Biological Activities of Wild Sparassis crispa Extracts (야생 꽃송이버섯 추출물의 생리활성)

  • Physiological activities of solvent extracts of Sparassis crispa were investigated for fibrinolytic, thrombin inhibitory, anti-oxidative and anti-inflammatory activities. The fibrinolytic activity was the highest in ethyl acetate extract (2.03 plasmin units/mL) followed by butanol extract (0.70 plasmin units/mL). The ethyl acetate extract exhibited the highest anti-oxidative activity as assessed by the 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging rate with a value of 95.94%. The chloroform extract showed thrombin inhibitory activity up to 83.87%. The chloroform extract also showed the highest anti-inflammatory effects on the production of nitric oxide (NO) in lipopolysaccharide (LPS)-stimulated RAW264.7 cells. These findings suggest that Sparassis crispa may be a useful material for development of drugs and functional foods.

Effect of Betula schmidtii Regel on Blood Pressure (한국산 생약제들의 혈압강하작용에 대한 연구(III) -박달수피의 혈압에 대한 작용-)

  • This study was undertaken to search for a new antihypertensive or hypotensive agents in natural crude products in Korea. Betula schmidtii Regel, a tree belonging to the Betulaceae family, is cultivated for the most part of our country, and their cortecies or tree milk has been used as folk remedy, which exerted good antiinflammatory effect on rheumatic fever, arthritis or rheumatism. The rats were treated with several extracts from Betula schmidtii Regel, that is, extracts of water, methanol from inner cortecies, extracts of ether, methanol, hexane, chloroform or butanol from outer and total cortecies. In this experiment, we measured the blood pressure and heart rate in anesthetized rats, and atropine, diphenhydramine, phentolamine, propranolol, epinephrine, hexamethonium and hydralazine were pretreated prior to Betula extracts to clarify the mechanism of the hypotensive action. The results of the experiment were as follows; 1. BOIE(Betula outer cortex water-insoluble extract) and BICE(Betula inner cortex chloroform extract) elevated the blood pressure and heart rate. 2. BIME(Betula inner cortex methanol extract), BIBE(Betula inner cortex butanol extract) and BIWE (Betula inner cortex water extract) significantly lowered the blood pressure and heart rate. 3. With the pretreatment of hydralazine, the hypotensive effect of Betula schmidtii Regel was significantly blocked. From the above results, it is suggested that the hypotensive effect of Betula schmidtii Regel is due to the direct vasodilation of blood vessel.

Screening of Antimicrobial activity of the Plantain (Plantago asiatica L.) extract (질경이(Plantago asiatica L.) 추출물의 항균성검색)

  • In order to develop a natural food preservative, freeze dried plantain (Plantago asiatica L.) was extracted with several solvents, and the antimicrobial activity was investigated. The methanol extract exhibited antimicrobial activites against five strains of bacteria such as Bacillus subtilis, Escherichia coli, Staphylococcus aureus, Listeria monocytogenes, and Vibrio parahaemolyticus. The methanol extract at the concentration of 1.0 mg/ml completely inhibited the growth of B. subtilis and V. parahaemolyticus. Antimicrobial activity of the ethylacetate fraction from the methanol extract was the strongest compared with those of other solvent fractions such as n-hexane, chloroform, n-butanol and water. The ethylacetate fraction showed the inhibitory effect at the concentration of 0.5 mg/disc on the growth of B. subtilis and V. parahaemolyticus.

  • The effect of Herricium erinaceus on the mutagenicity in salmonella assay and quinone reductase activity in hapalclc7 cells were studied. Antimutagenic as evaluated by Ames test, the extract and fractions of JHerricium erinaceus had no effects on the mutagenicity by themselves. However, methanol extract and fractions from Hericium erinaceus showed strong inhibitory effect on the mutagenesis induced by N-methyl -N'-nitor-N- nitroso-guanidine (MNNG) and benzo(a)pyrene (B(a)P). Among the solvent fractions of methanol extract, the hexane fraction, the chloroform fraction and the ethylacetate fraction exhibited stronger inhibitory activity against MNNG and B(a)P induced mutagenesis than butanol and water fractions. The methanol extract, the extract, the chloroform and the ethylacetate fractions of Hericium erinaceus induced the activity of quinone reductase, an anticarcinogenic marker enzyme, in murine hepalclc7 cells while the others did have little effect on the enzyme activity.

Selective in vitro cytotoxicity of Hypericum hookerianum towards cancer cell lines

  • The methanol extracts of the aerial parts, leaves and stem of Hypericum hookerianum were tested for in vitro cytotoxicity on selected normal and cancer cell lines and anti tumor activity using DLA cells. Cell viability and morphological changes were assessed. Among the three extracts tested, the stem extract of Hypericum hookerianum showed potent cytotoxicity against HEp-2 and RD cell lines. The $CTC_{50}$(concentration required to reduce viability by 50%) of this extract was found to be $2.02\;{\mu}g/ml$ for RD cell line, $10.25\;{\mu}g/ml$ for HEp-2 cell line and $100.06\;{\mu}g/ml$ for Vero cell line. In the clonogenic assay, no colony formation was observed up to a concentration of $100\;{\mu}g/ml$. In the short term cytotoxicity studies using DLA cells, 50% viability was observed in the concentration range of $50-100\;{\mu}g/ml$ for aerial parts, $100-200\;{\mu}g/ml$ for stem and more than $200\;{\mu}g/ml$ for leaf extracts of Hypericum hookerianum. In the long-term activity using HEp-2 cell line, no colony formation was observed over a concentration of 200 mg/ml for the stem extract. Hypericum hookerianum stem extract was fractionated into petroleum ether, chloroform, ethyl acetate and methanol soluble fractions. The petroleum ether and chloroform soluble fractions showed higher cytotoxic activity against HEp-2 cell line when compared to the other two fractions. The methanol stem extract of Hypericum hookerianum has the potential for further investigation in animal models to determine its anti-tumor activity and to identify its active principles.

  • A method was described for the analysis of Artemisia extract in cosmetic soap. Esculetin-6-methylether was used as indicator ingredient for analysis of Artemisia extract. It was isolated from the plant and purified with silicagel column. The structure was conformed with IR, NMR and MASS spectroscopy. The Artemisia extract in sample was isolated with chloroform and interfering com- pounds were eliminated with silicagel column. The Artemisia extract was determined by reverse phase high- performance liquid chromatography with a fluorescence detector and a solvent system of H2O/THF/MeOH. The recorveries of Artemisia extract were 93.9-106.1% in the cosmetic soap.

  • Mansam, the root of Codonopsis pilosula (Campanulaceae) has a action on blood metabolism. It has been known to possess anti leukemic effect by increasing of red blood cell, at same time, by decreasing white blood cell, with this connection, present study is aimed to clarify such potential anti leukenic effect by testing ethanol extract and chloroform extract against murine tumor model, lymphocytic leukemia P388. The data indicated that these extracts appeared to be inactive against this tumor line.

  • The inhibitory effects of various extracts from Chinese pepper on the mutagenicity and the growth of MG-63 human osteosarcoma cells were studied. Chinese pepper was extracted with methanol and then the methanol extract was further fractionated by using hexane, chloroform, ethyl acetate and butanol. The methanol extract of Chinese pepper revealed the strong antimutagenic activity on the aflatoxin B1(AFB1) and N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) in Ames mutagenicity test and SOS chromotest.

Effect of Butanol Fraction of Panax ginseng Head on Gastric Lesion and Ulcer

  • From our previous result that Panax ginseng head extract had inhibition of gastric damages, the extract was fractionated. Among the hexane, chloroform, butanol and water fractions, butanol fraction Showed the most potent inhibition of HCl.ethanol-induced gastric lesion, aspirin-induced gastric ulcer, acetic acid-induced ulcer and Shay ulcer. Butanol fraction showed significant increase in mucin secretion, and inhibited malondialdehyde (MDA) and $H^{+}/K^{+}ATPase$ activity in the stomach. This results indicate that the effectiveness of the fraction on gastric damages might be related to inhibition of acid secretion, increment of mucin secretion and antioxidant property.
