  • Anemarrhenae Rhizoma has been clinically used as a sedative drug in oriental medicine. We observed the sedative action and effect on the movement of isolated-ileum with each water extract of Anemarrhenae Rhizoma (F-I), saline solution treated preparation (F-II) and 25% ethanol treated preparation (F-III). The results obtained were summerized as follows; 1) Sedative actions were recognized with F-I, F-II and F-III. 2) Prolonged effect of sleeping time was recognized significantly with F-I, F-II and F-III. 3) Each fraction of Anemarrhenae Rhizoma showed the released action on the isolated-ileum of the mouse, rat, rabbit and guinea pig, and antagonic action against contraction induced by acetylcholine, barium chloride and histamine. Thus the mechanism in the released action of the smooth muscle was partly considered as the direct dependence of the smooth muscle.

  • In this study, we investigated the potential of Resina Pini (RP) for anti-inflammatory and analgesic agents to treat inflammatory diseases such as gingivitis and periodontitis. Crude RP (RP1), recrystallized RP (RP2), and Ramus Mori Albae-treated RP (RP3), plus their respective water extracts (RP1-WE, RP2-WE and RP3-WE) were prepared for in vitro and in vivo tests. We couldn't find any signs of heavy metals pollution in all the RP samples. RP2-WE exhibited the highest viability of human gingival fibroblasts (HGF) and the strongest scavenging activity on superoxide anion. RP1, RP2 and RP3, RP2 showed potent scavenging activity on DPPH free radical. RP2-WE displayed a stronger inhibition on hyaluronidase (HAase) activity and RP3 also displayed potent HAase inhibition. RP2-WE, RP3-WE, RP3 and RP2 were reduced admirably the production of $PGE_2$ in HGF. In addition, RP2-WE and RP3-WE exhibited potent inhibitory activities on arachidonic acid-induced ear edema in mouse. Moreover, RP-2 prevented completely acetic acid-induced writhing by 100.0% and RP1, RP3, RP1-WE and RP2-WE also exhibited excellent protective activities against writhing. While aminopyrine, the positive control, showed 76.9% analgesic effect at the same dose. Taken together, these results suggest that recrystallized aqueous extract of Resina Pini could be a promising drug for the treatment of periodontal diseases.

  • In order to investigate the effect of 'Whangryonhaedok-Tang' on the anti-inflammatory, ileum of mice, blood pressure, blood glucose, total cholesterol and antibacterial activity, those pharmacological actions were conducted. The results of this studies were summarized as follows; Rat hind paw edema induced by acetic acid was inhibited. In mice, tone of intestinal smooth muscle was suppressed with the treatment of sample and intestinal contractions induced by $BaCl_2$ was inhibited. Hypertensive effect was not observed in rabbits but was observed by the water extract of 'Whangryonhaedok-Tang' excluded Phellodendri Cortex in rabbits. The increased blood glucose levels in alloxandiabetic rabbits were significantly decreased by the oral administration of the sample I after 6 days. The total cholesterol levels in alloxan-induced rabbits were significantly decreased by the oral administration of the sample I after 9 days. Antibacterial activity was observed. In addition, we thought the pharmacological actions were significantly noted by the sample III (Methanol soluble fraction) because of the purification of effective components.

  • Allergic diseases are the result of Th2-dominated responses to single or multiple environmental allergens. Th2 cytokines regulate these mechanisms of allergic disease at many levels, including initiation, progression, and persistence. The effect of hwangryun-Hae-Dok-Tang (HRHDT) on tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) and interleukin-4 (IL-4) stimulated inflammation was investigated using 3-(4,5-dimethylthiazol-2-yl)-2,5- diphenyltetrazolium bromide assay, thymus and activation-regulated chemokine (TARC), eotaxin, regulated on activation normal T cells expressed and secreted (RANTES) immunoassay on the human bronchial epithelial microglial cells. From the present study, the crude extract of Hwangryun-Hae-Dok-tang suppressed the TNF-${\alpha}$ and IL-4 stimulated TARC, eotaxin, and RANTES production in the human bronchial epithelial A549 cells. Based on the present results, Hwangryun-Hae-Dok-tang may be useful in the treatment asthmatic allergy by inhibiting TARC, eotaxin, and RANTES chemokines.

  • The identification and characterization of antigens that elicit human T cell responses is an important step toward understanding of Leishmania major infection and ultimately in the development of a vaccine. Micropreparative SDS-PAGE followed by electro transfer to a PVDF membrane and elution of proteins from the PVDF, was used to separate 2 novel proteins from L. major promastigotes, which can induce antibodies of the IgG2a isotype in mice and also are recognized by antisera of recovered human cutaneous leishmaniasis subjects. Fractionation of the crude extract of L. major revealed that all detectable proteins of interest were present within the soluble Leishmania antigens (SLA). Quantitation of these proteins showed that their expression in promastigotes is relatively very low. Considering the molecular weight, immunoreactivity, chromatographic and electrophoretic behavior in reducing and non-reducing conditions, these proteins are probably 2 isoforms of a single protein. A digest of these proteins was resolved on Tricine-SDS-PAGE and immunoreactive fragments were identified by human sera. Two immunoreactive fragments (36.4 and 34.8 kDa) were only generated by endoproteinase Glu-C treatment. These immunoreactive fragments or their parent molecules may be ideal candidates for incorporation in a cocktail vaccine against cutaneous leishmaniasis.

  • IAA biosynthetic pathways in a basidiomycetous yeast, Rhodosporidiobolus fluvialis DMKU-CP293, were investigated. The yeast strain showed tryptophan (Trp)-dependent IAA biosynthesis when grown in tryptophan supplemented mineral salt medium. Gas chromatography-mass spectrometry was used to further identify the pathway intermediates of Trpdependent IAA biosynthesis. The results indicated that the main intermediates produced by R. fluvialis DMKU-CP293 were tryptamine (TAM), indole-3-acetic acid (IAA), and tryptophol (TOL), whereas indole-3-pyruvic acid (IPA) was not found. However, supplementation of IPA to the culture medium resulted in IAA peak detection by high-performance liquid chromatography analysis of the culture supernatant. Key enzymes of three IAA biosynthetic routes, i.e., IPA, IAM and TAM were investigated to clarify the IAA biosynthetic pathways of R. fluvialis DMKU-CP293. Results indicated that the activities of tryptophan aminotransferase, tryptophan 2-monooxygenase, and tryptophan decarboxylase were observed in cell crude extract. Overall results suggested that IAA biosynthetic in this yeast strain mainly occurred via the IPA route. Nevertheless, IAM and TAM pathway might be involved in R. fluvialis DMKU-CP293.

  • Ginger (Zingiber officinale Roscoe) is widely used as a common condiment for a variety of foods and beverages. In addition to its extensive utilization as a spice, the fresh or the processed rhizome is a useful crude drug in traditional Chinese medicine. It is considered to possess stomachic, carminative, stimulant, diuretic and antiemetic properties. Chemical studies on the pungent principles of ginger have been carried out by a number of investigators, and 6-gingerol and 6-shogaol as a major pungent substance have been isolated. In this study, the constituents inhibiting a drug metabolizing enzyme CYP3A4 from ginger were investigated. CYP3A4 is responsible for drug metabolism as heme-containing monooxygenases. As a result of experiment, 10-gingerol (lC$_{50}$ 5.75$\mu$M) isolated from EtOAc extract of ginger showed remarkable inhibitory activity compared to 6-gingerol ($IC_{50}$/ 14.56 $\mu$M) and zingerone ($IC_{50}$/ 379.63 $\mu$M). This paper describes the isolation, structure elucidation, and CYP3A4 inhibitory activity of these compounds. The structure of the compounds were identified by instrumental analysis such as LC-mass spectrometer and NMR.R.

  • The HCL hydrolyzate of the non-histone protein fractionated from the rat liver nuclei which have been incubated inthe presence of S-adenosyl-L-[methyl-$^{14}C$ ]-methionine shows at least four unidentified radioactive peaks on a basic amino acid analysis chromatogram. One of these unknown compounds (designated as compound 3) is also formed by the rat liver homogenated with the exogenous addition of an appropriate protein substrate. Since boiled rat liver homogenate or fresh homogenate in the absence of an exogenous protein substrate failed to form compound 3, its formation can be considered to be enzyme-catalyzed. The enzyme which yields compound 3 shows a preference of protein substrate in the order of reductively methylated hemoglobin > native > histone type II-A. The rat enzyme is nuclear in location associated with chromatin, and exhibits the highest activity in the liver among various rat organs. A compound 3-forming enzyme is also present in Neurospora crassa, since endogenous formation of the compound 3 can be demonstrated with the crude extract of this mold. The chemical identity of compound 3 is not yet known. However, it resisted to the following treatments; 6 N HCL and 0.1 N Na NaOH hydrolysis at $110^{\circ}C$, OR L-amino acid oxidase.
