합성배지에서 저온성 느타리(Pleurotus ostreatus 201)의 섬유소분해효소 생산조건 (Cultural Conditions of Pleurotus ostreatus 201 for the Production of Cellulolytic Enzymes in Synthetic Medium)

  • Cultural conditions and carbon sources affecting the productivity of cellulolytic enzymes of Pleurotus ostreatus 201 were examined in synthetic media. The optimum cultural temperature and initial pH for the production of enzymes were $25^{\circ}C$ and 5.5 in avicelase, and 3$0^{\circ}C$ and 5.0 In CMCase, and 3$0^{\circ}C$ and 6.5 in B-glucosidase. Among carbon sources, cellulose powder was the best for the production of avicelase, and Na-CMC for CMCase, and cellobiose for $\beta$-glucosidase. The optimum concentration of cellulose powder was 1.0% (w/v), and glucose depressed the production of enzymes remarkably.

  • 섬유소자원의 이용을 목적으로 담자균류인 T. trogii를 실험 균주로 하여 합성배지에서 cellulase 생성 최적 배양조건을 검토해 본 결과는 다음과 같다. 합성배지에서 T. trogii에 의한 cellulase 생산은 $30{\sim}35^{\circ}C,\;pH\;4.0{\sim}6.0,\;and\;11{\sim}15$일 배양시 가장 높았다. 탄소원은 Avicelase와 ${\beta}-glucosidase$의 경우 CMC를, CMCase는 cellulose를 탄소원으로 했을 때 최고의 생산성을 보였으며, CMC의 최적 농도는 세 효소 모두 3%이었다. 질소원은 ammonium tartrate 첨가시 cellulase 생산이 높았으며, 그 최적 농도로 Avicelase와 ${\beta}-glucosidase$는 0.03N, CMCase는 0.04 N이었다.

  • Alkaline cellulolytic enzymes from cultured medium of Coprinus cinereus 2249 were purified with gel and ion-exchange chromatography and characteristics of those enzyme proteins were investigated. A fiber length distribution and a crystallinity of cellulose and sugar composition of enzyme treated Mixed Office Wastepaper(MOW) and Unbleached Kraft Pulp(UKP) were analysed. The conclusion could summarized as follows; \circled1 Alkaline and acidic, endo- and exo-glucanases were purified from cultured medium of Coprinus cinereus 2249. \circled2 The approximate molecular weight of alkaline endo-glucanase was 42 kDa, and also that of alkaline exo-glucanase was 50 kDa. A fiber length distribution and a crystallization of cellulose and sugar composition of enzyme treated MOW and UKP were not so much changed with original paper and pulp.

  • Alkaline cellulolytic enzymes are prepared from Coprinus cinereus 2249. Recovery method of enzyme protein from cultured medium and effect factors on enzyme activity of protein were investigated. The results could summarized as follows, \circled1 Amount of enzyme protein from cultured medium was highest in incubation with shaking and addition of skim milk. \circled2 Protein from cultured medium was alkaline enzymatic protein which shows the highest activity at pH 9.0. \circled3 The most effective recovery method of enzyme protein was the precipitation of protein by addition of cultured medium of protein in ethanol. \circled4 The enzyme activity was enhanced by tween-80 and decreased with $Al_2(SO_4)_3$, $H_2O_2$et al, and was little changed with metal ions except $Hg^{++}$.

섬유소폐기물을 이용한 사상균 FJ1의 섬유소 분해효소의 고생산 (The High Production of Cellulolytic Enzymes using Cellulosic Wastes by a Fungus, strain FJ1.)

  • 섬유소 폐기물의 생물학적 당화를 위해 분리한 사상균 FJ1은 섬유소 분해효소의 생산성이 뛰어난 신규의 유망한 셀룰라아제 생산균주로 잠정적으로 평가되었다. 사상균 FJ1의 셀룰라아제 생산을 위한 배지조성은, 질소원으로서 0.1% peptone을 사용하였을 경우와, avicel 및 섬유소 폐기물인 볏짚을 혼합하여 사용한 경우 높은 효소생산성을 나타냈다. 경제적인 효소생산을 위한 기질로 섬유소 폐기물의 농도와 혼합비는 1%(w/v)농도에서 볏짚과 펄프를 각각 0.5%(w/v)와 0.5%(w/v)으로 사용하였을 경우 CMCase, xylanase, $\beta$-glucosidase, avicelase의 효소 활성은 각각 24.3, 38.7, 1.5, 0.6 unit/ml 이었다. 이러한 효소생산성은 섬유소 폐기물의 생물학적 당화기술에 크게 기여할 수 있으리라 사료된다.

Trichoderma inhamatum KSJ1의 반연속배양 방식에 따른 섬유소분해효소의 생산성 비교 (Comparison of cellulolytic enzyme productivities in various semicontinuous culture modes of Trichoderma inhamatum KSJ1)

  • 본 연구에서는 음식물쓰레기를 당화시키기 위해 섬유소분해효소를 효율적으로 배양하고자 먼저 refill하는 멘델배지의 농도를 0.5%로, 새로운 배지의 주입시간을 12시간으로 결정하였다. Flask 레벨에서는 fill-and-draw 방법으로 12시간 단위로 연속배양한 결과, amylase 활성은 300시간까지 1.0 U/mL 내외로 유지되었으며, FPase 활성은 156시간까지 0.40 U/mL 이상으로 유지되었다. 이때의 효소생산성은 amylase 3.49 U/L. hr, FPase 1.02 U/L. hr 이었다. 10 L에서는 batch, fed-batch, fill-and-draw 방법으로 효소를 생산한 결과 batch에서 가장 높은 효소생산성을 나타내었으며, 그다음은 fed-batch 이었다. Batch에서의 효소생산성은 amylase 42.30 U/L. hr, FPase 5.60 U/L. hr, fed-batch에서는 각각 23.03, 2.76 U/L. hr 이었다. 그리하여 T. inhamatum KSJ1을 이용한 섬유소분해효소의 연속배앙에서 10 L 생물반응기에서 fed-batch 빙법이 가장 효율적이었다.

Rice Straw-Decomposing Fungi and Their Cellulolytic and Xylanolytic Enzymes

  • Filamentous fungi colonizing rice straw were collected from 11 different sites in Korea and were identified based on characterization of their morphology and molecular properties. The fungi were divided into 25 species belonging to 16 genera, including 14 ascomycetes, one zygomycete, and one basidiomycete. Fungal cellulolytic and xylanolytic enzymes were assessed through a two-step process, wherein highly active cellulase- and/or hemicellulase-producing fungi were selected in a first screening step followed by a second step to isolate the best enzyme-producer. Twenty-five fungal species were first screened for the production of total cellulase (TC), endo-${\beta}$-1,4 glucanase (EG), and endo-${\beta}$-1,4 xylanase (XYL) using solid-state fermentation with rice straw as substrate. From this screening, six species, namely, Aspergillus niger KUC5183, A. ochraceus KUC5204, A. versicolor KUC5201, Mucor circinelloides KUC6014, Trichoderma harzianum 1 KUC5182, and an unknown basidiomycete species, KUC8721, were selected. These six species were then incubated in liquid Mandels' media containing cellulose, glucose, rice straw, or xylan as the sole carbon source and the activities of six different enzymes were measured. Enzyme production was highly influenced by media conditions and in some cases significantly increased. Through this screening process, Trichoderma harzianum 1 KUC5182 was selected as the best enzyme producer. Rice straw and xylan were good carbon sources for the screening of cellulolytic and xylanolytic enzymes.

몇 종류의 곰팡이에서 분리되는 Crude Cellulase의 다당류 분해능력의 조사 (Investigation of the Hydrolysis of Polysaccharides by Crude Cellulases prepared from Several Species of Fungi)

  • Crude cellulases freshly prepared from cultures of Aspergillus niger, Prnicillum motatum, Trichoderma vride 16273 and Trichoderma viride 16374 were assayed on 4 different substrates including Na-CMC, cellulose powder, starch and sucrose. Enzyme prepared from A. niger contained highly active hydrolytic enzymes of the 4 substrates assayed. P. notatum [yielded relatively lower amount of cellulase but the extracts were also highly reactive on starch and sucrose. Trichoderma viride 16274 yielded very little cellulase and invertase, but the extracts showed a high degree of amylase activity. Trichoderma viride 16374, however, yielded collulase comparable to that of Penicillium notatum, but lower activities of amylase and invertase were seen. Commercial cellulases prepared from Penicillium notatum (cellulase[K]) and Trichoderma viride(cellulase[J]) indicated enzyme activities closely parallel to the crude enzymes freshly prepared from fungus cultures. The optimum pH's of cellulolytic activities of cellulase[K] and cellulase[J] were 4.0 and 5.0 respectively. The optimum temperatures of the cellulolytic activities of cellulase[K] and cellualse[J] were 4.0 and 5.0 respectively. The optimum temperatures of the cellulolytic activities of cellulase [K] and cellulase [J] were $60{\circ}C$ and $50{\circ}C$ respectively. Assuming the average molecular weight of Na-CMC is about 115,000, the Km values of cellulase [K] and cellulase[J] were found to be $3.3{\times}10^{-5}/nM$ and $3.3{\times}10^{-4}/nM$ respectively.

Detection of Cellulolytic Activity in Ophiostoma and Leptographium species by Chromogenic Reaction

  • To understand the ability of producing cellulolytic enzyme activity in the sapstaining fungi, four species of Ophiostoma and two species of Leptographium were investigated in the culture media containing each of cellulose substrates such as CM-cellulose, Avicel and D-cellobiose and each of chromogenic dyes such as Congo-Red, Phenol Red, Remazol Brilliant Blue and Tryphan Blue. When the fungi were grown for $5{\sim}7$ days at $25^{\circ}C$, the formation of clear zone by chromogenic reaction around the margin of the fungal colony was demonstrated in all the culture media Congo-Red containing CM-cellulose. There was difference in the formation of clear zone among the dyes. Only Ophiostoma setosum and Leptographium spp. showed cellulolytic activity to the three substrates. Overall, the results of this study show that ophiostomatoid sapstaining fungi can produce cellulolytic enzymes.
