의적이지 않게 hexane 분획물이 처리 농도 범위(50, 100, 200, $300{\mu}g/mL$)에서 NO/iNOS, $PGE_2/COX-2$, 및 염증성 cytokine 생성을 유의적으로 억제하였다. iNOS와 COX-2 단백질 발현 억제 효과를 통해 NO와 $PGE_2$ 생성 억제에 영향을 끼치고 있음을 확인하였고, 염증성 cytokine 중에는 IL-6의 생성을 가장 강하게 억제함으로써 전체적으로 항염 활성에 영향을 미치고 있음을 확인할 수 있었다. 이러한 결과들로부터 레드비트 뿌리의 EtOAc 분획물에서의 항산화 효능 확인과 hexane 분획물의 세포내 항염 효과를 알 수 있었으며, 향후 유효 물질 동정을 통한 기전 연구를 하는 데 대한 기초자료로 활용할 수 있을 것이라 사료된다.
본 연구에서는 쇠비름의 활용가치를 높이고 기능성 소재로서의 이용 가능성을 타진하기 위해 쇠비름 물 및 에탄올 추출물의 항산화와 항염증 활성을 탐색하였다. DPPH 라디칼 소거능과 FRAP을 이용한 총 항산화 효능 평가 결과 에탄올 추출물이 물 추출물보다 항산화 활성이 우수한 것을 확인하였고, ABTS 라디칼 소거능은 $1,000{\mu}g/mL$의 농도에서만 쇠비름 물 추출물의 활성이 더 높았다. 한편, 세포독성을 살펴보기 위하여 쇠비름 물 및 에탄올 추출물을 이용해 대식세포에 MTT assay를 수행한 결과, $4,000{\mu}g/mL$의 이하 농도까지는 세포의 생존율에 영향을 미치지 않는 것을 확인하였다. $1.0{\mu}g/mL$ LPS로 염증반응을 유도시킨 세포에 대해 쇠비름 물 추출물에 비해 에탄올 추출물이 더 효과적으로 농도의존적인 NO와 ROS 생성 억제 효과가 있었다. 추가적으로 세포의 항염증 효능의 지표 단백질로 알려진 TNF-${\alpha}$, IL-$1{\beta}$, IL-6의 발현을 확인하였을 때 LPS 단독 처리군 대비 쇠비름 에탄올 추출물에서 3가지 단백질 모두의 발현양이 감소하는 것을 확인하였다. 이상의 결과를 종합하여 볼 때 쇠비름 에탄올 추출물은 물 추출물에 비해 높은 항산화와 항염증 활성을 가지는 것으로 확인되었으며, 추후 쇠비름의 유효성분 추출을 통한 항염증 물질의 연구와 염증 억제 성분의 분리 및 그 작용기전 연구에 기초 자료가 될 것으로 사료된다.

제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)

제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
