  • We developed a on-line gas control system for three gas components and investigated the effect of oxygen, carbon dioxide, and ethylene on polysaccharide production and cell growth in the suspension cultures of Aloe saponaria. Carbon dioxide(5% v/v) enhanced significantly polysaccharide production while the cell growth was not affected.

Effects of Protease-resistant Antimicrobial Substances Produced by Lactic Acid Bacteria on Rumen Methanogenesis

  • Effects of protease-resistant antimicrobial substances (PRA) produced by Lactobacillus plantarum and Leuconostoc citreum on rumen methanogenesis were examined using the in vitro continuous methane quantification system. Four different strains of lactic acid bacteria, i) Lactococcus lactis ATCC19435 (Control, non-antibacterial substances), ii) Lactococcus lactis NCIMB702054 (Nisin-Z), iii) Lactobacillus plantarum TUA1490L (PRA-1), and iv) Leuconostoc citreum JCM9698 (PRA-2) were individually cultured in GYEKP medium. An 80 ml aliquot of each supernatant was inoculated into phosphate-buffered rumen fluid. PRA-1 remarkably decreased cumulative methane production, though propionate, butyrate and ammonia N decreased. For PRA-2, there were no effects on $CH_4$ and $CO_2$ production and fermentation characteristics in mixed rumen cultures. The results suggested that PRA-1 reduced the number of methanogens or inhibited utilization of hydrogen in rumen fermentation.

Effect of Ethylene on Formations of Adventitious Roots, Trichomes, and Calli by NAA in Leaf segments of Arabidopsis thaliana (애기장대 잎 절편 배양시 NAA 농도에 따른 부정근, 모용 및 캘러스 형성에 미치는 에틸렌의 영향)

  • In order to study the role of ethylene on the formation of adventitious roots, trichomes and calli, the effects of 1-aminocyclopropane-l-carboxylic acid (ACC), ethephon, $CoCl_2$ and $AgNO_3$ were investigated in the leaf segments from ecotype Columbia of Arabidopsis thaliana. When the leaf segments were cultured on the media for forming adventitious roots (0.1 mg/L NAA), trichomes (2.0 mg/L NAA) and calli (10.0 mg/L NAA), and then each cultures was treated with 1-100 mg/L of ACC and ethephon, respectively. On the adventitious root-forming medium adventitious root formation was decreased, and trichomes were induced. And on the trichome-forming medium trichome formation was decreased, and calli were induced. In order hand each culture was treated with 1-100 mg/L of $CoCl_2$ and $AgNO_3$, respectively. On the adventitious root-forming medium adventitious roots was increased without trichome formation, and on the trichome-forming medium trichome formation was decreased, and adventitious roots were induced. However on the callus-forming medium treated with ACC, ethephon, $CoCl_2$ and $AgNO_3$, respectively, callus formation was inhibited and trichomes were induced in all cultures.

Anti-cancer Activity of Supernatant of Rahnella aquatilis AY 2000 Cocultured with Streptomyces griseus (Rahnella aquatilis AY 2000과 Streptomyces griseus의 공배양 상등액의 항암활성)

  • In order to produce a new antibiotic material against Jurkat T cells using horizontal gene transfer among microbes, co-cultures between soil bacteria AY2000 and the multiple antibiotic producer S. griseus was carried out. It showed that the highest active substance against Jurkat T cells was produced at 48 hr of co-culture time with MTT assay. Moreover, a morphological change of nuclear of Jurkat T cells treated with co-cultured substance was observed in DAPI staining. This result suggests that a new material was produced with co-culture supernatant, and that co-culture between microboes can develop new antibiotic materials.

  • Effects of ethylene inhibitors like silver nitrate $(AgNO_3)$, cobalt chloride $(CoCl_2)$ and Salicylic acid (SA) on multiple shoot induction and their impact on ethylene production using embryonal cotyledon cultures of Cucumis sativus L. were examined. The optimum concentration of $AgNO_3\;(40{\mu}M),\;CoCl_2\;(20{\mu}M)\;and\;SA\;(20{\mu}M)$, separately, induced maximum number of shoots on Murashige and Skoog's (MS) medium supplemented optimally with $4.44{\mu}M$ BA and $0.25{\mu}M$ NAA. Among the three ethylene inhibitors tested, $AgNO_3$ produced maximum number of shoots when compared to $CoCl_2$ and SA Ethylene production was monitored in all the treatments with $AgNO_3/CoCl_2/SA$ and it was observed that the treatment with $AgNO_3$ alone showed increase in ethylene production when compared to $CoCl_2$ and SA Even though ethylene concentration was the highest in $AgNO_3$ treated explants, maximum number of shoots was obtained.

Isolation of bovine Coronavirus from calves with diarrhea (소 Coronavirus 분리에 관한 연구)

  • Fecal samples were collected from 47 calves with diarrhea and 12 clinically normal co-h-abitants, and tested for virus using MDBK cell cultures. Three cytopathic viruses were isolated from 8 fecal samples obtained from diarrheic calves. The isolated viruses were neutralized by bovine coronavirus hyperimmune serum In plaque reduction assay and were detected in the cytoplasm of MDBK cell by bovine coronavirus hyperimmune serum using immunofluorescence staining. The viruses agglutinated mouse erythrocytes only among the various animal erythrocytes tested and new isolates were identified as bovine coronavirus.

Effect of a co-culture of scenedesmus dimorphus and nitrifiers on advanced wastewater treatment capacity (Scenedesmus dimorphus와 질산화 박테리아의 공배양이 하수고도처리능에 미치는 영향)

  • This study investigated the effect of a co-culture of Scenedesmus dimorphus and nitrifiers using artificial wastewater on the removal of ammonium, nitrate and phosphate in the advanced treatment. To test the synergistic effect of the co-culture, we compared the co-culture treatment with the cultures using S. dimorphus-only and nitrifiers-only treatment as controls. After 6 days of incubation, nitrate was removed only in the co-culture treatment and total amount of N removal was 1.3 times and 1.6 times higher in the co-culture treatment compared to those in the S. dimorphus- and nitrifiers-only treatments, respectively. In case of total amount of P, co-culture treatment removed 1.2 times and 12 times more P than the S. dimorphus -and nitrifiers-only conditions, respectively. This indicates that the co-culture improved removal rates for ammonium, nitrate, and phosphate. This further implies that there was no need for denitrification of nitrate and luxury uptake of P processes because nitrate and phosphate can be removed from the uptake by S. dimorphus. In addition, co-culture condition maintained high DO above 7 mg/L without artificial aeration, which is enough for nitrification, implying that co-culture has a potential to decrease or remove aeration cost in the wastewater treatment plants.

Effect of Chungpaesagan-tang on Ischemic Damage in Organotypic Hippocampal Slice Culture (청폐사간탕(淸肺瀉肝湯)이 뇌해마 조직배양의 신경세포 자연사에 미치는 영향)

  • Chungpaesagan-tang which is used for treating patients of brain in cerebrovascular disease frequently from clinical doctor has not reported about the effect of neuronal aptosis caused of brain ischemia. The aim of this study is to investigate effect of Chungpaesagan-tang protecting neuronal cells from being damaged by brain ischemia through using organotypic hippocampal slice cultures. We caused ischemic damage to organotypic hippocampal slice cultures by oxygen and glucose deprivation. And added Chungpaesagan-tang extract to cultures. thereafter we measured area percentage of propidium iodide (PI)-stained neuronal cell, lactate dehydrogenase (LDH) levels in culture media and Terminal deoxynucleotidyl transferase-mediated dUTP nick-end labeling (TUNEL)-positive cells. Area percentage of PI-stained neuronal cells and count of TUNEL-positive cells in CA1 and DG area of organotypic hippocampal slice culture were significantly decreased in pertinent density level of Chungpaesagan-tang extract. LDH levels in culture media of organotypic hippocampal slice culture were significantly decreased in pertinent density level of Chungpaesagan-tang extract. Within pertinent density level, Chungpaesagan-tang has cell protection effect that prevents brain ischemia damaging neuronal cells and apoptosis increasing.

Development of a Bottle-Free Multipurpose Incubator for Generating Various Bacterial Culture Conditions

  • The purpose of this study was to develop a multipurpose incubator, without the gas cylinders (bottles) which are required for $H_2$ and $CO_2$ supplementation. In our bottle-free multipurpose incubator, the $H_2$ and $CO_2$ were generated by chemical reactions induced within the chamber. The reaction between sodium borohydride and acetic acid at a molar ratio of 1:1 was used to generate $H_2$, according to the following formula: $4NaBH_4+2CH_3COOH+7H_2O{\rightarrow} 2CH_3COONa+Na_2B_4O_7+16H_2$, whereas the other reaction, citric acid and sodium bicarbonate at a 1:1 molar ratio, was used to generate $CO_2$, according to the following formula: $C_6H_8O_7+3NaHCO_3{\rightarrow}Na_3(C_6H_5O_7)+3H_2O+3CO_2$. Five species of obligate anaerobic bacteria, one strain of capnophilic bacterium, and one strain of microaerophilic bacterium were successfully cultured in the presence of their respective suitable conditions, all of which were successfully generated by our bottle-free multipurpose incubator. We conclude that, due to its greater safety, versatility, and significantly lower operating costs, this bottle-free multipurpose incubator can be used for the production of fastidious bacterial cultures, and constitutes a favorable step above existing anaerobic incubators.
