Bottle gourd (Lagenaria siceraria Standl.) has been used as a rootstock for the watermelon cultivation because of better growth ability at low temperature and avoidance from contamination of the soil disease. Since the genetic source for the elite rootstock is limited in nature, the genetic engineering method is inevitable to develop new lines especially to obtain the functionally important or multi-disease resistant bottle gourd. Recently, our lab has set up a successful system to transform the bottle gourd. in order to monitor the transformation process, GFP gene is used. Cotyledons of the inbred line 9005, 9006 and G5 were used to induce the shoot under the selection media with MS + 30 g/L sucrose + 3.0 mg/L BAP + 100 mg/L kanamycin + 500 mg/L cefotaxime + 0.5 mg/L $AgNO_3$, pH 5.8. The shoot was developed from the cut side of the explants after 3 weeks on the selection media. The shoot was incubated in the rooting media with 1/2 MS + 30 g/L sucrose + 0.1 mg/L IAA + 50 mg/L kanamycin + 500 mg/L cefotaxime, pH 5.8 and moved to pot for acclimation. Although the shoot development rate was depended on the genotype, the G5 was the best line to be transformed. Monitoring GFP expression from the young shoot under microscope could make the selection much easier to distinguish the transformed shoot from the non-transformed shoots.
A Macroscopic combination of two or more distinct materials is commonly referred to as a "Composite Material", having been designed mechanically and chemically superior in function and characteristic than its individual constituent materials. Composite materials are used not only for aerospace and military, but also heavily used in boat/ship building and general composite industries which we are seeing increasingly more. Regardless of the various applications for composite materials, the industry is still limited and requires better fabrication technology and methodology in order to expand and grow. An example of this is that the majority of fabrication facilities nearby still use an antiquated wet lay-up process where fabrication still requires manual hand labor in a 3D environment impeding productivity of composite product design advancement. As an expert in the advanced composites field, I have developed fabrication skills with the use of machinery based on my past composite experience. In autumn 2011, the Korea government confirmed to fund my project. It is the development of a composite sanding machine. I began development of this semi-robotic prototype beginning in 2009. It has possibilities of replacing or augmenting the exhaustive and difficult jobs performed by human hands, such as sanding, grinding, blasting, and polishing in most often, very awkward conditions, and is also will boost productivity, improve surface quality, cut abrasive costs, eliminate vibration injuries, and protect workers from exposure to dust and airborne contamination. Ease of control and operation of the equipment in or outside of the sanding room is a key benefit to end-users. It will prove to be much more economical than normal robotics and minimize errors that commonly occur in factories. The key components and their technologies are a 360 degree rotational shoulder and a wrist that is controlled under PLC controller and joystick manual mode. Development on both of the key modules is complete and are now operational. The Korean government fund boosted my development and I expect to complete full scale development no later than 3rd quarter 2012. Even with the advantages of composite materials, there is still the need to repair or to maintain composite products with a higher level of technology. I have learned many composite repair skills on composite airframe since many composite fabrication skills including repair, requires training for non aerospace applications. The wind energy market is now requiring much larger blades in order to generate more electrical energy for wind farms. One single blade is commonly 50 meters or longer now. When a wind blade becomes damaged from external forces, on-site repair is required on the columns even under strong wind and freezing temperature conditions. In order to correctly obtain polymerization, the repair must be performed on the damaged area within a very limited time. The use of pre-impregnated glass fabric and heating silicone pad and a hot bonder acting precise heating control are surely required.
Recently, the demand for the powdered products to process raw grains and vegetables has been expanded, and the growth possibility of their fields(markets) in the future will be highly valued with the westernization of living environment and the change of the dietary life. We have bought and analyzed the 111 products of raw grains and vegetables from the large-sized marts, markets and internet orders from March to October 2006. The rate of moisture content was $1.7{\sim}12.5%$. We also found out that the foods over 10% moisture content was 8 of those samples we bought or collected. Their ash rates were averagely $0.3{\sim}8.6%$. The number of those foods that the alien substances were detected was 2. The tar pigments, artificial sweeteners and sulfur dioxides of components that contained food additives were not detected. Each detection range of Cadmium, Lead and Arsenic of the injurious heavy metals was non-detectable $(nd){\sim}0.55,\;nd{\sim}4.52,\;nd{\sim}0.10mg/kg$, while the average detection content were 0.08, 0.48, 0.01 mg/kg. By investigating the contamination degree of the microflora, we discovered that the number of the aerobic plate count, B. cereus over 1,000 cfu/g and C. perfringens over 100 cfu/g was 36 (32.4%), 9 (8.1%) and zero, and that the whole sanitation condition of the products of powdered raw grains and vegetables circulated in the market was not so good.
The uranium removal efficiency of rhizofiltration was investigated in lab scale experiment. Three plants such as sunflower (Helianthus annuus L.), bean (Phaseolus vulgaris var.), and Indian mustard (Brassica juncea (L.) Czern.) were cultivated in artificially contaminated solution by uranium at 30 ${\mu}g$/L and 80 ${\mu}g$/L for 72 hours. The removal efficiencies of three cultivars were calculated from the ICP/MS analysis of uranium mass in solution and the plant. For Helianthus annuus L., more than 80% of initial uranium were removed from the solution and the uranium concentration of residual solution maintained lower than 10 ${\mu}g$/L. For Phaseolus vulgaris var. and Brassica juncea (L.) Czern., their uranium removal efficiencies ranged from 60 to 80%. Even the uranium concentration of solution was higher than 500 ${\mu}g$/L, these cultivars removed more than 70% of initial uranium by rhizofiltration, suggesting that the rhizofiltration has a great capability to remove uranium in the contaminated groundwater. The removal efficiency of rhizofiltration by using Brassica juncea (L.) Czern decreased from 83% to 42% with the increase of pH in solution. From the results of the analysis for the uranium accumulation in plants, 99% of uranium transferred into the plant from solution were accumulated in the root and only 1% of uranium existed in the shoot part (including leaves), suggesting that the cost and the time to treat massive grown plants after the rhizofiltration could be dramatically cut down because only their root parts needs to be treated. Finally, the genuine groundwater having high uranium concentration (81.4 ${\mu}g$/L), sampled from Daejoun area, was used in the experiment. The uranium removal efficiency of Helianthus annuus L. for the real groundwater was higher than 95%, investigating that the rhizofiltration is the very useful method to remediate uranium contaminated groundwater.
The non-magnetic materials with non-conductive showing high structure dispersity were developed on the base of natural quartz and lava-scoria which was collected from Je-ju island in Korea, and treated by methane-chemical technology those were obtained novel properties of magnetization through the analyzing. Depending on the processing conditions and subsequent applications the materials produced by strong methane-chemical reaction (MCR) in alcohol solution showed concurrently magnetic, dielectric and electrical properties. The obtained magnetic-electrical powders classified by aggregate complex of their features as segnetomagnetics, containing a dielectric material as a carrying nucleus, particularly the quartz on that surface one or more layers of different compounds were synthesized having thickness up to 10~50 nm and showing magnetic, electrical and other properties. It was confirmed in magnetizing process that powders of quartz and lava-scoria produced by MCR were better oil adsorbent as of oleophilic and floating matter on water surface although their specific gravities are comparably more than 1 in quartz or less than unity, as that of water, in lava-scoira. Therefore, it will be Possible and very useful to remove low density and light gravity oil spillage in difficult recovery from sea and inland water contamination spread on water surface, by marine accident and ship sinking accident occurring frequently in recent years, by way of magnetic adsorbent conveyer system in continuous, if it could be built up the mass Production system of water-floating magnetizable oleophilic adsorbent materials with use of iow cost and good Qualify lava-scoria spread on volcano district in Je-ju island. Background: Sputum decontamination with NALC-NaOH (N-acetyl-L-cysteine-sodium hydroxide) is known to better detect Mycobacterium tuberculosis (M. tb) by culture than that with using NaOH, which is widely used in Korean hospitals. In this report, sputum samples collected from pulmonary tuberculosis (TB) patients were treated with either NaOH or NALC-NaOH, and we compared the results of smear and culture positivity to determine whether the NALC-NaOH treatment method improves culture positivity in the sputum samples, and especially for those sputum samples that are smear negative and scanty. Methods: For each decontamination method, 436 sputum samples from pulmonary TB patients in the National Masan Tuberculosis Hospital were collected for this study. Sputum from a patient was collected two times for the first and second day of sampling time, and these samples were employed for the decontamination process by performing the 4% NaOH and NALC-2% NaOH treatment methods, respectively, for detecting M. tb by an AFB (Acid Fast Bacilli) smear and also by culture in solid Ogawa medium. Results: The NaOH and NALC-NaOH treatment methods did not significantly affect the AFB smear positivity of the sputum samples (33.0% vs 39.0%, respectively, p=0.078). However, the culture positive percents of M. tb in the Ogawa medium treated with NALC-NaOH and NaOH were 39.7% and 28.0%, respectively, which was a significantly different (p=0.0003). This difference in culture was more prominent in the sputum samples that were smear negative (the positive percents with NALC-NaOH and NaOH were 15.8% and 7.2%, respectively, p=0.0017) and scanty (NALC-NaOH and NaOH were 60.8% and 42.9%, respectively, p=0.036), but not for a smear that was 1+ or higher (p>0.05). Conclusion: NALC-NaOH treatment is better than NaOH treatment for the detection of M. tb by culture, but not by smear, and especially when the AFB smear is negative and scanty.
The purpose of this study was to examine the presence of Bacillus cereus, aerobic bacteria and coliforms in the raw material of infant formulas and investigate the manufacturing process in terms of microbial safety. Among ten kinds of raw infant formula material samples (n=20), Bacillus cereus appeared in two (n=4). Aerobic bacteria were not detected in raw infant formula material or maximum 4.15 log CFU/g. Eleven species of aerobic bacteria were isolated and 76% of them were Sphingomonas paucimobilis, Pseudomonas fluorescens, Rhizobium radiobactor, or Stenotrophomonas maltophilia. A Pearson's correlation analysis revealed that the most influential factors for detecting Bacillus cereus were aerobic bacteria and coliforms. In other words, when the measured values of aerobic bacteria and coliforms were higher, the possibility that Bacillus cereus would appear increased. In a regression model to predict Bacillus cereus, the rate of appearance was correlated with aerobic bacteria and coliforms, and its contribution rate for effectiveness was 86%. Improving microbial quality control by pasteurization, spray dry, popping and extrusion resulted in a decrease in the numbers of Bacillus cereus, aerobic bacteria and coliforms in the raw materials. The results suggest that a hazard analysis and critical control point system might be effective for reducing microbiological contamination.

A study on the electrolytic dissolution of SUS-304 and Inconel-600 specimen was carried out in neutral salt electrolyte to evaluate the applicability of electrochemical decontamination process for recycle or self disposal with authorization of large amount of metallic wastes contaminated with uranium compounds generated by dismantling a retired uranium conversion plant in Korea. Although the best electrolytic dissolution performance for the specimens was observed in a Na2s04 electrolyte, a NaNO$_3$ neutral salt electrolyte, in which about 30% for SUS-304 and the same for Inconel-600 in the weight loss was shown in comparison with that in a Na$_2$SO$_4$ solution, was selected as an electrolyte for the electrochemical decontamination of metallic wastes with the consideration on the surface of system components contacted with nitric acid and the compatibility with lagoon wastes generated during the facility operation. The effects of current density, electrolytic dissolution time, and concentration of NaNO$_3$ on the electrolytic dissolution of the specimens were investigated. On the basis of the results obtained through the basic inactive experiments, electrochemical decontamination tests using the specimens contaminated with uranium compounds such as UO$_2$, AUC (ammonium uranyl carbonate) and ADU (ammonium diuranate) taken from an uranium conversion facility were performed in 1M NaNO$_3$ solution with the current density or In mA/$\textrm{cm}^2$. it was verified that the electrochemical decontamination of the metallic wastes contaminated uranium compounds was quite successful in a NaNO$_3$ neutral salt electrolyte by reducing $\alpha$ and $\beta$ radioactivities below the level of self disposal within 10 minutes regardless of the type of contaminants and the degree of contamination.

BACKGROUND: Significant amount of bisphenol A has been released from the manufacturing process of plastics, epoxy resins, dental material and flame retardants. Bisphenol A has been detected at trace levels in wastewater, surface water, landfill leachate and drinking water. However, the residual survey of bisphenol A has not been performed in agricultural reservoir beside agricultural environment cultivating crops. This study was conducted to monitor the residual bisphenol A in national agricultural reservoirs and understand a level of contamination of bisphenol A in the agricultural environment in Korea. METHODS AND RESULTS: The water and water sediment were collected at agricultural reservoirs in Chungnam, Chungbuk, Kyunggi, Jeonnam, Jeonbuk, Kyungnam and Kyungbuk province. Bisphenol A was analyzed by the LC-MS/MS with triple quad 4500. The recovery of water and water sediment in the agricultural reservoirs showed the level of 95.7~97.2% and 91.5~100.3%, respectively. CONCLUSION: Bisphenol A was detected at the level of $0.05{\mu}g/L{\sim}0.18{\mu}g/L$ and $0.1{\mu}g/kg{\sim}34{\mu}g/kg$ in water and water sediment of the reservoirs, respectively. Based on the results, the residue of bisphenol A will be tried in the crops surrounding these agricultural reservoirs where bisphenol A detected above $10{\mu}g/kg$ of bisphenol A.
Improper disposal of petroleum and spills from underground storage tanks have created large areas with highly toxic contamination of the soil and groundwater. Methyl tert-butyl ether (MTBE) is widely used as a fuel additive because of its advantageous properties of increasing the octane value and reducing carbon monoxide and hydrocarbon exhausts. However, MTBE is categorized as a possible human carcinogen. This research investigated the Modified Photo-Fenton system which is based on the Modified Fenton reaction and UV light irradiation. The Modified Fenton reaction is effective for MTBE degradation near a neutral pH, using the ferric ion complex composed of a ferric ion and environmentally friendly organic chelating agents. This research was intended to treat high concentrations of MTBE; thus, 1,000 mg/L MTBE was chosen. The objectives of this research are to find the optimal reaction conditions and to elucidate the kinetic and mechanism of MTBE degradation by the Modified Photo-Fenton reaction. Based on the results of experiments, citrate was chosen among eight chelating agents as the candidate for the Modified Photo-Fenton reaction because it has a relatively higher final pH and MTBE removal efficiency than the others, and it has a relatively low toxicity and is rapidly biodegradable. MTBE degradation was found to follow pseudo-first-order kinetics. Under the optimum conditions, [$Fe^{3+}$] : [Citrate] = 1 mM: 4 mM, 3% $H_2O_2$, 17.4 kWh/L UV dose, and initial pH 6.0, the 1000 ppm MTBE was degraded by 86.75% within 6 hours and 99.99% within 16 hours. The final pH value was 6.02. The degradation mechanism of MTBE by the Modified Photo-Fenton Reaction included two diverse pathways and tert-butyl formate (TBF) was identified to be the major degradation intermediate. Attributed to the high solubility, stability, and reactivity of the ferric-citrate complexes in the near neutral condition, this Modified Photo-Fenton reaction is a promising treatment process for high concentrations of MTBE under or near a neutral pH.
