시판 초석잠 기원식물의 기억력개선효과 비교연구 (A Comparative Study of Memory Improving Effects of Stachys Rhizome and Lycopi Rhizome on Scopolamine-induced Amensia in mice)

  • Objectives : The purpose of this study was to characterize the effect of the Ethanolic extract of Stachys sieboldii and Lycopus lucidus on the learning and memory impairments induced by scopolamine. Methods : The genetic difference of Stachys sieboldii and Lycopus lucidus were observed with RAPD analysis. The cognition-enhancing effect of Stachys sieboldii and Lycopus lucidus was investigated using a passive avoidance test, Y-maze test and the Morris water maze test in mice. Drug-induced amnesia was induced by treating animals with scopolamine (1 mg/kg, i.p.). Results : As a result of RAPD analysis, Stachys sieboldii and Lycopus lucidus Radix was found to be genetically different and The results of learning memory analysis showed that Stachys sieboldii extract-treated group (500 mg/kg, p.o.) and the tacrine-treated group (10 mg/kg, p.o.) significantly ameliorated scopolamine-induced amnesia based on the Passive avoidance Y-maze test and Water maze test. And these results are same manner in DPPH radical scavenger effect and Acetylcholineseterase inhibition effect. These results suggest that Stachys sieboldii extract maybe a useful cognitive impairment treatment, and its beneficial effects are depending on the origin plants. Conclusions : Commercially available Stachys sieboldii Radix consists of two original plant, one of them people misuse. To clarify the origin of the plant Memory tests were performed. These results suggest that 80% Ethanol extract of Stachys sieboldii showed significant anti-amnestic and cognitive-enhancing activities related to the memory processes, and these activities were parallel to treatment duration and dependent of the learning models.

Comparative Study of White, Red, and Black Ginseng Extract on Improves the Learning and Memory Impairments by Increases of Synaptic Protein Expression in Scopolamine-induced Dementia Rats

  • To compare and analyze the improvement effects of white ginseng extract, red ginseng extract, and black ginseng extract on cognitive dysfunction and memory impairment caused by scopolamine in rats. In the cognitive behavioral test, the tendency of the SCOP+B group to overcome the escape time delay induced by scopolamine administration was observed, unlike the SCOP group. The frequency on plat form was significantly increased in the group treated with ginseng extracts compared to the SCOP group. As a result of measuring the duration time on goal quadrant, the time spent in the quadrant was significantly increased in the SCOP+B group compared to the SCOP group. In the hippocampus, the SCOP-treated group significantly decreased the activity of AChE compared to the normal group, but the ginseng extract-treated groups significantly increased it compared to the SCOP group. After sacrificing the rats after the behavioral test, the expression of PSD95 protein in the excised brain was significantly decreased in the SCOP group compared to normal, but it was observed that the SCOP+R and SCOP+B groups were significantly increased compared to the SCOP group. CREB1 protein expression was significantly increased in the SCOP+R group, and the expression of Cdk5 was significantly increased in the SCOP+B group. Ginseng extracts significantly restored the memory damaged by scopolamine suggesting that red ginseng increased the expression of CREB1 and PSD95 proteins, and black ginseng increased the protein expression of Cdk5 and PSD95 to induce memory recovery.

미토콘드리아 활성화를 통한 양파(Allium Cepa L.) 과육 및 과피의 Amyloid-β 유도성 인지손상에 대한 개선효과 (Ameliorative effect of onion (Allium Cepa L.) flesh and peel on amyloid-β-induced cognitive dysfunction via mitochondrial activation)

  • 본 연구에서는 양파(Allium Cepa L.) 과육과 과피를 이용하여 in vitro 신경세포 보호효과 및 Aβ로 유도된 인지기능 장애 마우스 모델에서의 개선효과를 검증하고자 하였다. 양파 과육 분획물(EOF)과 과피 분획물(EOP) 모두 PC-12 세포에서 Aβ로 유도된 세포 독성에 대하여 신경세포 보호효과(세포 내 산화적 스트레스 억제, 세포 생존율 및 세포막 보호효과)를 나타냈다. Aβ로 유도된 인지장애 마우스 모델에서의 행동실험(Y-미로, 수동회피 및 Morris 수중 미로 시험) 결과 또한 양파 과육 분획물(EOF)과 과피 분획물(EOP) 모두 효과적인으로 학습 및 기억능력을 개선시키는 것으로 나타났다. 행동실험 후 마우스 뇌조직에서의 산화적 스트레스에 대한 생체 방어 기작의 일종인 SOD 함량의 증가, oxidized GSH/총 GSH 및 MDA 함량 감소를 나타냄에 따라 산화적 스트레스에 대한 우수한 항산화효과가 긍정적인 영향을 미친 것으로 판단된다. 또한, 뇌조직으로부터 분리한 미토콘드리아에 대하여 막 전위(MMP) 보호 및 ATP 함량 증가를 나타냈으며, 미토콘드리아와 관련된 apoptosis 경로에서 BAX의 감소 및 cytochrome c 방출 억제를 통해 caspase 3/7의 활성을 억제하는 것으로도 나타났다. 결국, 양파 과육 분획물(EOF)과 과피 분획물(EOP)은 AChE의 활성 억제 및 ACh의 함량을 증가시킴으로써 효과적인 콜린성 시스템 보호효과를 나타냄에 따라, Aβ로 유도된 인지기능 장애를 예방할 수 있는 고부가가치 건강기능식품 소재로의 활용 가능성이 기대된다.

난청 고령자의 우울정도, 인지기능, 의사소통능력 및 정량뇌파 분석 연구 (A Study on Analysis of Depression, Cognition, Communication, and Quantitative Electroencephalogram in Hearing Impaired Elderly)

  • 본 연구는 난청 고령자의 우울정도, 인지기능, 의사소통능력 및 정량뇌파를 분석하고, 관련성을 규명할 목적으로 시도하였다. Y 시 소재의 S 청능재활 센터를 내원한 60세 이상의 난청 고령자 중 모집공고문을 통해 남성 37명, 여성 26명이 2020년 6월 20일부터 2020년 9월 3일까지 자발적으로 참여했다. 구조화된 설문지를 통해 대상자의 전반적인 특성, 우울정도, 인지기능을 평가하였다. 의사소통능력의 척도인 단어인지도는 한국표준 단음절어를 사용하여 진단용 청력검사기로 평가하였다. 정량뇌파는 전전두엽 Fp1과 Fp2에 건식 전극을 사용하여 2채널 뇌파 측정기로 측정하였다. 수집된 자료는 SPSS/WIN 25.0 프로그램으로 인구 사회학적 특성 파악을 위한 빈도분석, 변수간 Pearson's correlation분석, 3분할 집단 간 One-way ANOVA분석을 하였다. 결과를 요약하면 다음과 같다. 의사소통능력은 전전두엽 좌우 대칭성(**p<.01)과 정(+)적 상관관계를 보였고, 우뇌 정신적 산만 및 스트레스(*p<.05)와 부(-)적 상관관계를 보였다. 그러나 우울정도와 인지기능과는 유의한 상관관계는 없었다. 의사소통능력에 따른 3분할 집단별 차이 검정에서도 전전두엽 좌우 대칭성(**p<.01)이 우울정도와 인지능력보다 높은 수준의 상관관계를 보였다. 이상의 결과는 정량뇌파에서 측정된 전전두엽 좌우 대칭성이 난청 고령자의 의사소통능력의 강력한 생체적 지표 가능성을 시사 하였다.

농촌지역 독거노인 15인의 경도인지장애 및 치매의 한의학적 임상치료 효과 - 심(心)을 위주로 한 한의학적 치매 치료의 효용 - (Korean Medicine Clinical Effects On Light Cognitive Impairments And Dementia Of 15 Aged Men Living Alone In The Farm Village)

  • Objective : This study was designed in order to support the standpoint that dementia will be treated better by remedy for heart rather than for brain, liver and kidney in Korean Medicine. Methods : We investigated which of five viscera make dementia from original texts about it. Then, 15 aged men living alone in Yeongdong-gun, Chungcheongbuk-do were treated for dementia by herb remedies, acupuncture and moxa. After it, we verified through method, for example, SMMSE-DS, CDT, and etc. Results : We had results that the main viscera causing dementia is heart, and heart is related with gall bladder, spleen, and kidney according to philological method. Clinical testing in this viewpoint showed satisfactory effects on the diseases. Conclusion : From the results, we proved it could be a new way of remedy for dementia that main the viscera causing dementia is heart in Korean Medicine.

Phosphoproteomic Analysis of the Brain of Ovariectomized Adult Rat

  • Aging in females is associated with a reduced metabolic function, increased incidence of neurodegenerative diseases, and cognitive dysfunction, as a result of loss in gonadal function. The change can alter the states of phosphorylation on the proteins, which cause dramatic changes in the cellular location or activity of the proteins. In this study, the differential phosphorylation of the proteins responsible for the functions related to cognition was studied using the ovariectomized adult rats. Phosphoproteomic analysis using the cerebral and hippocampal tissues could identify 51 differentially phosphorylated proteins including 12 proteins for energy metabolism, 8 cytoskeletal proteins, 6 signaling proteins, and other functional proteins in the ovariectomized rats. Further, anti-oxidative enzymes, superoxide dismutase and peroxiredoxin-2, which are known to be inactivated by phosphorylation, were found to be differentially phosphorylated in the cerebellum and hippocampus of the ovariectomized rats, respectively. Many of the deactivated proteins by differential phosphorylation identified in this study were overlapped to those of Alzheimer's disease cases. These results will provide information for neurodegenerative learning and memory impairments in women as brought about by menopause.

  • 3-Methylcrotonyl-CoA carboxylase (MCC) is a biotin-dependent enzyme involved in leucine metabolism. We describe a patient with MCC deficiency who manifested with Reye syndrome-like illness with status epilepticus, metabolic acidosis, hypoglycemia, hyperammonemia, elevated liver enzymes and neurologic impairments after the viral gastroenteritis and then, has suffered from Lennox-Gastaut syndrome. Urinary organic acid analysis revealed increased excretion of 3-hydroxyisovaleric acid and 3-methylcrotonylglycine. This patient was managed with leucine restriction diet and supplementation of biotin and carnitine but was not so effective. He has suffered from neurologic sequelae such as Lennox-Gastaut syndrome, motor and cognitive impairement.

포공영 기원식물의 mouse 기억력 개선효과 비교 (A Comparative Study of Memory Improving Effects of Taraxaci herba on Scopolamine-induced Amnesia in Mouse)

Methods : The cognition-enhancing effect of Taraxacum officinale and T.coreanum was investigated using a passive avoidance test, the Morris water maze test and Y-maze test in mice. Drug-induced amnesia was induced by treating animals with scopolamine (1 mg/kg, i.p.). Results : The results showed that the Aug harvested T.offiicinale extract-treated group (200 mg/kg, p.o.) and the tacrine-treated group (10 mg/kg, p.o.) significantly ameliorated scopolamine-induced amnesia based on the Passive avoidance Y-maze test and Watermaze test. And these results are same manner in DPPH radical scavenger effect and Acetylcholineseterase inhibition effect. Conclusions : These results suggest that Taraxacum officinale extract maybe a useful cognitive impairment treatment, and its beneficial effects are depending on the collecting time and origin plants. As a result, Taraxacum officinale harvested in August improve memory most.

급성 일산화탄소 중독 환자에서 발생한 양하지 말초 운동신경병증 1례 (Motor Peripheral Neuropathy Involved Bilateral Lower Extremities Following Acute Carbon Monoxide Poisoning: A Case Report)

  • Carbon monoxide (CO) intoxication is a leading cause of severe neuropsychological impairments. Peripheral nerve injury has rarely been reported. Following are brief statements describing the motor peripheral neuropathy involved bilateral lower extremities of a patient who recovered following acute carbon monoxide poisoning. After inhalation of smoke from a fire, a 60-year-old woman experienced bilateral leg weakness without edema or injury. Neurological examination showed diplegia and deep tendon areflexia in lower limbs. There was no sensory deficit in lower extremities, and no cognitive disturbances were detected. Creatine kinase was normal. Electroneuromyogram patterns were compatible with the diagnosis of bilateral axonal injury. Clinical course after normobaric oxygen and rehabilitation therapy was marked by complete recovery of neurological disorders. Peripheral neuropathy is an unusual complication of CO intoxication. Motor peripheral neuropathy involvement of bilateral lower extremities is exceptional. Various mechanisms have been implicated, including nerve compression secondary to rhabdomyolysis, nerve ischemia due to hypoxia, and direct nerve toxicity of carbon monoxide. Prognosis is commonly excellent without sequelae. Emergency physicians should understand the possible-neurologic presentations of CO intoxication and make a proper decision regarding treatment.

Visual Perception in Autism Spectrum Disorder: A Review of Neuroimaging Studies

  • Although autism spectrum disorder (ASD) is a neurodevelopmental disorder characterized by social impairments, patients with ASD frequently manifest atypical sensory behaviors. Recently, atypical sensory perception in ASD has received much attention, yet little is known about its cause or neurobiology. Herein, we review the findings from neuroimaging studies related to visual perception in ASD. Specifically, we examined the neural underpinnings of visual detection, motion perception, and face processing in ASD. Results from neuroimaging studies indicate that atypical visual perception in ASD may be influenced by attention or higher order cognitive mechanisms, and atypical face perception may be affected by disrupted social brain network. However, there is considerable evidence for atypical early visual processing in ASD. It is likely that visual perceptual abnormalities are independent of deficits of social functions or cognition. Importantly, atypical visual perception in ASD may enhance difficulties in dealing with complex and subtle social stimuli, or improve outstanding abilities in certain fields in individuals with Savant syndrome. Thus, future research is required to elucidate the characteristics and neurobiology of autistic visual perception to effectively apply these findings in the interventions of ASD.
