Anthocyanin-Contents and Pigment Stability of Black Soybean by Different Extract Condition and Stabilizer (추출조건과 첨가물에 따른 검정콩의 안토시아닌 함량과 색소 안정성)

  • The purpose of this study was to analyze the anthocyanin contents of black soybean crude extracts derived using a countercurrent system and to compare the effects of stabilizers(${\beta}-cyclodextrin$, maltodextrin) and sugars(sucrose, maltose) on the color deterioration of the anthocyanin. When the extraction process was kept at 100$^{\circ}C$ for 120$\sim$180 min, only C3G (cyanidin-3-glucoside) was detected in the water extract. The C3G contents in the water extracts acquired at 8$^{\circ}C$, 60$^{\circ}C$, and 80$^{\circ}C$ were 2.38 ppm, 1.73 ppm, and 1.73 ppm, respectively. Sucrose and maltose retarded color deterioration of the crude pigment extract by the countercurrent method with methanol. Finally, the additions of maltodextrin or ${\beta}-cyclodextrin$ did not retard thermal color deterioration of the black soybean crude pigment extract.

Nutritional Components and Antioxidant Activities of Various Stachys Sieboldii Miq Parts (초석잠 부위별 영양성분 및 항산화효과)

  • This study was carried out to compare the major nutrient components and antioxidant activities of Stachys sieboldii Miq leaf and root powders. For proximate compositions, crude protein and crude fat contents of leaf powder were higher than those of root powder, whereas contents of crude ash and carbohydrates were lower in leaf powder. The content of glutamic acid was greater in amino acids of leaf and root powders, and contents of total amino acids and essential amino acids were higher in leaf powder compared with root powder. Root powder contained a higher level of total unsaturated fatty acids than leaf powder. Total contents of organic acids were higher in leaf powder; tartaric acid was the major organic acid in leaf powder, and malic acid was the major organic acid in root powder. The content of vitamin A was higher in leaf powder than in root powder. However, vitamin E content was higher in root powder than in leaf powder. Total mineral contents of leaf powder were higher than those of root powder, and mineral contents of leaf and root powders were in the order of K>Ca>Mg. Extract yields of leaf and root powders were 27.21% and 58.51%, respectively. Total polyphenols and total flavonoids of leaf extract were 236.35 mg/g and 1.90 mg/g, respectively, which were higher than those of root extract. The $IC_{50}$ values of leaf and root ethanol extracts based on DPPH hydroxyl scavenging were 0.69 mg/mL and 5934.31 mg/mL, respectively, and antioxidative activities of ethanol extracts from all Stachys sieboldii Miq parts dose-dependently increased. These results suggest that Stachys sieboldii Miq can be recommended as an edible functional food material.

Screening of Crude Drugs for Antioxidative Activity (생약추출물의 항산화 활성검색)

  • Based on DPPH radical scavenging activity and lipid peroxidation inhibitory activity, the MeOH extracts of 139 crude drugs were screened in order to search for antioxidants. Among tested samples, the extracts from the seed of Nelumbo nucifera, the fruit of Terminalia chebula var. gangetia, the root of Salvia miltiorhiza, the fruit of Ziziphus jujuba var. innermis, the root bark of Paeonia moutan, the fruit of Rubus coreanus, the fruit of Zanthoxylum schinifolium, the lignum of Caesalpinia sappan, the leaf of Pinus densiflora, the rhizome of Alpinia officinarum, the fruit of Syzygium aromaticum, the ramulus and uncus of Uncaria rhynchophylla, the root bark of Lycium chinense, and the fruit of Alpinia katsumadai showed a relatively strong antioxidative activity. Furthermore, the BuOH fraction from the extract of N. nucifera showed a potent activity in each assay. The isolation of bioactive compounds has been carried out and will be reported in the next paper.

Antioxidant and Antimicrobial Activities of The Extracts from Native Camellia japonica in Korea (국내 자생 동백나무(Camellia japonica L.) 추출물의 항산화 및 항미생물 활성)

  • This research has been undertaken to increase availiability of native Camellia japonica leaf and flower in Korea as a edible-medicinal resource. Chemical compositions, antioxidant and antimicrobial activities in different parts of camellia were investigated. Crude protein contained the highest in young leaves(14.22%) but less than 10% in different parts. The contents of crude fat and crude ash were 60.48% in seeds and 5.16% in mature leaves, respectively. Methanol extract of young leaf, flower, and flower bud in camellia showed strong antioxidant activity compared with different parts. Also, antioxidant activity of these was higher than that of BHT, but weaker than that of VtC. Antioxidant activity of various parts in camellia were in order of young leaf > flower bud > flower > mature leaf > stem > bark. Extracts of mature leaf and bark had a remarkable antibacterial activity(0 CFU/$m\ell$) on Bacillus subtilis. The young leaf extract displayed effective growth inhibition against B. subtilis, Candida albicans, and Trichosporon beigelii.

Nutritional Components and Antioxidant Activities of Solvent Extracts from White and Brown Flammulina velutipes (흰색과 갈색 팽이버섯(Flammulina velutipes)의 영양성분 및 용매별 추출물의 항산화 활성)

  • This study was conducted to determine the proximate compositions, nutritional components, and antioxidant effects of white and brown enoki mushrooms (Flammulina velutipes). The crude protein and carbohydrate contents were higher in the brown than white mushrooms, whereas the moisture, crude ash, crude lipid, and dietary fiber levels were lower. The mineral contents of the white mushroom was higher than levels obtained in the brown mushroom for the detected components (Ca, Cu, K, Mn, Na, and P). The amount of vitamin B3 in the brown mushroom was 1.51 mg/100 g, which was 4.5 times higher than that in the white mushroom. The major fatty acids detected were palmitic acid, linoleic acid, and α-linolenic acid. The total polyphenol and flavonoid contents were highest in 70% ethanol extracts of the white and brown mushrooms, respectively. For the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, a 70% methanol extract of the white enoki mushrooms showed an activity of 76.4% (p<0.05). For the ferric-reducing antioxidant power (FRAP) activity, a 70% methanol extract of the brown enoki mushrooms showed the highest value. Further, the total flavonoid contents were significantly correlated with the DPPH and FRAP activities.

Biological Activities of Crude Polysaccharides and Crude Saponins from Salicornia herbacea (함초 조다당체 및 조사포닌의 생리활성)

  • To develop Salicornia herbacea as a functional food material, crude polysaccharides and saponins were isolated from 80% ethanolic extracts of Salicornia herbacea using Diaion HP-20 column chromatography, and biological activities including anti-oxidation, anti-proliferation of cancer cells, and immune activities were investigated. The hydrogen-donating properties of crude polysaccharides and saponins were above 20% at $500{\mu}g/mL$ concentration. The reducing power of fractions increased in a dose-dependent manner. Crude polysaccharides and saponins at $500{\mu}g/mL$ inhibited more than 20% and 50% of proliferation by PC-3 and HT-29 human cancer cells, respectively. The anti-oxidative and anti-cancer properties of crude saponins were higher than those of crude polysaccharides. Also, proliferation in spleen cells and (nitric oxide) NO production in RAW264.7 macrophages treated with crude polysaccharides increased in a dose-dependent manner compared with the untreated control cells. However, crude saponins at concentration greater than $500{\mu}g/mL$ resulted in decreases in immune activities. Immune activities of crude polysaccharide were greater than those of crude saponins. These results indicate that Salicornia herbacea may be used as functional food materials.

Immune Stimulation and Anti-Metastasis of Crude Polysaccharide from Submerged Culture of Hericium erinaceum in the Medium Supplemented with Korean Ginseng Extracts (수삼추출물 첨가 배지에서 배양된 노루궁뎅이버섯 균사체 심부발효물 조다당획분의 면역 및 암전이 억제활성)

  • To find the new use of Korean ginseng and mushroom, crude polysaccharides were prepared from submerged cultures of Hericium erinaceum in the medium supplemented with Korean ginseng extracts. When we fractionated crude polysaccharides (HE-GE-CP-1, 3, and 5) from hot-water extracts of submerged cultures of H. erinaceum with ginseng extracts (1%, 3%, and 5% addition of total medium), the yields of HE-GE-CP-1, 3, and 5 were identified at 5.7, 5.1, and 4.8%, respectively. Among crude polysaccharide fractions, HE-GE-CP-5 was significantly higher (1.89-fold of the saline control) than those of HE-GE-CP-1 (1.64-fold) or HE-GE-CP-3 (1.76-fold) on mitogenic activity of splenocytes. HE-GE-CP-5 also had the more potent bone marrow cell proliferation (1.83-fold) rather than HE-CP or HE-GE-CP-1 or HE-GE-CP-3 (1.59- or 1.44- or 1.69-fold, respectively), and anti-metastatic activity as anti-cancer effect showed the highest prophylactic value (72.4% inhibition of tumor control) in 5% supplementation of ginseng extract. However, the lysosomal phosphatase of macrophage was significantly stimulated after HE-GE-CP-3 treatment (2.03-fold). In addition, the immunostimulating and anti-metastatic crude polysaccharide, HE-GE-CP-5, contained mainly neutral sugars (63.2%) with considerable amounts of uronic acid (19.3%) and a small amount of proteins (8.8%). HE-GE-CP-5 can stimulate immune system to inhibit tumor metastasis, and its anti-tumor metastasis may be associated with macrophages, splenocytes and Peyer's patch cells activation.

A Cytotoxic Activity of Panax Ginseng Extract Against Bome Cancer Cells In Vivo and In Vitro.

  • This study was devised to observe the cytotoxlc activities of petroleum-ether extract of Panax ginseng root(crude Gx) and its partially purified fraction from silicon acid column chromatography(7:3 CX) against sarcoma-180(5-180) and Walker carcinosarcoma 256(Walker 256) in vivo, and murine leukemic lymphocytes(L1210) and human rectal cancer cell(HRT-18) and human colon cancer cells(HT-29 and HCT-48) in vitro . Each cell-line was cultured in medium containing serial concentrations of the crude Gx or 7:3 Gx in vitro. A highly lipid soluble compound in the extract of Panax ginseng root was cytocidal to murine leukemic cells and human colon and rectal cancer cells in vitro In the meantime, ginseng saponin derivatives did not cytotoxic effects at its corresponding concentration. The growth rates of the cancer cells in medium containing ginseng extracts were inhibited gradually to a significant degree roughly in proportion to the increase of the extract concentration. The cytotoxic activity of 7:3 Gx was about 3 times more potent than that of crude Gx, one unit of cytotoxic activity against L121f cells being equivalent to 2.54$\mu\textrm{g}$ and 0.88 $\mu\textrm{g}$ for the crude Gx and 7:3 Gx, respectively. The Rf value of the active compound on silica -gel thin layer chromatography with petroleum-ether/ethyl ether/acetic acid mixture (90:10:1, v/v/v) as a developing solvent was 0.23. The survival times of mice inoculated with S-180 cells were extended about 1.5 to 2 times by the 7:3 Gx treatment compared with their control group. The significantly decreased hemoglobin values of rats after inoculation with Walker 256 were recovered to normal range by oral administration of the crude Gx. The synthetic levels of protein, DNA and RNA in human colon and rectal cancer cells were significantly diminished by treatment with the crude Gx, which can explain a part of the origin of its anticancer activity.

A Study on Separation of Thermolabile and Thermostable Pectinesterase from Valencia Orange (Valencia 오렌지에서 내열성 및 비내열성 Pectinesterase 분리 정제)

  • Pectinesterase(PE) has been definitively established as the causative agent for clarification of citrus juice and gelation of frozen concentrates. This enzyme is present in multiple forms in citrus fruit. Although representing a minor fraction of the total PE activity, thermostable pectinesterase(TSPE) accounts for the severe time/temperature processing treatment required to inactivate PE. This study was undertaken to separate and purify thermolabile pectinesterase(TLPE) and TSPE from the crude PE extracts of Valencia orange rag-pulp powder. The approach taken was to carry out the three kind of chromatographies of CM-Sephadex C-50, Phenyl Sepharose CL-4B and CM-Biogel A to the unheated crude PE or the heated crude PE. All of them used could increase the purity of PEs and, specially, Phenyl Sepharose CL-4B chromatography could separate crude PE as the mixture of PEs into two forms of TLPE and TSPE. The purified TLPE had specific activity of 1,005 units/mg, yield of 13.6% and purification of 35 fold, while the TSPE separated from the unheated crude PE showed specific activity of 3,115 units/mg, yield of 1.5% and purification of 100.5 fold, and another TSPE from the heated crude PE was found to be specific activity of 1,803 units/mg, yield of 15.4% and purification of 140 fold.

Physico-chemical attributes, sensory evaluation and oxidative stability of leg meat from broilers supplemented with plant extracts

  • This feeding trial was conducted to investigate the effects of Mentha arvensis (MA) and Geranium thunbergii (GT) in drinking water on physicochemical attributes, sensory qualities, proximate analysis and oxidative stability of broiler leg meat. One hundred and twenty broiler chicks were assigned to 1 of 4 dietary treatments for 5 weeks. The dietary treatments were 1) control, 2) T1 (0.1% 1 MA:1 GT), 3) T2 (0.1% 1 MA:4 GT), 4) T3 (0.1% 4 MA: 1 GT). The water holding capacity and cooking loss were improved (p < 0.05) in T2 and T3. The flavor, texture and acceptability of leg meat by consumers were significantly increased in T2 relative to the control (p < 0.05). The crude protein content was increased in T3 while the crude fat decreased in T2 (p < 0.05). Moreover, broilers supplemented with plant extracts had the lowest leg meat TBARS (thiobarbituric acid reactive substances) values after 2 weeks of storage as compared with the control. Total phenolic contents and 1-1-diphenyl 2 picrylhydrazyl (DPPH) activity were also better in the T2 group (p < 0.05) compared with the control, whereas 2,2-Azinobis-3 ethytlbenzothiazoline-6-sulfonic acid (ABTS+) remained unaffected. Overall, these results demonstrate that broiler drinking water with the inclusion of plant extract combination can be used to enhance the oxidative stability, shelf life and quality characteristics of broiler leg meat without compromising the growth performance.
