An Empirical Study on the Improvement of In Situ Soil Remediation Using Plasma Blasting, Pneumatic Fracturing and Vacuum Suction (플라즈마 블라스팅, 공압파쇄, 진공추출이 활용된 지중 토양정화공법의 정화 개선 효과에 대한 실증연구)

  • The in-situ remediation of a solidified stratum containing a large amount of fine-texture material like clay or organic matter in contaminated soil faces limitations such as increased remediation cost resulting from decreased purification efficiency. Even if the soil conditions are good, remediation generally requires a long time to complete because of non-uniform soil properties and low permeability. This study assessed the remediation effect and evaluated the field applicability of a methodology that combines pneumatic fracturing, vacuum extraction, and plasma blasting (the PPV method) to improve the limitations facing existing underground remediation methods. For comparison, underground remediation was performed over 80 days using the experimental PPV method and chemical oxidation (the control method). The control group showed no decrease in the degree of contamination due to the poor delivery of the soil remediation agent, whereas the PPV method clearly reduced the degree of contamination during the remediation period. Remediation effect, as assessed by the reduction of the highest TPH (Total Petroleum Hydrocarbons) concentration by distance from the injection well, was uncleared in the control group, whereas the PPV method showed a remediation effect of 62.6% within a 1 m radius of the injection well radius, 90.1% within 1.1~2.0 m, and 92.1% within 2.1~3.0 m. When evaluating the remediation efficiency by considering the average rate of TPH concentration reduction by distance from the injection well, the control group was not clear; in contrast, the PPV method showed 53.6% remediation effect within 1 m of the injection well, 82.4% within 1.1~2.0 m, and 68.7% within 2.1~3.0 m. Both ways of considering purification efficiency (based on changes in TPH maximum and average contamination concentration) found the PPV method to increase the remediation effect by 149.0~184.8% compared with the control group; its average increase in remediation effect was ~167%. The time taken to reduce contamination by 80% of the initial concentration was evaluated by deriving a correlation equation through analysis of the TPH concentration: the PPV method could reduce the purification time by 184.4% compared with chemical oxidation. However, the present evaluation of a single site cannot be equally applied to all strata, so additional research is necessary to explore more clearly the proposed method's effect.

  • The land area, which is on the inner side of the water line, is controlled to a certain degree due to the purposes of space utilities controllers' respective roles, but the sea area is not so much controlled as the land area. However, as interest in the security and utility of the marine space, as well as that in the consistent development of the land space, has increased, there has been increase of the requirement for information about marine data and environment. The object of this study, Busan, plays various roles such as a harbor and bay, a tourist resort, and a port unlike coastal seas of other areas, and thus needs systematic control because it has numerous environmental factors. However, there are limitations in the number of data about Busan coastal sea constantly provided while the acquisition of data from main points and many areas is important for the analysis of marine contamination in Busan coastal sea area. Thus, using nautical charts with numerical values in preparing increase of the demand of them according to the degree of information requirement for the analysis of the changes in coastal areas, this study constructs property data and space data by combining vertex data about Busan's 17 coastal sea areas provided by NFRDI, and tries to present the analytical techniques for grasping the environmental conditions and continuity of the coastal areas and methods of their utilities, using GIS Geostatistical Analysis Technique.

A study on the contamination level of pathogenic microorganisms in beef distribution stages (소고기의 유통 단계별 병원성 미생물 오염도에 관한 연구)

  • Contamination levels of pathogenic microorganisms in 145 cases of beef, which were distributed in Gwangju province, had been investigated in each distributed stage and also monitored by general bacterial count and E coli count index. General bacterial count of beef from the slaughterhouse was 10$^4$cfu/g less than the level of promotion(10 cfu/$\textrm{cm}^2$) and E coli count index was also under the level of 10$^2$cfu/$\textrm{cm}^2$ recommended level of the ministry of agriculture and forestry. Pathogenic microorganisms were detected from 23.2% of samples in the consumption stage, 12.5% in the slaughtering stage and 5.6% in the transporting and processing stage. Staphylococcus aureus was isolated in the largest number and its ratio was 9.0%, listeria monocytogenes 5.5% and salmonella spp 1.4%. There were no samples that bacteria had been detected dually. E coli O157:H7 and campylobacter jejuni were not isolated. In raw and chilled beef, isolation rate of pathogenic microorganisms were 13.3% and 16.5% each. Especially in raw beef, L monocytogenes was. isolated in 3 samples among 30 cases (10%) and S aureus in one sample (3.3%). According to a scale of meat store, isolation rates of pathogenic microorganisms were different. It was 28.6% in the small-scale meat store and 16.7% in the large-scale meat store each. Four cases (16.7%) of S aureus were isolated in the large-scale meat store and seven cases (20.0%) of L monocytogenes and 2 cases (5.7%) of salmonella spp were isolated in the small-scale meat store. S aureus was isolated in two places among 10 feeding facilities of the elementary school. This result shows that the sanitation of elementary school feeding facilities is so poor and more careful policy consideration is needed. Eleven strains of S aureus isolated showed ${\beta}$-hemolysis on blood agar, 1 strain ${\alpha}$-hemolysis, and 1 strain ${\gamma}$-hemolysis. Isolated strains of L monocytogenes were reconfirmed in 560 bp by PCR. Conclusively, these results show that the sanitary condition in the stages of slaughtering, transportation-processing and consumption influences the degree of pathogenic microorganisms contamination in beef severely It is necessary to apply thoroughly hazard analysis critical control point in a process of beef distribution and also to develop rapid test methods for microorganism diagnosis. This effort is very important for the supply of safe and clean meat from farm to table and helpful for the improvement of public health.

Hydrogeochemistry of Groundwaters at the Gogum island area in Jeonnam, Korea (전남 고금도 지역 지하수의 수리지구화학)

  • The object of this study is to investigate the contamination degree and the interpretation of sea water intrusion phenomena with hydrogeochemical and hydrogen-oxygen stable isotope of coastal aquifer in the Gogum area, Korea. The physical characteristics of groundwaters is the neutral pH condition and transitional Redox environments, and groundwater is affected by sea water & surface water. The chemical properties of groundwaters are showing an increase in contamination owing to the sea water intrusion, waste water from the surface and agricultural chemicals. In the case of chloride, 6 samples of the groundwater in the study area are in excess of the drinking water standard. The Piper diagram shows the contamination in GG-4 and 14 by sea water intrusion. GG-3, 7 and 13 dominate the Na-HCO$_{3}$ type water and regional (GG-14) is indicated to dominate the Na-Cl type water such as sea water. According to the Sl (saturation index), sea water is oversaturated with respect to calcite and dolomite, GG-3, 14 and 18 are approaching the saturation state. The hydrogen-oxygen stable isotope ratio of groundwaters originates in the meteoric water, and groundwaters of GG-1, 5 and 14 display high oxygen isotope value due to surface water trespass and sea water intrusion. The result of this study, GG-14 is contaminated by sea water intrusion, groundwaters expected GG-3, 7 and 13 is in progress to artificial pollution and sea water intrusion.

Causes of Bacterial Growth in Gels and Gel Containers Used for Ultrasonography (초음파 검사용 젤과 젤 용기의 세균증식 원인)

  • This study aimed to investigate the causes of bacterial growth to prevent infection caused by ultrasound gel and gel containers in contact with patients during ultrasonography. To investigate bacterial contamination during manufacturing or storage, we cultured ultrasound gels originally supplied from three manufacturers. To analyze bacterial growth according to the lapse of time and frequency of use of the ultrasound gel container, the gel and container were cultured at regular intervals every week for 4 weeks. In addition, to determine the source of infection, the examiner's hand was inspected with hand plate and the degree of bacterial contamination was measured before the test. As a result of the study, bacteria were not detected in the gel provided at the initial supply, and in the gel and gel container used repeatedly for 4 weeks, the same bacteria residing on the skin were identified in the examiner's hand, such as Staphylococcus epidermidis, Micrococcus luteus, Leuconostoc mesenteroid spp cremoris, Kocuria rhizophila, and etc. Separated strains were classified as those of the low- or non-pathogenicity; however, most of these strains may render fatal consequences to patients of lower level of immunity due to acquired tolerance to antibiotics. At week 1, when the number of tests was the highest, 44 colonies were identified, and at week 4, when the number of tests was the lowest, 4 colonies were identified. As r=0.994, it was found that the number of colonies increased as the number of tests increased. In conclusion, it was confirmed that the cause of the infection was not the ultrasound gel, but the examiner's hand. The ultrasound gel or gel container may be contaminated by skin flora of examiner's hands, which can cause opportunistic infection in patients with low immunity.   • Compomer that release fluoride could be used on proximal caries of child effectively. But oral cavity is always wet, so saliva inhibits bonding of tooth and compomer. When the saliva exist on bonding, it can be occured microleakages. The purpose of this study was to evaluate the influence of salivary contamination on compomer restoration and degree of microleakage according to restoration methods. Dyract $AP^{(R)}$ and prime and $bond^{(R)}$ NT was applied by the manufacture s instructions. Elipar Trilight was applied for light curing. Saliva pool was made for reconstruction of oral cavity. Two premolar was embedded in acrylic resin. After class II cavity preperation, Dyract $AP^{(R)}$ was restored under several condition, the specimen was thermocycled 500 times with 30 second dwell time. 0.5% methylene blue was used for microleakage test. Micoleakage was measured by the ratio of the infiltration length to occlusal and gingival side interface. Data were analyzed statistically using Kruskal Wallis Test, Mann-Whitney Test. The Result were as follows ; 1. In occlusal side, there were no statistical differences. 2. In gingival side, there were no statistical differences in Group III ($ContactMatrix^{TM}$, Rubber dam, $Oraseal^{(R)}$), Group IV (No saliva contamination). 3. In gingival side, there were no statistical differences in Group I$(ContactMatrix^{TM})$, II($ContactMatrix^{TM}$, Rubber dam). 4. In gingival side, there were statistical differences in Group I$(ContactMatrix^{TM})$, II($ContactMatrix^{TM}$, Rubber dam).

Mating System in Seed Orchard of Japanese Red Pines Revealed by DNA Markers (DNA 표지에 의한 채종원내 소나무 교배양식 구명)

  • To investigate the mating system of clones in the seed orchard of Japanese red pine, parameters of mating system, such as outcrossing rates, number of potential pollen contributors, and degree of pollen contamination, were estimated in the seed orchard of Japanese red pines on the basis of DNA data including 4 nSSR and 6 cpSSR markers. Estimates of outcrossing rates were ranged from 94.9 to 100% with an average of 98.9% on the basis of the analysis of cpSSR haplotypes. They were ranged from 90.3% to 100% with an average of 95.9% on the basis of the analysis of nSSR genotypes. However, cross checking of both DNA markers revealed that the seeds presumed to be products of self pollination were turned out to be generated by pollination between mother tree and other tree (i.e., 100% of cumulative outcrossing rate). Estimates of pollen contamination ranged from 43.6% (Gangwon-10) to 56.4% (Gangwon-12) with the average of 48.9%. On the basis of pooled cpSSR haplotype of each seed, maximum number of 21 pollen contributors were verified from the seeds reproduced by Kyungbuk-38. Minimum number of 13 pollen contributors were verified in Gangwon-10. Mean of 16.2 pollen contributors were verified from a total of 5 mother trees. In conclusion, considering pretty high outcrossing rates between clones within a seed orchard, it may be expected that a fairly good genetic potential of the seeds, produced in '77 plot of the seed orchard of Japanese red pines at Anmyeon island, may be guaranteed. Observed results from the analysis of mating system of Japanese red pines in a '77 plot of the seed orchard may also provide useful information for the establishment and management of the seed orchard of the progressive generation.

Geochemical Characteristics and Contamination of Surface Sediments in Streams of Gwangju City (광주광역시 하천의 표층퇴적물에 대한 지구화학적 특성과 오염)

  • In order to investigate the geochemical characteristics of surface sediments in streams of Gwangju City, sediment samples from the main stream of Yeongsan river, Hwangryong river, and Gwangjucheon in the city were collected and analyzed for grain size and metal and organic carbon contents. The sediment types of the streams widely vary from pebble to mud. The metal contents in the sediments from Yeongsan river and Hwangryong river are mainly dependent on the grain size of the sediments and the geology around the streams, while the sediments in Gwangiucheon are controlled by organic matter contents from the domestic sewage. The enrichment factor (Ef) and index of geoaccumulation (Igeo) representing the degree of metal contamination in the sediments are relatively low in the mainstream of Yeongsan river and Hwangryong river. However, those of Gwangjucheon show EF values of P=8.30, Cu=5.54, Zn=14.28 and Pb=7.41 and Igeo values of P=3.78, Cu=2.79, Zn=3.66 and Pb=1.59. The heavy metal contamination is especially significant near the area where the Seobangcheon and Donggyecheon branches of Gwanjucheon, join. Therefore, it is suggested that the metallic contaminations of these small streams are significantly influenced by the domestic sewage of the city.

The Changes on Total Bacterial Counts and Coliform Counts of Human Milk vs Formula Milk Stored under Cold and Room Temperature Conditions (냉장 및 실온 보존에 따른 모유와 환원조제분유 중의 총균수와 대장균수의 변화)

  • Human milk is normally contaminated with various microorganisms, which seem to produce no ill effects. A contamination of hand-expressed and pump-expressed human milk is a major concern in the collection of storage milk. In this study we compared milks collected by two methods, hand expression and suction breast pump, to quantify differences in the degree of bacterial contamination. Thirty-one samples had been manually expressed. The mean of total bacterial counts was 10,600 CFU/mL (range: 360 ∼59,200 CFU/mL) and coliform counts was 43 CFU/mL (range: 20 ∼ 1,060 CFU/mL) in these samples. Whereas in the 118 breast pump-expressed samples, the mean of total bacterial counts was 20,200 CFU/mL (range: 240 ∼ 492,000 CFU/mL) and coliform counts was 158 CFU/mL (range: 4∼10,600 CFU/mL). There was no bacterial growth when the samples were incubated for 10 days at 4$\^{C}$. We also compared total bacterial growth in colostrum and in matured human milk for 24 hr at 20$\^{C}$ and 30$\^{C}$. Although bacterial growth had not shown for 24 hr at 20$\^{C}$, but shown slight growth in colostrum and rapidly increase in matured human milk for 24 hr at 30$\^{C}$. The coliform bacteria in all samples, particulary in formula milk, had grown at 20$\^{C}$ and 30$\^{C}$.

Hydrogeochemistry of shallow groundwaters in western coastal area of Korea : A study on seawater mixing in coastal aquifers (서해 연안지역 천부지하수의 수리지구화학 : 연안 대수층의 해수 혼입에 관한 연구)

  • Salinization is an important environmental problem encountered in coastal aquifers. In order to evaluate the salinization problem in the western coastal area of Korea, we have performed a regional hydrochemical study on shallow well groundwaters (N=229) collected within 10 km away from the coastline. The concentrations of analyzed solutes are very wide in range, suggesting that the hydrochemistry is controlled by several processes such as water-rock interaction, seawater mixing, and anthropogenic contamination. Based on the graphical interpretation of cumulative frequency curves for some hydrochemical parameters (esp., $Cl^{-}$ and ${NO_3}^-$), the collected water samples were grouped into two major populations (1) a background population whose chemistry is predominantly affected by water-rock interaction, and (2) an anomalous population which records the potential influences by either seawater mixing or anthropogenic pollution. The threshold values obtained are 34.7 mg/l for $Cl^{-}$ and 37.2 mg/l for ${NO_3}^-$, Using these two constituents, groundwaters were further grouped into four water types as follows (the numbers in parenthesis indicate the percentage of each type water) : (1) type 1 waters (38%) that are relatively poor in $Cl^{-}$ and ${NO_3}^-$, which may represent their relatively little contamination due to seawater mixing and anthropogenic pollution; (2) type 2 waters (21%) which are enriched in $Cl^{-}$, Indicating the considerable influence by seawater mixing; (3) ${NO_3}^-$-rich, type 3 waters (11%) which record significant anthropogenic pollution; and (4) type 4 waters (30%) enriched in both $Cl^{-}$ and ${NO_3}^-$, reflecting the effects of both seawater mixing and anthropogenic contamination. The results of the water type classification correspond well with the grouping on a Piper's diagram. On a Br x $10^4$versus Cl molar ratio diagram, most of type 2 waters are also plotted along or near the seawater mixing line. The discriminant analysis of hydrochemical data also shows that the classification of waters into four types are so realistic to adequately reflect the major process(es) proposed for the hydrochemical evolution of each water type. As a tool for evaluating the degree of seawater mixing, we propose a parameter called 'Seawater Mixing Index (S.M.I.)’ which is based on the concentrations of Na, Mg, Cl, and $SO_4$. All the type 1 and 3 waters have the S.M.I. values smaller than one, while type 2 and type 4 waters mostly have the values greater than 1. In the western coastal area of Korea, more than 21% of shallow groundwaters appear to be more or less affected by salinization process.
