Successful Development of Cordyceps bassiana Stromata from Beauveria bassiana

  • A specimen of Beauveria bassiana was collected from Yang-yang of Gangwon province, Korea in October 2006. Conidial isolates were prepared from the specimen by the dilution method and inoculated in brown rice medium for fruiting body production. After nearly two months incubation for perithecial stromata developed from single isolates as well as from their combinations. They were determined as Cordyceps bassiana by observing the stromatal characters and their conidial structures. This is the first report of the development of C. bassiana from B. bassiana cultures.

  • Many Cordyceps spp. were collected for excellent strains selection at near Wanju-kun from May to October, 2000. They were identified. As the resulting, They were 2 genera and 4 species. 6 strains were got through 4 species culture. 6 strains are Cordyceps militaris f. albino, C, gracilaides, C. militaris and Isaria japoica. Among them, 4 species used for induction of artifitial fruiting body. 3 strains of them were selected for excellent strains. excellent strains are Cordyceps militaris and Isaria japoica.

  • Quality characteristics of kochujang made with Aspergillus oryzae-and/or Cordyceps sp.-inoculated koji were investigated. Protease activity of Cordyceps sp.-inoculated koji was higher than that of A. oryzae-inoculated one. Sensory evaluation showed that kochujang made with mixture of A. oryzae-and Cordyceps sp.-inoculated koji (70 : 30, w/w) was superior to others.

  • A Cordyceps species was found with a Beauveria anamorph state on larval insect cadavers on Obong mountains in Gangwon Provinces, Republic of Korea. Cultures from discharged ascospores formed an anamorph identifiable as Beauveria bassiana. This teleomorph-anamorph connection was also confirmed by the in vitro production of fertile ascomata from conidial cultures with morphology like that of field-collected specimen. This is the first report of in vitro production of a teleomorph for any Beauveria species. The Cordyceps species has been conspecified as Cordyceps bassiana, a species described from China with B. bassiana anamorph.

Effect of Light Conditions on Production of Cordycepin of Cordyceps militaris (광조건이 Codyceps militaris의 Cordycepin 생산에 미치는 영향)

  • The effect of light on production of cordycepin (3'-deoxyadenosine) was investigated using Cordyceps militaris. Among Cordyceps spp. tested, C. militaris KCTC 6064 was the best for cordycepin production, After 120-hr cultivation of C. militaris KCTC 6064 at $24^{\circ}C$ in YMG broth with light (1,000 lux), 51.6 mg/l of cordycepin was produced. In general, production of cordycepin with light illumination was better than that without light. Cordycepin production under light illumination was enhanced up 96-hr cultivation, whereas after 96-hr, cordycepin production was decreased.

  • This study was carried out to investigate infection process, symptoms and $LD_{50}$ values of the entomopathogenic fungus cordyceps, Paecillomyces japonica, on the domestic silkworm, Bombyx mori. Susceptibility of infection in the silkworms by cordyceps, Paecillomyces japonica, was 100% in $10^{7}$ block, 96% in $10^{6}$ block, 76% in $10^{5}$ block, 44% in $10^{4}$ block, 28% in $10^{3}$ block and 8% in $10^{2}$ block. Cordyceps, Paecillomyces japonica, was highly infectious to the silkworms. A pathogenicities of cordyceps, Paecillomyces japonica, may be highly virulent because of the low resistance or high susceptibility of the silkworms. Dosage of the pathogen of $LD_{50}$ was $3.78{\times}10^{3}\;spores/mm^{3}$.

Comparison of Sensory and Mechanica] Properties of Breads with Paecilomyces japonica and Cordyceps militaris powder by Storage Time and Temperature (눈꽃과 번데기 동충하초 첨가 식빵의 저장기간과 온도에 따른 관능적.기계적 특성 비교)

  • The quality characteristics of breads with different concentrations(2%, 4%) of Paecilomyces japonica (J2, J4) and Cordyceps militaris (M2, M4) powder were compared in terms if sensory and mechanical properties. In the sensory test, J2 and M2 groups gained good scores in taste, texture and odor acceptance as control grourp. Paecilomyces japonica-added group showed higher moisture and bigger volume than that of control group, and Cordyceps militaris group showed lower moisture content and decreasing volume. 'L' values of J and M groups was lower than control, and the longer the storage period, the lower the lightness. The more Paecilomyces japonica was added, the higher the 'a' value was, but Cordyceps militaris showed an opposite trend. The hardness of the bread was the lowest in J2 group and the highest in M4 group, and the longer tile storage period. the higher the hardness was (p<.001). Overall, the addition of 2% Paecilomyces japonica showed a similar tendency with control group, and showed good scores especially in acceptance, moisture and volume. Paecilomyces japonica appeared to be more desirable than Cordyceps militaris.

  • Traditionally, Cordyceps species have been used as a part of herbal medicine in Oriental countries, including Korea for internal health, vigor and to cure different diseases related to heart, lung etc. In recent years, research on artificial fruiting of some species of the genus Cordyceps including C. militaris has been carried out extensively because of their medicinal value. Instability observed in the in vitro fruiting of C. militaris is reported in the present study.

  • Objectives : This study was performed to investigate the effects of Cordyceps Sinensis (CS) and Cordyceps Militaris (CM) on anti oxidation in the livers of ${CCl_4}-treated$ rats. Methods : Hepatotoxicity in rats was induced by carbon tetracWoride. $CCl_4-induced$ rats were administered with the extract of CS and CM. Results : In vitro, CS and CM didn't affect levels of lipid peroxide and the activities and type conversion ratio of xanthine oxidase. However, hydroxyl radicals and DPPHL radicals were decreased. In vivo, in the ${CCl_4}-treated$ rats, lipid peroxide, the activities and type conversion ratio of xanthine oxidase and superoxide radicals were increased but superoxide dismutase was decreased. After CS and CM were administered to ${CCl_4}-treated$ rats, levels of lipid peroxide, the activities and type conversion ratio of xanthine oxidase and superoxide radicals were decreased but superoxide dismutase was increased. Conclusions : These results suggest that CS and CM decrease the activities of free-radical-generating enzymes which form lipid peroxide and increase the activities of oxygen free radical scavenging enzymes.

Studies of the Cordyceps militaris Extract Administration on the Metabolic Enzyme Activities in Hypercholesterolemia (동충하초 엑스의 고콜레스테롤혈증 대사효소 활성 변동에 관한 효과)

  • The objective of this study was to investigate the effects of the feeding Cordyceps militaris extract on the improvement of the free fatty acid, lipid peroxide, creatinine and enzyme (creatine phosphokinase; CPK, lactate dehydrogenase; LDH, alkaline phosphatase; ALP, lecithin cholesterol acyltransferase; LCAT) activities in the sera of dietary hypercholesterolemic rats (SD strain, male) fed the experimental diets for 5 weeks. Concentrations of free fatty acid, lipid peroxide and CPK, LDH, ALP activities in sera were fairly reduced in the Cordyceps militaris extract administration group (CHE) than in the hypercholesterolemic diet group (CHD). However, no significance was found in the effect of an creatinine concentration among the groups. The LCAT activity in serum was increased in the Cordyceps militaris extract administration (CHE) than in the hypercholesterolemic diet group (CHD). From these results, Cordyceps militaris extracts were effective on the improvement of the lipid components and metabolic enzyme activities in sera of dietary hypercholesterolemic rats.
