Characteristics of Chungkookjang that Enhance the Flavor and GABA Content in a Mixed Culture of Bacillus subtilis MC31 and Lactobacillus sakei 383 (Bacillus subtilis MC31와 Lactobacillus sakei 383의 혼합배양으로 향상된 풍미와 GABA 함량을 지닌 청국장의 품질 특성)

  • Chungkookjang has several functional properties, such as fibrinolytic activity, anticancer effects, and antioxidant effects. However, children do not like Chungkookjang because of its foul odor. A mixed culture of Bacillus subtilis MC31 and Lactobacillus sakei 383 was used to improve the production of GABA in Chungkookjang and its flavor. Most of the foul odor of Chungkookjang was removed. The slime content and viscosity of Chungkookjang fermented in the mixed culture were similar to those of commercial Chungkookjang when B. subtilis MC31 and Lactobacillus sakei 383 were inoculated in a 1:1 ratio. The maximum GABA content was obtained when Chungkookjang was fermented with B. subtilis MC31 and L. sakei 383, which was fermented at $37^{\circ}C$ for 72 hr. During the period of fermentation, the viable cell number of B. subtilis MC31 reached a peak (log 9.13 CFU/g) at six days, and L. sakei 383 reached a peak (log 6.78 CFU/g) at two days. The moisture, crude ash, crude protein, crude fat, and crude fiber contents were 61.71%, 2.05%, 17.54%, 8.36%, and 1.95%, respectively. The amino-type nitrogen content of Chungkookjang fermented by B. subtilis MC31 and L. sakei 383 was less than Chungkookjang fermented by B. subtilis MC31 alone. The ammonia-type nitrogen and reducing sugar content of the Chungkookjang fermented by B. subtilis MC31 and L. sakei 383 were higher than that of steamed soybean. The glutamic acid and GABA content detected with an amino acid analyzer were 1.40 mg/g and 0.47 mg/g, respectively. These results suggest that fermentation with B. subtilis MC31 and L. sakei 383 in a 1:1 ratio removes more of the foul odor and increases the GABA content compared with single fermentation.

Monitoring of Color Changes and Organoleptics Properties of Chungkookjang Products during Storage for Shelf-life Establishment (청국장 제품의 유통기한 설정을 위한 저장중의 색도변화 및 관능적 특성 monitoring)

  • In order to examine the changes of commercial value during storage conditions and shelf-life of Chungkookjang, response surface methodology of SAS program was applied to monitor orgenoleptic properties and color changes to determine optimum storage conditions. Lightness (L) value of vacuum-packed Chungkookjang showed under the 1% significance probability with high influence in all conditions such as sterilized temperature, storage time and storage temperature. Redness (a) value of vacuum-packed Chungkookjang was increased during highly sterilized and storage temperatures, and was individually influenced. One of the generally packed sample also showed that significance value was influenced under 10%, during high storage temperature. Yellowness (b) value of vacuum-packed Chungkookjang showed the significance probability under 1% during storage time and temperature, and under 5% during sterilized temperature. Also, stational point showed saddle type in both packaging samples. In case of vacuum-packed Chungkookjang, compatibility of odor was influenced by sterilized temperature under 10% significance probability. But generally packed samples have an effect on storage time under 1% significance probability effect on storage time, and sterilized temperature under 5%. In case of vacuum-packed Chungkookjang, compatibility of taste was highly concerned with storage time under 1% significance probability, whereas generally packed samples were concerned with storage temperature under 5% significance probability. Overall palatability of vacuum-packed samples showed maximum value of 5.15 on sterilized temperature ($61.92^{\circ}C$), storage temperature ($6.79^{\circ}C$) and storage time (56.48 days) during storage.

The Bacterial Biological Response Modifier Enriched Chungkookjang Fermentation (면역증강물질 강화 청국장 발효)

  • The aim of this study was to produce Chungkookjang-a food produced through fermentation with Bacillus licheniformis E1-that contains an increased concentration of a bacterial biological response modifier (B-BRM). Unfortunately, sensory studies have indicated that B. licheniformis E1-fermented Chungkookjang is unacceptable for commercial use. We isolated another bacterial strain from this food product: B. subtilis S2. The optimum time and temperature for Chungkookjang fermentation with B. licheniformis E1 and B. subtilis S2 were 48 hr and $40^{\circ}C$, respectively. Sensory studies showed that Chungkookjang fermented by both B. licheniformis E1 and B. subtilis S2 was more acceptable than B. licheniformis E1 only. The amino nitrogen and crude protein content of the product were 359 mg% and 45.6% respectively. Additionally, it was confirmed that the proliferation of mouse splenic lymphocytes increased significantly, when the cells were treated with the BRM from Chungkookjang fermented using the mixture of bacterial strains in vitro. These results suggest that the enriched Chungkookjang may help patients who are medically in need of potentiation of lymphocytes proliferation.

Flavor Improvement of Chungkookjang by Addition of Yucca (Yucca shidigera) Extract (유카(Yucca shidigera) 추출물 첨가에 의한 청국장의 풍미 개선에 관한 연구)

  • This study was carried out to investigate the effect of yucca extract on the flavor development in Chungkookjang fermented by Bacrillus sp. b01. The changes in the contents of amino-type N, ammonia type N, volatile compounds and organic acids, and those in the activities of ${\alpha}-amylase$ and protease were also determined with aging period. The amount of amino-type N increased gradually with time and was slightly higher in Chungkookjang containing yucca extract than in control. The content of ammonia-type N in Chungkookjang decreased by the addition of yucca extract. During aging, a little higher amylase activity was detected in Chungkookjang containing yucca extract. However, the amylase activity was the lowest in Chungkookjang containing 1 mg/g of yucca extract. The protease activity was slightly higher in Chungkookjang containing yucca extract. The organic acids, such as citrate, acetate, malate, fumarate, and succinate, were detected. The yucca extract reduced the production of tetramethylpyrazine which was responsible for the unpleasant odor, but enhanced the production of 2,5-dimethylpyrazine and 2,6-dimethylpyrazine which contributed to the taste. Sensory evaluation showed that the addition of yucca extract of 0.5 mg/g significantly improved the flavor and taste of products.

Characteristics of Chungkookjang Produced by Bacillus subtillus MC31 (B. subtilis MC 31를 이용한 청국장의 품질특성)

  • Chungkookjang was fermented by B. subtilis MC31, a ${\gamma}$-amino butyric acid (GABA) producing microorganism. The characteristics of Chungkookjang were investigated while fermenting. Twenty four amino acids were detected in Chungkookjang, leucine was the highest of them all. Total cell populations of B. subtilis MC31 phase were between log $9.52{\pm}0.5$ ~ log $9.049{\pm}0.5$ CFU/g at stationary phase. Contents of moisture, crude ash, crude protein, crude lipid and crude fiber are $61.07{\pm}0.01%$, $1.52{\pm}0.01%$, $17.66{\pm}0.04%$, $8.96{\pm}0.03%$ and 2.61%, respectively. Contents of ammonia type nitrogen, amino type nitrogen and reducing sugar were increased during fermentation at $40^{\circ}C$ for 72 hr, however those of titratable acidity and total sugar were decreased. pH was slowly alkalized during fermentation. Viscous substance and protease contents in Chungkookjang were $4.7{\pm}0.05%$ and $0.519{\pm}7.36$ g/l, apiece. When the fibrin plate and Robbin method for fibrinolytic activity were applied, B. subtilis MC31 showed high activity. These results suggested that B. subtilis MC31 is suitable to be used as a starter to enhance the quality of Chungkookjang.

Antioxidant Activity of Substances Extracted by Alcohol from Chungkookjang Powder (분말청국장에서 알코올로 추출한 물질의 항상화능)

  • It is previously reported that Bacillus licheniformis B1 strain isolated from nature was successfully used for Chungkookjang fermentation. Antioxidant activity of its powder was determined in this study. Sephadex G-75 gel filtration chromatography was performed, far soluble fractions of the powder extracted by distilled water. The soluble fractions were separated into large and small fractions. The substance 1,1-diphenyl-2-picrylhydrazyl (DPPH) was used as an electron acceptor. Antioxidant activity was found in the small fractions. Five% solution of the Chungkookjang powder was the most effective in the extraction of antioxidant substances from the powder. It was proven in this study that strong antioxidant activity still remained in the Chungkookjang powder.

Chungkookjang Fermentation of Mixture of Barley, Wormwood, Sea Tangle, and Soybean (보리, 쑥, 다시마, 대두 혼합물의 청국장 발효)

  • Barley, wormwood, sea tangle, and soybean were fermented by Bacillus licheniformis B 1 to make Chungkookjang with better flavor and aroma. Maximal protease activity in mixed grains was observed one day after inoculation. pH increased to 8.4 two days after inoculation. Browning material derived from interaction between sugar and amino acids increased 20-fold. Thus, it is proved that Chungkookjang can be made in the mixed grains. Antioxidant activities of mixed fermented grains dissolved in ethanol or methanol (0.2 and 1 %) increased depending on their concentrations. Antioxidant activities were determined using 1,1-diphenyl-2-picrylhydrazyl (DPPH). One % of powder Chungkookjang dissolved in methanol showed highest antioxidant activity. Systolic blood pressure of hypertensive volunteers who took 20 g of mixed fermented powder decreased on average by 10 mmHg in 2 h. Preference of mixed fermented soybean containing barley, wormwood, sea tangle to fermented soybean was demonstrated by t-test analysis. Mixed fermented grains can be developed as a functional food to lower hypertension.

Changes in Nitrogenous Compounds of Soybean during Chungkookjang Koji Fermentation (청국장(淸國醬) 발효중(醱酵中) 질소화합물(窒素化合物)의 변화(變化))

  • In order to study the flavor quality of Chungkookjang, the changes in nitrogenous compounds, nucleotides and their related compounds, free amino acids, amino acid composition and fatty acids were analysed during Chungkookjang Koji fermentation. Koji was prepared with Bacillus natto isolated from Japanese natto. Insoluble nitrogenous was rapidly decreased, whereas PAA (peptide, amino, ammonia) nitrogen were slightly increased during the fermentation of Chungkookjang Koji. The content of extracted nitrogen and free amino acid nitrogen were rapidly increased until 48 hours fermentation of Chungkookjang Koji and then decreased. The contents of ADP, ATP, AMP and inosine in raw soybean were abundant. The contents of ADP, ATP and AMP were decreased while inosine and hypoxanthine were increased during the fermentation of Chungkookjang Koji. The free amino acids analyzed in this experiment were not changed in composition but changed in amounts during the fermentation of Chungkookjang Koji. The contents of alanine, valine, isoleucine and phenylalanine were continually increased during the fermentation of Chungkookjang Koji. The contents of lysine, histidine, arginine, glutamic acid, glycine, methionine and tyrosine were increased until 48 hours fermentation and then decreased gradually. The increase in the contents of aspartic acid, threonine, serine, proline and cystine were fluctuated. In raw soybean, amino acid composition such as glutamic acid, serine and proline were dominant amino acid and amounts those were 63.8% of the total amino acids. The contents of aspartic acid, proline, glycine, alanine, cystine, leucine and tyrosine were continually decreased during the fermentation of Chungkookjang Koji, arginine and methionine were increased until 48 hours fermentation of Chungkookjang Koji and then decreased gradually. The increase of threonine and serine were fluctuated. Eight kinds of fatty acids were detected from raw soybean, but 10 kinds of fatty acids detected from Chungkookjang Koji. Palmitic, oleic and linoleic acid were identified as the major fatty acid of raw soybean and Chungkookjang Koji, and amounts of those were estimated above 80% of the total fatty acids.
