The influence of deficient(7${\mu}M$) and suffcient(1000${\mu}M$) magnesium(Mg) supply on the content of mg, chlorophyll, protein and the activity of superoxide dismutase(SOD), scavengers of superoxide radical($O_2$), and ascorbate peroxidase(AP), $H_2O_2$scavenging enzyme, and glutathione redutase(GR) were studied in pumpkin(Cudurbita moscata $D_{UCHESNE}$) plants over a 11-day period. Over the 11 days Period of growth in nutrient solution with sufficient and deficient Mg supply, the contents of Mg, chlorophyll protein and the activities of the antioxidative enzymes remained more or less constant in Mg-sufficient leaves. In Mg-deficient leaves, the contents of Mg, chlorophyll and protein was seriously decreased with time, however the activities of SOD, AP and GR highly enhanced compared to those of Mg-sufficient. The results indicated the stimulative effect of Mg deficiency on toxic oxygen species and scavenging enzymes in plants.
The effects of decursinol and decursin on chloroplast-mediated electron transport and phosphorylation in barley seedlings were investigated in comparison with coumarin in the dark or light. The changes of CP-complexes were also studied. Decursinol, decursin and coumarin caused marked inhibitory effects on germination of seed and electron transport and phosphorylation activity of seedlings. The following order of inhibitory effectiveness was exhibited; decursinol>coumarin>decursin. Loss of chlorophyll and decrease of electron transport activity were retarded in the dark, but were reversely accelerated in the light by these three chemicals. The changes of CP-complex patterns were also similar to effects on chlorophyll content and the electron transport activity. These opposite effect in the dark and light suggest that these three chemicals act as natural growth retardants rather than cytokinins or growth inhibitors.

Changes in physiological and biochemical patterns in lucerne plants caused by the presence of 'Candidatus Phytoplasma australasia', which is one of the significant pathogens causing yield losses in lucerne plants, were investigated. Significant differences were evident in total chlorophyll, chlorophyll a, chlorophyll b, and protein amounts between 'Ca. Phytoplasma australasia'-positive and negative lucerne plants. Stress-related metabolites such as phenol, malondialdehyde, and proline accumulations in 'Ca. Phytoplasma australasia'-positive plants were remarkably higher than those of phytoplasma-negative plants. As a response to disease attack, phytoplasma-positive plants exhibited higher antioxidant enzymes (peroxidase and catalase) and nonenzymatic metabolite responses such as jasmonic and salicylic acids. We state that partial disease responses were revealed for the first time to breed resistant lucerne lines infected by 'Ca. Phytoplasma australasia'.
Heat shock protein 70 (HSP70) is known as molecular chaperone, the fundamental protein participating in various processes, from nascent protein synthesis to protection of proteins during abiotic stresses and developmental programs. However, their biological functions in plants are not yet well known. Here, NtHSP70-1 (AY372069), HSP70 of Nicotiana tabacum induced by heat stress was investigated. To analyze the protective role of NtHSP70-1, transgenic tobacco plants, which constitutively overexpressed NtHSP70-1 as well as contained either the vector alone or having NtHSP70-1 in the antisense orientation, were constructed. The altered NtHSP70-1 levels in plants were confirmed by western blotting and transgenic sense lines exhibited tolerance to heat stress. Seedlings with the constitutively expressed NtHSP70-1 grew as green or healthy plants after heat stress. In contrast, transgenic vector or antisense lines exhibited yellowing of leaves or some delay in growth, which finally led to death. Evaluation of chlorophyll contents of heat-shocked transgenic tobacco seedlings indicated that NtHSP70-1 contributes to thermotolerance by preventing chlorophyll synthesis in plants.

Trampling stress in turfgrass fields caused by traffics often occurs in zoysiagrass (Zoysia japonica) fields including golf course. In order to know the influences of trampling stress on the growth and development of turfgrass, leaf and root growth, chlorophyll fluorescence, chlorophyll content and 2-DE protein analysis were conducted in the turfgrass plants subjected to various levels of trampling stress from 0 to $9,420J\;day^{-1}$ day. Shoot growth was more highly inhibited by trampling stress than root growth. Although root growth was affected by trampling with weak intensity, the highest root length was observed in the turfgrass treated with weak trampling ($1,570J\;day^{-1}$). Chlorophyll fluorescence (Fv $Fm^{-1}$) was drastically lowered by trampling stress with moderate intensity. Leaf number showed similar tendency with leaf greenness. The number was decreased as the trampling intensity was increased. Shoot dry weight was decreased showing a similar tendency with the result of shoot length. The specific protein expressions under weak trampling were related to the functions of stress amelioration. Heat shock 70 kDa protein 10 and oxygen-evolving enhancer protein 1 were the proteins increased positively under trampling stress.
The objectives of this study focused on measuring chlorophyll fluorescence related to drought stress comparing some parameters. Almost parameters were declined although they were not significant on the basis of mean values of fluorescence of total leaf area. While the ratio of fluorescence intensity variable chlorophyll ($F_V$) to fluorescence intensity maximal chlorophyll ($F_M$) was not changed, the effective quantum yield of photochemical energy conversion in photosystemII (${\Phi}PSII$) and chlorophyll fluorescence decrease ratio ($R_{fd}$) were slightly reduced, indicating inhibition of the electron transport from quinone bind protein A ($Q_A$) to quinone bind protein B ($Q_B$). Some parameters such as non-photochemical quenching rate ($NPQ_{_-LSS}$) and coefficients of non-photochemical quenching of variable fluorescence (qN) in mid-zone of leaf and near petiole zone leaf were significantly enhanced within 4 days after drought stress, which can be used as physiological stress parameters. Decrease in ${\Phi}PSII$ could was significantly measured in all leaf zones. In conclusion, three parametric evidences for chlorophyll fluorescence responses such as ${\Phi}PSII$, NPQ, and qN insinuated the possibility of photophysiological indices under drought stress.
The multifunctional triple gene block protein 1 (TGB1) of the Potexvirus Alternanthera mosaic virus (AltMV) has been reported to have silencing suppressor, cell-to-cell movement, and helicase functions. Yeast two hybrid screening using an Arabidopsis thaliana cDNA library with TGB1 as bait, and co-purification with TGB1 inclusion bodies identified several host proteins which interact with AltMV TGB1. Host protein interactions with TGB1 were confirmed by biomolecular fluorescence complementation, which showed positive TGB1 interaction with mitochondrial ATP synthase delta' chain subunit (ATP synthase delta'), light harvesting chlorophyll-protein complex I subunit A4 (LHCA4), chlorophyll a/b binding protein 1 (LHB1B2), chloroplast-localized IscA-like protein (ATCPISCA), and chloroplast ${\beta}$-ATPase. However, chloroplast ${\beta}$-ATPase interacts only with $TGB1_{L88}$, and not with weak silencing suppressor $TGB1_{L88}$. This selective interaction indicates that chloroplast ${\beta}$-ATPase is not required for AltMV movement and replication; however, TRV silencing of chloroplast ${\beta}$-ATPase in Nicotiana benthamiana induced severe tissue necrosis when plants were infected by AltMV $TGB1_{L88}$ but not AltMV $TGB1_{L88}$, suggesting that ${\beta}$-ATPase selectively responded to $TGB1_{L88}$ to induce defense responses.
The response of Spirulina (Arthrospira) platensis to high salt stress was investigated by incubating the cells in light of moderate intensity in the presence of 0.8 M NaCl. NaCl caused a decrease in photosystem II (PSII) mediated oxygen evolution activity and increase in photosystem I (PSI) activity and the amount of P700. Similarly maximal efficiency of PSII (Fv/Fm) and variable fluorescence (Fv/Fo) were also declined in salt-stressed cells. Western blot analysis reveal that the inhibition in PSII activity is due to a 40% loss of a thylakoid membrane protein, known as D1, which is located in PSII reaction center. NaCl treatment of cells also resulted in the alterations of other thylakoid membrane proteins: most prominently, a dramatic diminishment of the 47-kDa chlorophyll protein (CP) and 94-kDa protein, and accumulation of a 17-kDa protein band were observed in SDS-PAGE. The changes in 47-kDa and 94-kDa proteins lead to the decreased energy transfer from light harvesting antenna to PSII, which was accompanied by alterations in the chlorophyll fluorescence emission spectra of whole cells and isolated thylakoids. Therefore we conclude that salt stress has various effects on photosynthetic electron transport activities due to the marked alterations in the composition of thylakoid membrane proteins.
To understand the responses of grapevines in response to cold stress causing the limited growth and development, differentially expressed genes (DEGs) were screened through transcriptome analysis of shoots from 2 grapevine cultivars ('Campbell Early' and 'Muscat Baily A') kept at -$2^{\circ}C$ for 4 days. In gene ontology analysis of DEGs from 'Campbell Early', there were 17,424 clones related with biological process, 28,954 with cellular component, and 6,972 with molecular function genes in response to freezing temperature. The major induced genes included dehydrin xero 1, K-box region and MADS-box transcription factor family protein, and MYB domain protein 36, and inhibited genes included light-harvesting chlorophyll B-binding protein 3, FASCICLIN-like arabinoogalactan 9, and pectin methylesterase 61 in 'Campbell Early' grapevines. In gene ontology analysis of DEGs from 'Muscat Baily A', there were 1,157 clones related with biological process, 1,350 with cellular component, and 431 with molecular function gene. The major induced genes of 'Muscat Baily A' included NB-ARC domain-containing disease resistance protein, fatty acid hydrozylase superfamily, and isopentenyltransferase 3, and inhibited genes included binding, IAP-like protein 1, and pentatricopeptide repeat superfamily protein. All major DEGs were shown to be expressed differentially by freezing temperature in real time-PCR analysis. Protein domain analysis using InterPro Scan revealed that ubiquitin-protein ligase was redundant in both tested grapevines. Transcriptome profile of shoots exposed to cold can provide new insights into the molecular basis of tolerance to low-temperature in grapevines, and can be used as resources for development new grapevines tolerant to coldness.
Blue green algaes (NIES 19, 39, 46) have been cultivated to product sigle cell protein. After 7 days of cultivation under 5000 (W/M$^{2}$) of ligth intensity, final cell concentration was 2.8 (g/L) and chlorophyll a was 4.9 (mg/L). When initial concentration of NaHCOC was 1.7% and NaNO$_{3}$ was0.25%, maximum cell concentration was obtained. Spirulina platensis NIES 39 showed faster growth rate than NIES 46. While most 39 sedimented, most 46 floated.
