The adsorption ability of dyes on chitin, a natural polymer was investigated for decolorization of dye wastewater. Chitin was manufactured in lab by decalcification in dilute aqueous hydrochloric acid solution and deproteination in dilute aqueous sodium hydroxide solution with shrimp shells. Absorbance of residue solution of dyebaths after dye adsorptions of chtin were measured in varieties of dye concentration and dipping periods. Three kinds of Direct dyes -C.I. Direct Red 81 (red 81), C.I. Direct Brown l(brown 1) and C.I. Direct Green 26(green 26) - were used. Red 81 and brown 1 have smaller molecular weight than green 26. The results from experimentals were of]tamed as follows: 1) Adsorption of green 26 was improved in dyebath by addition of salt, but red 81 and brown 1 were not found any improvements. 2) Smaller size of chitin particles could be absorbed much more speedy. In this experiments, the smallest range of chitin particle size was $250\mu{m}$ and less. 3) The most efficient amount of chitin for 20m1 of dyebath was 0.2g, 4) Both of red 81 and brown 1 showed good and speedy adsorption abilities as dyestuffs of over 90 percent in just one minute in dyebath of 0.01% dye concentration. But green 26 was absorbed slowly because of its large molecular weight. It took 40minutes to absorb dyestuffs of over 90 percent in dyebath of 0.01% dye concentration.

Nanofibrous membranes of poly(D,L-lactic acid)/chitin blend were prepared by electro spinning for a barrier of guided tissue regeneration. A miscible solution was obtained by the blending chitin-salt complex into 1-methyl-2-pyrrolidone solution of poly(D,L-lactic acid). The properties of the blend were examined for nanofibrous fabrication. The viscosity of the blend solution was increased significantly due to chain entanglement despite the low ratio of chitin to poly(D,L-lactic acid). An interaction between two polymeric compositions was confirmed by Fourier transform infrared spectroscopy. X-ray diffraction detected an appreciably ordered microstructure in the nanofiber of the blend. A membrane of thinner nanofibers was fabricated by electro spinning the chitin blend. The permeability of the membranes was examined using bioactive model compounds.
Packed bed column과 continuous stirred tank reactor에서 고정화된 glucoamylase의 catalytic activity를 비교하였다. Settling chamber를 이용한 연속애화, 당화 공정을 사용하여 액화된 전분으로부터 포도당을 연속적으로 생산하였다. . Chitin에 고정화된 glucoamylase에 의한 당화실험에 있어서는 dextrin의 농도가 100g/l일 때, 체류시간 20분 동안 20의 당화수율을 나타냈다.
This study was performed to investigate the effects of Chitosan on the nickel poisoning in rats. In the study, 150 male Sprague-Dawley were used. The experimental groups were divided into four: A (30 mg/L nickel), B (30 mg/L nickel+0.2% Chitin, Chitosan and Dithiocarbamate Chitosan), C (30 mg/L nickel+0.4% Chitin, Chitosan and Dithiocarbamate Chitosan), D (30 mg/L nickel+0.8%Chitin, Chitosan and Dithiocarbamate Chitosan). The results were as flows; 1. The nickel concentration in the livers of the control group (A) was $0.153{\sim}0.186\;mg/kg$ but the nickel concentration in the livers of the experimental decreased during the experimental period (P<0.05). 2. Metallothionenin levels in rat liver were $2.77{\sim}3.25\;ug/g$ wet,wt in control group (A), but were $2.89{\sim}3.51\;ug/g$ wet,wt (B), $2.97{\sim}3.62\;ug/g$ wet,wt (C), $2.68{\sim}3.68\;ug/g$ wet,wt (D). Respectively in the experimental groups. The experimental groups were inclined to increase compare to the control group (P<0.05). In conclusion, this study revealed a preventive effect of Chitin, Chitosan and Dithiocarbamate Chitosan against nickel toxicity.
The Arabidopsis gene AtLEC (At3g15356) gene encodes a putative 30-kDa protein with a legume lectin-like domain. Likely to classic legume lectin family of genes, AtLEC is expressed in rosette leaves, primary inflorescences, and roots, as observed in Northern blot analysis. The accumulation of AtLEC transcript is induced very rapidly, within 30 min, by chitin, a fungal wall-derived oligosaccharide elictor of the plant defense response. Transgenic Arabidopsis carrying an AtLEC promoter-driven ${\beta}$-glucuronidase (GUS) construct exhibited GUS activity in the leaf veins, secondary inflorescences, carpel heads, and silique receptacles, in which no expression could be seen in Northern blot analysis. This observation suggests that AtLEC expression is induced transiently and locally during developmental processes in the absence of an external signal such as chitin. In addition, mechanically wounded sites showed strong GUS activity, indicating that the AtLEC promoter responds to jasmonate. Indeed, methyl jasmonate and ethylene exposure induced AtLEC expression within 3-6 h. Thus, the gene appears to play a role in the jasmonate-/ethylene-responsive, in addition to the chitin-elicited, defense responses. However, chitin-induced AtLEC expression was also observed in jasmonate-insensitive (coi1) and ethylene-insensitive (etr1-1) Arabidopsis mutants. Thus, it appears that chitin promotes AtLEC expression via a jasmonate- and/or ethylene-independent pathway.
Chitin deacetylase(CDA; EC 3.5.1.41)는 키틴의 N-acetamide bonds를 가수분해하여 이를 키토산으로 전환시키는 효소다. 한편, 키토산은 의약, 화장품, 식품, 농업 등의 분야에서 다양하게 응용되는 고분자 다당류이다. 본 논문에서는 미생물 유래 CDA의 분포, 분석법, 효소적 특성, 기질 특이성, 작용기작, 유전자의 구조, 생물학적 역할, 응용 등의 최신 지견을 기술하고자 하였다. 미생물 CDA가 세포벽 형성과 식물-미생물 상호작용에 관여한다는 연구결과들을 제시하였으며, CDA의 유전자 구조를 다양한 acetylated poly/oligo-saccharides를 탈아세틸화하는 family 4 carbohydrate esterase의 유전자 구조와 비교하였다. 키틴의 탈아세틸화로 키토산을 제조하는 과정에 CDA의 활용 가능성과, CDA를 포함한 고활성의 키틴 대사효소들을 분비하는 곤충 병원균의 활용 가능성도 살펴보았다.

In the course of searching for potent chitin synthase 1 inhibitors from natural resources, Streptomyces sp. A6705 was found to exhibit potent inhibitory activity against the chitin synthase 1 from C. albicans (CaCHS1p). As a result, the inhibitor was isolated and identified using a series of chromatographies. Through chemical analyses with UV spectrophotometry, MS spectrometry, and various NMR techniques, the inhibitor was identified as N,N-bis(2-phenylethyl)urea. The compound exhibited strong inhibitory activity against the chitin synthase 1 from C. albicans with an $IC_{50}$ of 14 ${\mu}g$/ml, representing a similar inhibitory activity to that of the well-known chitin synthase inhibitor, polyoxin D ($IC_{50}$ 15 ${\mu}g$/ml). However, the compound showed no inhibitory activity against the chitin synthase 2 of Saccharomyces cerevisiae up to 280 ${\mu}g$/ml, which is structurally and functionally analogous to CaCHS 1 p. In addition, the compound exhibited weak antifungal activities against Cryptococcus neoformans and Rhizoctonis solani.
An antifungal chitinase, ChiCW, produced by Bacillus cereus 28-9 is effective against conidial germination of Botrytis elliptica, the causal agent of lily leaf blight. ChiCW as a modular enzyme consists of a signal peptide, a catalytic domain, a fibronectin type-III-like domain, and a chitin-binding domain. When two C-terminal domains of ChiCW were truncated, $ChiCW{\Delta}FC$ (lacking the chitin-binding domain and fibronectin type III-like domain) lost its antifungal activity. Since $ChiCW{\Delta}C$ (lacking the chitin-binding domain) could not be expressed in Escherichia coli as $ChiCW{\Delta}FC$ did, a different strategy based on protein engineering technology was designed to investigate the involvement of the chitin-binding domain of ChiCW ($ChBD_{ChiCW}$) in antifungal activity in this study. Because ChiA1 of Bacillus circulans WL-12 is a modular enzyme with a higher hydrolytic activity than ChiCW but not inhibitory to conidial germination of Bo. elliptica and the similar domain composition of ChiA1 and ChiCW, the C-terminal truncated derivatives of ChiA1 were generated and used to construct chimeric chitinases with $ChBD_{ChiCW}$. When the chitin-binding domain of ChiA1 was replaced with $ChBD_{ChiCW}$, the chimeric chitinase named ChiAAAW exhibited both high enzyme activity and antifungal activity. The results indicate that $ChBD_{ChiCW}$ may play an important role in the antifungal activity of ChiCW.
미곡저장중 곰팡이 침해 여부를 chitin 정량법으로 판정하기 위하여 상이한 수부조건(14.5, 18.0, 22.3, 26.9%)에서 26$^{\circ}C$를 유지하면서 4 주동안 저장한 후 미곡(통일)에 침해한 곰팡이 및 화학성분의 변화를 1주 간격으로 조사, 분석한 결과를 요약하면 다음과 같다. 1) 표면살균 후에 나타난 곰팡이는 Aspergillius sp., Penicilium sp., Rhizopus sp., 였으며 Asperg-illius sp.가 대부분이었다. 2) 통일쌀에 함유된 Chitin의 함량은 25$\mu\textrm{g}$/g-Dry.wt이었으나 수분의 함량이 높아지고 저장기간이 길어짐에 따라 증가하였다. 29.6%의 수분함량에서 4주간에 생성된 Chitin의 량은 1955$\mu\textrm{g}$/g.Dry.wt이었다. 3) 비환원당의 함량은 수분의 증가와 저장기간이 길어짐에 당라 감소하였다. 4) 지방산도는 수분의 증가와 저장기간이 길어짐에 따라 증가하였다. 5) 이상의 결과로 미곡 저장중 곰팡이 침해에 의하여 민감하게 변하는 Chitin 졍량법이 이용될 수 있을 것으로 사료된다.
