Colletotrichum Diversity within Different Species Complexes Associated with Fruit Anthracnose in South Korea and Their Fungicides In-Vitro Sensitivity (국내 과실 탄저병을 일으키는 종 복합체와 종 다양성 및 살균제 감수성)

  • Anthracnose, caused by the Colletotrichum genus, comprises a significant number of plant pathogens and poses a major threat to fruit production worldwide, including South Korea. Colletotrichum species were identified associated with anthracnose in fruits such as apple, persimmon, plum, peach, jujube, walnut, and grape. A polyphasic approach, including morphology, multigene phylogenetics, and pathogenicity testing, was used. Additionally, the in-vitro sensitivity of identified Colletotrichum species to common fungicides was also evaluated. A total of nine Colletotrichum species within two complexes, namely gloeosporioides and acutatum, have been identified as the causal agents of anthracnose in common fruits in South Korea. In the gloeosporioides complex, we found Colletotrichumaenigma, C. fructicola, C. gloeosporioides, C. horii, C. siamense, and C. viniferum. Meanwhile, in the acutatum complex, C. fioriniae, C. nymphaeae, and C. orientalis were identified. Notably, C. fructicola, C. siamense, C. fioriniae, and C. nymphaeae were reported for the first time from apple, C. siamense, C. fioriniae and C. nymphaeae from plum, C. siamense, C. fructicola, and C. fioriniae frompeach, C. siamense and C. horii from persimmon, C. fioriniae from Omija (Schisandra), C. orientalis from walnut, C. nymphaeae from jujube, and C. aenigma, C. fructicola, and C. siamense fromgrape. Fungicide sensitivity tests revealed significant variation in the EC50 values among specific Colletotrichum species when exposed to different fungicides. Moreover, the same Colletotrichum species isolated from different host plants displayed varying sensitivity to the same fungicide.

Nematicidal Activity of Streptomyces flavogriseus KRA15-528 to Meloidogyne incognita (Meloidogyne incognita에 대한 Streptomyces flavogriseus KRA15-528의 살선충활성)

  • Plant disease caused by root-knot nematode is a major problem in crop production. Using of chemical pesticides, one of the most efficient methods to control nematodes, have raised issues in toxicity to humans and animals and environmental pollution. In this study, to select actinomycete strains that have potential to serve as a microbial agent for control of nematodes, we investigated nematicidal activity of culture broth from 670 Streptomyces isolates. A culture filtrate of KRA15-528 isolate that was identified as S. flavogriseus on the basis of 16S rRNA sequence analysis, showed strong nematicidal activity against second stage of juveniles of Meloidogyne incognita and inhibited egg hatching; exposure to 10% of culture filtrate resulted in 71% juvenile mortality at 48 hours afters treatment and suppressed egg hatching by 54% at 9 days after treatment. When the KRA15-528 culture filtrate was partitioned with ethyl acetate and butanol, ethyl acetate layer exclusively showed strong activity; 91%, 53%, 30% of mortality at 1,000, 500, $250{\mu}g/ml$, respectively. Additionally, the culture filtrate suppressed gall formation on cucumber plant by M. incognita with no phytotoxicity. These results suggest that S. flavogriseus KRA15-528 has potential to serve as a microbial nematicide for the control of root-knot nematode disease.

Effect of Microbial Agent on Control of Tomato Gray mold and Powdery mildew (미생물제 처리에 의한 토마토의 잿빛곰팡이병과 흰가루병의 방제효과)

  • In vivo experiments were carried out to investigate the effect of microbial agents on tomato disease occurrences in a sustainable tomato farm in Gong-ju, Chung-nam during 2 years (2010-2011). Two kind of commercial microbial agents (EXTN-1 and Cillus suspension concentrate) and 2 kind of microbial agents (Bacillus subtilis strain 'B4' and 'B17') developing by National academy of agricultural science were used in this study. In the 1st experimental year, the microbial agents were sprayed on leaves for 2-4 times at 1-2 weeks interval from 3 weeks after transplanting, and then disease occurrence were observed. As a result, the control efficacies of commercial EXTN-1 and Cillus suspension concentrate (SC) against gray mold were 59.6% and 50.5% in the treatment of 4 times foliar application at 1 week interval from 3 weeks after transplanting, and the control efficacy of commercial EXTN-1 SC against gray mold was 55.4% in the treatment of 3 times foliar application at 1 week interval from 4 weeks after transplanting. However, the control efficacies of EXTN-1 and Cillus SC were not enough for tomato crop protection against powdery mildew. In the 2nd experimental year, the microbial agents were treated following by the planned combination schedule of transplanting stage drip-watering, early stage root irrigation and early blooming stage foliar application. The control efficacies of EXTN-1 SC and B17 treatment following by transplanting stage drip-watering, 2 times root irrigation at 2 weeks interval root irrigation and 4 times foliar application at 1 weeks interval for gray mold management were 57.0 and 55.1%, respectively. In the case of same treatment for gray mold management mentioned at above, the control efficacies of EXTN-1 SC, B4 and B17 treatment for powdery mildew management were 50.5, 51.3 and 52.5%, respectively.

The Tresnds of Artiodactyla Researches in Korea, China and Japan using Text-mining and Co-occurrence Analysis of Words (텍스트마이닝과 동시출현단어분석을 이용한 한국, 중국, 일본의 우제목 연구 동향 분석)

  • Artiodactyla, which is an even-toed mammal, widely inhabits worldwide. In recent years, wild Artiodactyla species have attracted public attention due to the rapid increase of crop damage and road-kill caused by wild Artiodactyla such as water deer and wild boar and the decrease of some species such as long-tailed goral and musk deer. In spite of such public attention, however, there have been few studies on Artiodactyla in Korea, and no studies have focused on the trend analysis of Artiodactyla, making it difficult to understand actual problems. Many recent studies on trend used text-mining and co-occurrence analysis to increase objectivity in the classification of research subjects by extracting keywords appearing in literature and quantifying relevance between words. In this study, we analyzed texts from research articles of three countries (Korea, China, and Japan) through text-mining and co-occurrence analysis and compared the research subjects in each country. We extracted 199 words from 665 articles related to Artiodactyla of three countries through text-mining. Three word-clusters were formed as a result of co-occurrence analysis on extracted words. We determined that cluster1 was related to "habitat condition and ecology", cluster2 was related to "disease" and cluster3 was related to "conservation genetics and molecular ecology". The results of comparing the rates of occurrence of each word clusters in each country showed that they were relatively even in China and Japan whereas Korea had a prevailing rate (69%) of cluster2 related to "disease". In the regression analysis on the number of words per year in each cluster, the number of words in both China and Japan increased evenly by year in each cluster while the rate of increase of cluster2 was five times more than the other clusters in Korea. The results indicate that Korean researches on Artiodactyla tended to focus on diseases more than those in China and Japan, and few researchers considered other subjects including habitat characteristics, behavior and molecular ecology. In order to control the damage caused by Artiodactyla and to establish a reasonable policy for the protection of endangered species, it is necessary to accumulate basic ecological data by conducting researches on wild Artiodactyla more.

Monitoring the Reoccurrence of Fire Blight and the Eradication Efficiency of Erwinia amylovora in Burial Sites of Infected Host Plants Using Sentinel Plants (미끼식물을 이용한 화상병 감염 기주 매몰지 내 화상병균 제거 효율 검증 및 병 재발 모니터링)

  • The fire blight caused by Erwinia amylovora (Ea) was first reported in 2015 in Korea, and the disease has rapidly spread to 22 regions until 2021. In Korea, all host plants in the apple and pear orchards where fire blight occurred should be eliminated and buried by the Plant Protection Act. To prevent the spread of the disease, all burial sites were prohibited from planting the new host plants for the next three years. To confirm the eradication efficiency of Ea and the reoccurrence of fire blight, the surveillance facilities were established on three burial sites from 2019 to 2020 in Anseong-si, Gyeonggi-do, and Chungju-si, Chungcheongbuk-do. As host plants, five apple trees of fire blight-susceptible cultivar 'Fuji', were planted in each facility. All facilities were enclosed with fences and nets and equipped with two CCTVs, motion sensors, and several other sensors for recording weather conditions to monitor the environment of the sentinel plants in real-time. The sentinel plants were checked for the reoccurrence of fire blight routinely. Suspicious plant parts were collected and analyzed for Ea detection by loop-mediated isothermal amplification polymerase chain reaction and conventional polymerase chain reaction. Until November 2022, Ea has not been detected in all sentinel plants. These results might support that the burial control of infected plants in soil works efficiently to remove Ea and support the possibility to shorten the prohibition period of host plant establishment in the burial sites.

Studies on Garlic Mosaic Virus -lts isolation, symptom expression in test plants, physical properties, purification, serology and electron microscopy- (마늘 모자이크 바이러스에 관한 연구 -마늘 모자이크 바이러스의 분리, 검정식물상의 반응, 물리적성질, 순화, 혈청반응 및 전자현미경적관찰-)

  • Garlic (Allium sativum L.) is an important vegetable crop for the Korean people and has long been cultivated extensively in Korea. More recently it has gained importance as a source of certain pharmaceuticals. This additional use has also contributed to the increasing demand for Korean garlic. Garlic has been propagated vegetatively for a long time without control measures against virus diseases. As a result it is presumed that most of the garlic varieties in Korea may have degenerated. The production of virus-free plants offers the most feasible way to control the virus diseases of garlic. However, little is known about garlic viruses both domestically and in foreign countries. More basic information regarding garlic viruses is needed before a sound approach to the control of these diseases can be developed. Currently garlic mosaic disease is most prevalent in plantings throughout Korea and is considered to be the most important disease of garlic in Korea. Because of this importance, studies were initiated to isolate and characterize the garlic mosaic virus. Symptom expression in test plants, physical properties, purification, serological reaction and morphological characteristics of the garlic mosaic virus were determined. Results of these studies are summarized as follows. 1. Surveys made throughout the important garlic growing areas in Korea during 1970-1972 revealed that most of the garlic plants were heavily infected with mosaic disease. 2. A strain of garlic mosaic virus was obtained from infected garlic leaves and transmitted mechanically to Chenopodium amaranticolor by single lesion isolation technique. 3. The symptom expression of this garlic mosaic virus isolate was examined on 26 species of test plants. Among these, Chenopodium amaranticolor, C. quince, C. album and C. koreanse expressed chlorotic local lesions on inoculated leaves 11-12 days after mechanical inoculation with infective sap. The remaining 22 species showed no symptoms and no virus was recovered from them whet back-inoculated to C. amaranticolor. 4. Among the four species of Chtnopodium mentioned above, C. amaranticolor and C. quinoa appear to be the most suitable local lesion test plants for garlic mosaic virus. 5. Cloves and top·sets originating from mosaic infected garlic plants were $100\%$ infected with the same virus. Consequently the garlic mosaic virus is successively transmitted through infected cloves and top-sets. 6. Garlic mosaic virus was mechanically transmitted to C, amaranticolor when inoculations were made with infective sap of cloves and top-sets. 7. Physical properties of the garlic mosaic virus as determined by inoculation onto C. amaranticolor were as follows. Thermal inactivation point: $65-70^{\circ}C$, Dilution end poiut: $10^-2-10^-3$, Aging in vitro: 2 days. 8. Electron microscopic examination of the garlic mosaic virus revealed long rod shaped particles measuring 1200-1250mu. 9. Garlic mosaic virus was purified from leaf materials of C. amaranticolor by using two cycles of differential centrifugation followed by Sephadex gel filtration. 10. Garlic mosaic virus was successfully detected from infected garlic cloves and top-sets by a serological microprecipitin test. 11 Serological tests of 150 garlic cloves and 30 top-sets collected randomly from seperated plants throughout five different garlic growing regions in Korea revealed $100\%$ infection with garlic mosaic virus. Accordingly it is concluded that most of the garlic cloves and top-sets now being used for propagation in Korea are carriers of the garlic mosaic virus. 12. Serological studies revealed that the garlic mosaic virus is not related with potato viruses X, Y, S and M. 13. Because of the difficulty in securing mosaic virus-free garlic plants, direct inoculation with isolated virus to the garlic plants was not accomplished. Results of the present study, however, indicate that the virus isolate used here is the causal virus of the garlic mosaic disease in Korea.

Yield and Dry Matter at Developed Hybrid Corns Using CNU and SK Inbred Lines (사료용 교잡종 옥수수 연구 -III. CNU와 SK계통을 이용한 교잡종 옥수수의 건물중 및 종실 수량)

  • In order to develop the crude forage maize hybrids with high dry matter and grain yield, several hybrids evaluated as superior hybrids at three regions in 2001. They were planted and surveyed again for second selection in 2002. The selected hybrids showed large difference in some major characteristics according to planting areas and dates(Suwon ; April 25, Daejeon ; May 15 and Milyang ; May 25), but insect and disease injury were similar to check. Fresh and dry weight of the H67 hybrid at Suwon and Daejeon was the highest among hybrids including check, but low in Milyang due to raining after flowering. Grain yield was higher than check in H15 at Suwon, H45 and H37 at Daejeon and H67 at Milyang.

Biological control of Gray Mold Rot of Perilla Caused by Botrytis cinerea II. Formulation of Antagonistic Bacteria and Its Control Effect (들깨 잿빛곰팡이병의 생물학적 방제 II. 미생물농약의 제조 및 그 방제효과)

  • An antagonistic bacteria, Bacillus licheniformis Nl strain which effectively inhibited mycelial growth of gray mold rot pathogen, Botrytis cinerea was isolated from the rhizosphere of perilla crop. Powder soy formulation by B. lichentfomis Nl strain as a biocontrol agent was developed far the first time and estimated its control effect on perilla leaves in this study. First of all, far the mass production of antifungal metabolites of B. lichentfomis Nl strain in flask liquid culture, the most effective carbon and nitrogen source were selected as glucose and tryp-tone, respectively, For the formulation, vegetative biomass of B. licheniformis Nl strain from 5-day-old liquid culture in nutrient broth added glucose and tryptone was mixed with soy flour, rice flour glucose, FeSo$_4$~7$H_2O$, and MnCl$_2$. 4$H_2O$, and dried and pulverized. In plastic house test, powder soy formulation effectually controlled gray mold rot as the control value of 93.1 %, was more effective than chemical fungicide, benomyl showing the control value of 86.1%. Thus, development of powder soy formulation of B. lichentfomis Nl will aid large-scale application of biological control in field trials.

Genomics and Molecular Markers for Major Cucurbitaceae Crops (주요 박과작물의 유전체 및 분자마커 연구 현황)

  • Watermelon and melon are economically important Cucurbitaceae crops. Recently, the development of molecular markers based on the construction of genetic linkage maps and detection of DNA sequence variants through next generation sequencing are essential as molecular breeding strategies for crop improvement that uses marker-assisted selection and backcrossing. In this paper, we intended to provide useful information for molecular breeding of watermelon and melon by analyzing the current status of international and domestic research efforts on genomics and molecular markers. Due to diverse genetic maps constructed and the reference genome sequencing completed in the past, DNA markers that are useful for selecting important traits including yield, fruit quality, and disease resistances have been reported and publicly available. To date, more than 16 genetic maps and loci and linked markers for more than 40 traits have reported for each watermelon and melon. Furthermore, the functional genes that are responsible for those traits are being continuously discovered by high-density genetic map and map-based cloning. In addition, whole genome resequencing of various germplasm is under progress based on the reference genome. Not only by the efforts for developing novel molecular markers, but application of public marker information currently available will greatly facilitate breeding process through genomics-assisted breeding.

Comparative Assay Fungus Population and Resistant Genes about Magnaporthe grisea between Korea and China (한국·중국 벼 도열병균 생리형 분석 및 품종 저항성 특성 비교)

  • One hundred isolates of Magnaporthe grisea from Korea and China were characterized for pathogenicity using eight Korean differential varieties(KDV), six Chinese differential varieties(CDV) and six near isogenic lines(NILs) developed in China. The restriction length polymorphism of M. grisea isolates from each country also was analyzed using MGR586 as a probe. One hundred Korean isolates classified into 17 races on KDV were grouped into 29 pathotypes on Chinese near isogenic lines(NILs). Virulence of 46% of Korean isolates against all the six Chinese NILs indicated that the current six Chinese NILs alone was not enough to be used as differential varieties in Korea. Especially, susceptibility of the BL1 carrying resistance gene Pi-b to 70% of tested Korean isolates suggested that BL1(Pi-b) may not be a useful resistance source to Korean blast. Based on the virulence assays of M. grisea populations from each country were divided into two groups. About 50% of Chinese isolates showed similarity to the 30% of the Korean isolates. Especially, the isolates from northern part of China, where Japonica rice cultivars were grown, showed high similarity to the Korean isolates, while isolates from southern part of China, where Indica rice were mainly grown, showed low similarity to Korean isolates. The genome RFLPs of Korean isolates were quite different from those of southern part of China using MGR586 as a probe. These data indicated that the physiological and genetical characteristics of M. grisea population might be determined by strong interaction with cultivated rice.
