Effects of Korean Herbal Bathing Extracts Composition on Th2 Cytokine Production in MC/9 Mast Cells (한방 입욕제 조성물이 MC/9 mast cell에서의 Th2 cytokine 발현에 미치는 영향)

  • Objectives The purpose of this study is to investigate the effects of korean herbal bathing extracts composition 1 and composition 2 on Th2 cytokine production in MC/9 mast cells. Methods The effects of composition 1, 2 was analyzed by ELISA and Real-time PCR in MC/9 mast cells. Levels of IL-5, IL-13 were measured using enzyme-linked immunosorbent assays(ELISA). mRNA levels of IL-4, IL-5, IL-6, IL-13 were analyzed with Real-time PCR. Results Composition 1, 2 inhibited the IL-5, IL-13 production significantly(p<.001) in comparison to PI-control group at concentration of 100 ${\mu}g/mL$, 200 ${\mu}g/mL$. Composition 1, 2 inhibited the IL-4, IL-5, IL-13 mRNA expression significantly in comparison to PI-control group at concentration of 100 ${\mu}g/mL$, 200 ${\mu}g/mL$. Composition 1 inhibited the IL-6 mRNA expression significantly in comparison to PI-control group at concentration of 200 ${\mu}g/ml$. Composition 2 inhibited the IL-6 mRNA expression significantly in comparison to PI-control group at concentration of 100 ${\mu}g/mL$, 200 ${\mu}g/mL$. Conclusions These results indicate that composition 1, 2 has the effect of decreasing the Th2 cytokine production in the MC/9 mast cell.

  • When predicting the concentration of fine dust using deep learning, there is a problem that the characteristics of a high concentration of 81㎍/m3 or more are not well reflected in the prediction model. In this paper, a comparison through predictive performance was conducted to confirm the results of reflecting the characteristics of fine dust in the high concentration area according to the deep learning algorithm. As a result of performance evaluation, overall, similar levels of results were shown, but the RNN model showed higher accuracy than other models at concentrations of "very bad" based on AQI. This confirmed that the RNN algorithm reflected the characteristics of the high concentration better than the DNN and LSTM algorithms.

  • This study investigates the evaluation on simple comparison and seeking hidden figures works under LED illumination conditions according to three types of color temperatures (7000K, 5000K, 3000K) in the classroom, Since the objective of this study is to develop an electric lighting conditions suitable for students in the classroom, the performance of concentration and visual discrimination by students under three types of LED illumination conditions were analyzed. The 4 kinds of simple tests concerning with concentration and 2 types of tests for visual discrimination were developed, and test results under natural light and LED illumination by 3 types of color temperatures were analyzed. There were differences on concentration as well as visual discrimination performance in most cases by t-test ($p{\leq}0.05$). For further analysis among three types of color temperatures and natural light, ANOVA tests were performed. The test results shows that illumination condition plays an important role when students try to concentrate on simple comparison of characters, figures or colors and seek hidden figures. The experimental results of this study might be applied to designing better luminous environment.

The Concentration of Serum Lipids and Protein Electrophoresis Fractions in Thyroid Disease Patients

  • We analyzed and compared the concentration of total cholesterol (CHOL), high density lipoprotein (HDL) cholesterol, low density lipoprotein (LDL) cholesterol and triglyceride (TG) in serum and the serum protein electrophoresis fractions of thyroid disease patients. In comparison with the average of reference, our data showed that the average concentration of CHOL, LDL cholesterol and TG in hyperthyroidism patients were decreased significantly, but HDL cholesterol was increased significantly. In hypothyroidism patients, CHOL, HDL cholesterol, LDL cholesterol and TG were all increased significantly. In comparison of the concentration of lipids in each patient to reference range, 28.3% of hyperthyroidism patients showed abnormally low level of total cholesterol. In the patients with hypothyroidism, the percentage of patients showed abnormally high level of CHOL, HDL cholesterol, LDL cholesterol and TG were 37.7%, 10%, 68.8% and 49.1%, respectively. In our studies of serum protein electrophoresis, the average of ${\alpha}_2$-globulin and $\gamma$-globulin in hyperthyroidism patients were increased and $\beta$-globulin was decreased significantly. In hypothyroidism patients, the average of $\gamma$-globulin was increased and $\beta$-globulin was decreased significantly. In comparison of protein fractions of each patient to reference range, 38.3% and 50.0% of hyperthyroidism patients showed abnormally high levels of ${\alpha}_2$-globulin and $\gamma$-globulin, but 73.3% of patients showed abnormally low level of $\beta$-globulin. In hypothyroidism patients, 70.4% of patients were abnormally decreased in $\beta$-globulin and 63.9% of patients were abnormally increased in $\gamma$-globulin. These data suggest that the concentrations of CHOL, HDL cholesterol, LDL cholesterol and TG are not critical data for clinical interpretation of hyperthyroidism, but the levels of them are useful for interpretation of hypothyroidism patients. Our results of serum protein electrophoresis suggest that the concentration of serum protein electrophoresis fractions can be useful to understand the thyroid disease.

COMPARISON OF THE FIRE SUPPRESSION PERFORMANCE OF HALON REPLACEMENT AGENTS

  • HFC-227ea and HCFC Blend A were evaluated using full-scale fire tests to obtain information on their fire suppression performance, drop-in capability, thermal decomposition products and physical behaviour of the agent such as its flow characteristics in the piping system. Also, full-scale tests were conducted with Halon 1301 to provide a basis for comparison. Halon 1301, at concentrations of 5% to 7.5%, showed effective total-flooding fire- extinguishing performance for all test scenarios. HFC-227ea, at a design concentration of 7.6% or higher, and HCFC Blend A, at a design concentration of 12%, extinguished all fires in the test facility, however, these agents produced higher concentrations of acid gases than Halon 1301. The quantity of the acid gases generated during fire suppression was dependent on agent concentration, agent discharge time, fire type and size as well as extinguishment time.

The Study on the Cytotoxicity of Compounds of Bee Venom for Herb-Acupuncture (봉독(蜂毒) 약침액(藥鍼液)의 세포독성(細胞毒性)에 관(關)한 연구(硏究))

  • Objective : This study was undertaken to determine which compound of Bee Venom for herb-acupuncture has cytotoxicity on mouse mast cell line. Methods : We compared crude bee venom and its compounds such as melittin, mast cell degranulating peptide (MCD peptide), apamin with control groups on cytotoxicity by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. Results & Conclusion : 1. Crude bee venom showed significant cytotoxic effect(p<0.01) in 1 hour treatment with $1{\mu}g/m{\ell}$ in comparison with control group in 1 hour treatment with low concentration of $10-4{\mu}g/m{\ell}$, $10-3{\mu}g/m{\ell}$, $10-2{\mu}g/m{\ell}$, $10-1{\mu}g/m{\ell}$ and $1{\mu}g/m{\ell}$, but it showed no significant cytotoxic effect in 6 hours treatment. 2. Melittin group showed no significant cytotoxic effect in comparison with control group in 1 and 6 hours treatment with low concentration of $10-4{\mu}g/m{\ell}$, $10-3{\mu}g/m{\ell}$, $10-2{\mu}g/m{\ell}$, $10-1{\mu}g/m{\ell}$ and $1{\mu}g/m{\ell}$. 3. MCD peptide and Apamin group showed no significant cytotoxic effect in comparison with control group in 1 and 6 hours treatment with low concentration of $10-4{\mu}g/m{\ell}$ $10-3{\mu}g/m{\ell}$, $10-2{\mu}g/m{\ell}$, $10-1{\mu}g/m{\ell}$ and $1{\mu}g/m{\ell}$. 4. Crude bee venom showed significant cytotoxic effect(p<0.01) in 1 and 6 hours treatment in comparison with control group in 1 and 6 hours treatment with high concentration of $10{\mu}g/m{\ell}$, $20{\mu}g/m{\ell}$ and $102{\mu}g/m{\ell}$. 5. Melittin group showed significant cytotoxic effect(p<0.01) in 1 hour treatment in comparison with control group in 1 hour treatment with high concentration of $10{\mu}g/m{\ell}$ but it showed no significant cytotoxic effect in 6 hours treatment. 6. Crude bee venom and its compounds have more cytotoxic effect in 1 hour treatment than in 6 hours treatment. It means cytotoxicity tends to decrease according to the treatment time.

Analysis of Measurement Error for PM-10 Mass Concentration by Inter-Comparison Study (비교 측정을 통한 PM-10 질량농도의 오차 분석)

  • Particular attention is paid to the validity of PM-10 measurement using beta attenuation method. In this study, the inter-comparison studies were undertaken for PM-10 sampler with different measuring methods. The three beta attenuation monitoring instruments (BAM1020, FH C-14, and DIGS-150) and filter based gravitational method using High Volume Air Sampler were collocated and mass concentrations were compared from August to November, 2006. The results of the investigation show that beta attenuation monitoring instruments are comparable with gravitational method within 10% of measurement error. For specific conditions under high humidity and high concentrations, the inter-comparison results also showed a relatively good agreement and this can be explained as the effect of inlet heating of beta attenuation instrument. Subsequently, this study shows that beta attenuation monitoring instruments can be equivalently used with reasonable validity when compared gravimetric method.

Comparison of the Stress Concentration Factors for GFRP Plate having Centered Circular Hole by Three Resource-Conserving Methods

  • Fiber reinforced plastic (FRP) composites have drawn increasing attentions worldwide for decades due to its outstanding properties. Stress concentration factor (SCF) as an essential parameter in materials science are critically considered in structure design and application, strength assessment and failure prediction. However, investigation of stress concentration in FRP composites has been rarely reported so far. In this study, three resource-conserving analyses (Isotropic analysis, Orthotropic analysis and Finite element analysis) were introduced to plot the $K_T^A-d/W$ curve for E-glass/epoxy composite plate with the geometrical defect of circular hole placed centrally. The plates were loaded to uniaxial direction for simplification. Finite element analysis (FEA) was carried out via ACP (ANSYS composite prepost module). Based on the least squares method, a simple expression of fitting equation could be given based on the simulated results of a set of discrete points. Finally, all three achievable solutions were presented graphically for explicit comparison. In addition, the investigation into customized efficient SCFs has also been carried out for further reference.

High concentration ratio approximation of linear effective properties of materials with cubic inclusions

  • This paper establish a high concentration ratio approximation of linear elastic properties of materials with periodic microstructure with cubic inclusions. The approximation is derived using first few terms of power series expansion of the solution of the equivalent eigenstrain problem with a homogeneous eigenstrain approximation. Viability of the approximation at high concentration ratios is proved by comparison with a numerical solution of the homogenization problem. To this end some theoretical result of symmetry properties of the homogenization problem are given. Using these results efficient numerical computation on a reduced computational domain is presented.

A Fermentation Strategy for Anti-MUC1 C595 Diabody Expression in Recombinant Escherichia Coli

  • The development of fermentation conditions for the production of C595 diabody fragment (dbFv) in E. coli HB2151 clone has been explored. Investigations were carried out to study the effect of carbon supplements over the expression period, the comparison of C595 dbfv production in synthetic and complex media, the influence of acetic acid upon antibody production, and comparison of one-stage and two-stage processes operated at batch or fed-batch modes in bioreactor. Yeast extract supplied during expression yielded more antibody fragment than any other carbon supply. The synthetic medium presented higher specific productivity (0.066 mg dbFv $g^{-1}$ dry cell weight) when compared to the complex medium (0.044 mg dbFv $g^{-1}$ DCW). The comparison of fermentation strategies demonstrated that (1) one-stage fed-batch fermentation performed higher C595 dbFv production than that operated in batch mode which was significantly affected by acetate concentration; (2) a two-stage batch operation could enhance C595 dbFv production. It was found that a concentration of 12.3 mg $L^{-1}$ broth of C595 dbFv and a cell concentration of 10.8g $L^{-1}$ broth were achieved at the end of two-stage operation in 5-L fermentation.
