Characterization of artificially induced zinc-tolerant yeast mutants (아연 저항성 갖는 인위적으로 유도된 효모 돌연변이체의 특성)

  • Bioremediation is a technique using microorganisms to clean up contaminated pollutants including heavy metals. It is well known that yeasts have a high capacity to remove a wide range of metals by biosorption. Therefore, this study was focused on to obtain yeast mutant that has strong tolerance to zinc (Zn), one of representative heavy metals. The Zn resistant yeast mutant (ZnR) was induced and isolated by growing yeast cells in media containing 1 mM $ZnCl_2$ and gradually increasing the concentration until 80 mM $ZnCl_2$, in which cells were adapted and survived. The induced ZnR cells showed strong tolerance to Zn stress compared with control cells. Moreover, the ZnR cells showed increased tolerance to cadmium and nickel stress but decreased tolerance to copper stress. The increased tolerance of ZnR cells to Zn stress was due to mutation of genes. This study can be useful in bioremediation of heavy metals as the metal tolerant microorganism was artificially induced in short time.

Distribution of trace metals in the deep ocean waters of the East Sea (동해심층수 개발해역의 미량금속 분포)

  • In order to develop the deep ocean water, we performed to study the characteristics of vertical distribution of dissolved trace metals(Cd, Co, Cu, Ni, Pb, Zn) from Apr. to Oct., 2005 in the East Sea. Total six sampling sites were selected in Gangwon-Do and Gyeongsanbuk-Do. Accuracy of the analytical procedures was assessed by the SRM(CASS-4) for dissolved metals in seawater. The mean recoveries of CASS-4 ranged from 89.4% for Co to 99.8% for Cd. In this study, the dissolved metal concentrations varied with space, time and element. The metal concentrations showed wide range in the surface. Cd, Ni and Zn showed a nutrient-type profile with surface depletion and enrichment at depth. However, Co, Cu and Pb were irregular in the vertical distribution. All metal concentrations studied in this study are lower than the criteria of Korean drinking water.

Viable Bacterial Cell Patterning Using a Pulsed Jet Electrospray System

  • In the present study, drop-on-demand two-dimensional patterning of unstained and stained bacterial cells on untreated clean wafers was newly conducted using an electrospray pulsed jet. We produced various spotted patterns of the cells on a silicon wafer by varying the experimental conditions, such as the frequency, flow rate, and translational speed of the electrospray system in a two-dimensional manner. Specifically, the electrospray's pulsed jet of cell solutions produced alphabetical patterns consisting of spots with a diameter of approximately $10{\mu}m$, each of which contained a single or a small number of viable bacteria. We tested the viability of the patterned cells using two visualization methods. This pattering technique is newly tested here and it has the potential to be applied in a variety of cell biology experiments.

Particle Size Distribution Analysis of Mineral Dust in Polar Snow Using a Coulter Counter (쿨터카운터(Coulter Counter)를 이용한 극지 눈시료 중 광물성 먼지의 입자크기분포 분석)

  • Mineral dust in the polar snow plays an important role both in the climate system of the Earth and in global biogeochemical cycles. Analysis of the concentration and the particle size distribution of mineral dust has been carried out in the snow from the Antarctic surface snow and the Greenland snowpit. Among the various particle size determination techniques, a Multisizer 3 Coulter Counter in a class 100 clean bench counted all particles between 1.1 and $30.0{\mu}m$ with a $50{\mu}m$ aperture tube. The aperture tube size, the concentration of electrolytes and the accuracy of the particle size distribution were determined in this study. The number concentrations from the Antarctic surface snow were 81,843 particles $mL^{-1}$, but those from the Greenland snowpit were 10,666 particles $mL^{-1}$. In the volume distribution, the distributions of mineral dust in both the Antarctic surface snow and the Greenland snowpit showed lognormal distribution in the size interval 1.1 to $6.0{\mu}m$ with the mode, 3.562 and $3.836{\mu}m$, respectively. The analysis technique using a coulter counter for mineral dust could be used for reconstructing paleoclimates from polar ice cores.

  • The generation of small hydropower as compared to other different developed environmental methods produces one of the clean energies. In such manner, various application system development through IT technique is being developed for an advanced small hydropower energy resources data mining. However, existing data analysis of New & Renewable Information System for small hydropower resources application is incomplete therefore it limits expressing these information on the Web. Thus for positive usage of small hydropower resources, a more systematic and precise analysis system should be built. This study seeks to develop a map of the domestic small hydropower resources problems to further improve small hydropower resources, developed through Package Tool which can accurately evaluate a wide range of small hydropower basin in a short period of time. Small hydropower Package Tool was calculated using existing Analysis System small hydropower resources which did not provide diverse capabilities resulting to 840 standard basin classified by A and facility capacity, etc., and to assume a 40% annual capacity, expected annual electricity production was calculated. Small hydropower for the national water system of small hydropower resources potential calculated in terms of resources for the development of small hydropower will be utilized as basic data.

  • Nitridation technique has been receiving much attention for the formation of a thin nitrided buffer layer on which high quality nitride films can be formedl. Particularly, gallium nitride (GaN) has been considered as a promising material for blue-and ultraviolet-emitting devices. It can also be used for in situ formed and stable passivation layers for selective growth of $GaAs_2$. In this work, formation of a thin nitrided layer is investigated. Nitrogen electron cyclotron resonance(ECR)-plasma is employed for the formation of thin nitrided layer. The plasma source used in this work is a compact ECR plasma gun3 which is specifically designed to enhance control, and to provide in-situ monitoring of plasma parameters during plasma-assisted processing. Microwave power of 100-200 W was used to excite the plasma which was emitted from an orifice of 25 rnm in diameter. The substrate were positioned 15 em away from the orifice of plasma source. Prior to nitridation is performed, the surface of n-type (001)GaAs was exposed to hydrogen plasma for 20 min at $300{\;}^{\circ}C$ in order to eliminate a native oxide formed on GaAs surface. Change from ring to streak in RHEED pattern can be obtained through the irradiation of hydrogen plasma, indicating a clean surface. Nitridation was carried out for 5-40 min at $RT-600{\;}^{\circ}C$ in a ECR plasma-assisted molecular beam epitaxy system. Noise Removal Using Complex Wavelet and Bernoulli-Gaussian Model (복소수 웨이블릿과 베르누이-가우스 모델을 이용한 잡음 제거)

  • Orthogonal wavelet tansform which is generally used in image and signal processing applications has limited performance because of lack of shift invariance and low directional selectivity. To overcome these demerits complex wavelet transform has been proposed. In this paper, we present an efficient image denoising method using dual-tree complex wavelet transform and Bernoulli-Gauss prior model. In estimating hyper-parameters for Bernoulli-Gaussian model, we present two simple and non-iterative methods. We use hypothesis-testing technique in order to estimate the mixing parameter, Bernoulli random variable. Based on the estimated mixing parameter, variance for clean signal is obtained by using maximum generalized marginal likelihood (MGML) estimator. We simulate our denoising method using dual-tree complex wavelet and compare our algorithm to well blown denoising schemes. Experimental results show that the proposed method can generate good denoising results for high frequency image with low computational cost.

  • In this paper, we propose a robust speech reinforcement technique to enhance the intelligibility of the degraded speech signal under the ambient noise environments based on soft decision scheme incorporating a speech absence probability (SAP) with speech reinforcement gains. Since the ambient noise significantly decreases the intelligibility of the speech signal, the speech reinforcement approach to amplify the estimated clean speech signal from the background noise environments for improving the intelligibility and clarity of the corrupted speech signal was proposed. In order to estimate the robust reinforcement gain rather than the conventional speech reinforcement method between speech active periods and nonspeech periods or transient intervals, we propose the speech reinforcement algorithm based on soft decision applying the SAP to the estimation of speech reinforcement gains. The performances of the proposed algorithm are evaluated by the Comparison Category Rating (CCR) of the measurement for subjective determination of transmission quality in ITU-T P.800 under various ambient noise environments and show better performances compared with the conventional method.

  • The diagnostic management of patients with chest pain remains a clinical challenge. Magnetocardiography (MCG) has been proposed as a new non-invasive method for detection of myocardial ischemia. To date, however, MCG technique is not intensively introduced for clinical use. One of the main reasons might be the absence of statistically valid and diagnostically clean criteria, which can determine the presence of certain heart disease. In this work, we suggested a new method to classify the diagnostic value of MCG for the detection of coronary artery disease (CAD) in patients with chest pain. MCG was recorded for three groups (healthy subjects and patients without and with CAD) by means of the 64 channel SQUID gradiometer system installed at a hospital. Using four parameters, which were found to be significantly different between groups, we evaluated a probability, in which parameters can be classified into each group based on the distribution function of the parameter in each group. For all parameters, sum of probabilities was compared between groups to determine the presence of CAD. Our classification method shows that the MCG can be a useful tool to predict the presence of CAD with sensitivity and specificity of higher than $80\%$ each.

  • Felt, is one of the Korean traditional woolen fabrics, that is thermally excellent and durable, so it has been widely used for various living goods from the ancient time to the modern society. Felt makes various changes in colors and shapes, does not change easily even after a long-term use, and is easy to clean. In this study, the origin and history of felt were researched. Also, different kinds and shapes of felt and the currently used manufacturing technique, and how the Korean traditional pattern-applied cultural products were developed using felt were examined as well. The method of this study were as follows. First, felt manufacturing techniques used after investigating the precedent studies, books, records and literatures of museums, the existing relics of felt were examined. Second, the conditions of the traditional pattern-applied cultural products were found out through the precedent studies after examination of kinds and symbols of the Korean traditional patterns. Third, patterns that were applied to cultural products were selected and designed with new patterns. Fourth, the past techniques were conducted with the following three methods by referring to the range of cultural products manufacturing; )color felt made with one color, pattern felt, and pattern-embroidered felt. Fifth, the following products combined with decoration and practical application by applying felt techniques were used. This study was aiming at the development of practical cultural products made with the traditional wool, felt, which can receive a good response from all age groups. It is allegedly required to develop various cultural products through studies and the rediscovery of the national traditional materials. In the future, it is necessary to study in such a way that diversification and differentiation are made through approaches and attempts with various techniques.
