숙지황과 가시오가피 복합추출물(OPB)이 난소절제 흰쥐의 골밀도 및 골대사에 미치는 영향 (The Effect of Herbal Formulation (OPB), Rehmannia Glutinosa Libosch and Eleutherococcus Senticosus Max Extracts on Bone Density and Bone Biochemical Marker in Ovariectomized Rats)

  • 본 연구는 숙지황과 가시오가피 복합추출물 (OPB)이 난소절제 흰쥐의 골밀도 및 골대사 변화에 미치는 영향을 관찰하기 위하여 수행하였다. 13주된 20마리의 Sprague-Dawley 암컷 흰쥐를 대조군과 실험군으로 나누었고, OPB의 골밀도 및 골대사 변화에 대한 효과를 관찰하기 위하여 난소절제 후 3일 후부터 OPB를 100 mg/kg씩 8주 동안 경구투여 하였다. pQCT 방식으로 골밀도, 골함유량, 골강도를 측정하였고, 혈청 중 CTx (C-telopeptide of type I collagen)의 변화를 관찰하였다. 난소절제 후 대조군의 골밀도는 $-29.8{\pm}3.0%$로 현저히 감소하였으며, 이러한 감소는 OPB의 투여에 의해 $-21.4{\pm}2.3%$로 억제되었다. 골강도의 변화에서는 anti-fracture 값과 anti-twisting 값이 대조군에 비해 OPB 처리군에서 유의성 있는 차이를 나타냈다. 또한 혈청 중 골흡수 지표인 CTx 농도는 대조군에 비해 OPB 처리군에서 낮게 나타났다. 이상의 결과로 숙지황과 가시오가피의 복합 추출물인 OPB가 난소를 절제한 흰쥐에서 골 소실을 억제하므로 임상에서 난소절제 후 또는 폐경 후 골다공증의 개선제로 이용될 수 있으리라 여겨진다.

Comparison of Molecular Characteristics of Extended Spectrum ${\beta}$-lactamase Producing Escherichia coli Strains Isolated from Patients with Urinary Tract Infections between 2 Time Periods of 1989 and 2010 at Gangwon Province in Korea

  • Etiological agents of extended spectrum ${\beta}$-lactamase (ESBL) producing uropathogenic Escherichia coli (UPEC) have become a major problem in urinary tract infections. The purpose of this study was to compare the molecular characteristics of ESBL producing UPEC strains isolated from 1989 and 2010. A total of 301 strains of UPEC clinical isolates was collected from Korean healthcare facility in 1989 (126 strains) and in 2010 (175 strains). UPEC clinical isolates were analyzed by multiplex polymerase chain reaction method (ESBL related bla genes and phylogenetic groups) and amplified fragment length polymorphism (AFLP). Among 301 isolates, ESBL producing UPEC were 8 strains (6.3%) in 1989 isolates and 35 strains (20%) in 2010 isolates. The rate of bla genes in ESBL producing UPEC from 1989 isolates and 2010 isolates were $bla_{TEM}$ (75% and 85.7%), $bla_{CTX-M}$ (0% and 91.4%), $bla_{OXA}$ (25% and 20%), $bla_{PER}$ (0% and 2.9%). The distribution of phylogenetic groups in 1989 isolates and 2010 isolates were A (37.5% and 11.4%), B2 (12.5% and 51.4%), and D (50% and 37.1%). The most prevalent ESBL related bla gene and phylogenetic group were $bla_{CTX-M}$ (91.4%) and B2 (51.4%) in 2010 isolates, while $bla_{CTX-M}$ was not detected in 1989 isolates. Among 43 ESBL producing UPEC were grouped into 12 clusters up to 76% of genetic similarities by AFLP analysis. During past twenty one years, the rate of the ESBL producing UPEC strains in 2010 isolates was increased than that of in 1989 isolates. Also, the most prevalent ESBL related bla gene has been changed from $bla_{TEM}$ to $bla_{CTX-M}$.

경북지역의 닭으로부터 CTX-M-14 생성 장내세균 분리동정 (CTX-M-14 Producing Enterobacteriaceae Isolated from Chickens at Gyeongsang Provinces)

  • 지난 반세기 동안 세균 감염증 치료 또는 성장촉진을 목적으로 가금류에게 광범위하게 항균제를 사용해 왔다. 그러나 항균제 내성의 증가는 사람에게 보편적으로 사용되는 항균제들에 대한 내성을 유도하여 치료를 위한 항균제 선택에 제약을 주어 치료에 어려움을 가중시키고 있다. 본 연구에서는 경상도 지역에서 사육된 닭으로부터 분리된 장내세균을 대상으로 ${\beta}-lactam$, quinolone, 및 aminoglycoside 계열 항균제에 대한 내성유전자의 빈도를 조사하였다. 그리고 항균제 감수성 검사를 시행하여 내성유전자와의 관련성을 알아보았다. 본 연구에서 총 43균주의 장내세균이 40마리의 닭의 맹장으로부터 분리되었으며 디스크확산법을 이용하여 항균제 감수성 검사를 시행하였다. 그리고 중합효소연쇄반응과 염기서열분석을 통해 플라스미드 매개 항균제 내성 유전자를 조사하였다. 총 43균주 중 2균주가 $bla_{CTX-M-14}$ 유전자를 포함하고 있었으며 qnrS 및 aac(6')-Ib-cr 유전자도 각각 2균주와 5균주에서 확인되었다. 총 43균주를 대상으로 항균제 감수성을 조사한 결과 cefepime, ceftazidime, 및 cefaclor를 제외한 모든 항균제에 대해 0.0%부터 23.3%까지의 낮은 감수성률을 보였다. 본 연구에서는 닭으로부터 분리된 대장균에 플라스미드 매개 항균제 내성유전자가 확산되어 있음을 확인하였다. 항균제 내성 유전자의 확산을 막기 위해서는 지속적인 항균제 내성유전자의 조사와 감시가 필요할 것으로 사료된다.

  • 임상검체에서 분리되는 그람음성 막대균의 제 3세대 cephalosporin에 대한 내성율의 증가는 임상적으로 심각한 문제가 되고 있다. 3세대 cephalosporin 및 monobactam계 항균제에 대한 내성은 주로 Extended-Spectrum ${\beta}$-Lactamase(ESBL)의 생성에 기인한다. 따라서 ESBL 유전자의 정확한 검출은 병원내의 감염경로 파악을 위한 감시 및 역학조사를 위해 필수적이다. 본 연구는 2012년 2월부터 8월까지 대전, 충남, 충북지역의 대학병원으로부터 ESBL 생성 Klebsiella penumoniae 46균주를 분리하여 Clinical and Laboratory Standards Institute (CLSI)에 따라 ceftazidime (CAZ)과 CAZ/clavulanate (CLA)를 이용한 combination disk test (CDT) 방법에 의해 표현형을 조사하고, 유전형 특이 프라이머를 이용한 multiplex PCR을 수행하여 유전형을 검출하였다. CDT 결과 42균주가 ESBL생성균주로 확인되었다. PCR 결과, 46균주 모두 TEM형이었으며, 37균주는 SHV형, 14균주는 CTX-M형으로 나타났으며 10균주가 TEM, SHV, CTX-M 유전자를 모두 가지고 있었다. Multiplex PCR에 의한 유전형 검출 방법은 임상에서 분리한 ESBL생성 K. penumoniae균주의 감별과 검출에 유용한 방법으로 사료된다.

Detection of CTX-M and Clonal Analyses using MLST in Cefotaxime Resistant $Escherichia$ $coli$ Isolated from the Han-River, Korea

  • Bacteria resistant to various antibiotics have recently become an issue of the utmost importance. Resistant strains are not uncommon, even in municipal drinking water sources. The health threat posed by resistant, pathogenic bacteria has serious ramifications for both public health and agriculture. In this study, we isolated antibiotic resistant bacteria from water samples from the Han River, Korea, which is contaminated by the wastewater from many industrial complexes, hospitals, agricultural and animal husbandry estates, and from wastewater treatment facilities. We determined the degrees of resistance to various antibiotics exhibited by the isolated strains. The similarities between the isolated $E.$ $coli$ strains were examined, using the pulsed field gel electrophoresis and multilocus sequence typing, in order to trace their origins and to explore the syntechnic adaptations and pathogenicity of the various strains and relate these to their genetic sequence. A total of 25 $E.$ $coli$ strains were isolated from six stations along the Han River. All the 25 strains exhibited resistance to ampicillin. We also investigated resistance to amoxicillin, clavulanic acid, cefazolin, cofoxitin, cefotaxime, cefpodoxime, ceftriaxone, cefepime, nalidixic acid, aztreonam, ciprofloxacin, streptomycin, gentamicin, chloramphenicol and imipenem. Based on the ESBL detection, 14 strains belonged to the ESBL producing strains. The number of the clonal complex producing strains was 5 among the 14 isolated strains. The 5 strains were included in the 168, 23, 38, 469, 156 clonal complex, respectively. The rest 9 strains were not included in the clonal complex, but showed independent STs.

가금병원성대장균의 항생제 내성 및 분자유전학적 역학분석

  • This study was conducted to investigate O group serotyping, antimicrobial drug resistance and distribution of extended spectrum ${\beta}$-lactamase of 203 Escherichia coli(E. coli) isolated from poultry in Korea during the period from April in 2003 to December 2005. The serogroup of 69.4% of isolates was determinated ; O 78(32.5%), O88(7.9%). O15(6.9%) and O141(6.4%) were the most common. These E. coli isolates showed resistance to nalidixic acid(92.6%), streptomycin(81.8%), ampicillin(77.3%), ciprofloxacin(70.9%), sulfisoxazole(66.5%) and trimethoprim(58.1%), respectively. The bla CTX$_{-M-3\;like}$(2 strains) and bla$\;_{CMY-2}$(2 strains) genes producing extended spectrum ${\beta}$ - lactamase(ESBL) were detected in four wild strains resistant to the third generation cephalosporin, respectively. The presence of the ESBL genes was confirmed in all transconjugants by PCR analysis with primers encoding CTX-M-3 like types or CMY-2.

국내 가금유래 병원성 대장균의 extended-spectrum β-lactamase(ESBL) 특성 조사 (Survey of extended-spectrum β-Lactamase (ESBL) in pathogenic Escherichia coli isolated from poultry in Korea)

  • This study was conducted to investigate incidence of extended-spectrum ${\beta}$-lactamase (ESBL) producing strains and characteristics of ESBL gene in pathogenic Escherichia coli isolated from poultry during the period from April 2003 to December 2005 in Korea. Among 203 isolates, 4 isolates (3 from broilers and 1 from layer) were confirmed as ESBL producing strains by double disk synergy test, polymerase chain reaction and sequencing for ${\beta}$-lactamase genes. $bla_{CTX-M-15}$ and $bla_{CMY-2}$ were detected in these 4 isolates and were transferred to recipient by conjugation, respectively. Also, these ESBL producing strains were associated with multiple drug resistance. In conclusion, these results exhibit incidence of CTX-M and CMY-2 ${\beta}$-lactamase in pathogenic E coli from poultry in Korea, and clinically important meaning in human. And they also suggest the needs for rapid and broad surveillance to monitor ESBL genes and R plasmid transferring resistant gene in poultry.

G Protein Mediated Hatching Regulation in the Mouse Embryo

  • Hatching occurred in the time dependent manners and strictly controlled. Although, the hatching processes are under the control of muti-embryotrophic factors and the expressed G proteins of cell generate integrated activation, the knowledge which GPCRs are expressed during hatching stage embryos are very limited. In the present study, which G proteins are involved was examined during blastocyst development to the hatching stage. The early-, expanded-, and lobe-stage blastocysts were treated with various $G_{\alpha}$ activators and H series inhibitors, and examined developmental patterns. Pertusis toxin (PTX) improved the hatching rate of the early-stage blastocyst and lobe-formed embryos. Cholera toxin (CTX) suppressed the hatching of the early-stage blastocyst and expanded embryos. The effects of toxins on hatching and embryo development were changed by the H7 and H8. These results mean that PTX mediated GPCRs activation is signaling generator in the nick or pore formation in the ZP. In addition, PTX mediated GPCR activation induces the locomotion of trophectoderm for the escaping. CTX mediate GPCRs activation is the cause of suppression of hatching processes. Based on these data, it is suggested that various GPCRs are expressed in the periimplantation stage embryos and the integration of the multiple signals decoding of various signals in a spatial and temporal manner regulate the hatching process.

골대사의 생화학지표 (Biochemical Markers of Bone Turnover)

  • Biochemical markers of bone turnover has received increasing attention over the past few years, because of the need for sensitive and specific tool in the clinical investigation of osteoporosis. Bone markers should be unique to bone, reflect changes of bone loss, and should be correlated with radiocalcium kinetics, histomorphometry, or changes in bone mass. The markers also should be useful in monitoring treatment efficacy. Although no bone marker has been established to meet all these criteria, currently osteocalcin and pyridinium crosslinks are the most efficient markers to assess the level of bone turnover in the menopausal and senile osteoporosis. Recently, N-terminal telopeptide (NTX), C-terminal telopeptide (CTX) and bone specific alkaline phosphatase are considered as new valid markers of bone turnover. Recent data suggest that CTX and free deoxypyridinoline could predict the subsequent risk of hiP fracture of elderly women. Treatment of postmenopausal women with estrogen, calcitonin and bisphosphonates demonstrated rapid decrease of the levels of bone markers that correlated with the long-term increase of bone mass. Factors such as circadian rhythms, diet, age, sex, bone mass and renal function affect the results of biochemical markers and should be appropriately adjusted whenever possible. Each biochemical markers of bone turnover may have its own specific advantages and limitations. Recent advances in research will provide more sensitive and specific assays.

  • 저자들은 최근 급성골수성백혈병 환자로부터 다제내성대장균 균주 KBN10P04869를 분리했다. 균주는 4,457개의 단백질 코딩 유전자, 88개의 운반 RNA, 22개의 리보솜 RNA를 포함하는 5,104,264 염기쌍으로 구성되고, $^{bla}CMY-2$, $^{bla}TEM-1$, $^{bla}CTX-M-15$, $^{bla}NDM-5$, $^{bla}OXA-18$를 포함한 다제내성유전자를 가지고 있다. 저자들은 이 균주의 총유전체를 보고하는 바이다.
