Studies on the Tick Killing and Repellent Effects of Two Korean Indigenous Crude Drugs, Radix Jingyu and Fructus Ponciri (두가지 국산생약(國産生藥), 진규와 지실(枳實)의 소진드기 구제(驅除)(살충(殺蟲) 및 기피(忌避)) 효력(效力)에 관한 연구(硏究))

  • Decoctions of two Korean indigenous crude drugs. Radix Jingyu(the dried root of Aconitum pseudo-laeve Nakai var. erectum Nakai form. Genuinum Nakai) and Fructus ponciri (the transversely sliced and dried unripe fruit of Poncirus trifoliata Rafinesque), are being utilized by the inhabitants in certain parts of southeastern district of Korea with an empirical belief that these serve as effective insecticides for cattle ticks. Of the two species of cattle ticks so far identified in Korea, Boophilus microplus and Haemaphysalis bispinosum, the former was employed in this experiment as the species occupies nearly 99 percent of the tick population. The results of the experiments herein reported seen to indicate that although the killing power of aqueous extracts of the drugs is minimal, the repellent effect against the ticks, especially of Radix Jingyu, is excellent. It may be of interest to note that the aqueous extract of Radix Jingyu has not proved to be inferior to any of the most effective cattle repellents commercially available. Further studies on these Korean indigenous crude drugs as cattle tick repellents are indicated to find whether active ingredients can be extracted in a hope to materialize an extensive application of these drugs in the field.

A study on the extracting characteristics of velvet antlers using kyenegum protease (계내김(鷄內金)에서 추출한 protease를 이용한 녹용(鹿茸) 추출 특성 연구)

  • Objective : Kyenegum has been frequently used for characterizing digestive symptoms in the traditional and oriental medicines. This study was conducted to investigate the characteristics of extracts from velvet antlers using the 4 different kinds of extracting methods. Methods : The extracts of velvet antlers were extracted using a $65^{\circ}C$ DW (9hrs), a Kyenegum crude enzyme, a $121^{\circ}C$ DW (2hrs), and a Kyenegum protease. To evaluate the characteristic of velvet antler extracts, we examined the brix, soluble solid, amino acid, mineral composition, and collagen protein. Results : As a result of the comparisons of velvet antlers extracted by the traditional extraction and the crude enzyme of kyenegum, the brix and soluble solid showed the higher contents for kyenegum enzymes. Also, mineral contents of the extracted velvet antlers were higher, particularly in Ca and P for those. The contents of collagen protein, hydroxyproline and hydroxylysine, were found to be more than twice in kyenegum protease compared with other extracting methods. Conclusion : These results indicated that the Kyenegum crude enzyme and protease are very effective to extract of velvet antlers.

Biochemical and Histological Charaeteristics of Inferior Red Ginseng (불량홍삼(내백삼)의 생화학적 및 조직학적 특성)

  • In order to investigate the inferior factor of red ginseng quality, the contents of various chemical components, physico-chemical properties and arrangement state of ginseng cells were observed. Contents of total reducing sugar, reducing sugar, crude protein, crude fibre and specific gravity of inside white part of red ginseng were less than those of normal part. But differences in content of crude saponin, HPLC pattern of ginsenosides and reducing ability for DP P H(1,1-dipheny 1-2-picrylhydrazyl) between normal and inside white part of red ginseng were not found. The optical density of 1 water extract of normal part of red ginseng did not differ from that of inside white 1 part of red ginseng, but the visible and UV absorbance of acid hydrolyzate of normal red ginseng showed higher than those of inside white part of red ginseng. The differences in the internal color and tissue of normal and inside white part of red ginseng were easily found with naked eye, and by the microscopic fractography, the orangement state of ginseng cell in the inside white part of red ginseng was less dense than that in normal red ginseng.

Identification of Allergens in Pork Meat (돼지고기 중 알레르기 유발성분의 동정)

  • The pork meat has been reported as one of the food occurring allergic reactions predominantly to korean. To identify the potential food allergens in pork meat, sera were collected from 25 allergic patients to the pork meat and 10 allergic patients not to pork meat as well as 5 normal subjects after skin prick test and open food challenge test. Crude extracts were prepared by blending raw pork meat in phosphate buffered saline (pH 7.0) and the heat treatment on crude extracts was carried to characterize sensibility of the allergens to heat. ELISA was performed to determine specific IgE antibody levels of allergic patients to pork meat, and resulted in twofold higher mean value than that of tolerated patients. Extracted proteins from pork meat was separated with SDS-PAGE followed by immunoblotting using sera from pork sensitive patients and control subjects, respectively. The IgE binding response to pork meat by immunobots correlated with quantitative specific IgE value of each person. Immunoblots showed four prominent IgE-binding bands (66, 60, 50, 44 kDa) in crude extract, but two bands of those (60, 44 kDa) were heat-labile. These results suggest that most prominent allergens from pork meat are four components(66, 60, 50, 44 kDa) in korean and the heat treatment on allergen is additional parameter to characterize allergen.

  • From crude drug preparation(Soshiho-Tang) ginseng sapogenins were identified by TLC and $ginsenoside-Rb_1$ was determined quantitatively by HPLC. Panaxadiol, pandaxatriol, acid-hydrolysates of ginseng saponin, were identified by TLC with benzene/acetone(4 : 1, v/v). Rf values of which were measured as 0.26 and 0.14, respectively. The content of $ginsenoside-Rb_1$ was determined by HPLC on $Lichrosorb-NH_2$ column with $CH_3CN/H_2O/n-BuOH$(80 : 20 : 10, v/v). Its recovery rate in the extract granules, was as relatively low as $19.8{\pm}1.4%$ compared to the content in raw red ginseng.

Effects of Ganoderma lucidum Extract on Production of Fatty Acids by Rhodotorula glutinis (영지 열수추출물이 Rhodotorula glutinis의 지방산 생성에 미치는 영향)

  • Rhodotorula glutinis was inoculated in the medium containing 0 ($S_0$), 0.01 ($S_1$), 0.1 ($S_2$) and 1.0% ($S_3$) Ganodoma lucidum extract and incubated in a shaking incubator at $30^{\circ}C$ for 8 days and the cell growth, sugarity, lipid content, and fatty acid composition were measured to investigate the effects of G. lucidum extract on the growth and biosynthesis of fatty acids by oleaginous yeast, Rhodotorula glutinis. After the 8 day incubation, the cell growth of $S_1,\;S_2\;and\;S_3$ increased 1.6, 1.7 and 2.1 times, respectively, than that of $S_0$. Sugar consumption of incubating medium was decreased but the crude lipid content in R. glutinis was increased with increase of the amount of G. Lucidum extract. Myristic, palmitic, stearic, oleic and linoleic acids were identified by GC as the major fatty acids in the crude lipid produced by R. glutinis and the content of unsaturated fatty acids (38.6 mg/g) was greater than that of saturated fatty acids (22.3 mg/g). As the G. lucidum extract concentration increased, fatty acids contents were increased except myristic acid, and the most increase occurred at the addition of 0.1% while they were considerably decreased in the case of the addition of 1.0% G. lucidum extract.

A Study on the Cytotoxic Activity of Garlic(Allium Sativum) Extract Against Cancer Cells (마늘중 지용성 성분의 암세포증식 억제효과 연구)

  • This study was devised to observe the cytotoxic activities of garlic extracts against various cancer cells, that is, murine leukemic lymphocyte(L1210 and P388) and human rectal(HRT-18) and colon cancer cells(HCT-48 and HT-29) in vitro, and murine ascitic tumor cell(S-180) in vivo. Each cell-line except S-180 was cultured in medium containing serial concentration of the garlic extract in vitro. Inhibitory effect n the growth rate of the cancer cells was stronger in extracts of petroleum ether than that of ethanol. A lipid soluble compound in the extracts of garlic was cytocidal to murine leukemic cells, human rectal and colon cancer cells in vitro. The growth rates of the cancer cells in medium containing garlic extracts were inhibited gradually to a significant degree in proportion to the increase of the extract concentration. The cytotoxic activity of garlic active fraction from TLC was about 2.3 times more potent than that of crude garlic extract, one unit of cytotoxic activity against L1210 cells being equivalent to 4.2$\mu\textrm{g}$ and 1.8$\mu\textrm{g}$ from the crude garlic and active fraction, respectively. The Rf value of the active fraction on silica-gel TLC was 0.18 in condition that petroleum ether/ethyl ether/acetic acid mixture(90:10:1, v/v/v) was used as a developing solvent. The survival times of mice inoculated with S-180 cells were extended about 1.5 to 2 times in the groups treated with garlic extract(through i.p. and oral administration) compared with their control group(no garlic extract treatment). Observations were carried out on S-180. Ethanol extracts of garlic injured markedly tumor cells within 3 hours after injection.

  • A study was designed to produce a self-stable and clear Red Ginseng wine using ethanol solution for Red Ginseng extraction and low temperature treatment on the extract before packaging. Extraction of Red Ginseng at high temperature using low ethanol concentration caused an increase in the amount of crude Red Ginseng extract and a decrease of purified Red Ginseng extract. Extraction with 75% ethanol at $50{\sim}70^{\circ}C$, followed by dilution of the extract to the ethanol concentration of final product, resulted minimum precipitation in the Red Ginseng wine. Low temperature treatment at $4^{\circ}C$ retained all of the saponin content in crude extract while $-20^{\circ}C$ treatment caused a significant loss of saponin along with precipitation during purification.

  • This experiment was carried out to determine the relationships between maturity and content of the chemical components in flue-cured tobacco. The results were as follows : 1) Among the chemical components, the contents of total nitrogen, ammonia nitrogen, total alkaloids, nicotine, total volatile base, petroleum-ether extract, crude fiber, crude ash, water soluble ash, and chlorine decreased significantly, and those of total sugar, reducing sugar, and starch increased as leaf maturity progressed until the stage of ripeness, but, afterwards, a reverse tendency appeared. 2) There was a significant relationship between maturity and the contents of chemical components, such as total nitrogen, protein nitrogen, ammonia nitrogen, total volatile base, and the correlation coefficients of them were -0.743, -0.789, -0.797, and -0.642, respectively.

  • The in vitro anticoagulant and fibrinolytic activities of crude extract from insects which have been used as traditional medicines. The extracts of Formica, Huechys and Eupolyph-aga/Steleophaga prolonged activated partial thromboplastin time and thrombin time compared to the value of the control. The fibrinolytic activity of insect extracts was also tested by fibrin plate method. We found that the extracts of Cicadae Periostracum, Eupolyphaga/Steleophdga, Mantidis $O{\ddot{o}}otheca$ and Huechys directly could hydrolyze the fibrin clot without the activation of plasminogen by plasminogen activators.
