Phenolic Antioxidants Isolated from Mulberry Leaves

  • In this study, the crude methanol extract of mulberry leaves was fractionated with chloroform, ethyl acetate, n-butanol, and water, successively. The antioxidant activities of the fractions were examined with the 2-deoxyribose oxidation and linoleic acid peroxidation methods. The ethyl acetate fraction showed the strongest antioxidant activity. From it we isolated chlorogenic acid, caffeic acid, quercetin $3-O-{\beta}-D-glucopyranoside$, and kaempferol $3-O-{\beta}-D-glucopyranoside$ with preparatory octadecyl silane-high performance liquid chromatography (ODS-HPLC), and identified the compounds by nuclear magnetic resonance (NMR) and fast atom bombardment mass (FAB-MS) analyses. Overall, quercetin $3-O-{\beta}-D-glucopyranoside$ showed the strongest antioxidant activity by both the 2-deoxyribose oxidation and rat liver microsome peroxidation methods.

  • Methanol extracts of eighty Chinese medicinal plants were investigated for platelet-activating factor (PAF) receptor binding inhibitory activity using rabbit platelet. Extracts of Cratoxylon ligustrinum, Kalimeris indica, Euonymus japonica, Ophiopogon japonicus, Gleditsia sinensis, Clausena lansium, Agave sisalana were found to exhibit significant inhibitory effects. Chloroform partition of the Methanol extract of Kalimeris indica was further fractionated by column chromatography to afford one strong active subfraction with 93.6% inhibition at a concentration of $100{\mu}g/ml$.

  • The herb of Melampyrum roseum var. hirsutum Beauv. was extracted with methanol and the extract was fractionated with n-hexane, chloroform, ethylacetate and n-buthanol. Repeated column chromatography of silica gel, Sephadex LH-20 and Lobar-A lichroprep Si 60 led to the isolation of five compounds from n-hexane layer and ethylacetate layer. These compounds were identified as apigenin, linarin, pectolinarin, wogonin and mussaenoside, respectively, by the physicochemical and spectral properties.

  • An alkaloidal component was extracted from the root of Aconitum koreanum R. $R_{AYMOND}$ of the commercial origin. Crude extract was obtained in ca. 2% yield out of dried root with ether-methanol (10:1). White amorphous compound was precipitated out from the HCl solution upon the addition of ammonia water, and crude base was obtained by extraction of its mother liquid with chloroform. This crude base was purified by the alumina-column chromatographic method. A pure alkaloidal component(500mg) was yielded from its crude base. Its pure base comes as needle crystals melting at $250{\sim}252^{\circ}C$ (decomp.), $[\alpha]^{20}_D$ D+9.82 (methanol) and Anal. calcd. for $C_{19}H_{27}O_{8}N$.

  • Alcoholic extract of Spathodea campanulata P. aerial parts, and two of the isolated fractions from celite column showed strong antioxidant activity (92, 94 & 89% RSA, Radical Scavenging Activity). Phytochemical investigation of chloroform/EtOAc faction of this column led to the isolation of phenolic acids, caffiec acid (1), and ferulic acid (2), fraction EtOAc/MeOH on further fractionation afforded 3 Flavonoids, kampferol 3-O-glucoside (3), quercetin 3-methyl ether (4) and 8-methoxy kampferol 3-O-glucoside (5). The isolated constituents were identified by co chromatography with authentic samples, TLC, PC., UV, MS and $^1H-NMR$. Also the lipoidal matter of the plant was studied. The unsaponifiable matter was found to be mixture of hydrocarbons from $(C_{14}-C_{28})$, cholesterol, campasterol, stigmasterol, and ${\alpha}-amyrin$. Fatty acid methyl esters were found to contain 12 fatty acids. The fatty acids containing $C_{18}$ farmed ca.65% of the total mixture.

Separation of Antioxidant compounds from Persimmon Leaves (감잎(시엽(枾葉))의 항산화 성분 분리)

  • This study was carried out to separate and identify the antioxidative substances in persimmon leaves. The antioxidative substances in persimmon leaves were extracted by methanol. The extract was fractionnated by SEP-PAK cartridge colum. From these results five fractions(F-I${\sim}$V) were obtained. Antioxidative activity of each fractions was examined by the DPPH methord. The F-II, III and IV showed antioxidative activity and among them F-II and F-III showed the strongest. Five frations were separated by TLC using ethylacetate : chloroform : formic acid : $H_2O$(8 : 1 : 1 : 1 v /v) as the solvent. From these results were obtained spots of Rf 0.71, 0.35 and 0.25. This spots were scraped from the plate and extracted by methanol. The extracts thuse obtained were used for examination of identify by TLC, UV /VIS-spectrophotometer and HPLC. Among them spot of Rf 0.71 were demonstrated to catechin and the spots of Rf 0.35 and 0.25 was suggested to polyphenol substances.

  • Attempts were made to investigate sterol components of Calvatia saccatum (Vahl.) Fr. which grows wildly in Korea. Its carpophores were collected in Gyeong Gi Province and extracted with chloroform and methanol. Two compounds were isolated from the extract and one of these compounds was identified as ergosterol by TLC, GLC and chemical tests.

Anthraquinones with Immunostimulating Activity from Cassia tora L.

  • Many of plants had been reported having immunostimulating activity. This study reports the immunostimulating activity of Cassia tora L. (Leguminosae) seed, by means of solvent extraction method. Ethanol extract and solvent fractions, n-hexane, chloroform, ethylacetate, n-butanol and aqueous layer of Cassia tora L. seed were tested for immunostimulating activity in vitro. The ethylacetate-soluble fraction caused significant inhibition on the production of nitric oxide by murine macrophages (RAW 264.7), and mouse splenocytes were also stimulated at the concentration of 10 pgfmL. Three anthraquinones, chrysophanol (1), isochrysophanol (2) and aloe-emodin (3) with immunostimulating activity were isolated from the ethylacetate-soluble fraction of Cassia tora L. seed through activity-monitored fractionation and isolation method. These results permit Cassia tora L. to be useful as one the of natural immunostimulating crops.

Anti-HIV Activity of Dehydroaltenusin- a Metabolite from a Streptomyces sp.

  • Dehydroaltenusin (1) was isolated from the chloroform extract of the culture filtrate of a Streptomyces sp. and its structure was determined from spectral data as well as by comparison with published values. In an XTT-based in vitro anti-HIV assay, dehydroaltenusin effectively inhibited the cytopathic effects of HIV infection at a concentration of $1-5\;{\mu}g/mL$.

  • To elucidate chemical structure of the wood extracitives, softwood Larix leplolepis Gorden) metal was extracted with 95% ethanol at room temperature for 72 hours. The extract was fractionated with organic solvents such as n-hexane, chloroform. ether, and ethylacetate. From the n-hexane soluble fraction of the extratives, flavonoid and sterol compounds were isolated and identified as taxifolin(5,7,3',4'-tetrahydroxyflavanonol)(I) and ${\beta}$-sitosterol(II) by UV, IR, $^1$H-NMR spectroscopy and Mass spectrometry.
