  • This study was focused on the purification, characterization and promotion mode of an anticoagulant polysaccharide from Hizikia fusiforme. The anticoagulant crude polysaccharide(HF 0) was obtained by using hot water extraction at 100oC for 3 hrs after homogenizing desalted Hizikia fusiforme. The anticoagulant polysaccharide(HF 2 3 1a) was purified from the crude extract(HF 0) through stepwise gradient ethanol precipitation(HF 2), DEAE Toyopearl 650C(HF 2 3), Sephadex G 75(HF 2 3 1), Sepharose CL 6B(HF 2 3 1a) chromatography and HPLC to homogeneity. HF 2 3 1a was estimated at 5.3$\times$105 Da molecular weight and composed of fucose(51.92%), galactose(19.34%), mannose(13.92%), xylose (7.14%), arabinose(3.95%) and rhamnose(3.78%), and comprimised 29.7 % sulfate residue. The sulfated anticoagulant polysaccharide from HF 2 3 1a was proposed to inhibit via the intrinsic pathway and common pathway in the blood coagulation. The HF 2 3 1a exhibited the anticoagulant activity by activating an antithrombin III and the activity depended on the concentration of HF 2 3 1a. Acute toxicity of HF 2 in mice was not detected. Only 14 of 33 control mice(11.4%) that had taken saline survived for 30 min after injecting thrombin(100 NIH unit/ml).

  • Dried samples of Ligustrum japonicum were extracted twice: with methylene chloride and with methanol (MeOH), respectively. The combined crude extracts were successively fractionated into n-hexane, 85% aqueous methanol (85% aq.MeOH), n-butanol (n-BuOH), and water fractions by liquid-liquid partition. Antioxidant activities of crude extract and its solvent fractions were evaluated by measuring authentic $ONOO^-$, and $ONOO^-$ generated from 3-morpholinsydnonimine (SIN-1) as well as degree of occurrence of intracellular ROS in HT 1080 cells, and genomic DNA oxidation. The 85% aq.MeOH and n-BuOH fractions exhibited the good antioxidant activity. Further purification of the 85% aq.MeOH fracition led to the isolation of Oleanolic acid (1), Maslinic acid (2), and Ursolic acid (3). All compounds showed the significant antioxidant effects in all assay systems.

  • In this study, lotus root(Nelumbo nucifera G.) and omija(Schizandrae fructus), which have been used in oriental medicine and folks remedy, were examined to apply to functional foods. We prepared yanggaeng with lotus root, sugar, oligosaccharide, agar, omija extract solution and analyzed the nutritional composition(moisture, protein, fat, ash, crude fiber, carbohydrate, free sugar and minerals). Also we investigated texture profile and evaluated sensory characteristics of developed yanggaengs and 8 commercial ones. Potassium and crude fiber contents of yanggaeng with lotus root were higher than the commercial yanggaengs. Cohesiveness, springiness and gumminess of lotus root yanggaeng were the same levels of commercial yanggaengs. Sensory evaluation with the ones, showed that the lotus yanggaeng was more desirable than the commercial ones. Yanggaeng with lotus had good scores in texture profile and sensory evaluation compared with commercial yanggaengs. These results demonstrated that lotus root hadsufficient values to use a foodstuff for yanggaeng.

  • An antimicrobial substance-producing microorganism was isolated from soil samples. Based of the taxonomic characteristics of its morphological, cultural, physiological properties and 16s rRNA sequence alignment, this microorganism was identified as Pseudomonas aeruginosa, and we named Pseudomonas aeruginosa EL-KM. The optimal culture condition for production of antimicrobial substance was 1% mannitol, 0.4% yeast extract, 0.5% Nacl, 0.2% $K_2SO_4$, 100$\mu$M $MgSO_4$.$7H_2O$, 10$\mu$M $CaCl_2$.$2H_2O$, 1$\mu$M $FeSO_4$.$7H_2O$, 1$\mu$M $MnSO_4$.$4-5H_2O$, initial pH 7 and 200 rpm at 3$0^{\circ}C$. The purification of the antimicrbial substance was performed by silica gel column chromatographys, and fraction with TLC $R_f$ 0.77 value represented good antimicrobial activity. The crude antimicrbial substance was stable within a pH range of 3-10 and temperature range of 4$^{\circ}C$-121$^{\circ}C$ autoclaving. This crude antibacterial substance acted as bacteriolytic agent against Vibrio cholerae non-Ol ATCC 25872, and also exhibited excellent properties, when the substance was demonstrated against many other gram-positive, gram-negative bacteria, yeast and fungi.

  • The effect of Capricornidis Cornu (C.C) on the amount of total cholesterol, triglycerides, and phospholipid in serum from rabbit were observed. The preparations were extracted by methanol, and extracted samples were oraly adminstrated to the rabbits fasted for three days. After three days starvation, the amounts of total cholesterol and triglycerides in serum of the rabbit were markedly increased, while phospholipid was slightly decreased. The amount of total cholesterol and triglycerides were gradually decreased by adminstration of the extracted samples and were returned to the control leveles after 15 days. However, the amount of phospholipid was increased to higher than the control levels by adminstraion of the drug. According to the above results, it would be concluded that the extract of C.C. and crude drug preparation containing C.C. influences for the recovery of the increased amounts of total cholesterol and triglycerides by abnormal metabolism to the normal on rabbit.

  • Rhodococcus erythropolis amidase was expressed in Escherichia coli cells. The crude amidase in the cell-free extract was immobilized using the cross-linked enzyme aggregate (CLEA) method. The crude amidase was mixed with bovine serum albumin and then precipitated with ammonium sulfate. The resultant precipitant was subsequently cross-linked with glutaraldehyde. Scanning electron microscopy revealed that this co-CLEA had a ball-like shape with a diameter of approximately $1\;{\mu}m$. This co-CLEA evidenced hydrolytic activity toward a variety of amide substrates. The amidase co-CLEA evidenced an optimum temperature of $60^{\circ}C$ and an optimum pH of 8.0, results that were similar to those of the soluble amidase. The reaction stability of the co-CLEA was increased. That is, it was stable up to $50^{\circ}C$ and in a pH range of 5.0-12.0. Additionally, the co-CLEA could be recovered by centrifugation, and retained 96% activity after 3 repeated cycles. This amidase co-CLEA may prove useful as a substitute for soluble amidase as a biocatalyst in the pharmaceutical and chemical industries.
