To test the cytoprotective effect of sophorae radix (SR) against hydrogen peroxide (H₂O₂)-induced cytotoxicity, we investigated the cell viability using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) assay in the presence of ethyl acetate subfractions of SR water extracts In H9c2 cells. And to clarify the cytoprotective mechanism of SR extracts, we evaluated the cellular glutathione (GSH) contents in the presence of subfraction 1, 2, 3, and 4 of SR ethyl acetate soluble fractions. Among 1 -12 subfractions of SR ethyl acetate soluble fractions, 1, 2, 3 and 4 subfractions have an efficacy inhibiting the cytotoxicity induced by H₂O₂ in H9c2 cells. Also, the protective effects of 1, 2, 3 and 4 subfractions of SR ethyl acetate soluble fractions resulted from the anti-oxidant effects. These results suggest that ethyl acetate soluble fractions of SR water extracts is effective in the prevention of H₂O₂-induced cytotoxicity and 1, 2, 3 and 4 subfraction of ethyl acetate soluble fractions possess the anti-oxidant component.
With the rapid developments in nanotechnology, an increasing number of nanomaterials have been applied in various aspects of our lives. Recently, pharmaceutical nanotechnology with numerous advantages has growingly attracted the attention of many researchers. Zinc oxide nanoparticles (ZnO-NPs) are nanomaterials that are widely used in many fields including diagnostics, therapeutics, drug-delivery systems, electronics, cosmetics, sunscreens, coatings, ceramic products, paints, and food additives, due to their magnetic, catalytic, semiconducting, anti-cancer, anti-bacterial, anti-inflammatory, ultraviolet-protective, and binding properties. The present review focused on the recent research works concerning role of ZnO-NP on inflammation. Several studies have reported that ZnO-NP induces inflammatory reaction through the generation of reactive oxygen species by oxidative stress and production of inflammatory cytokines by activation of nuclear factor-${\kappa}B$ ($NF-{\kappa}B$). Meanwhile, other researchers reported that ZnO-NP exhibits an anti-inflammatory effect by inhibiting the up-regulation of inflammatory cytokines and the activation of $NF-{\kappa}B$, caspase-1, $I{\kappa}B$$kinase{\beta}$, receptor interacting protein2, and extracellular signal-regulated kinase. Previous studies reported that size and shape of nanoparticles, surfactants used for nanoparticles protection, medium, and experimental conditions can also affect cellular signal pathway. This review indicated that the anti-inflammatory effectiveness of ZnO-NP was determined by the nanoparticle size as well as various experimental conditions. Therefore, the author suggests that pharmaceutical therapy with the ZnO-NP is one of the possible strategies to overcome the inflammatory reactions. However, further studies should be performed to maximize the anti-inflammatory effect of ZnO-NP to apply as a potential agent in biomedical applications.

"Two-cysteine" peroxiredoxins are antioxidant enzymes that exert a cytoprotective effect in many models of oxidative stress. However, under highly oxidizing conditions they can be inactivated through hyperoxidation of their peroxidatic active site cysteine residue. Sulfiredoxin can reverse this hyperoxidation, thus reactivating peroxiredoxins. Here we review recent investigations that have shed further light on sulfiredoxin's role and regulation. Studies have revealed sulfiredoxin to be a dynamically regulated gene whose transcription is induced by a variety of signals and stimuli. Sulfiredoxin expression is regulated by the transcription factor AP-1, which mediates its up-regulation by synaptic activity in neurons, resulting in protection against oxidative stress. Furthermore, sulfiredoxin has been identified as a new member of the family of genes regulated by Nuclear factor erythroid 2-related factor (Nrf2) via a conserved cis-acting antioxidant response element (ARE). As such, sulfiredoxin is likely to contribute to the net antioxidative effect of small molecule activators of Nrf2. As discussed here, the proximal AP-1 site of the sulfiredoxin promoter is embedded within the ARE, as is common with Nrf2 target genes. Other recent studies have shown that sulfiredoxin induction via Nrf2 may form an important part of the protective response to oxidative stress in the lung, preventing peroxiredoxin hyperoxidation and, in certain cases, subsequent degradation. We illustrate here that sulfiredoxin can be rapidly induced in vivo by administration of CDDO-TFEA, a synthetic triterpenoid inducer of endogenous Nrf2, which may offer a way of reversing peroxiredoxin hyperoxidation in vivo following chronic or acute oxidative stress.
Objectives : This study was designed to investigate the cellular and molecular mechanisms of anti-inflammatory activity of the water extract of Polygala tenuifolia Willd. (Pt-WE). Methods : Using lipopolysaccharide (LPS)-stimulated murine RAW264.7 cells, we examined inflammatory mediators such as nitric oxide (NO), inducible NO synthase (iNOS), cyclooxygenase (COX)-2 and prostaglandin $E_2$ ($PGE_2$). Also, the inhibitory effect of Pt-WE on the activity of activator protein 1 (AP-1) and upstream signaling molecules was evaluated. To assess the protective effect of Pt-WE on hydrochloride/ethanol (HCl/EtOH)-induced gastric ulcer in mice, we compared Pt-WE (200 mg/kg) with ranitidine (50 mg/kg) treated mice's gastric mucosa, based on gross observations. Results : Pt-WE inhibited LPS-induced production of NO, $PGE_2$ in a dose-dependent manner, without causing cytotoxicity. Pt-WE suppressed AP-1 activation by reducing generations of both c-Jun and c-Fos. In addition, Pt-WE inhibited the p-MKK 4/7 (mitogen-activated protein kinase kinase 4/7) and p-JNK (c-Jun N-terminal kinase) 1 in LPS-stimulated RAW264.7 cells. HCl/EtOH-induced gastric ulcer lesions were inhibited by pre-treatment of Pt-WE based on gross observations. In addition, Pt-WE decreased the phosphorylation level of JNK. Conclusions : These results demonstrate that Pt-WE has anti-inflammatory and gastroprotective effects. Thus, Pt-WE may be used widely in treatment of not only neurodegenerative diseases but also inflammatory diseases.
본 연구에서는 마를 건조 후 분말화하여 실험에 사용하였으며, 건조 마 분말에 대한 생리활성에 대한 연구로 건조된 마를 유기용매로 추출하여 마 추출물과 분획물들의 $H_2O_2$에 의해 유도된 산화적 스트레스 및 인체 암세포(HT1080 인체 섬유육종세포, HT-29 인체 결장암세포)에 대한 증식 억제 효과에 대해 검토하고자 하였다. 마는 항비만, 항변비, 항돌연변이, 혈당 및 혈중 콜레스테롤 감소 등의 활성이 있어 건강식품으로 알려져 있다. $H_2O_2$에 의해 유도된 산화적 스트레스 저해효과를 알아보기 위하여 DCFH-DA assay를 행하였다. 건조 마 A+M 추출물을 농도별로 인체 섬유육종세포(HT1080)에 처리하였을 때 농도 의존적으로 세포 내 활성산소종을 크게 억제시켰다. 각 분획물들 중 85% aq. MeOH 분획물은 다른 분획물과 비교하여 높은 지질 과산화물 생성 억제 효과를 보였다. 인체 암세포에 대한 증식 억제 실험에서 A+M 및 MeOH 추출물은 농도 의존적으로 인체 섬유육종 및 결장암세포의 증식을 억제하였다. 마 추출물로부터 얻어진 n-hexane, 85% aq. MeOH 및 n-butanol (n-BuOH) 분획물들은 첨가농도 0.5 mg/ml 이상 처리했을 때 이들 두 암세포의 증식을 유의적으로 억제하였다(p<0.05). 본 연구 결과로 부터 건조 마의 암세포 증식 억제 효과는 마의 A+M 추출물이 MeOH 추출물에 비해 높았고, 분획물들의 경우에는 water 분획물을 제외한 분획물들의 암세포 억제 효과가 높았다. 특히 85% aq. MeOH 분획물은 인체 암세포 증식 억제 효과뿐만 아니라 세포 내 활성산소종 감소시키는 효과가 높아 이들 분획물에 활성물질이 있을 것으로 추정된다.

Prostaglandin $E_2$ ($PGE_2$) promotes tumor-persistent inflammation, frequently resulting in cancer. Curcumin is a diphenolic turmeric that inhibits carcinogenesis and induces apoptosis. $PGE_2$ inhibits curcumin-induced apoptosis; however, the underlying inhibitory mechanisms in colon cancer cells remain unknown. The aim of the present study is to investigate the survival role of $PGE_2$ and whether addition of exogenous $PGE_2$ affects curcumininduced cell death. HCT-15 cells were treated with curcumin and $PGE_2$, and protein expression levels were investigated via Western blot. Reactive oxygen species (ROS) generation, lipid peroxidation, and intracellular glutathione (GSH) levels were confirmed using specific dyes. The nuclear factor-kappa B ($NF-{\kappa}B$) DNA-binding was measured by electrophoretic mobility shift assay (EMSA). $PGE_2$ inhibited curcumin-induced apoptosis by suppressing oxidative stress and degradation of PARP and lamin B. However, exposure of cells to the EP2 receptor antagonist, AH6809, and the PKA inhibitor, H89, before treatment with $PGE_2$ or curcumin abolished the protective effect of $PGE_2$ and enhanced curcumin-induced cell death. $PGE_2$ activates PKA, which is required for cAMP-mediated transcriptional activation of CREB. $PGE_2$ also activated the Ras/Raf/Erk pathway, and pretreatment with PD98059 abolished the protective effect of $PGE_2$. Furthermore, curcumin treatment greatly reduced phosphorylation of CREB, followed by a concomitant reduction of $NF-{\kappa}B$ (p50 and p65) subunit activation. $PGE_2$ markedly activated nuclear translocation of $NF-{\kappa}B$. EMSA confirmed the DNA-binding activities of $NF-{\kappa}B$ subunits. These results suggest that inhibition of curcumin-induced apoptosis by $PGE_2$ through activation of PKA, Ras, and $NF-{\kappa}B$ signaling pathways may provide a molecular basis for the reversal of curcumin-induced colon carcinoma cell death.
목적: 활성산소종의 하나인 $H_2O_2$는 DNA, RNA에 직접적인 손상을 주어 유전자를 변형시키고 세포고사를 일으켜 각종 질환을 일으키는 발병인자로 알려져 있다. 본 연구는 결막 세포주에서 일어나는 $H_2O_2$에 의한 세포고사에서 EGCG의 보호효과를 알아보고자 실험하였다. 방법: 세포의 생존율은 MTT assay로 확인하였고, 세포고사의 형태적인 특징은 DNA 단절화을 통해 확인하였으며, EGCG의 자유라디칼 제거능력을 측정하기 위해 DPPH free radical 소거능을 실시하고, 세포내 활성산소량의 평가를 위한 DCFH-DA assay를 시행하였다. 세포내의 mRNA 발현은 RTPCR을 시행하여 밴드 양상을 확인하였다. 결과: 세포생존율과 프리라디칼 소거능은 EGCG의 처리 농도 의존적으로 증가하였고, DNA 분절화와 세포내 활성산소량은 감소하였다. mRNA 합성에서 bcl-2, bcl-xL의 발현은 증가하였고, bax 발현은 감소되었다. 결론: EGCG는 $H_2O_2$로 세포고사를 유도한 결막 세포주에서 항산화효과를 나타냈으며, EGCG가 anti-apoptosis 관련한 mRNA의 발현을 증가시켜 결막 세포주의 산화적 손상을 억제함으로서 세포보호효과를 나타냈음을 확인할 수 있었다.
본 연구에서는 피로 회복 또는 원기 회복에 효능이 있는 것으로 알려진 홍경천과 홍삼을 이용하여 홍경천-홍삼 복합 발효물의 산화적 손상 억제 효과를 평가하고자 $H_2O_2$로 산화적 스트레스를 유도시킨 C2C12 근육세포에 홍경천-홍삼 복합 발효물의 처리한 후, 세포의 morphology, cell viability 및 항산화 효소들의 유전자 발현 양상을 비교, 분석하였다. 홍경천-홍삼 복합 발효물은 C2C12 근육세포의 cell viability를 유의적으로 증가시켰으며, Cu/Zn-SOD, Mn-SOD 및 GPx 등과 같은 세포내 항산화 효소의 발현을 증가시킬 뿐만 아니라, 근육세포 분화의 주요 전사인자인 Myo D의 발현 또한 증가시키는 것으로 나타났다. 이상의 결과로, 홍경천-홍삼 복합 발효물은 세포내 항산화 효소 시스템을 증가시켜 외부로부터의 산화적 손상에 대한 방어효능을 갖는 것으로 나타났으며, 향후 in vivo 시스템 이용한 추가적인 연구가 수행된다면, 홍경천-홍삼 복합 발효물을 이용한 항피로 건강기능식품의 소재개발이 가능할 것으로 판단된다.
본 연구에서는 일반 메밀과 쓴 메밀의 제분 분획 별(겉껍질, 겨, 가루) 에탄올 추출물의 항산화 성분 함량을 분석하고, 항산화 활성을 시험관과 세포 수준에서 확인하였다. 그 결과, 일반 메밀보다 쓴 메밀에서 총 폴리페놀 및 총 플라보노이드 함량이 월등히 높았으며, 항산화 물질로 주된 성분은 루틴으로 확인되었고, 에탄올 추출과정에서 루틴이 퀘세틴으로 일부 분해되는 것으로 나타났다. 또한 식용 부위인 가루 분획보다 겨 분획에서의 항산화 성분 함량이 높은 것으로 분석되어 겨 분획의 항산화 기능성 소재로 개발의 필요성을 제시하였다. 한편 시험관 및 세포수준에서 항산화 효과를 확인한 결과, 일반 메밀보다 쓴 메밀에서의 항산화 활성이 유의적으로 높았으며, 특히 쓴 메밀 겨 분획의 항산화력이 우수한 것으로 나타났다. 메밀 추출물을 산화스트레스 유발 전, 후 처리한 경우 모두에서 항산화 활성이 유사한 경향으로 나타나, 메밀 추출물의 산화스트레스 예방 및 치료 효과 가능성을 제시하였다.
말채나무 열수추출물의 항산화 활성을 알아보기 위해 HepG2 세포에 TBHP로 산화 스트레스를 유도한 뒤 말채나무 열수추출물의 세포 보호효과, ROS 생성억제, 지질과산화 억제 및 GSH 생성에 미치는 영향에 대해 살펴보았다. 말채나무 열수추출물은 HepG2 세포에 TBHP로 산화 스트레스를 유도한 뒤 나타나는 세포독성에 대해 농도 의존적으로 유의하게 세포 보호효과를 보였으며, ROS 생성과 과산화물에 대한 지표로서 측정한 MDA도 말채나무 열수추출물에 의해 효과적으로 억제되었다(P<0.05). 또한 항산화 성분으로 생체 내에서 산화 및 환원반응에 중요한 역할을 하며 항산화 효소인 GSH-Px, GST에 전자공여체로 작용하는 GSH의 생성촉진 효능에서도 말채나무 열수추출물은 산화 스트레스로 감소된 GSH의 생성을 농도 의존적으로 촉진시켰다(P<0.05). 산화 스트레스에 의해 활성이 증가된 항산화 효소(CAT, SOD, GSH-Px, GR)도 말채나무 열수추출물의 처리로 감소하는 경향을 보였다. 이러한 결과로 미루어 보아 말채나무 열수 추출물은 인체 내에서 질병과 노화를 일으키는 원인 물질인 활성산소에 대해 강한 항산화 활성을 나타냄에 따라 보다 다양한 형태의 소재로 활용될 수 있을 것으로 생각된다.

제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
