Assaying Mitochondrial COI Sequences and Their Molecular Studies in Hexapoda, PART I: From 2000 to 2009 (육각강에서 보고된 미토콘드리아 COI 염기서열과 이들을 이용한 분자 연구 논문 분석, 파트 I: 2000년~2009년)

  • Since 2000, a large number of molecular studies have been conducted in Hexapoda with generating large amount of mitochondrial sequences. In this study, to review mitochondrial COI sequences and their molecular studies reported in Hexapoda from 2000 to 2009, 488 molecular studies conducted based on 58,323 COI sequences were categorized according to 26 orders and the positions of COI sequences (5', 3', and entire regions). The numbers of molecular studies in which the three regions utilized varied largely among the 26 orders; however, seven orders showed preferred positions of COI sequences in the researches: Diptera and Orthoptera revealed the largest number of studies in the 5' region; while, Coleoptera, Phthiraptera, Odonata, Phasmatodea, and Psocoptera, showed the largest number of studies in the 3' region. From comparing 84 molecular studies published before 2000, we observed the possibilities that molecular studies in Coleoptera, Diptera, Phthiraptera, and Phasmatodea from 2000 to 2009 had been followed classical studies using the positions of COI sequences well-known until 1999. This study provides useful information to understand the overall trends in COI sequence usages as well as molecular studies conducted from 2000 to 2009 in Hexapoda.

Major character analysis of CAX 1 (cation exchanger 1) transgenic rice plants in In Vivo (CAX 1 형질전환체 벼의 In Vivo에서 주요특성 분석)

  • This study was carried out to develop transgenic rice cultivars with the CAX1 (accession no. U57411) gene. We successfully selected the transgenic rice plants over-expressing the Arabidopsis H+/$Ca^{2+}$ antiporter CAX1 (accession no. U57411) gene in T6 generation. The brown rice of the CAX1 expressing rice contained 13.4~68.0 % more calcium $(Ca^{2+})$ than that of the wild type and 5 lines were selected based on the phenotypes compared to the control cultivar at the GMO field. The CAX1 expressing transgenic rice plants were similar in phenotype to the wild type during the whole growth period. Also these selected 4 lines appeared to be resistant to blast, cold and water solution compared with the wild type. Difference in 1,000 grain weight of brown rice was observed among each line but grain shape did not show any morphological alternations. These results suggest the enhanced Ca-substrate specificity of CAX1 exchanger in donor plant. Therefore, intact CAX1 exchanger can be functionally useful for $Ca^{2+}$ nutrient enrichment of rice with reduced accumulation of undesirable cation.

Growth and Yield Comparisons among Barley Varieties with Different Resistance to Barley Mosaic Virus (보리 모자이크 바이러스 저항성 정도가 상이한 보리 품종간 생육 및 수량 비교)

  • Viral diseases, especially Barley yellow mosaic virus (BaYMV) and Barley mud mosaic virus (BaMMV), have been most serious in barley fields. In this study, we investigated the effect of different level of resistance to viral diseases on the plant growth and yield in barley. Various viral disease symptoms on leaves of overwintered plants were similar between medial-resistant and susceptible varieties of Saessalbori and Baegdong. In diagnosis of virus infection, BaYMV and BaMMV were detected in Saessalbori and Baegdong, but not in the resistant variety, Naehanssalbori. Plant height was restrained about $11{\sim}12cm$ prior to heading in Saessalbori and Baegdong comparing to Naehanssalbori. Even if both varieties were medial resistant to virus diseases, Saessalbori was different from Baegdong in heading date and culm length due to its recovery from viral damages prior to heading. Both medial-resistant and susceptible varieties were quite different from the resistant variety in yield components such as heading date, number of spikes and culm length when evaluated in the virus-infected or non-infected field. Baegdong delayed 7 days in heading date and reduced by more than 50% in culm length and spike numbers as compared to Naehanssalbori. On the other hand, Saessalbori showed similar heading date, but was shorter by 20% in culm length than Naehanssalbori. Three varieties tested in the non-infected field over two years were not significantly different for yield potential with ranges of $340{\sim}405kg/10a$. However, significant yield reduction (P<0.01) was observed in Saessalbori and Baegdong with ranges of $108{\sim}288kg/10a$ as compared to Naehanssalbori (391 kg/10a) when tested in the virus-infected field. Yield potentials of Saessalbori and Baegdong reduced by 35 and 63%, respectively in the virus-infected field as compared to those in the non-infected field. Our results showed that damages from virus diseases were significant on the early plant growth to yield and its components in barley.

Growth Characteristics and Yield of Collected Boxthon(Lycium chinense Mill.) Varieties (구기자 수집종(蒐集種)의 생육특성(生育特性)과 수양(收量))

  • This research was carried out to obtain the field resistance to disease and pest, and high yielding of new Boxthorn (Lycium chinense Mill.) varieties. Growth characteristics, yield potential, correlation of trait which was related the yield and path coefficient of the domestic native varieties. two of cultivated varieties and four of introduced varieties were investigated. In growth characters of collected varieties, the stem length of collected varieties in China were slightly long. Leaf area in Jindo, Kumsan native and Yusong 1 and 2 were wider than that of other varieties. Resistance to pest iniury was very strong at China 1, Japan 1 and Yusong 2 in eriophyidmite, and also resistance to anthrax anthracnose were showed less than 10% of the rate of attack fruit at Japan 1 and China 1. Flowering stage was more early in Chungyang native than that of other varieties. Fruit size of Kumsan native, Yusong 2 were large size fruit varieties. Fruit setting number was most abundant at 1,623 in Yusong 2 which was collected variety. And dry yields of fruit per lOa was more cmparatively abundant in Yusong 2 at 86% than those of Chungyang native. The high correlation cofficienct character with fruit yield were fruit setting and branching number and leaf area. Direct effect by path anaslysis were fruit setting number (Py=0.45), and branching number was showed indirect effect.

Identification of Quantitative Trait Loci Associated with Anthracnose Resistance in Chili Pepper (Capsicum spp.) (고추 탄저병 저항성 관련 양적형질 유전자좌 분석)

  • Pepper ($Capsicum$ spp.) anthracnose caused by $Colletotrichum$ $acutatum$ is a destructive disease susceptible to areas where chili peppers are grown. $Capsicum$ $baccatum$ var. $pendulum$ (Cbp) is resistant to anthracnose and has actively been used for interspecific hybridization for the introgression of resistance gene(s) into cultivated chili peppers. The goals of this study were to determine the inheritance of resistance to anthracnose within $Capsicum$ $baccatum$ and to map quantitative trait loci (QTLs) for the anthracnose resistance. A genetic mapping population consisting of 126 $F_2$ plants derived from a cross between $Capsicum$ $baccatum$ var. $pendulum$ (resistant) and $Capsicum$ $baccatum$ 'Golden-aji' (susceptible) was used for linkage mapping. The linkage map was constructed with 52 SSRs, 175 AFLPs, and 100 SRAPs covering 1,896cM, with an average interval marker distance of 4.0cM. Based on this map, the number, location, and effect of QTLs for anthracnose resistance were studied using plants inoculated in the laboratory and field. A total of 19 quantitative trait loci (2 major QTLs and 16 minor QTLs) were detected. Two QTLs ($An8.1$, $An9.1$) showed 16.4% phenotypic variations for anthracnose resistance after wounding inoculation. In addition, five minor QTL loci ($An7.3$, $An7.4$, $An4.1$, $An3.1$, $An3.2$) showed a total of 60.73% phenotypic variations of anthracnose resistance in the field test. Several significant QTLs were also detected and their reproducibility was confirmed under different inoculation conditions. These QTLs are now being confirmed with different breeding populations. Markers tightly linked to the QTLs that are reliable under different environmental conditions will help to determine the success of marker-assisted selection for anthracnose -resistant breeding programs in chili pepper.

Occurrence of Viral Diseases in Barley Fields and Responses of Resistant Genes to BaYMV-Ik and BaMMV (맥류 바이러스병 발생 현황 및 BaYMV-Ik와 BnMMV에 대한 저항성 유전자의 반응)

  • The major symptom such as yellowish and mosaic spots in overwintering barley were mostly caused by viruses such as Barley yellow mosaic virus (BaYMV) and Barley mild mosaic virus (BaMMV) in the nation-wide for four years. The result showed that more than $78\%$ collected samples were infected by the viruses. The incidence of Ba YMV was more than $70\%$, and relatively uniformly distributed in the southern regions of barley fields in Korea. However the incidence of BaYMV in Gyeonggi Province was as low as $19\%$ compared to $65\~85\%$ in the rest of regions. Occurrence of BaMMV varied depending on investigated regions such as $20\~40\%$ in Jeonbuk, Jeonnam, Gangwon and Gyeongnam, and a lower infection in Gyeongbuk, Chungnam and Gyeonggi Provinces. In this result, $60\%$ of BaMMV was found to be in the southwest regions of Korea such as Jeonbuk and Jeonnam Provinces. Over all, both BaYMV and BaMMV were thought to be dominantly casual agents in overwintering barley by either solely or mixed infections. Soil-borne wheat mosaic virus(SBWMV) occurred at most $14\%$ in Gyeonggi and Barley yellow dwarf virus-MAY (BYDV­MAV) was found only one place in Jeonbuk, suggesting that SBWMV and BYDV-MAV were not significant diseases in Korea. Exotic genetic resources that possess different resistant genes to BaYMV and BaMMV were tested to identify the responses to the viruses occurred in Iksan. According to the ELISA results, BaYMV and BaMMV were infected in some plant materials but SBWMV was not identified. Any resistant gene was not effective to BaYMV-Ik (Insan strain) and BaMMY. Ishukushirazu (rym 3) and Chosen (rym 3), Tokushima Mochi Hadaka (rym 4y) and Hakei 1-41 (rym 5a) showed resistant response with little symptoms to BaYMY. The other five accessions possessing rym 1+5, rym 2, rym 4m, rym 5 and rym 9, respectively, were resistant to BaMMV. Various symptoms were observed in the tested plant materials such as not only yellowish and mosaic symptoms mostly but also necrotic spot, tissue necrosis, leaf stripe and leaf curling. However, it was difficult to find any relationship between resistant genes and specific symptoms.

Symptoms of Cucumber Virus Diseases Occurred in Sangju and Gurye in 2006 and 2007 (2006년과 2007년 상주와 구례에서 발생한 오이 바이러스병의 병징 특성)

  • Virus diseases occurring on cucumber was surveyed at main plantation areas of 'Sangju' and 'Gurye' in 2006 and 2007. Viral infection rate on cucumber was ranged from 14% to 90% in fields and the average infection rate was 46% at Sangju area. Cucumber cultivated at Gurye area had viral incidence ranged 9-100% and averaged 48%. The majorly infected viruses were Cucumber green mottle mosaic virus, Zucchini yellow mosaic virus, Papaya ring spot virus and Watermelon mosaic virus-2, and their infection rates were 23.5%, 13.0%, 9.0% and 2.0%, respectively in 2006 and 2007. Mixed infection rate of duplex, triplex and tetraplex was 31.5%, 7.5% and 2.5%, respectively. The rate of infection type was 47.7% by single infection, 31.5% by double infection and 88.7% by both infection types. Important viruses involved in mixed infection types were Zucchini yellow mosaic virus, Cucumber green mottle mosaic virus, Papaya ring spot virus and Broad been wilt virus2, orderly. Symptom in single infection was almost same on 'Dadagi' line and 'Chicheong' line of cucumber cultivars. Cucumber green mottle mosaic virus produced various symptoms of chlorotic spot, vein chlorosis and vein wrinkle and so on. Main symptoms of vein chlorosis, and severe mosaic and malformation were induced by Zucchini yellow mosaic virus. Mild symptoms were occurred relatively by Papaya ring spot virus and Watermelon mosaic virus2.

Quantitative Analysis of Resveratrol in Mulberry Leaves (뽕잎의 품종별.시기별 resveratrol 함량 변이)

  • Resveratrol has been associated with reduced cardiovascular disease and reduced cancer risk. This phytochemical has been reported in a number of plant species including grapes, peanuts and pine trees in response to stress such as fungal infection, heavy metal ions or UV irradiation. The objective of this study was to determine the resveratrol contents in leaves of mulberry varieties at different collecting times. Quantitative analysis of 16 cultivars showed a range of $102{\sim}466{\mu}g/100g$ on dry weight basis (which is equivalent to $25{\sim}116.5{\mu}g/100g$ on fresh weight basis). Resveratrol contents in mulberry leaves was higher in autumn than spring, and higher in fully matured leaves than in juvenile leaves. Among the tested samples, 'Kaeryangppong', 'Sugeppong' and 'Cheongilppong' collected in the middle of October showed high resveratrol contents of $838{\mu}g/100g$, $803{\mu}g/100g$, $800{\mu}g/100g$ on dry weight basis, respectively. Especially mulberry leaves dried in the shade showed of $1,030{\mu}g/100g$ on dry weight basis in resveratrol content, this result may contribute to utilization of mulberry leaves.

Variation of Rice Production for Two Decades before and after Breeding Tongil Variety in Korea (수도 통일품종 육성보급 전후 20년간의 생산성 변이)

  • The variability of rice productivity during last 2 decades (1961-1980) of ten years before and after the introduction of"Tongil" was reviewed from the epochal, regional and varietal points of view. During that period the cultivated area of paddy rice have remained almost unchanged, while the total rice production have got elevated from 3, 463 million metric tons in 1961 to 6.006 million metric tons in 1977, recording 73.4% increase. This remarkable increase in rice production is considered to be attributable much to the development and release of new high yielding variety, "Tongil", coupled with the amelioration of cultural techniques. However, in 1978 Tongil type varieties experienced the epidemic outbreak of blast disease due to the shifted race population of blast fungus and in 1980 recorded poor rice production as low as in 1960's due to the unfavorable weather stress throughout the rice growing season, giving rise to many problems awaiting solutions for securing the stabilized high production of rice. The rice yield has continued the gradual increase during last two decades but its difference between farmer and research organization have got wider from 79kg/10a during 1960 to 1971 to 101kg/l0a during 1972 to 1980, and also the inter-regional differences have been increased from 50-60kg/10a to 80kg/10a during those periods. Therefore, this proves that we have raised the upper boundary of rice yield by increasing the yield potential of rice variety but have not changed those absolute deviations. Estimates indicate that the increased rice production during that period was indebted 40 percent to the varietal improvement and 13 percent to the ameliorated agro-technologies, and the rest, 47 percent, could be ascribed to the other factors besides varieties and cultural technologies such as the improved agricultural environments, etc. Of course, even though it cannot be expected to unify the cultural environments and the cultural technologies, provided that much efforts are to be endeavored to minimize the yield difference of 20 percent between farmer and research organizations and the inter-regional yield difference of 20 percent, much increased rice production can be expected to be achieved with the current level of cultural technology and the yielding potential of the present rice varieties. In order to expedite the above effects on rice production the followings are to be put into practices consitently and steadfastly. 1. Reinforcement of breeding for varieties with high yielding potential and less susceptible to climatic-stress and pests, and of basic physicoecological studies of rice plant for improving the cultural technologies. 2. Continuous endeavor to secure the stabilized cultural environments by improving the soil fertility and increasing the drainage and irrigation facilities. 3. Political back-up to encourage the farmers' incentives for production 4. Precise surveys for agricultural statistics to facilitate the long-term planninge long-term planning.

Relationships of Physiological Activity and Anatomical Structure to the Wilting Phenomena in Rice Plant 2. Relationships between the anatomical structure and wilting phenomena of rice variety "Yushin" (수도품종의 위조현상과 생리 및 형태해부학적 구조와의 관련성에 관한 연구 제2보 유신벼의 위조현상발생과 형태해부학적 구조와의 관계)

  • One of the physiological disease, sudden wiliting of Yushin variety suggested that low sunlight, excessive nitrogen application, and highly reduced soil condions either singly or combined, might be possible causes of the disorder. Some visual symptom of sudden wilting are discoloration of leaves, development of nodal roots above the soil surface, total root rot, and lodging. Those observations led to the hypothesis that suffocation of root tissues was a direct cause of the wilting. The oxygen transport characteristics of Yushin, IR262 and Tongil were examined by two methods. First, Soil-cultured plants of the three varieties were subjected to paraffin treatment to decrease the oxygen supply from the air to root tissues through the soil-water system, liquid paraffin was applied to the water surface in the pots at panicle formation stage. In this experiment, sudden wilting was observed of Yushin and IR262 at about a week after the treatment, but Tongil remained green and healthy. Wilting-resistant variety Tongil had higher oxygen release, whereas the susceptible Yushin and IR262 had lower oxygen release. Second, the number and size of the air spaces in each internode were investigated in the 5th internode from the top, all three varieties have a similar number of air spaces, although the air spaces of Thongil were larger. In the 4th internode, Tongil had plenty air spaces, Yushin and one of the Yushin's parents IR262 had scanty or none. The observations indicated that the ability of Yushin and IR262 for oxygen transport is very limited compared with that of Tongil. The limited oxygen supply due to poor development of air space in internode of rice plants may cause suffocation of root tissues, weaken metabolic activity of the tissues, and induce root rot, subsequently inducing sudden wilting and lodging under unfavorable weather, soil and cultural conditions.
