As a long-term goal for the development of new prescriptions for hyperlipidemia, Cheungsimdodamtang(CSDDT) was examined in the present study using a rat model in which the hyperlipidemia was induced. The major parameters related to lipid metabolism were investigated and the key findings are summarized below. 1. CSDDT-treated animal group showed only a 4.5% body weight increase compared to normal animal group whereas hyper- lipidemia-induced control group increased body weight by 13.6% compared to normal animals. 2. CSDDT-induced animal group showed a significant decrease in cholesterol levels beginning one week after CSDDT treatment, compared to hyperlipidemia-induced control group. 3. Total cholesterol levels in CSDDT-treated animal group were significantly decreased compared to the hyperlipidemia-induced control group. 4. Glucose levels in CSDDT-treated animal group were significantly decreased compared to the hyperlipidemia-induced control group. 5. Triglyceride levels in CSDDT-treated animal group were significantly decreased compared to the hyperlipidemia-induced control group. 6. SGOT and SGPT levels in CSDDT-treated animal group were significantly decreased compared to the hyperlipidemia-induced control group. 7. HDL-cholesterol levels were significantly increased in CSDDT -treated animal group compared to the hypeiIipidemia-induced control group. 8. LDL-cholesterol levels were significantly decreased in CSDDT-treated animal group compared to the hyperlipidemia-induced control group.
This study was conducted to examine the effects of dietary xylooligosaccharides on lipoprotein lipase (LPL) activitiy in epididymal adipose tissue and lipid composition in serum of rats fed normal or high fat diet. Male Sprague-Dawley male rats weighing 100 $\pm$ 10 g were randomly divided into four groups, two normal diets and two high fat diets containing 1% cholesterol and 10% lard. Two normal diets were classified into a basal diet (normal group) and that with 10% xylooligosaccharide diet (NX group). The high fat diet groups were classified into a HF group without xylooligosaccharides diet and HFX group supplemented with 10% xylooligosacchride diet. Experimental diets were fed ad libitum to the rats for 4 weeks and then they were sacrificed. Body weight, epididymal weight and abdominal weight in HF group were hevier than the those of normal group, but HFX group was significantly reduced compared to HF group. Relative body weight to epididymal weight and relative body weight to abdominal weight in HF group were increased to 50%, 51%, respectively, compared to normal group, but HFX group was reduced 22%, 16%, respectively, compared to HF group. The levels of serum triglyceride, total cholesterol, LDL-cholesterol and atherogenic index in HFX groups were significantly lower than those of HF group, whereas HDL-cholesterol levels were significant increased. Triglyceride contents of epididymal adipose tissue in HF group was increased to 39%, compared to normal group, but HFX group was reduced to 15.8%, compared to HF group. Cholesterol contents of epididymal adipose tissue in HF group was increased 121%, compared to normal group, but HFX group was reduced to 26%, compared to HF group. The activity of LPL in epididymal adipose tissue was increased to 259% in HF group, compared to normal group and HFX group was reduced to 66%, compared to HF group. These result of this study suggested that improved lipid metabolism observed in rats fed xylooligosaccharides may be caused by an alteration of LPL activity in epididymal adipose tissue and lipid composition in serum.
This study was conducted to investigate whether dietary factors, normal fat and genistein leads to beneficial improvement of lipid metabolism and oxidative stress in adult hyperlipidemic male rats. Seven wk-old male SD rats were fed high fat diet (15% fat, 1% cholesterol) for 4 wks for induction of hyperlipidemic model rat. Weight-matched rats were then assigned to four groups according to dietary fat level (7% or 15% fat) and genistein contents (0 or 320 mg/kg diet). Food intake was significantly decreased by both high fat intake and genistein supplementation compared with normal fat intake and genistein no supplementaion. But weight gain was significantly decreased by genistein supplementation in normal fat intake compared with the other groups. Total lipid, total cholesterol and triglyceride in serum and liver were significantly decreased by normal fat intake compared with high fat intake. But total cholesterol in liver was significantly increased by genistein supplementation in both high fat and normal fat intake. TBARS in serum and liver was less produced by normal fat intake compared with high fat intake but TBARS in liver was significantly increased by genistein supplementation compared with genistein no supplementation in normal fat intake. Glutathione reductase activity in erythrocytes was significantly reduced by genistein supplementation in normal fat intake compared with the other groups. Glutathione peroxidase and glutathione reductase activities in liver were significantly inhibited by normal fat intake compared with high fat intake. Catalase activity in liver was significantly increased by genistein supplementation compared with genistein no supplementation in high fat intake. Nitrite was significantly decreased by normal fat intake compared with high fat intake. These results suggest that normal fat intake has the treatment effect against risk factors related with cardiovascular disease by reducing lipid profiles, lipid peroxidation. And genistein shows action as a antioxidant replacing antioxidant enzymes but also may act as prooxidant causing the production of TBARS.
Background: The vasoconstrictive effect of epinephrine in local anesthetics affects the heart, which leads to hesitation among dentists in injecting local anesthetics into patients with cardiovascular disease. Due to its vasoconstrictive effects, the present study investigated the effects of vasopressin administration on cardiac function in rats. Methods: Experiment 1 aimed to determine the vasopressin concentration that could affect cardiac function. An arterial catheter was inserted into the male Wistar rats. Next, 0.03, 0.3, and 3.0 U/mL arginine vasopressin (AVP) (0.03V, 0.3V, and 3.0V) was injected into the tongue, and the blood pressure was measured. The control group received normal saline only. In Experiment 2, following anesthesia infiltration, a pressure-volume catheter was placed in the left ventricle. Baseline values of end-systolic elastance, end-diastolic volume, end-systolic pressure, stroke work, stroke volume, and end-systolic elastance were recorded. Next, normal saline and 3.0V AVP were injected into the tongue to measure their effect on hemodynamic and cardiac function. Results: After 3.0V administration, systolic blood pressures at 10 and 15 min were higher than those of the control group; they increased at 10 min compared with those at baseline. The diastolic blood pressures at 5-15 min were higher than those of the control group; they increased at 5 and 10 min compared with those at baseline. The preload decreased at 5 and 10 min compared to that at baseline. However, the afterload increased from 5 to 15 min compared with that of the control group; it increased at 10 min compared with that at baseline. Stroke volume decreased at 10 and 15 min compared with that of the control group; it decreased from 5 to 15 min compared with that at baseline. Stroke work decreased from 5 to 15 min compared with that of the control group; it decreased from 5 to 15 min compared with that at baseline. Conclusion: Our results showed that 3.0 U/mL concentration of vasopressin resulted in increased blood pressure, decreased stroke volume and stoke work, decreased preload and increased afterload, without any effect on myocardial contractility.
The purpose of this study was to investigate the effects of group music therapy on the emotional stabilisation and Self-Efficacy of juvenile delinquents. The subjects, who were from age 15 to 19 as juvenile delinquents, received juvenile protection Order one (1) and three (3) or Order (4) and were placed in custody designated by the Gwang Ju Family Court. The 24 subjects were divided into the experimental group (N=12), and the control group (N=12) at random. The experimental group was given 14 group music therapy session for 90 minutes each, two times a week, and the control group was not given any. Emotional stabilisation and the Self-Efficacy scores for each adolescent was compared before and after the experiment. Statistical methods used for data analysis were mean, the standard deviation, and the ANCOVA. Results are as follows. The experimental group was significantly increased in emotional stabilisation compared with the control group. The experimental group was significantly decreased in aggressive behavior compared and in stress compared with the control group. The experimental group was significantly increased in self-efficacy compared, in self-confidence compared and in self control efficacy compared with the control group. These results suggest that group music therapy are positive effects on improving the emotional stabilisation and the Self-Efficacy of juvenile delinquents.
In order to study the effects of Jamyong-oil(蠶?油) on the diabetic rats induced by streptozotocin, during 15days rats were administered Jamyong-oil after streptozotocin injection(50mg/kg). On 2nd, 9th and 15th day, I investigated the levels of serum glucose, serum total cholesterol, serum triglyceride, serum amylase, and body weight. The last day, I killed rats and investigated hepatic lipid peroxide, tissue amylase. The results were obtained as follows: 1. Serum glucose levels of the sample group decreased effectively (p < 0.05) at the 15th day as compared with the control group. 2. Serum total cholesterol levels of the sample group decreased effectively (p < 0.05) at the 9th and 15th day as compared with the control group. 3. Serum triglyceride levels of the sample group decreased effectively (p < 0.05) at the 15th day as compared with the control group. 4. Activities of the serum amylase of the sample group increased respectively at the 9th (p < 0.05) and 15th (p < 0.01) day as compared with the control group. 5. Body weights of the sample group increased effectively (p < 0.05) at the 15th day as compared with the control group. 6. Hepatic lipid peroxide levels of the sample group decreased significantly (p < 0.01) as compared with the control group. 7. Tissue amylase levels of the sample group increased but not effective as compared with the control group.
In order to investigate the effects according to the concentration(o, 30, 60 and $120{\mu}g/350{\sim}400g$: saline group, I-group, II-group and III-group) of Labiatae Herba Prunellae Acua-acupuncture on the bilateral loci Taechung(LR3) in hyperthyroidism rats induced by sodium triiodothyronine injection, triiodothyronine(T3), thyroxine(T4), thyroid- stimulating hormone(TSH) and total cholesterol were observed. The results were summarized as follows; 1. The increases of the serum T3 were inhibited significantly by II-group and III-group compared with the control group. 2. The decreases of the serum T4 were inhibited significantly by I-group, II-group and III-group compared with the control group. The I-group and II-group compared with the saline group is significantly different. 3. The decreases of the serum TSH were inhibited significantly by II-group and III-group compared with the control group. The II-group compared with the saline group is significantly different. 4. The decreases of the serum total cholesterol were inhibited significantly by I-group, II-group and III-group compared with the control group. The II-group and III-group compared with the saline group is significantly different.
The effect of treatment with Citrus junos fractions (citron 3W, citron 3H, citron 4W and citron 4H) upon rat hepatocytes exposed to alcohol was investigated. We compared the serum biochemistry of rats administered both alcohol and Citrus junos fractions to control rats treated with alcohol alone. The effects of Alanine amino transferase (ALT) were significantly lower in the citron 3H extract group compared with the negative control group (p<0.05) and other experimental groups were not significantly low but a little low compared to negative control group. The levels of triglyceride (TG) were significantly low in all experimental groups compared with negative control group. Especially triglyceride level of citron 3H was lowest near to normal control group. The concentration of total cholesterol was significantly high in negative groups compared with normal control group but in all experimental groups, the concentration of total cholesterol was similar to that of negative control group. Total cholesterol of the citron 4W group was somewhat low compared with negative control group. In contrast, activities of alcohol dehydrogenase (ADH) were significantly higher in all experimental groups compared with the negative control (p<0.05) group. These data suggest that Citrus junos fractions may represents an excellent candidate for protection of rat hepatocytes from alcohol-mediated damage.
The purpose of this study was to investigate the effects of green tea catechin on mixed function oxidase system (MFO), lipofuscin contents, carbonyl value, oxidative damage and the antioxidative defense system in lung of microwave exposed rats. Experimental groups were divided to normal group and microwave exposed group. The microwave exposed groups were subdivided into three groups: catechin free diet (MW-0C) group, 0.25% catechin (MW-0.25C) group and 0.5 % catechin (MW-0.5C) group according to the levels of dietary catechin supplementation. The rats were irradiated with microwave at frequency of 2.45 GHz for 15 min. Experimental animals were sacrificed at 6th day after microwave irradiation. The contents of cytochrome P$_{450}$ contents in MW-0C group was increased to 95% , compared with normal group. MW-0.25C and MW-0.5C groups were reduced to 16% and 31%, respectively, compared with MW-0C group. The activity of NADPH-cytochrome P$_{450}$ reductase in MW-0C group was increased to 44%, compared with normal group. MW-0.25C and MW-0.5C groups were reduced to 12% and 17%, respectively, compared with MW-0C group. The activity of superoxide dismutase (SOD) in MW-0C group was decreased to 21 %, compared with normal group. MW-0.25C and MW-0.5C group were significantly (p < 0.05) increased, compared with MW-0C group. The activity of glutathione peroxidase (GSHpx) in MW-0C group was significantly decreased, compared with normal group. MW-0.25C and MW-0.5C groups were recovered to the level of normal group. The thiobarbituric acid reactive substances (TBARS) content in MW-0C group was increased to 34 %, compared with normal group. Catechin supplementation groups were maintained the level of normal group. The levels of caybonyl value in MW-0C group was increased to 21 %, compared with normal group. MW-0.25C and MW-0.5C groups were reduced to 14% and 12%, respectively, compared with MW-0C group. The lipofuscin contents in MW-0C group were increased to 23.4 %, compared with normal group. That of MW-0.5C group was significantly reduced, compared with MW-0C group. In conclusion, MFO system was activated and the formation of oxidized protein, lipofuscin was increased and antioxidative defense system was weakened of lung tissue in microwave exposed rats, thus oxidative damage was increased. But it was rapidly recovered to normal level by green tea catechin supplementation.n.
The purpose of this study was to investigate the effects of glucuyonic acid (isolated from xylan) on the antioxidative defense systems of red gastrocnemius in rats after aerobic exercise. The glucuronic acid was isolated from xylan. Male Sprague-Dawley vats weighing 150$\pm$10 g were randomly assigned to one normal group and three exercise training groups. The exercise training groups were classified as T (glucuronic acid-free diet), TU (250mg glucuronic acid/kg bw) and 2TU (500mg glucuronic acid /kg bw) according to the level of glucuvonic acid supplementation. The rats in the normal group were confined to a cage for 4 weeks. The rats in the exercise training groups ran on a treadmill for 30 min/day, 5 days/week at a speed of 28 m/min (7% incline) for 4 weeks. Glutamate oxaloacetate transaminase (GOT) activity in the exercise training groups increased significantly compared with that of the normal group. That of the TU and 2TU groups decreased significantly compared with that of the T group. Xanthine oxidase (XOD) activity in the T group increased significantly to 74% compared with that of the normal group. That of the 2TU group decreased to 42% compared with that of the T group, thus recovering to a normal level. Superoxide dismutase (SOD) activity in the T group decreased to 32% compared with that of the normal group. That of the TU and 2TU groups increased to 28% and 34%, respectively, compared with that of the T group. Glutathione peroxidase (GSHpx) activity in the T group decreased to 16% compared with that of the normal group, but that of the TU group increased to 17% compared with that of the T group. Glutathiones transferase (GST) activity in the T group decreased to 11% compared with that of the normal group, but that of the TU and 2TU groups Increased to 28% and 31%, respectively, compared with that of the T group. The contents of thiobarbituric acid reactive substances (TBARS) in the T group increased to 81% compared with that of the normal group, but the glucuronic supplementation group recovered to the normal level. In conclusio, the effects of glucuronic acid on red gastrocnemius in rats engaged in exercise training would appear to be to reduced lipid peroxidation of tissue as an antioxidative defense mechanism.
