  • It was prevously shown that 'Kamisamhwang-Tang' had been examined for effects of the anticonvulsion, antipyretic, analgesic and sedative. To investigate the effect on cardiovascular system and antibacterial activities, pharmacological studies have been carried out with each extract. The results of this investigation were summarized as follows: 1) 'Kamisamhwang-Tang' depressed central convulsion induced by strychnine, picrotoxine and caffeine in mice. 2) Antipyretic, analgesic and sedative action were observed. 3) Hypotensive effect was observed in rabbits. 4) Antibacterial activities was observed.

Effects of Roughage Neutral Detergent Fiber on Dairy Performance under Tropical Conditions

  • $Holstein\;{\times}\;indigenous$ multiparous dairy cows were offered diets with increasing roughage neutral detergent fiber (NDF) contents to determine the effects on intake, milk yield and compositions. Roughage NDF contents were 15, 18, 21 and 24% dry matter (DM), and concentrate NDF content was 10% DM. Experimental treatments were isonitrogenous and isocaloric diets. Maximum and minimum temperature humidity index during the experimental period were 85.9 and 76, respectively. Intakes of DM, crude protein and net energy, 4% fat corrected milk, milk protein and average daily gain decreased with increasing roughage NDF contents (p<0.05). Intakes of ether extract (p<0.01) and dietary NDF (p>0.05) and milk fat (p<0.01) increased with increasing roughage NDF contents. The results support the conclusion that higher DM intake, optimal milk yield and compositions can be maintained with lower roughage NDF diets for dairy cows under tropical conditions.

  • Biosurfactants produced by Pseudomonas aeroginosa KK-7 were purified and their properties were studied. The bacterial surfactant was seperated into two sorts of biosufactants (Type I, 11) by silica gel column chromatograpgy. On the basis of physiochemical analysis, Type I was found to be mixture of two glycolipids with M.W. 800, and Type II was peptide with M.W. 1300. The Type 11 biosurfactant was compose of glutamic acid, proline, glycine, leucine, histidine. The crude extract was used to dertermine some properties as a surfactant. The biosurfactant had the properties as stronger emulsification agent and a stronger stabilizing agent emulsion than any other surfactants tested.

  • Carica papaya is an important medicinal plant used in the management of different disease conditions. Phytochemical screening was carried out using different chemical test, Percentage yield and total phenolic content was evaluated using Folin Ciocalteu method in different solvent system; methanol, ethanol, ethyl acetate, n-butanol and n-hexane respectively. The phytochemical screening of the studies showed the presence of flavonoids, saponins, tannins, terpenoids, glycosides, steroids, carbonhydrate, anthraquinone and alkaloids. The percentage yield of crude extract and total polyphenol content was high in methanol, ethanol and ethyl acetate when compared with n-butanol and n-hexane. The studies show that Carica papaya leave extracts is a potent source of secondary metabolites. The use of the plant in the management of diseases is justified.

Isolation and Physiological Characterization of Bacillus clausii SKAL-16 Isolated from Wastewater

  • An alkaliphilic bacterium, Bacillus clausii SKAL-16, was isolated from soil that had been contaminated with vegetable oil. The optimal pH and general pH range for bacterial growth was 8, and 7 to 10, respectively. The bacterium could grow on tributyrin and glycerol, but could not grow on acetate and butyrate. The SKAL-16 strain excreted butyric acid during growth on tributyrin, and selectively ingested glycerol during growth on a mixture of butyric acid and glycerol. The SKAL-16 generated intracellular lipase, but did not produce esterase and extracellular lipase. The DNA fragment amplified with the chromosomal DNA of SKAL-16 and primers designed on the basis of the esterase-coding gene of Bacillus clausii KSM-KI6 was not identical with the esterase-coding gene contained in the GenBank database. Pyruvate dehydrogenase, isocitrate dehydrogenase, and malate dehydrogenase activities were detected in the cell-free extract (crude enzyme).

Role of Unstable Phenanthrene-Degrading Pseudomonas species in Natural Attenuation of Phenanthrene-Contaminated Site

  • An unstable yet efficient phenanthrene-degrading bacterium strain Ph-3 was isolated from a petroleum-contaminated site at the Mathura Oil Refinery, India. The strain was identified as Pseudomonas sp. using a polyphasic approach. An analysis of the intermediates and assays of the degradative enzymes from a crude extract of phenanthrene-grown cells showed a novel and previously unreported pattern of 1, 2-dihydroxy naphthalene and salicylic acid production. While strain Ph-3 lost its phenanthrene- degrading potential during successive transfers on a rich medium, it maintained this trait in oligotrophic soil conditions under the stress of the pollutant and degraded phenanthrene efficiently in soil microcosms. Although the maintenance and in vitro study of unstable phenotypes are difficult and such strains are often missed during isolation, purification, and screening, these bacteria constitute a substantial fraction of the microbial community at contaminated sites and play an important role in pollutant degradation during biostimulation or monitored natural attenuation.

  • Isocitrate lyase from crude extract of Saccharomycopsis lipolytica ATCC44601 and MX9-11RX8 temperature-sensitive mutant was purified about 54 times and 87 times, respectively by ammonium sulfate fractionation, Toyo peal HW-55F gel filtration and DEAE-Cellulose ion exchange chromatography, The molecular weight of the purified isocitrate lyase from this yeast was estimated to be 230, 000 by gel filtration on Sephadex G-200, and SDS-polyacrylamide Eel electrophoresis showed that the enzyme consisted of four identical or similar subunits with a molecular weight of 59, 000 and the enzyme showed optimum activity at pH 6.9.

Numerical Identification of Streptomyces fIaveus Producing Antibiotic Substances Inhibitory to Plant Pathogenic Fungi

  • The actinomycete strain A 11 was antagonistic to plant pathogenic fungi Phytophthora capsid and Magnaporthe grisea. Based on the diaminopimelic acid (DAP) type and morphological characteristics examined by scanning electron microscopy, the strain A 11 was confirmed to belong to the genus Streptomyces. Based on Willcox probability and similarity level, the strain A 11 was numerically identified as Streptomyces flaveus using TAXON program of Ward and Goodfellow. Antibiotic production of S. flaveus strain A 11 was most favorable when cultured on glycerol yeast extract peptone (GYP) agar for 20 days at $28^{\circ}C$. The crude antibiotics from solid GYP agar cultures of the strain A 11 were most effective against Phytophthora capsici and Sclerotinia sclerotiorum among the fungi tested. Antifungal activity of the antibiotics against Alternaria solani, Botryosphaeria dothidea, Cercospora capsici, Magnaporthe grisea, and Rhizoctonia solani was somewhat high, whereas Colletotrichum gloeosporioides and Fusarium oxysporum f. sp. cucumerinum were rarely inhibited even at high concentrations.

  • Ohyaksungisan, combined preparation of crude drugs, has been used for hemiplegia, arthralgia and paralysis in traditional Korean medicine. The anti-inflammatory activity of the aqueous extract from Ohyaksungisan(OSSE) was investigated on acetic acid-induced edema and adjuvant arthritis in rats. Acute toxicity and analgesic action in mice were also examined. Its anti-inflammatory activity on 5% acetic acid-induced edema and adjuvant arthritis was observed with oral administration. The acute toxicity showed 10% mortality at 2400 mg/kg (p.o), but was not showed at 1200 mg/kg (i.p). OSSE was showed to have significant analgesic action (P<0.05) at 150 mg/kg and this action was strengthened at 300, 600 mg/kg. The anti-inflammatory effect was showed significant preventive effect on the hind paw edema from 90 min. and the adjuvant arthritis, when orally administered for 19 days. showed significant inhibitory effect on the hind paw edema from the 5th day.

  • Studies on the stem bark extracts of Garcinia cuneifolia have furnished a new xanthone cuneifolin (1) and the triterpene stigmasterol (2). Structures for these compounds were elucidated based on NMR, 2D NMR, MS and GCMS data. Larvicidal activity screening of the crude bark extract using the larvae of Aedes aegypti indicated the larvae to be susceptible to these extracts. $LC_{50}$ values of the bioassays show the extracts to be moderately toxic to the larvae of Aedes aegypti.
