남부지방(南部地方)에서 율무의 재식밀도(栽植密度)에 따른 몇가지 형질(形質)및 수량변화(收量變化) (Effects of Planting Density on Growth and Yield of Coix lachryma-jobi L. var. mayeun STAPF in Southern-Region of Korea)

  • 남부지방(南部地方)의 율무 재배체계(栽培體系) 확립(確立)을 위한 기초(基礎) 자료(資料)를 얻고자 재식밀도(栽植密度) 차이에따른 생육(生育)과 수량(收量)을 검토(檢討)했던바 다음과 같은 결과(結果)를 얻을 수 있었다. 1. 초장(草長), 경직경(莖直徑), $1m^2$ 당(當) 경수(莖數)및 $1m^2$ 당(當) 엽수(葉數) 등의 모든 형질(形質)이 $60{\times}15cm$ 재식(栽植)에서 가장 우수(優秀)하였다. 2. 100립당(粒重), 1 l 당(重), 건경중(乾莖重)및 종실종(種實種)등의 수량(收量)이 $60{\times}15cm$ 재식(栽植)에서 가장 우수(優秀) 하였다. 3. 재식밀도(栽植密度)와 품종간(品種間)에 따른 농업형질(農業形質) 들의 분산분석(分散分析) 에서도 유의성(有意性)이 인정되었다.

The effects of cytokinin and plating density on protoplast culture of sunflower

  • Sunflower (Helianthus annuus L.) protoplasts were isolated from seven-day-old etiolated hypocotyls of 10 A line and four-week-old fully expanded young leaves of PI 441983 line in vitro seedlings using an enzymatic method. Purified protoplasts were collected by filtration and floatation in sucrose solution. Semi-solid protoplast culture was performed using the L4 regeneration protocol with various culture media and plating densities to achieve the highest efficiencies for protoplast culture of hypocotyl and mesophyll protoplasts of 10 A and PI 441983 lines, respectively. The concentrations in liquid L'4M medium and different plating densities were evaluated in two types of cytokinins, the adenine-type 6-benzyladenine (BA) and the phenylurea-type thidiazuron (TDZ). The highest colony formation was achieved in both sunflower lines when 0.5 mgL-1 BA and 0.5 mgL-1 TDZ were applied with a high plating density (3 × 105 protoplasts mL-1). These conditions led to 38.45% and 39.40% colony formation for hypocotyl protoplasts of the 10 A line and mesophyll protoplasts of the PI 441983 line, respectively. Moreover, many hypocotyl protoplast-derived colonies developed into micro-calli. In addition, superior development of both sunflower protoplasts was observed with all plating densities when BA was used in combination with TDZ. This finding will be applicable to future sunflower hybrid production via somatic hybridization.

Factors Affecting Agrobacterium tumefaciens-mediated Transformation of Panax ginseng C.A. Meyer

  • A protocol for the production of transgenic Panax ginseng C.A. Meyer was established via Agrobacterium tumefaciens-mediated genetic transformation of direct somatic embryos. A number of conditions related to the co-cultivation were tested with respect to maximizing transformation efficiency. The results showed that pH of the co-cultivation medium (5.7), the bacterial growth phase (optical density; $OD_{600}$ = 0.8), co-cultivation period (3 days), and acetosyringone concentration $(100\;{\mu}M)$ had positive effects on transformation. Selected plantlets were cultured on the medium at an elevated hygromycin level(30 mg/l). Integration of the transgenes into the P. ginseng nuclear genome was confirmed by PCR analysis using hpt primers and by Southern hybridization using hpt-specific probe. The transgenic plantlets were obtained after 3-month cultivation and did not show any detectable variation in morphology or growth characteristics compared to wild-type plants.

Effect of Plant Density on Growth Responses and Yield in Yacon

  • This study was conducted to determine the optimal planting distance in cultivation of yacon (Polymnia sonchifolia Poeppig & Endlicher). Plug seedlings were planted with 6 different plant densities of 80 $\times$ 60, 80 $\times$ 50, 70 $\times$ 50, 70 $\times$ 45, 70 $\times$ 40 and 60 $\times$ 45 cm. The plant height and the petiole length were increased with increasing the planting distance. The tallest plant height of yacon was 165.4cm with the plant density of 80 $\times$ 60 cm. However, branch number per plant, leaf number on main stem and stem diameter were not significantly difference among planting densities. Tuberous root was harvested 31.42 tons/ha in 70 $\times$ 50 cm spacing. The ratio of heavier tuberous roots than 200 g to total tuberous roots decreased significantly according to increase of planting density. Fresh weights of shoot and root, contain the crown bud, were decreased, as planting distance was shorter. Tuberous root number was fewer but its weight was heavier in wide planting than in dense planting. We think that optimal planting density is about 30,000 plants/ha, if it were to be 70 cm row spacing, intrarow spacing should calculate about 47cm.

Effect of Planting Density and Nitrogen Level on Growth and Yield in Heavy Panicle Weight Type of Japonica Rice

  • To investigate the effects of planting density and nitrogen level on growth and yield potential of newly bred heavy panicle japonica rice with large grain (Iksan 435 and Iksan 438) or many spikelets per panicle(HR14022-21-8-4 and HR14022-21-8-6), four heavy panicle type rices and two many panicle type rices(Dongjinbyeo and Donganbyeo) as the checks were planted under standard planting density (30$\times$15 cm) and dense planting density (15$\times$15 cm) with two nitrogen levels of standard nitrogen level(110 kg h $a^{-1}$) and heavy nitrogen level(165 kg h $a^{-1}$). Effective tiller rate decreased in dense planting or heavy nitrogen, when compared to standard nitrogen and planting, while leaf area index and to dry weight increased in dense planting or heavy nitrogen. Tiller numbers and panicle numbers were more increased by dense planting than heavy nitrogen, whereas spikelet numbers were more increased by heavy nitrogen than dense planting. Ripened grain ratio was slightly lower only in dense planting. 1,000 grain weight in brown rice was not significantly different in dense planting or heavy nitrogen. Milled rice yield was highest in heavy nitrogen with standard planting for heavy panicle type rice, while yield for many panicle type rice was highest in heavy nitrogen with dense planting, suggesting that many panicle type rice possesses higher adapt-ability for dense planting than heavy panicle type rice. Path coefficient analysis revealed that top dry weight, spikelet number and grain weight were the greatest positive contributors to yield, whereas tiller number was negative to yield.d.

구기자나무의 재식밀도 및 예취높이가 가공용 잎 생산에 미치는 영향 (Effects of Planting Density and Cutting Height on Production of Leaves for Processing Raw Materials in Goji Berry)

  • Background: The leaves of the goji berry (Lycium chinense Mill.) are used as raw materials in processing and by replace fruits to some extent. The reason is that the leaves are cheaper, however, betaine content is higher than in the fruits. These experiments were conducted to determine the planting density and cutting height for producing a large number of leaves. Methods and Results: The cultivar 'Myeongan' with many branches was used. When the shoot height reached 50 cm - 70 cm, harvesting was possible four times a year. The time to next harvest was approximately 38 days after regeneration of new shoots. Leaf quantity was in the order of 1st > 2nd > 4th > 3rd harvest. Insect damage occurred during the third harvest in late July and early August, therefore, eco-friendly control was necessary. The total yield was higher at the planting density 60 cm × 30 cm than that of 60 cm × 20 cm or 60 cm × 40 cm. The yield at cutting for shoot height of 60 cm was increased by 6.3 percent compared to that of 50 cm, At the cutting height of 70 cm, harvest was difficult owing to hardening of stems and thorns. Betaine content, an indicator component of goji berry, was not significantly different according to planting densities and cutting height. Conclusions: The ideal cutting period to produce leaves of goji berry for processing is when the shoots grow to approximately 60 cm, and the leaves can be harvested 4 times a year. The dried-leaf yield was highest at the planting density of 60 cm × 30 cm.

  • Based on the use of Acorus gramineus SOLAND (AG) for the treatment of stroke in traditional Korean medicine, the present study was carried out to evaluate neuroprotective effects of ${\alpha}$-asarone after transient global cerebral ischemia using rat 4-vessel occlusion (4VO) model in rats. ${\alpha}$-Asarone (5 mg/kg) administered intraperitoneally significantly protected CA1 neurons against 10 min transient forebrain ischemia as demonstrated by measuring the density of neuronal cells stained with Cresyl violet. ${\alpha}$-Asarone significantly reduced hippocampal neuronal cell death by 85.2% where as its isolated single compounds from AG compared with a vehicle-treated group.

Hydraulic Property and Solute Breakthrough from Salt Accumulated Soils under Various Head Pressures

  • Salt accumulated soil should be reclaimed to lower salt level for crop production. This study was carried out to investigate the characteristics of water flow and transport of mono and divalent solutes on salt accumulated soils with different head pressures. Saturated hydraulic conductivity was measured by constant and falling head methods with maintaining different head pressures. Saturated hydraulic conductivity was influenced by bulk density and organic matter contents in soils, but it had different elusion patterns between saline and sodic soil. While the quantity of water necessary for reclamation could be varies with soil type, it was considered that the supply of one pore volume of water was affordable and economic. Additional head pressure significantly increased the volume of leachate at a given time and it was more effective at low organic matter soils. The results indicate that additional head pressure would be one of the best irrigation practices on desalination method for salt accumulated soils.
