대추 추출물과 이를 첨가한 호상 요구르트의 항산화능 (Antioxidant Activity of Jujube and Curd Yogurt Addition to Jujube)

  • The purpose of this study was to evaluate the jujube hot-water extraction and antioxidant. After extraction of hot-water jujube, general composition analysis and functional tests were performed. The results of general composition analysis showed 22.33% of moisture content, 0.71% of crude lipid, 5.21% of crude protein, and 3.87% of ash. From DPPH test results, in concentrations of $1,000{\mu}g/mL$ of jujube extracts, electron donating ability was shown with 68.24%. The SOD ability in concentrations of $1,000{\mu}g/mL$ was 13.12%. The nitrite scavenging ability of jujube extracts was 11.79% at $1,000{\mu}g/mL$. The yogurt with 0, 1, 2, 3 and 4% of jujube extracts was made, and the general composition analysis and the functional tests were performed. The results of the general composition analysis of jujube yogurt, showed 74.71~76.56% of moisture contents, 1.31~3.38% of crude lipid, 2.13~3.40% of crude protein, and 1.18~1.28% of ash. The DPPH test results showed 46.33% for 1% added jujube extract, 53.78% for 2% added jujube extract, 90.87% for 3% added jujube extract, and 89.58% for 4% added jujube extract. The SOD ability showed 4.93% for 1% added jujube extract, 7.28% for 2% added jujube extract, 11.38% for 3% added jujube extract, and 11.50% for 4% added jujube extract.

Arginase II Inhibitory Activity from Crude Drugs

  • Arginase competitively inhibits nitric oxide synthase (NOS) via use of the common substrate L-arginine. Arginase II has recently reported as a novel therapeutic target for the treatment of cardiovascular diseases such as atherosclerosis. In our experiment, the EtOH extracts of four-hundreds extracts drugs were investigated for the arginase inhibitory activity. Among them, four extracts exhibited over 50% inhibition of arginase II activity compared to control at a concentration of 150${\mu}g/ml$. In particular, the seed of Arctium lappa, gum-resin of Boswellia carterii, aerial part of Artemisia apiacea and rhizome of Cyperus rotundus inhibited arginase II activity, with $IC_{50}$ values of 118.4, 135.4, 123.9 and 86.7${\mu}g/ml$, respectively. In addition, four plant extracts showed less than 20% inhibition of arginase I activity at 150${\mu}g/ml$. These plants might be the potential candidate materials in the development of the novel atherosclerosis drug.

녹차, 오룡차 및 홍차 추출물의 항산화효과 (Antioxidative Effect of Tea Extracts from Green Tea, Oolong Tea and Black Tea)

  • Antioxidative effect of tea extracts from green tea(steamed, roasted), oolong tea and black tea was investigated. Antioxidative activities of the crude catechin fraction were the most effective in oolong tea which contained the highest level of (-)-epigallocatechin gallate(EGCg). The water-soluble fraction obtained from oolong tea exhibited binding with more ferrous than copper ions. It showed a synergistic effect when used with an antioxidant such as BHT(0.5mg) and $\alpha$-tocopherol(2mg). Especially, this synergistic effect was exhibited more with BHT than with $\alpha$-tocopherol. Also steamed green tea, roasted green tea and oolong tea showed remarkable free radical scavenging action. The SC50(the concentration of a water-soluble fraction which is required to scavenge 50% of 100$\mu$M 1, 1-diphenyl-2-picryl hydrazyl radicals) of steamed and roasted green tea, oolong tea and black tea extracts were 11.3$\mu\textrm{g}$/ml, 11.4$\mu\textrm{g}$/ml, 12.7 $\mu\textrm{g}$/ml and 14.9$\mu\textrm{g}$/ml, respectively. It is assumed that the antioxidative activity of tea extracts is due to inhibition of peroxidation free radical scavenging and binding action of ferrous ions by mainly tea polyphenol compounds.

Olfactory Responses of Male and Female Red-spelled Newts to Sex Pheromones from the Opposite Sex

  • Functional characterization of sex pheromones in olfactory responses is essential for the study of chemical communications in amphibians. Using Y-maze olfactory preference tests, we have investigated the olfactory res-ponses of male and female red-spotted newts, Notophthalmus viridescens, to the crude extracts of the opposite sex's genial and cloacal pheromones. Both male and female cloacal pheromone extracts caused the opposite sex to initiate olfactory responses by leaving the starting area in the Y-maze, but only subject males exposed to female cloacal pheromone extracts completed olfactory choice by entering the side arm of the Y-maze which received the pheromones. For genial pheromone extracts, only female genial pheromone extracts induced initial olfactory responses from test males. Neither male nor female genial pheromone extracts made the oppo-site sex complete olfactory choice. Pre-exposure of test females to male pheromone extracts increased the likelihood of initial olfactory responses. The latency for initial olfactory responses of test females that were previously exposed to male genial extracts was significantly shorter than that of control females.

추출용매에 따른 조릿대 잎 추출물의 항산화활성 (Antioxidant Activities of Bamboo (Sasa Borealis) Leaf Extract according to Extraction Solvent)

  • 본 연구에서는 조릿대 잎의 물과 70% 에탄올 조추출물 2종 및 70% 에탄올 조추출물의 분획 5종에 함유된 총 폴리페놀과 총 플라보노이드 함량을 정량하고, 이들 조추출물과 분획의 항산화 활성을 검증하고자 전자공여능, SOD 유사활성, 환원력 및 지질과산화물 생성억제능을 측정하였다. 조추출물 2종의 수율은 물과 70% 에탄올 각각 8.5%와 11.4%이었다. 분획 5종의 수율은 5.1-0.6%이었으며, aqueous, n-butanol, n-hexane, chloroform, ethyl acetate 분획 순으로 높았다. 총 폴리페놀 함량은 물과 70% 에탄올 추출물은 두 조추출물 간에 유의한 차이가 없었으나 총 플라보노이드 함량은 70% 에탄올 추출물이 물 추출물보다 유의하게 많았다. 분획 5종의 총 폴리페놀 함량은 chloroform, ethyl acetate, n-butanol, aqueous, n-hexane 분획 순으로 많았고, chloroform 분획은 타 분획에 비해 유의성 있게 많았다. 각 분획별 총 플라보노이드 함량은 ethyl acetate, chloroform, n-hexane, n-butanol, aqueous 분획 순으로 함량이 높았으며, chloroform과 ethyl acetate 분획은 타 분획에 비해 유의적으로 많았다. 물과 70% 에탄올 조추출물은 모두 BHT에 비해, 유의성 있게 높거나 또는 같은 수준의 지질과산화물 생성억제능을 보였으며, 동일한 수준의 SOD 유사활성을 나타내었다. 다만, 전자공여능과 환원력은 BHT에 비해 낮았다. 물과 70% 에탄올 조추출물 간에 이들 항산화 활성은 다르지 않았다. 다만, 전자공여능만 일부 농도에서 70% 에탄올 조추출물이 물 조추출물에 비해 유의하게 높았다. 분획 5종은 모두 BHT에 비해 유의성 있게 높거나 같은 수준의 지질과산화물 생성억제능을 보였다. Chloroform과 ethyl acetate 및 n-butanol의 세 분획은 전자공여능과 SOD 유사활성도 BHT에 비해 유의하게 높았다. 다만, 환원력은 분획 5종 모두 BHT보다 낮았다. 조추출물 2종과 분획 5종의 전자공여능이나 SOD 유사활성, 환원력 및 지질과산화물 생성억제능 모두 본 연구에서 실험한 농도 범위에서, 농도 의존적으로 증가했거나 또는 증가하는 추세를 보였다. 이러한 연구결과에 기초해 조릿대 잎의 물 또는 70% 에탄올의 조추출물이나 70% 에탄올의 조추출물의 chloroform과 ethyl acetate 및 n-butanol 분획은 천연 항산화제로서 특히, 지질의 과산화를 방지하는 용도로서의 활용 가치가 있다고 결론지을 수 있다. 또한 건강기능성 소재로서의 식품산업 분야에 특히, 지질과산화물 생성억제능과 SOD 유사활성이 BHT보다 우수했고, 전자공여능을 보인 점은 이들 추출물이나 분획이 생체 내에서 활성산소종을 제거하고 생체막의 구성성분의 과산화를 방지하는 등 건강기능성 작용을 수행할 것이란 점을 시사한다.

매실농축액을 첨가한 보리다식의 품질 특성 (Quality Characteristics of Barley Dasik added with Maesil Extracts)

  • This study assessed the quality of barley Dasik manufactured, according to traditional methods, with various levels of Maesil (Prunus mume) extracts at weight percentages of 0% (BD1), 10% (BD2), 20% (BD3), 30% (BD4), 40% (BD5). The Dasik were analysed with proximate analysis, a sugar content analysis, Hunter's color value test, mechanical characteristics test and a sensory evaluation. Increased ratios added Maesil extracts led to the moisture content, crude ash, and crude fat to significantly increase (p<.001). 10% (BD2) had significantly higher levels of crude protein (p<.01). Sugar contents was highest in 0% (BD1) and decreased as the amount of added Maesil extracts increased. This increase also led to higher L-values and b-values of chromaticity (p<.001) and the a-value of 0% (BD1) was significantly higher than the sample groups (p<.001). Changes in texture profile analysis, hardness and gumminess values were highest in the control group and the values of those in the sample group decreased with increasing amounts of Maesil extract (p<.001). It should be noted that, among the samples evaluated, barley Dasik that contained 10% Maesil extract (BD2) had the highest commercial value.

다시마 추출액의 항산화 및 항염증 활성에 대한 효모 발효의 영향 (Effect of Yeast Fermentation on the Antioxidant and Anti-inflammatory Activity of Sea Tangle Water Extract)

  • To examine the effective use of seaweeds, sea tangle (Laminaria japonica) was extracted with water and the resultant extracts were fermented with Saccharomyces cerevisiae. Four strains of S. cerevisiae were cultured in aqueous extracts from sea tangle. S. cerevisiae SC-2, which was isolated from a traditional Korean fermented food (Meju), was selected for further study based on the results of a sensory evaluation. No significant differences in proximate compositions, such as moisture, crude protein, crude fat, and crude ash, of the sea tangle extracts before and after fermentation were observed. The reducing sugar decreased as the fermentation period increased, and the contents of some free amino acids were also affected by S. cerevisiae SC-2 fermentation. However, the content of glutamic acid, which is a major taste compound in sea tangle extract, was not affected by fermentation for up to 36 hr by the SC-2 strain. To determine the antioxidant activity of fermented sea tangle extract (fermented for 36 hr by SC-2 strain), the radical scavenging activities of 1,1-diphenyl-2-picrylhydrazyl (DPPH), superoxide, and nitric oxide were investigated and xanthin oxidase inhibition assay was performed. The antioxidant activity increased by 8 to 35%. The greatest enhancement of antioxidant activity was seen in the superoxide radical scavenging assay with $100\;{\mu}g/mL$ of raw and fermented sea tangle extract. The anti-inflammatory activity of fermented sea tangle extract was also enhanced. The fermented sea tangle extract showed 34.2% inhibitory activity against nitric oxide synthesis versus 11.9% for raw sea tangle extract at $100\;{\mu}g/mL$ concentration. These results suggest that fermented aqueous extracts from sea tangle are a useful resources.

요소분해효소 기반 식물추출액을 이용한 광산폐기물 내 중금속 오염 저감 (Bioremediation of Heavy Metal Contaminated Mine Wastes using Urease Based Plant Extract)

  • Acid mine drainage occurrence is a serious environmental problem by mining industry, it usually contains high levels of metal ions, such as iron, copper, zinc, aluminum, and manganese, as well as metalloids of which arsenic is generally of the greatest concern. An indigenous plant extract was used to produce calcium carbonate from Canavalia ensiformis as effective biomaterial, and its ability to form the calcium carbonate under stable conditions was compared to that of purified urease. X-ray diffraction and scanning electron microscopy were employed to elucidate the mechanism of calcium carbonate formation from the crude plant extracts. The results revealed that urease in the plant extracts catalyzed the hydrolysis of urea in liquid state cultures and decreased heavy metal amounts in the contaminated soil. The heavy metal amounts were decreased in the leachate from the treated mine soil; 31.7% of As, 65.8% of Mn, 50.6% of Zn, 51.6% of Pb, 45.1% of Cr, and 49.7% of Cu, respectively. The procedure described herein is a simple and beneficial method of calcium carbonate biomineralization without cultivation of microorganisms or further purification of crude extracts. This study suggests that crude plant extracts of Canavalia ensiformis have the potential to be used in place of purified forms of the enzyme during remediation of heavy metal contaminated soil.

삼채부위별 에탄올 추출물의 생리활성탐색 (Physiological Activities of Ethanol Extracts from Different Parts of Allium hookeri)

  • 삼채뿌리와 잎이 가진 기능성 식품소재로서의 활용도를 높이고자 다양한 생리활성을 평가하였다. 삼채뿌리의 조사포닌, 폴리페놀, 플라보노이드 함량은 각각 4.28 mg/g, 18.27 mg/g, 0.95 mg/g, 삼채잎의 조사포닌, 폴리페놀, 플라보노이드 함량은 각각 4.17mg/g, 35.91 mg/g, 2.79 mg/g으로 나타났다. 삼채뿌리 및 잎 부위의 70% 에탄올 추출물의 DPPH 라디칼 소거능 $IC_{50}$으로 23.01 mg/mL, 4.82 mg/mL로 나타났으며, hydroxyl radical 소거능 $IC_{50}$ 값이 각각 40.82 mg/mL, 27.24 mg/mL를 나타났다. ${\alpha}$-Amylase 저해 활성은 삼채뿌리 및 잎 부위 에탄올 추출물 10 mg/mL를 처리했을 때 억제활성이 각각 61.38%, 61.01%로 나타났다. 삼채뿌리 및 잎 부위의 아질산염 소거능을 측정한 결과, 삼채추출물의 농도가 증가함에 따라 그 활성 역시 증가하는 경향으로 나타났다. 이상의 결과로 삼채뿌리보다는 삼채잎 부위에 항산화 및 항당뇨 활성이 나타났으며, 삼채잎을 활용한 식품 소재로써 개발할 가치가 매우 높은 것으로 판단된다.

청각(Codium fragile) 추출물의 항산화성 및 생리활성 (Comparison of Antioxidant and Physiological Activities of Different Solvent Extracts from Codium fragile)

  • The present study investigated the chemical composition, and antioxidant and physiological activities of the Korean marine algae, Codium fragile. The solvent extracts from C. fragile were prepared using 70% ethanol, 80% methanol, and distilled water. Based on the general chemical composition, carbohydrate, crude protein, crude lipid, crude ash, and moisture were 74.22%, 16.73%, 0.66%, 4.39%, and 4.00%, respectively. Calcium, magnesium, sodium and potassium were the main minerals. The extraction yield range of the solvent extracts was 3.51-9.76%. The ranges of total polyphenol and flavonoid contents were approximately 10.97-13.76 mg/g and 8.00-8.69 mg/g, respectively. The ABTS [2,2'-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid)] radical scavenging activity, reducing power and FRAP (ferric reducing antioxidant power) activities were the highest in the ethanol extract, while methanol extract exhibited the strongest nitrite oxide scavenging activity. On the other hand, tyrosinase, elastase, and xanthine oxidase inhibitory activities of the ethanol and methanol extracts were higher than those of the water extract. Furthermore, the ethanol extract exhibited the highest β-secretase inhibitory activity. The results indicate that C. fragile can be used as an antioxidant and a functional ingredient in food and pharmaceutical products.
