Optimization of Ethanol Extraction Conditions from Propolis (a Bee Product) Using Response Surface Methodology (반응표면분석법을 이용한 프로폴리스의 에탄올 추출조건 최적화)

  • A central composite design was used to optimize extraction of propolis materials using ethanol. The independent variables in extraction experiments were ethanol concentration (50, 60, 70, 80, 90%, v/v) and extraction time (1, 2, 3, 4, and 5 h). Higher ethanol concentration and shorter extraction time increased total polyphenol content, but total polyphenol concentration began to decrease when ethanol concentration was higher than 80% (v/v). Ethanol concentration was more important than extraction time in optimization of total polyphenol content in propolis extracts. Electron-donating ability increased with ethanol concentration and shorter extraction time, with ethanol concentration being of greater significance. Antioxidant ability in extracts was optimal at an ethanol concentration of 65 - 75% and with an extraction time of 2.2 - 3.6 h. Nitrite-scavenging ability was increased with use of higher ethanol concentration and shorter extraction time. Total flavonoid content was maximized with an ethanol concentration of 68 - 82% and an extraction time of 2.4 - 3.7 h. Total flavonoid content was affected by both ethanol concentration and extraction time. By superimposition of contour plots, an ethanol concentration of 72 - 82% and an extraction time of 2.2 - 3.3 h were optimal for preparation of propolis extracts.

Distributions of Water Temperature and Salinity in the Korea Southern Coastal Water During Cochlodinium polykrikoides Blooms (C. polykrikoides 적조 발생시의 한국 남해안의 수온 및 염분 분포)

  • In order to elucidate the cause of Cochlodinium polykrikoides blooms in the Korea southern coastal water, we investigated observational data of water temperatures and salinities in summer and winter, obtained from the stoppage of ship by NFRDI (National Fisheries Research and Development Institute) as well as composite images by NOAA from 1995 to 2008. Cochlodinium polykrikoides blooms occurred when water temperature was approximately $25.0{\sim}26.0^{\circ}C$ and salinity was 31.00 psu on average in Narodo neighboring seas. Different thermohaline fronts were observed between the Korea southern coastal water and the open sea water in summer and winter, respectively. That is, in winter four fronts were observed between the Korea southern coastal water with low temperature and low salinity, intermediate water originated from Tsushima Warm Current, Tsushima Warm Current with high temperature and high salinity, and the China coastal water with low temperature and low salinity. In contrast, in summer two fronts were observed between the Korea southern coastal water with low temperature and high salinity, Tsushima Warm Current with high temperature and low salinity, and the China coastal water with high temperature and high salinity. These thermohaline fronts also proved to be formed by two water masses with a different physical property, in terms of T-S diagrams. Consequently, we noticed that C. polykrikoides blooms occurring in Narodo neighboring seas in summer had a close relationship with thermohaline fronts observed between the Korea southern coastal water and Tsushima Warm Current.

Development of National R&D Information Navigation System Based on Information Filtering and Visualization (정보 필터링과 시각화에 기반한 국가R&D정보 내비게이션 시스템 개발)

  • This paper aim; to develop the National R&D Information Navigation System(NRnDINS) that is convenient and easy to use by the researchers on the basis of information filtering and visualization by converging and integrating the three types of the contents, namely, paper, report and project at the stage of development of the information system An information system is developed by establishing ontology and RDF on the three types of contents, and by applying information filtering and semantic search technology after having created the prototype for the screen by reflecting the user needs analysis and information visualization elements surveyed at the previous stage of information service planning. In this paper, to make the measure for information filtering, R&D navigation index is prosed and implemented, and NRnDINS capable of integrated search of the R&D contents through information visualization is developed. Also, for the testing of the developed system, the preference survey for its design by 1m persons and usability test of the system by 10 users are performed The result of the survey on the preference for the design is affirmative with 85% of the subjects finding it favorable and the composite receptivity is good with the score of 87.2 the results of the usability test. However, it was also found that further development of the personalization functions is needed. It is hoped that the R&D navigation index of the proposed and implemented in this paper would present quantitative objectivity and will induce further development of other information filtering index of contents in the future.

Multiscale Analysis on Expectation of Mechanical Behavior of Polymer Nanocomposites using Nanoparticulate Agglomeration Density Index (나노 입자의 군집밀도를 이용한 고분자 나노복합재의 기계적 거동 예측에 대한 멀티스케일 연구)

  • In this study, multiscale analysis in which the information obtained from molecular dynamics simulation is applied to the continuum mechanics level is conducted to investigate the effects of clustering of silicon carbide nanoparticles reinforced into polypropylene matrix on mechanical behavior of nanocomposites. The elastic behavior of polymer nanocomposites is observed for various states of nanoparticulate agglomeration according to the model reflecting the degradation of interphase properties. In addition, factors which mainly affect the mechanical behavior of the nanocomposites are identified, and new index 'clustering density' is defined. The correlation between the clustering density and the elastic modulus of nanocomposites is understood. As the clustering density increases, the interfacial effect decreased and finally the improvement of mechanical properties is suppressed. By considering the random distribution of the nanoparticles, the range of elastic modulus of nanocomposites for same value of clustering density can be investigated. The correlation can be expressed in the form of exponential function, and the mechanical behavior of the polymer nanocomposites can be effectively predicted by using the nanoparticulate clustering density.

Optimal Fermentation Conditions of Hyaluronidase Inhibition Activity on Asparagus cochinchinensis Merrill by Weissella cibaria

  • This study was conducted to evaluate the hyaluronidase (HAase) inhibition activity of Asparagus cochinchinesis (AC) extracts following fermentation by Weissella cibaria through response surface methodology. To optimize the HAase inhibition activity, a central composite design was introduced based on four variables: the concentration of AC extract ($X_1$: 1-5%), amount of starter culture ($X_2$: 1-5%), pH ($X_3$: 4-8), and fermentation time ($X_4$: 0-10 days). The experimental data were fitted to quadratic regression equations, the accuracy of the equations was analyzed by ANOVA, and the regression coefficients for the surface quadratic model of HAase inhibition activity in the fermented AC extract were estimated by the F test and the corresponding p values. The HAase inhibition activity indicated that fermentation time was most significant among the parameters within the conditions tested. To validate the model, two different conditions among those generated by the Design Expert program were selected. Under both conditions, predicted and experimental data agreed well. Moreover, the content of protodioscin (a well-known compound related to anti-inflammation activity) was elevated after fermentation of the AC extract at the optimized fermentation condition.

Optimization of Preparation Condition on Oriental Melon Jam by Response Surface Methodology (반응표면 분석에 의한 참외잼의 제조조건 최적화)

  • This study was carried out to establish the preparation condition of muskmelon jam. A central composite design was applied to investigate effects of muskmelon paste content(40, 45, 50, 55, 60 g), fructose ratio of sugar(20, 35, 50, 65, $80\%$) and pectin addition(l, 2, 3, 4, 5 g). The maximum sugar content was 61.48 $^{\circ}$Brix in 41.04 g of muskmelon paste content, $48.10\%$ of fructose ratio of sugar and 2.12 g of pectin content. The maximum value of softness was 2.71 g in 45.06 g of muskmelon paste content, $79.46\%$ of fructose ratio of sugar and 2.71 g of pectin addition. The minimum value of jelly strength was $0.04\;g{\cdot}cm$ in 47.80 g of muskmelon paste content, $63.0\%$ of fructose ratio of sugar and 1.99 g of pectin addition. The maximum value of organoleptic overall palatability was 5.89 in 55.65 g of muskmelon paste content, $73.19\%$ of fructose ratio of sugar and 2.42 g of pectin addition. The optimum conditions predicted for each corresponding physicochemical and organoleptic properties of muskmelon jam were 55.2 g(muskmelon paste content), $76.3\%$(fructose ratio of sugar) and 2.5 g(pectin addition).

FLUORIDE RELEASE AND MICROHARDNESS OF GIOMER ACCORDING TO TIME (Giomer의 불소 유리 양상 및 미세경도에 관한 연구)

  • The aim of this study was to evaluate the fluoride release and microhardness of Beautifil II as giomer(Group I), F2000 Compomer as compomer(Group II), GC Fuji II LC Capsule as resin-modified glass ionomer(Group III) and $Filtek^{TM}$ Z350 as composite resin(Group IV) according to time. Forty discs(5 mm diameter and 2 mm height) were prepared for each material. Each disc was immersed in 3 ml of de-ionized water within polyethylene tube and stored at $37^{\circ}C$. Evaluations were performed by pH/ISE meter for analysis of fluoride release and hardness testing machine for analysis of microhardness over 31 days. The results can be summarized as follows : 1. For all groups except group IV, the greatest fluoride release was observed after the first day of the study period and then dramatically diminished over time. On the 7th day of the study period, fluoride release level was stabilized. 2. Group III showed the highest fluoride release among test groups and then group II, group I were followed. Significant difference in cumulative fluoride release over 31 days was found between each groups. Group IV showed no fluoride release during study period. 3. Group IV showed the highest microhardness among test groups and then group I, group II, group III were followed. Significant difference in microhardness was found between each group, except between group I and group II. 4. After 31 days, microhardness was slightly diminished in every group. However, no significant difference was found.

A Study on Dissolve and Remove Analysis of Organic Chemicals Using a Mixed Method of Advanced Oxidation and Micro Filtering - Water Drinking Point - (고도산화와 정밀여과의 혼성공법을 이용한 유기화학물질의 분해 및 제거분석에 관한 연구 - 먹는 물을 중심으로 -)

  • It is impossible to remove toxic organic substances that are recognized as a cancer caused suspicious element in drinking water using the conventional water purification method. This study introduces groundwater into a reaction chamber as an effective amount of water to process this water using a mixed method of AOP oxidation and M/F membrane and purifies it as a desirable level by artificially mixing such toxic substances in order to effectively process the water. Based on this fact, this study configures an optimal operation condition. The VOCs existed in toxic substances was investigated as a term of phenol and toluene, and agricultural chemicals were also investigated as a term of parathion, diazinon and carbaryl. The experiment applied in this study was performed using a single and composite soolution. In the operation condition applied to fully dissolve and remove such substances, the amount of $H_2O_2$ injected in the process was 150 mL of a fixed quantity, the value of pH was configured as $5.5{\sim}6.0$, the temperature was controlled as a range of $12{\sim}16^{\circ}C$, the dissolved amount of ozone was applied more than 5.0 mg/L, the reaction time was determined as an optimal condition, such as $30{\sim}40$ minutes, and the segregation membrane in the same reactor was determined for acquire water drinking of large quantity using a pore size of $0.45{\mu}m$ M/F membrane.

A poroelastic model for ultrasonic wave attenuation in partially frozen brines (부분 동결된 소금물에서의 초음파감쇠에 대한 다공성탄성 모델)

  • Although there are many possible mechanisms for the intrinsic seismic attenuation in composite materials that include fluids, relative motion between solids and fluids during seismic wave propagation is one of the most important attenuation mechanisms. In our previous study, we conducted ultrasonic wave transmission measurements on an ice-brine coexisting system to examine the influence on ultrasonic waves of the unfrozen brine in the pore microstructure of ice. In order to elucidate the physical mechanism responsible for ultrasonic wave attenuation in the frequency range of 350.600 kHz, measured at different temperatures in partially frozen brines, we employed a poroelastic model based on the Biot theory to describe the propagation of ultrasonic waves through partially frozen brines. By assuming that the solid phase is ice and the liquid phase is the unfrozen brine, fluid properties measured by a pulsed nuclear magnetic resonance technique were used to calculate porosities at different temperatures. The computed intrinsic attenuation at 500 kHz cannot completely predict the measured attenuation results from the experimental study in an ice-brine coexisting system, which suggests that other attenuation mechanisms such as the squirt-flow mechanism and wave scattering effect should be taken into account.

Optimization of Microwave-assisted Extraction Conditions for Total Catechin and Electron Donating Ability of Grape Seed Extracts (포도씨 추출물의 총 카테킨 함량과 전자공여능에 대한 마이크로웨이브 추출조건 최적화)

  • Microwave-assisted extraction (MAE) of grape seeds was performed under the different conditions based on a central composite design for independent variables of microwave power ($0{\sim}120\;W$), ethanol concentration ($0{\sim}100%$) and extraction time ($1{\sim}5\;min$). Response surface methodology (RSM) was used to predict the optimum extraction conditions for three dependent variables in grape seed extracts: total yield, total catechin and electron donating ability. Determination coefficients ($R^2$) of regression equations for the three dependent variables were higher than 0.9 (p < 0.01). The optimal MAE conditions to yield the maximum value of total catechin (434.16 mg%) were 122.76 W microwave power, 42.88% ethanol and 4.67 min extraction time. The superimposed contour maps for maximizing the three dependent variables indicated that the MAE condition ranges were 75150 W, 4060% ethanol and 3.55.0 min. The predicted values at the optimized conditions (6.72% total yield, 408.65 mg% total catechin, and 83.33% electron donation ability) were similar to the experimental values. The optimized MAE (112.5 W, 50% EtOH, 4.2 min) was more efficient than the conventional solvent extraction using 80% EtOH, $60^{\circ}C$ for 3h and 150 rpm.
