Role of the de novo Ceramide and Arachidonic Acid in Paclitaxel-Induced Apoptosis

  • Recently, several reports suggest that ceramide formation has been implicated in the apoptosis signaling in response to chemotherapeutic agents. In this study, to enhance paclitaxel-mediated cytotoxicity and endogenous ceramide levels, we blocked ceramide metabolism using an inhibitor of glucosylceramide synthase, l-phenyl-2- dacanoylamino-3-morpholino-l-propanol(PDMP) and SM synthase, D609. Exposure of human breast cancer cells to paclitaxel accumulated de novo ceramide synthesis by enhancement of SPT activity 1.2-fold, whereas ceramide synthase activity was not altered. (omitted)

  • Ceramide is an important lipid messenger involved in mediating a variety of cell functions including apoptosis. Previously, we have shown that ceramide induces Bax translocation which is associated with cytochrome c release from the mitochondria. In this study, we show that p38 MAP kinase is involved in ceramide-induced Bax translocation. In human leukemic cells, ceramide stimulated the phosphorylation of p38 MAP kinase. Preincubation of cells with SB203580, a specific inhibitor of p38 inhibited DNA fragmentation induced by cell-permeable ceramide. (omitted)

Activation of Akt/PKB at Serine 473 by N-acetylphytosphingosine (NAPS) and $C_{2}-ceramide$ Reduces Melanin Synthesis in B16F10 Mouse Melanoma Cells

  • Sphingolipid metabolites regulate many aspects of cell proliferation, differentiation, and apoptosis. In the present study, we have assessed the effects of the novel phytosphingosine derivative, N-acetylphytospingosine (NAPS), on the depigmentation of murine B16F10 melanoma cells, and have also attempted to identify the possible signaling pathway involved, in comparison with $C_{2}-ceramide$. NAPS and $C_{2}-ceramide$ both inhibited the growth of the B16F10 cells in a dose-dependent manner. Melanin content and tyrosinase activity were significantly reduced in response to treatment with NAPS and $C_{2}-ceramide$ at concentrations in a range between $1-5\;{\mu}M$. However, the levels of tyrosinase mRNA, as well as the levels of tyrosinase related protein-1 (TRP-1) and tyrosinase related protein-2 (TRP-2) genes and the level of tyrosinase protein remained unaffected by treatment with either NAPS or $C_{2}-ceramide$. We also attempted to determine the signaling pathway exploited by NAPS and $C_{2}-ceramide$. Interestingly, the phosphorylation of Akt/PKB at serine 473 by NAPS was reduced at the 5 minute mark, whereas $C_{2}-ceramide$ induced the phosphorylation of Akt/PKB at serine 473. Finally, Akt/PKB activity in the NAPS-treated cells was elevated in comparison with the untreated cells. LY294002, a specific PI3-K inhibitor which is located upstream of Akt/PKB, inhibited the phosphorylation of Akt/PKB, but induced an increase in melanin synthesis. These results suggest that the activation of Akt/PKB at serine 473 is related with the suppression of melanin production in the B16F10 mouse melanoma cells. Therefore, the mechanisms exploited by NAPS and $C_{2}-ceramide$ responsible for the depigmentation of B16F10 cells were concluded to involve the inhibition of melanosomal tyrosinase activity.

Preparation of Hydrated Liquid Crystalline Vesicle Using Mutual Self-Association between Ceramide and Phospholipid (인지질/세라마이드 혼합물의 상호적 자가회합 특성을 활용한 수화 액정형 베시클 제조)

  • An attempt was made to prepare hydrated liquid crystalline vesicles by utilizing a mutual self-assembly between phospholipid and ceramide. When an edge activator was added to the mixture of phospholipid and ceramide, it was possible to prepare a vesicle containing a high content of poorly soluble ceramide. In this study, we investigated changes in structural and thermal behaviors of the hydrated liquid crystalline phase according to the mixed composition between phospholipid, ceramide, and sodium deoxycholate. Also, both the droplet size of the vesicles with different composition and stability of the vesicle dispersion solution were observed. From results of the experiments, by increasing the ratio of ceramide up to 70% in the mixture between phospholipid and ceramide, a formulation containing 3.5 wt% of ceramide in the vesicle dispersion solution could be obtained.

Protective Effect of Ginsenoside Rgl on H2O2-Induced Cell Death by the Decreased Ceramide Level in LLC-PK1 Cells

  • Ceramide has been involved in celt death and acted as a lipid mediator of stress responses. Elevation of ceramide level was reported to occur in oxidative stress and lead to cell death in many cell types. This study was undertaken to elucidate a protective role of ginsenoside Rgl in cell death induced by oxidative stress. When LLC-PK1 cells were treated with $H_2O_2$ at a concentration of $400{\mu}M$ for 5 hr, cell death was observed and a released LDH activity indicative of cytotoxicity was Increased. $H_2O_2$ exposure to LLC-PK1 cells was shown to elevate the content of total ceramide by approximately 200% compared to control cells. Ceramide level was hypothesized to be a key to a reversal of cell death to survival. Ginsenoside Rgl at the concentrations ranging from 12.5 to $250{\mu}M$ protected LLC-PK1 cells from cell death induced by $H_2O_2\;at\;400{\mu}M$ for 5 hr, and decreased the ceramide level relative to $H_2O_2$. Ginsenoside Rgl inhibited neutral human ceramidase by 71% of controls, while sphingomyelinase was not inhibited. These results suggest that ginsenoside Rgl show the protection against cell death via the modulation of ceramide metabolism, and ceramide may be a promising therapeutic target for human diseases related to cell death.

Liquid Crystal Emulsions Containing High Content Ceramides for Improved Skin Barrier Functions

  • In this work, we fabricated liquid crystal (LC) emulsions with fatty alcohol in order to stabilize high content ceramide in cosmetic formulation. We investigated the role of fatty alcohol and surfactant in the formation of higher order structure. As a result, we found that they play a crucial role to form higher order structure. SAXS study shows that ceramide can be incorporated up to 3% in cosmetic formulation with higher order structure and its stability was maintained up to 12 weeks at room temperature. According to WAXS study, the higher order structure can suppress the re-crystallization of ceramide in cosmetic formulation. Finally, we performed in vivo skin barrier recovery test for the damaged skin. LC emulsions with ceramide and O/W emulsions show significant effect in skin barrier recovery at D 1, D 2 and D 6 compared to the untreated condition. While only LC emulsions show significant skin recovery effect at D 14. We expect that LC emulsions are the promising skin carrier to stabilize ceramide and LC emulsions with ceramide can improve the skin barrier function.

Skin Absorption and Physical Property of Ceramide-added Ethosome (세라마이드 함유 에토좀의 물성과 피부흡수)

  • In order to delivery biotin to skin, ethosomes containing both biotin and ceramide were researched by using high pressure homogenizer. Biotin was utilized as a drug and ceramide NP was utilized as a supporter of bilayer. The biotin was entrapped in aqueous core, while ceramide NP was packed in the bilayer of the ethosomes. Looking at the physical properties of vesicles containing ceramide NP, the sized was 80~130 nm, the polydispersity index was 0.09~0.16, and the zeta potential was -40~-49 mV. In vesicles without ceramide NP, the size was 124.80±1.46 nm, and the zeta potential and polydispersity index were -45.48±1.27 mV and 0.088±0.018, respectively. Therefore, the ethosome with ceramide NP has improved physical properties of vesicles compared to the ethosome without ceramide NP. Skin absorption rates of ethosomes with ceramide NP were 6.13~14.98%, while skin absorption rate of ethosome without ceramide NP was 7.08% at 12 h. In conclusion, ethosomes containing ceramide NP not only improved the skin absorption efficiency, but had also a positive effect on the stability of vesicles.

  • Ceramide, a potential cell death marker formed by sphingomyelinase, is involved in the expression of cyclooxygenase-2 (COX-2). This study examines the effect of C2-ceramide (C2), a cell-permeable ceramide analog, on the LPS-inducible COX-2 expression and signaling pathways. C2 did not induce COX-2, but potentiated LPS-inducible COX-2 expression in Raw264.7 cells, whereas dihydro-C2 was inactive.(omitted)

  • Galactosyl ceramides paly important roles in biological system as immunomodulator and essential constituents of membranes and cell walls. An efficient synthesis of $\alpha$-galactosyl ceramide, KRN 7000, derived from marine sponge Agelas mauritianus as accomplished via a short reaction involving the coupling ceramide moiety and trichloroacetimidate as glycosylation donor. We could synthesize $\alpha$-galactosyl ceramide stereoselectivity without $\beta$-anomer formation on a multigram scale.
